The actin cytoskeleton plays a role in mobility of many different organelles in plant cells, including chloroplasts, mitochondria, Golgi, and peroxisomes. While progress has been made in identifying the myosin motors involved in trafficking of various plant organelles, not all of the cargoes mobilized by different members of the myosin XI family have yet been identified. The involvement of myosins in chloroplast positioning and mitochondrial movement was demonstrated by expression of a virus-induced gene silencing （VIGS） construct in tobacco. When VIGS with two different conserved sequences from a myosin Xl motor was performed in plants with either GFP-labeled plastids or mitochondria, chloroplast positioning in the dark was abnormal, and mitochondrial movement ceased. Because these and prior obser- vations have implicated a role for myosins and the actin cytoskeleton in plastid and stromule movement, we searched for myosin tail domains that could associate with plastids and stromules. While a yellow fluorescent protein （YFP） fusion with the entire tail region of myosin XI-F was usually found only in the cytoplasm, we observed that an Arabidopsis or Nicotiana benthamiana YFP：：myosin XI-F tail domain homologous to the yeast myo2p vacuole-binding domain associated with plastids and stromules after transient expression in N. benthamiana. Taken together, these observations implicate myosin motor proteins in dynamics of plastids and stromules.

Human norovirus is the leading cause of foodborne illness *** and quantification of norovirus commonly involves the use of reverse transcriptase quantitative polymerase chain reaction(RT-qPCR);however,the presence of inhibitory compounds in foods limit detection and accurate *** some studies have been done on PCR inhibitors from foods,many of them are over a decade old and do not investigate inhibition in contemporary one-step RT-qPCR-based detection *** purpose of this work was to quantify the degree of inhibition that occurs from inhibitory compounds found in produce(pectin)and mollusks(hemocyanin,glycogen)—foods commonly associated with norovirus ***-qPCR reactions containing different amounts of genomic bacteriophage MS2 RNA,a norovirus surrogate,were spiked with different concentrations of pectin(0.0625%–0.25%w/V),glycogen(1.25%–10%),and hemocyanin(0.0625%–0.25%).Past research has implicated glycogen as an inhibitory compound in oysters;however,even high levels of glycogen(10%)had no significant effect(P>0.05)on ***,both pectin and hemocyanin caused complete inhibition at 0.25%,with no significant inhibition observed at 0.0625%(Pdant in the hemolymph of mollusks and previously untested as a PCR *** work demonstrates that pectin and hemocyanin should be considered when testing produce and mollusk samples with PCR-based methods.

Industrial woven meshes are composed of metal materials and are often used in construction,industrial and residential activities or *** objective of this work is defect detection in industrial fabrics in the quality control *** order to overcome the limitations of manual methods,which are often tedious and time-consuming,we propose a strategy that can automatically detect defects in micrometric steel meshes by means of a Convolutional Neural *** database used for such a purpose comes from real problem data for anomaly detection in micrometric woven *** detection is performed through supervised classification with a Convolutional Neural Network using a VGG19 *** define a pipeline and a strategy to tackle the related small amount of *** includes i)augmenting the database with translation,rotation and symmetry,ii)using pre-trained weights and iii)checking the learning curve behaviour through *** obtained results show that,despite the small size of our databases,detection accuracy of 96%was reached.

Recent studies investigating possible causes of male subfertility have largely focused on how lifestyle or environmental factors impact on the process of spermatogenesis, Markedly, fewer studies have investigated those risk factors that result in reduced sperm quality, such as poor sperm motility. The speed at which sperm swim is a major predictor of fertility and is extremely variable in human populations. It has been hypothesized that offspring sex may be adaptively manipulated to maximize the offspring＇s reproductive fitness （e.g., parents with genes for good male fertility traits, such as high sperm speed, would produce primarily sons and fewer daughters because the offspring will inherit advantageous male fertility genes）. Conversely, parents with poor male fertility genes would produce primarily daughters, We tested whether there was an association between how fast a man＇s sperm swam and the sex bias of his siblings in a sample of men attending clinic for fertility investigations with their partner and with a wide range of semen characteristics, including sperm speed. We found that the sex bias of a man＇s siblings is associated with his sperm speed; men with female-biased siblings had significantly slower sperm （judged using computer-assisted sperm analysis （CASA）） than men from male-biased sibships. This observation suggests family composition is an important factor that needs to be considered in future edidemiological and clinical studies of human fertility,

Mouse models of cancer enable researchers to learn about tumor biology in complicated and dynamic physiological systems. Since the development of gene targeting in mice, cancer biologists have been among the most frequent users of transgenic mouse models, which have dramatically increased knowledge about how cancers form and grow. The Chinese Journal of Cancer will publish a series of papers reporting the use of mouse models in studying genetic events in cancer cases. This editorial is an overview of the development and applications of mouse models of cancer and directs the reader to upcoming papers describing the use of these models to be published in coming issues, beginning with three articles in the current issue.

The absence of ultrahigh pressure(UHP)orogenic eclogite in the geological record older than c.0.6 Ga is problematic for evidence of subduction having begun on Earth during the Archean(4.0-2.5 Ga).Many eclogites in Phanerozoic and Proterozoic terranes occur as mafic boudins encased within low-density felsic crust,which provides positive buoyancy during subduction;however,recent geochemical proxy analysis shows that Archean continental crust was more mafic than previously thought,having greater proportions of basalt and komatiite than modern-day ***,we show via petrological modelling that secular change in the petrology and bulk composition of upper continental crust would make Archean continental terranes negatively buoyant in the mantle before reaching UHP *** or delaminated Archean continental crust passes a point of no return during metamorphism in the mantle prior to the stabilization of coesite,while Proterozoic and Phanerozoic terranes remain positively buoyant at these *** orogenic eclogite may thus readily have formed on the Archean Earth,but could not have been exhumed,weakening arguments for a Neoproterozoic onset of subduction and plate ***,isostatic balance calculations for more mafic Archean continents indicate that the early Earth was covered by a global ocean over 1 km deep,corroborating independent isotopic evidence for large-scale emergence of the continents no earlier than c.3 *** findings thus weaken arguments that early life on Earth likely emerged in shallow subaerial ponds,and instead support hypotheses involving development at hydrothermal vents in the deep ocean.

Sequencing data from 10 species show that a plant hexokinase （HXK） family contains 5-11 genes. Functionally, a given family can include metabolic catalysts, glucose signaling proteins, and non-catalytic, apparent regulatory enzyme homologs. This study has two goals. The first aim is to develop a predictive method to determine which HXK proteins within a species have which type of function. The second aim is to determine whether HXK-dependent glucose signaling proteins occur among more primitive plants, as well as among angiosperms. Using a molecular phylogeny ap- proach, combined with selective experimental testing, we found that non-catalytic HXK homologs might occur in all plants, including the relatively primitive Selaginella moellendorffi. We also found that different lineages of angiosperm HXKs have apparent conserved features for catalytic activity and for sub-cellular targeting. Most higher-plant HXKs are expressed predominantly at mitochondria, with HXKs of one lineage occurring in the plastid, and HXKs of one monocot lineage occurring in the cytosol. Using protoplast transient expression assays, we found that HXK glucose signaling pro- teins occur likely in all higher plants and in S. moellendorffi as well. Thus, the use of glucose by plant HXK isoforms in metabolism and/or as a regulatory metabolite occurs as widespread, conserved processes.

Gastroesophageal reflux disease:A review of surgical decision making

Gastroesophageal reflux disease(GERd) is a very common disorder with increasing prevalence. It is estimated that up to 20%-25% of Americans experience symptoms of GERd weekly. Excessive reflux of acidic often with alkaline bile salt gastric and duodenal contents results in a multitude of symptoms for the patient including heartburn, regurgitation, cough, and dysphagia. There are also associated complications of GERd including erosive esophagitis, Barrett's esophagus, stricture and adenocarcinoma of the esophagus. While first line treatments for GERd involve mainly lifestyle and non-surgical therapies, surgical interventions have proven to be effective in appropriate circumstances. Anti-reflux operations are aimed at creating an effective barrier to reflux at the gastroesophageal junction and thus attempt to improve physiologic and mechanical issues that may be involved in the pathogenesis of GERd. The decision for surgical intervention in the treatment of GERd, moreover, requires an objective confirmation of the diagnosis. Confirmation is achieved using various preoperative evaluations including: ambulatory p H monitoring, esophageal manometry, upper endoscopy(esophagogastroduodenoscopy) and barium swallow. Upon confirmation of the diagnosis and with appropriate patient criteria met, an antireflux operation is a good alternative to prolonged medical therapy. Currently, minimally invasive gastroesophageal fundoplication is the gold standard for surgical intervention of GERd. Our review outlines the many factors that are involved in surgical decisionmaking. We will review the prominent features that reflect appropriate anti-reflux surgery and present suggestions that are pertinent to surgical practices, based on evidence-based studies.

Invasive alien plant species have threatened the integrity of ecosystems and threatened biological integrity throughout the world. In the Nyika National Park, a number of alien invasive plant species have been reported. These invasive alien plants are reported to be very difficult that have caused adverse ecological, economic and social impact. The main objective of this study is to assess the environmental impact of plant invasive alien species in Nyika National Park in order to ascertain means of addressing the possible effects on the biodiversity. This study adopted an exploratory stance in order to answer the hypotheses where purposive and simple random sampling was used. Purposive sampling was only used to the staff of Nyika while simple random sampling was used to collect data from households surrounding Nyika National Park. The results revealed that invasive alien plant species have some ecological impacts of on the biodiversity of the Nyika National Park like change in physical habitats such as loss of native habitats, alteration of ground water regime, drying of rivers, loss of native species and alteration of biomass. The study would therefore be important to policy makers in guiding and development of policies and guidelines governing eradication of invasive alien plant species. The department of National Parks and Wildlife (dNPW) can form partnership with various stakeholders with a clearly stated intention of developing a relationship, based on collaboration to enhance the control of invading alien plant species and the best management practices.
