Temporal pattern of humoral immune response in mild cases of COVID-19

BACKGROUND Understanding the humoral response pattern of coronavirus disease 2019(COVID-19)is one of the essential factors to better characterize the immune memory of patients,which allows understanding the temporality of reinfection,provides answers about the efficacy and durability of protection against severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),and consequently helps in global public health and vaccination *** the patients who became infected with SARS-CoV-2,the majority who did not progress to death were those who developed the mild COVID-19,so understanding the pattern and temporality of the antibody response of these patients is certainly *** To investigate the temporal pattern of humoral response of specific immunoglobulin G(IgG)in mild cases of *** Blood samples from 191 COVID-19 real-time reverse transcriptase-polymerase chain reaction(RT-qPCR)-positive volunteers from the municipality of Toledo/Paraná/Brazil,underwent two distinct serological tests,enzyme-linked immunosorbent assay,and detection of anti-nucleocapsid *** samples and clinicoepidemiological data of the volunteers were collected between November 2020 and February *** assays were performed in duplicate and the manufacturers'recommendations were strictly *** data were statistically analyzed using multiple logistic regression;the variables were selected by applying the P<0.05 *** Serological tests to detect specific IgG were performed on serum samples from volunteers who were diagnosed as being positive by RT-qPCR for COVID-19 or had disease onset in the time interval from less than 1 mo to 7 *** time periods when the highest number of participants with detectable IgG was observed were 1,2 and 3 *** was observed that 9.42%of participants no longer had detectable IgG antibodies 1 mo only after being infected with SARS-CoV-2 and 1.57%were also IgG negative at less than 1 *** 5 mo,3.14%of volunteers were IgG n

The gain of transcription factor binding sites(TFBS) is believed to represent one of the major causes of biological *** we used strategies based on comparative genomics to identify 21,822 TFBS specific to the human lineage(TFBS-HS),when compared to chimpanzee and gorilla genomes. More than 40%(9,206) of these TFBS-HS are in the vicinity of 1,283 genes.A comparison of the expression pattern of these genes and the corresponding orthologs in chimpanzee and gorilla identified genes differentially expressed in human tissues. These genes show a more divergent expression pattern in the human testis and brain, suggesting a role for positive selection in the fixation of TFBS gains. Genes associated with TFBS-HS were enriched in gene ontology categories related to transcriptional regulation, signaling, differentiation/development and nervous ***, genes associated with TFBS-HS present a higher expression breadth when compared to genes in general. This biased distribution is due to a preferential gain of TFBS in genes with higher expression breadth rather than a shift in the expression pattern after the gain of TFBS.

Photodynamic therapy in cancer treatment - an update review

Cancer remains a worldwide health problem, being the disease with the highest impact on global health. Even with all the recent technological improvements, recurrence and metastasis still are the main cause of death. Since photodynamic therapy (PDT) does not compromise other treatment options and presents reduced long-term morbidity when compared with chemotherapy or radiotherapy, it appears as a promising alternative treatment for controlling malignant diseases. In this review, we set out to perform a broad up-date on PDT in cancer research and treatment, discussing how this approach has been applied and what it could add to breast cancer therapy. We covered topics going from the photochemical mechanisms involved, the different cell death mechanisms being triggered by a myriad of photosensitizers up to the more recent-on-going clinical trials.

The search for cancer biomarkers is frequently based on comparisons between tumors and adjacent-to-tumor samples. However, even after histological confirmation of been free of cancer cells, these adjacent-to-tumor samples might harbor molecular alterations which are not sufficient to cause them to look like cancer, but can differentiate these cells from normal cells. When comparing them, potential biomarkers are missed, and mainly the opportunity of finding initial aberrations presents in both tumors and adjacent samples, but not in true normal samples from non-cancer patients, resulting in misinterpretations about the carcinogenic process. Nevertheless, collecting adjacent-to-tumor samples brings trumps to be explored. The addition of samples from non-cancer patients opens an opportunity to increase the finds of the molecular cascade of events in the carcinogenic process. Differences between normal samples and adjacent samples might represent the first steps of the carcinogenic process. Adding samples of non-cancer patients to the analysis of molecular alterations relevant to the carcinogenic process opens a new window of opporttmides to the discovery of cancer biomarkers and molecular targets.

A decade without diagnosis: Sheehan’s syndrome

Sheehan’s Syndrome (SS) occurs as a result of ischemic pituitary necrosis due to severe post-partum hemorrhage. We report a 41-year-old woman with a history of severe postpartum hemorrhage 18 years prior to presentation. During this period of time, she experienced severe fatigue. Three months prior to being admitted to our hospital, the patient started to notice alopecia, generalized edema and cognitive impairment. After proper investigation, laboratory tests and clinical findings indicated panhypopituitarism. In addition to neuroimaging and past medical history, SS was diagnosed and treatment started. We emphasize the importance of thorough investigation, further diagnosis and management of this condition (especially in third world countries), since SS is a neurological and endocrinological emergency and potentially life threatening.

Analysis of Daily Rainfall in São Carlos/SP, Brazil over 1979-2017 Using Laplace Trend Test

Rainfall expresses one of the most complex climate factors in Southeastern Brazil. Understanding the dynamics and temporal trends of rainfall represents a significant challenge due to regional and even global mechanisms, such as FS (Frontal Systems) and the SACZ (South Atlantic Convergence Zone), and the interaction with the Atlantic and Pacific Oceans. The present study aimed at analyzing the pluviometric tendencies in São Carlos/SP, in the countryside of São Paulo State. Laplace trend test was used to comprehend the temporal evolution of daily rainfall in the region in the historical series 1979-2017, in seven pluviometric stations (climatological or surface stations). Significant fluctuations in interannual trends and between seasons were observed. However, it was noted that the beginning of the 1980s showed positive trends, whereas, as of the year 2000, most of the stations demonstrated negative trends, indicating a reduction in daily rainfall volume due to the great tropical climatic variability of Brazil. Emphasis should also be given to the regional and local effects, such as elevation and urbanization, respectively, which corroborate such differences among the analyzed stations. This methodology is of considerable value for the observation of pluviometric trends, and future studies can validate such a tool in climatological studies.

Evaluation of Visible Losses and Damage to the Ratoon Cane in the Mechanized Harvesting of Sugarcane for Different Displacement Speeds

This study aimed at quantitatively evaluating the sugarcane losses and the damage caused to the ratoon cane while using the Case IH A8800 harvester in different displacement speeds;it was conducted in the municipality of Campos dos Goytacazes, in Rio de Janeiro, Brazil. Three speeds were used as treatment (2 km·h-1, 3 km·h-1, and 4.5 km·h-1), and each treatment was composed of six rows of harvested ratoon, each with a length of 290 m. In order to evaluate the quantitative losses in t·ha-1 and the percentage of losses, the remaining sugarcane, left on the field after harvesting, was collected. The sampling frame was set to every 50 m, dividing the borders by 40 m;the measurement for each sampling area was of 20 m2, with five repetitions. In order to evaluate the damage caused to the ratoon canes, we chose a visual methodology to classify the damage degrees, ranging from one to four. There was no significant difference in losses when comparing different speeds. Therefore, it is more advantageous and economically viable to use the speed of 4.5 km·h-1, which collects more in less time.

The Assessment of Medical Students’ Chronotypes

Objective: This study aimed to investigate the circadian typology of medical students at public universities from one of the northeastern states in Brazil, active in the morning and afternoon shifts, checking the compatibility between their study time, productivity, income and chronotype. Methods: To obtain the results, the assessment of the chronotype in 342 medical students of both sexes was considered. Each participant was submitted to the questionnaire developed by Horne and Ostberg, adapted by Cardinali, Colomberk and Rey. It’s composed of 9 questions, providing the general ranking of each individual. Based upon scored, the chronotypes were classified. Results: The sample was composed of 49.12% (168) male and 50.87% (174) female subjects, a similar gender distribution of enrolled students during data collection time. By applying the questionnaire, the prevalent rating among the medical students as moderately morning (29.82%) and intermediate (43.85%) has been relevant. The academic performance assessment in two morning-intermediate groups (8.16) had no significant distance from others considered afternoon active (8.20). Conclusion: The health sciences student, especially Medicine, faces constant adaptation of scheduling, in the attempt of fulfilling the requirements throughout the course, causing chronogram desynchronization effects and forced readaptation to each different school semester.

The aim of this work was to estimate the adaptability and stability of grain yield per hectare and percentage of crude protein of maize grains combined in an index, and to establish a multicharacter selection through mixed models based on an objective character and 15 auxiliary traits. The trials were conducted in the 2013/2014 agricultural year in four growing environments of the Rio Grande do Sul, BR state. The experimental design was randomized blocks arranged in a factorial scheme, being four growing sites × 15 single cross maize hybrids, arranged in three repetitions. The genotypic index, composed by the grain yield and the crude protein percentage in the grains, is the best selection strategy to achieve maize superior genotypes. The multivariate genotypes selection, considering grain yield and crude protein, is efficient. The genotypes FORMULA TL®, AS1656PRO®, P30F53Hx®, LG6304YG®and 30F53 are more adapted and stable for grain yield and percentage of crude protein, in the conditions of this study. The mixed models were efficient to employ the multicharacter selection and to contribute for maize genetic breeding.
