Epigenetics of gastroenteropancreatic neuroendocrine tumors:A clinicopathologic perspective

Gastroenteropancreatic neuroendocrine tumors(GEP-NETs) are a heterogeneous group of rare tumors whose site-specific tumor incidence and clinical behavior vary widely. Genetic alterations associated with familial inherited syndromes have been well defined; however, the genetic profile of sporadic tumors is less clear as their tumorigenesis does not appear to be controlled by classic oncogenes such as P53, RB, or KRAS. Even within GEP-NETs, there are no common oncogenic drivers; for example, dAXX/ATRX mutations are strongly implicated in the tumorigenesis of pancreatic but not small bowel NETs. Accordingly, the dysregulation of epigenetic mechanisms has been hypothesized as a potential regulator of GEPNET tumorigenesis and has become a major focus of recent studies. despite the heterogeneity of tumor cohorts evaluated in these studies, it is obvious that there are methylation patterns, chromatin remodeling alterations, and microR NA and long non-coding RNA(lncR NA) differential expression profiles that are distinctive of GEPNETs, some of which are correlated with significant differences in clinical outcomes. Several translational studies have provided convincing data identifying potential prognostic biomarkers, and some of these have demonstrated preliminary success as serum biomarkers that can be used clinically. Nevertheless, there are many opportunities to further define the mechanisms by which these epigenetic modifications influence tumorigenesis, and this will provide better insight into their prognostic and therapeutic utility. Furthermore, these findings form the foundation for future studies evaluating the clinical efficacy of epigenetic modifications as prognostic biomarkers, as well as potential therapeutic targets.

Acceptability of Kangaroo Mother Care (KMC) by Mothers with Low-Birth-Weight Babies at Arthur davison Children’s Hospital, in Ndola, Zambia

Background: Kangaroo Mother Care is a simple safe method used to care for low-birth-weight babies. Low-birth-weight is a global public health issue that pose significant challenge to perinatal care systems. Globally, complications due to low-birth-weight are the leading cause of neonatal mortality, resulting in an estimated 1 million deaths annually. Kangaroo Mother Care (KMC) is a low-cost method of care for low-birth-weight infants in areas with inadequate incubators and power outages with positive outcomes. Objectives: To assess factors influencing acceptability of Kangaroo Mother Care (KMC) in NICU at Arthur davison Children’s Hospital in Ndola, Zambia. Methodology: A cross sectional quantitative analytical study design was used. The study was conducted at Arthur davison Children’s Hospital (AdCH) in Ndola, Zambia. The purposive sampling method was used to select the study participants and a total of 129 mothers with Low Weight Babies A were selected to participate structured closed ended questionnaire was used to collect data from the participants using an interview. data was analyzed using a Statistical Package for Social Scientists (SPSS) software version 26. Cross tabulations were done to determine association of variables using a Chi square (x2) test at 95% confidence interval and were assumptions where not met, fishers exact test was used. Multivariate binary logistic regression analysis was used to quantify the relationship between the dependent and independent variables. Results: Most of the respondents had male babies 77%, 69% of the babies weighed 1000 g - 1400 g, majority 79% had normal delivery, 71% of the respondents were multigravida and most of the respondents 79% were married. The study reviewed that 85% of those who attained secondary education accepted KMC, 74% of the respondents without monthly income accepted KMC, 80% of the respondents had positive attitude and accepted KMC. 82% of the respondents who had positive perception accep

Relational and Euclidean Temporal Space

In mathematics, space encompasses various structured sets such as Euclidean, metric, or vector space. This article introduces temporal space—a novel concept independent of traditional spatial dimensions and frames of reference, accommodating multiple object-oriented durations in a dynamical system. The novelty of building temporal space using finite geometry is rooted in recent advancements in the theory of relationalism which utilizes Euclidean geometry, set theory, dimensional analysis, and a causal signal system. Multiple independent and co-existing cyclic durations are measurable as a network of finite one-dimensional timelines. The work aligns with Leibniz’s comments on relational measures of duration with the addition of using discrete cyclic relational events that define these finite temporal spaces, applicable to quantum and classical physics. Ancient formulas have symmetry along with divisional and subdivisional orders of operations that create discrete and ordered temporal geometric elements. Elements have cyclically conserved symmetry but unique cyclic dimensional quantities applicable for anchoring temporal equivalence relations in linear time. We present both fixed equivalences and expanded periods of temporal space offering a non-Greek calendar methodology consistent with ancient global timekeeping descriptions. Novel applications of Euclid’s division algorithm and Cantor’s pairing function introduce a novel paired function equation. The mathematical description of finite temporal space within relationalism theory offers an alternative discrete geometric methodology for examining ancient timekeeping with new hypotheses for Egyptian calendars.

Nano-and micro-particles are being increasingly used to tune interfacial frictional properties in diverse applications,from friction modifiers in industrial lubrication to enhanced biological fluids in human osteoarthritic ***,we assessed the tribological properties of a simulated synovial fluid enriched with non-spherical,poly lactic-co-glycolic acid(PLGA)microparticles(μPL)that have been previously demonstrated for the pharmacological management of osteoarthritis(OA).Three different μPL configurations were fabricated presenting a 20μm×20μm square base and a thickness of 5μm(thin,5H μPL),10μm(10H μPL),and 20μm(cubical,20H μPL).After extensive morphological and physicochemical characterizations,the apparent Young’s modulus of the μPL was quantified under compressive loading returning an average value of~6 kPa,independently of the particle ***,using a linear two-axis tribometer,the static(μ_(s))and dynamic(μ_(d))friction coefficients of the μPL-enriched simulated synovial fluid were determined in terms of particle configuration and concentration,varying from 0(fluid only)to 6×10^(5) μPL/*** particle morphology had a modest influence on friction,possibly because the μPL were fully squeezed between two mating surfaces by a 5.8 N normal load realizing boundary-like lubrication ***,friction was observed to depend on the dimensionless parameterW,defined as the ratio between the total volume of the μPL enriching the simulated synovial fluid and the volume of the fluid *** coefficients of friction were documented to grow withWreaching a plateau of μ_(s)~0.4 and μ_(d)~0.15,already at Ω~2×10^(-3).Future investigations will have to systematically analyze the effect of sliding velocity,normal load,and rigidity of the mating surfaces to elucidate in full the tribological behavior of μPL in the context of osteoarthritis.

Background The study objective was to test the hypothesis that low crude protein(CP)diet with crystalline amino acids(CAA)supplementation improves Lys utilization efficiency for milk production and reduces protein turnover and muscle protein *** lactating multiparous Yorkshire sows were allotted to 1 of 2 isocaloric diets(10.80 MJ/kg net energy):control(CON;19.24%CP)and reduced CP with“optimal”AA profile(OPT;14.00%CP).Sow body weight and backfat were recorded on d 1 and 21 of lactation and piglets were weighed on d 1,14,18,and 21 of *** d 14 and 18,a subset of 9 sows(CON=4,OPT=5)was infused with a mixed solution of 3-[methyl-2H3]histidine(bolus injection)and[13C]bicarbonate(priming dose)first,then a constant 2-h[13C]bicarbonate infusion followed by a 6-h primed constant[1-13C]lysine *** blood and milk sampling were performed to determine plasma and milk Lys enrichment,Lys oxidation rate,whole body protein turnover,and muscle protein *** Over the 21-d lactation period,compared to CON,sows fed OPT had greater litter growth rate(Pd to CON,sows fed OPT had greater efficiency of Lys(Pd whole-body protein turnover efficiency(Pd to CON,sows fed OPT tended to have lower whole body protein breakdown rate(P=0.069).Muscle protein breakdown rate did not differ between OPT and CON(P=0.197).Conclusion Feeding an improved AA balance diet increased efficiency of Lys and reduced whole-body protein turnover and protein *** results imply that the lower maternal N retention observed in lactating sows fed improved AA balance diets in previous studies may be a result of greater partitioning of AA towards milk rather than greater body protein breakdown.

Pressure vessel priming analysis for a regulated liquid propulsion system

An investigation was performed to determine the pressure transient levels and time required to reach pressure equilibrium in a regulated liquid propulsion system due to the priming of a propellant tank during system *** experimental fluid flow test setup was designed and fabricated using a pressure vessel to house distilled water as a propellant simulant for liquid hydrazine and high-pressure gaseous nitrogen as the pressurant.A snubber orifice was installed downstream of the high-pressure gaseous nitrogen bottles to protect a regulator from high-pressure transients,prior to the regulation of incoming gas to beginning of life propulsion system propellant tank *** were conducted to measure pressure transients and time to pressure equilibrium by varying initial system pressures,snubber orifice diameter,and system ullage volume.A mathematical model was developed and compared against the experimental regulated system priming results to predict the system pressures and time to reach system pressure equilibrium during the pressurization period for regulated propulsion systems given initial conditions.

The TP53 gene is well known to be the most frequently mutated gene in human *** addition to mutations,there are > 20 different coding region single-nucleotide polymorphisms (SNPs) in the TP53 gene,as well as SNPs in MdM2,the negative regulator of *** of these SNPs are known to alter p53 pathway *** makes p53 rather unique among cancer-critical genes,*** coding regions of other cancer-critical genes like Ha-Ras,RB,and PI3KCA do not have non-synonymous coding region SNPs that alter their function in *** next frontier in p53 biology will consist of probing which of these coding region SNPs are moderately or strongly pathogenic and whether they influence cancer risk and the efficacy of cancer *** challenge after that will consist of determining whether we can tailor chemotherapy to correct the defects for each ofthese *** we review the SNPs in TP53 and MdM2 that show the most significant impact on cancer and other *** also propose avenues for how this information can be used to better inform personalized medicine approaches to cancer and other diseases.

Are trends in SeaWiFS chlorophyll time-series unusual relative to historic variability

For selected locations in the Atlantic and Pacific Ocean, we compared surface ocean chlorophyll time series extracted from SeaWiFS imagery from 1997-2004 with the results of an ocean coupled circulation and biogeochemical model covering the period 1958-2004. during the 1997-2004 time period, linear trends in model and satellite time series were significantly correlated at most of the 44 sites we studied. Eleven sites were selected for further study, and we used the longer time series of the model to assess whether trends observed during the SeaWiFS period at these 11 sites were unusual in relation to those observed over the longer historical period covered by the model. The results show that the trends observed during the SeaWiFS period were not unusual and fell well within the range in magnitude of linear trends observed in other 8-year periods of model output. This result implies that the SeaWiFS satellite ocean color time series is not yet sufficiently long, on its own, to directly observe any long term changes in phytoplankton chlorophyll that may be occurring in the surface waters of the open ocean as a result of increased ocean stratification linked to global climate changed.

Evaluation of effects of 1,3-dinitrobenzene on sperm motility of hamster using computer assisted semen analysis (CASA)

Aim: To evaluate the effects of 1,3-dinitrobenznene (mdNB) on sperm motility of hamster and to correlate the re-sults with the fertility. Methods: Adult male hamsters were gavaged with one of the 3 dose regimes of mdNB(1.5 mg daily for 4 weeks ,1.5 mg one day a week for 4 weeks and 1.0 mg 3 days a week for 4 weeks). Computerassisted semen analysis (CASA) was used to analyse the sperm motility parameters, curvilinear velocity (VCL) andstraight line velocity (VSL) of sperm in distal corpus epididymides and distal cauda epididymides. In vitro fertilisationwas carried out only for 1.5 mg mdNB daily group to determine the sperm fertilising capacity. Results: There wasa significant reduction in sperm velocity parameters at weeks 3 and 4 after treatment, which was correlated with a de-cline in sperm fertility. Conclusion; Sperm velocity parameters may be used to determine the effect of a toxic insulton the sperm function.

Effects of acut and chronic doses of methoxy acetic acid on hamster sperm fertilising ability

Aim: To evaluate the effects of acute and chronic doses of methoxy acetic acid (MAA) on in vitro fertilisation byhamster sperm and to correlate the data with the testicular damage. Methods: Adult male hamsters were gavagedwith 3 single doses (0, 80, 160 and 650 mg/kg) and 3 chronic doses (0, 8, 32 and 64 mg/kg daily for 5 weeks) ofMAA in distilled water. After treatment hamsters were killed at weekly intervals and spermatozoa recovered from thedistal cauda epididymides were used to assess the fertilising capacity in vitro. The testes were processed for histologicalexamination. Results; Acute doses showed a significant reduction in sperm fertilising ability from week 3 and 4 af-ter treatment and with the chronic doses, the effects were more extensive and persistent. The results were in correpon-dence with the testicular damages observed. Conclusion: It is evident that both acute and chronic doses of MAAcan impair the sperm function by damaging one or more cell populations in the testis.(Asian J Androl 2001 Sep; 3: 209- 216)
