Spain has been one of the main global pandemic epicenters for coronavirus disease 2019(COVID-19).Here,we analyzed>41000 genomes(including>26000 high-quality(HQ)genomes)downloaded from the GISAID repository,including 1245(922 HQ)sampled in *** aim of this study was to investigate genome variation of novel severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)and reconstruct phylogeographic and transmission patterns in *** analysis suggested at least 34 independent introductions of SARS-CoV-2 to Spain at the beginning of the *** lineages spread very successfully in the country,probably favored by super-spreaders,namely,A2 a4(7.8%),A2 a5(38.4%),A2 a10(2.8%),B3 a(30.1%),and B9(8.7%),which accounted for 87.9% of all genomes in the Spanish *** distinct feature of the Spanish SARS-Cov-2 genomes was the higher frequency of B lineages(39.3%,mainly B3 a+B9)than found in any other European *** B3 a,B9,(and an important sub-lineage of A2 a5,namely,A2 a5 c)most likely originated in Spain,the other three haplogroups were imported from other European *** B3 a strain may have originated in the Basque Country from a B3 ancestor of uncertain geographic origin,whereas B9 likely emerged in *** time of the most recent common ancestor(TMRCA)of SARS-CoV-2 suggested that the first coronavirus entered the country around 11 February 2020,as estimated from the TMRCA of B3 a,the first lineage detected in the ***,earlier claims that the D614 G mutation is associated to higher transmissibility is not consistent with the very high prevalence of COVID-19 in Spain when compared to other countries with lower disease incidence but much higher frequency of this mutation(56.4% in Spain vs.82.4% in rest of Europe).Instead,the data support a major role of genetic drift in modeling the micro-geographic stratification of virus strains across the country as well as the role of SARS-CoV-2 super-spreaders.

Background: Nutritional value of proteins in feed ingredients can be negatively affected by hydrothermal processing, which causes large variation in the bioavailability of amino acids(AA) and negatively affects animal productive performance. Supplementation of exogenous proteases could increase the rate of digestion of damaged proteins, thereby increasing overall AA digestibility and bioavailability. The aim was to determine the effect of exogenous protease supplementation on the apparent ileal digestibility(AID) of crude protein(CP) and AA of soybean meals(SBM) with different degrees of hydrothermal processing in ***: The experiment involved a 3 × 2 factorial arrangement, with SBM processing time(commercial SBM or autoclaved for 30 or 60 min at 120 ℃) and protease supplementation(not supplemented and supplemented) as factors. Protease was included at three times the recommended dose(0.06%) and the experimental diets were fed from 15 to 21 ***: The interaction between the effects of SBM processing and protease supplementation was significant for the AID of CP(P = 0.01), Trp(P = 0.01), Gly(P = 0.03) and Pro(P = 0.03), and also for the average daily gain(P = 0.01)and feed conversion ratio(P = 0.04). Increasing the processing time of SBM decreased(P < 0.0001) the AID of all amino acids, whilst the effect of protease supplementation was only significant for the AID of Phe(P = 0.02) and Tyr(P = 0.01).Conclusions: Exogenous protease supplementation at three times the commercial dose does not seem to offset the negative effects of hydrothermal processing of SBM on the apparent ileal digestibility of CP and amino acids or performance of broilers. Whilst positive numerical improvements of digestibility and performance(ADG and FCR)were noticed with protease supplementation at relatively mild processing levels, negative results were obtained with the harsh-processed meals.

A frozen mummy was found in Cerro Aconcagua (Argentina)in 1985(Fig.1a-c).Archaeologists identified this mummy as a seven-year-old Inca sacrifice victim living at the time of the Inca Civilization about 500years ago [1].The child was sacrificed following an Incan ritual known as *** complete mitochondrial DNA (mtDNA) genome of this mummy was recently analyzed in Gomez-Carballa et al.[2].This mitogenome belongs to a new sub-branch of the Native American phylogeny named C1bi dated to approximately 14 thousand years ago (loja).Information retrieved from a large database of mtDNA profiles indicated the existence of a few closely related haplotypes in Peru (including one haplotype retrieved from ancient DNA of an individual belonging to the Wari Empire)and the Aymaras from *** these data suggested a Peruvian Inca origin for this C1bi haplotype,in good agreement with archaeological reports [1].

A chemical method for removing calcium sulfate saturated solutions(0.016 mol/L CaSO_(4))using barium chloride(BaCl_(2)∙2H_(2)O)and sodium phosphate(Na_(3)PO_(4))was experimentally *** main interest is to remove these ions from the solution through the precipitation of two solid species:sulfate(SO_(4)^(2−))as barite(BaSO4),and calcium(Ca^(2+))as hydroxyapatite(Ca_(5)(PO_(4))_(3)OH).Additionally,a solid/liquid separation method(i.e.,flotation)was explored,using oleic acid and dodecylamine as *** results show that,the chemical treatment of saturated solutions at 60℃,pH 11.5 and using 3.9 g/L BaCl_(2)·2H_(2)O and 1.6 g/L Na_(3)PO_(4),promotes the precipitation of barium sulfate and calcium-deficient hydroxyapatite(Ca_(10−x)(HPO_(4))_(x)(PO_(4))_(6−x)(OH)_(2−x)),with residual concentrations of calcium and sulfate below 0.10 and 5 mg/L,*** residual calcium concentration increases to 28 mg/L when using the same amount of reactants,at temperature and pH values below those *** highest flotation recovery of hydroxyapatite with oleic acid at pH 9.5 was about 80%,while that of barite floated with dodecylamine at pH 6.5 was about 90%.

AIM: To study whether matrix metalloproteinase-9 (MMP-9) is a key factor in epithelial damage in the dextran sodium sulphate (DSS) model of colitis in ***: MMP-9-deficient and wild-type (wt) mice were given 5% DSS in drinking water for 5 d followed by recovery up to 7 d. On d 5 and 12 after induction of colitis, gelatinases, MMP-2 and MMP-9, were measured in homogenates of colonic tissue by zymography and Western blot, whereas tissue inhibitor of metalloproteinases (TIMPs) were measured by reverse zymography. The gelatinolytic activity was also determined in supernatants of polymorphonuclear leukocytes (PMN) isolated from mice blood. Moreover, intestinal epithelial cells were stimulated with TNF-α to study whether these cells were able to produce MMPs. Finally, colonic mucosal lesions were measured by microscopic examination. RESULTS: On d 5 of colitis, the activity of MMP-9 was increased in homogenates of colonic tissues (0.24 ± 0.1 vs 21.3 ± 6.4, P < 0.05) and PMN from peripheral blood in wt (0.5 ± 0.1 vs 10.4 ± 0.7, P < 0.05), but not in MMP-9-deficient animals. The MMP-9 activity was also up-regulated by TNF-α in epithelial intestinal cells (2.5 ± 0.5 vs 14.7 ± 3.0, P < 0.05). Although colitis also led to increase of TIMP-1 activity, the MMP-9/TIMP-1 balance remained elevated. Finally, in the MMP-9-deficient colitic mice both the extent and severity of intestinal epithelialinjury were significantly attenuated when compared with wt mice. CONCLUSION: We conclude that DSS induced colitis is markedly attenuated in animals lacking MMP-9. This suggests that intestinal injury induced by DSS is modu-lated by MMP-9 and that inhibition of this gelatinase may reduce inflammation.

The aim of this study was the synthesis of silver nanoparticle using Annona diversifolia *** silver nanoparticles obtained were analyzed by spectroscopic methods and dynamic light scattering *** inhibition of AgNPs was evaluated against Bacillus cereus,Klebsiella pneumoniae and Enterobacter *** results showed that AgNPs have high values at 3 keV and particle size between 45 to 58 nm with a homogenous *** AgNPs showed growth inhibition against Klebsiella pneumoniae and Enterobacter *** studies are needed to confirm the potential antimicrobial of different AgNP from *** in Gram negative and Gram positive bacteria.

Group-specific functional patterns of mitochondrion-related organelles shed light on their multiple transitions from mitochondria in ciliated protists

Adaptations of ciliates to hypoxic environments have arisen independently several *** on mitochondrion-related organelle(MRO)metabolisms from distinct anaerobic ciliate groups provide evidence for understanding the transitions from mitochondria to MROs within *** deepen our knowledge about the evolutionary patterns of ciliate anaerobiosis,mass-culture and single-cell transcriptomes of two anaerobic species,Metopus laminarius(class Armophorea)and Plagiopyla ***(class Plagiopylea),were sequenced and their MRO metabolic maps were *** addition,we carried out comparisons using publicly available predicted MRO proteomes from other ciliate classes(i.e.,Armophorea,Litostomatea,Muranotrichea,Oligohymenophorea,Parablepharismea and Plagiopylea).We found that single-cell transcriptomes were similarly comparable to their mass-culture counterparts in predicting MRO metabolic pathways of *** patterns of the components of the MRO metabolic pathways might be divergent among anaerobic ciliates,even among closely related ***,our findings indicate the existence of group-specific functional relics of electron transport chains(ETCs).Detailed group-specific ETC functional patterns are as follows:full oxidative phosphorylation in Oligohymenophorea and Muranotrichea;only electron-transfer machinery in Armophorea;either of these functional types in Parablepharismea;and ETC functional absence in Litostomatea and *** findings suggest that adaptation of ciliates to anaerobic conditions is group-specific and has occurred multiple *** results also show the potential and the limitations of detecting ciliate MRO proteins using single-cell transcriptomes and improve the understanding of the multiple transitions from mitochondria to MROs within ciliates.
