Dysfunctional stem and progenitor cells impair fracture healing with age

Successful fracture healing requires the simultaneous regeneration of both the bone and vasculature;mesenchymal stem cells (MSCs) are directed to replace the bone tissue, while endothelial progenitor cells (EPCs) form the new vasculature that supplies blood to the fracture site. In the elderly, the healing process is slowed, partly due to decreased regenerative function of these stem and progenitor cells. MSCs from older individuals are impaired with regard to cell number, proliferative capacity, ability to migrate, and osteochondrogenic differentiation potential. The proliferation, migration and function of EPCs are also compromised with advanced age. Although the reasons for cellular dysfunction with age are complex and multidimensional, reduced expression of growth factors, accumulation of oxidative damage from reactive oxygen species, and altered signaling of the Sirtuin-1 pathway are contributing factors to aging at the cellular level of both MSCs and EPCs. Because of these geriatric-specific issues, effective treatment for fracture repair may require new therapeutic techniques to restore cellular function. Some suggested directions for potential treatments include cellular therapies, pharmacological agents, treatments targeting age-related molecular mechanisms, and physical therapeutics. Advanced age is the primary risk factor for a fracture, due to the low bone mass and inferior bone quality associated with aging;a better understanding of the dysfunctional behavior of the aging cell will provide a foundation for new treatments to decrease healing time and reduce the development of complications during the extended recovery from fracture healing in the elderly.

Preoperative Feeding in Single Ventricle Neonates is Predictive of Shorter Time to Goal Feed

Background:Patients with single ventricle anatomy are at increased risk of growth failure and *** cardiac centers,there is little standardization of feeding practices in this complex *** hypothesized that initiation of our center’s preoperative feeding protocol would result in decreased gastrostomy tube(G-tube)use,decreased length of stay and would not result in increased Necrotizing Enterocolitis(NEC)***:A single institution review of 52 patients who had undergone stage I single ventricle palliative repair was *** diagnoses were hypoplastic left heart syndrome(39%),atrioventricular canal(15%),and other(46%).Postoperative parameters such as time to goal feed and need for gastrostomy tube(G-tube)were compared among preoperatively fed and non-preoperatively fed *** to goal feed was calculated as time from first postoperative enteral feed to goal volume of 100 mL/***:Of the 26 patients who met inclusion criteria for preoperative feeding,22 patients(85%)were fed prior to *** proportional hazard ratio revealed that age at surgery(p=0.047)and being preoperatively fed(p=0.001)were associated with reaching goal feed *** analysis revealed that being preoperatively fed made a patient twice as likely to reach goal feed sooner(p=0.047).Univariable logistic regression revealed that days on total parenteral nutrition(p=0.018),length of hospitalization(p=0.008),and time to 1st postoperative feed(p=0.020)were significantly associated with higher odds of needing a G-tube *** logistic regression did not show any predictors of postoperative G-tube ***,there was a trend towards lower G-tube usage in the preoperatively fed ***:Implementing a standardized preoperative feeding protocol in single ventricle neonates can result in significantly shorter time to goal feed in preoperatively fed *** is beneficial for institutions to begin implementi

Extending the neurocircuitry of behavioural inhibition:a role for the bed nucleus of the stria terminalis in risk for anxiety disorders

Behavioural inhibition is a biologically based risk factor for anxiety *** with behavioural inhibition are shy,cautious and avoidant of new *** research on behavioural inhibition has focused on the amygdala as an underlying neural substrate and has identified differences in amygdala function and volume;however,amygdala findings have yet to lead to meaningful interventions for prevention or treatment of anxiety *** bed nucleus of the stria terminalis(BNST)is a prime candidate to be a neural substrate of behavioural inhibition,given current evidence of BNST function and development in human research and animal *** with behavioural inhibition have an increased startle response to safety cues and an increased cortisol response to social evaluative situations,both of which are mediated by the *** rodents,activation of the BNST underlies contextual fear responses and responses to uncertain and sustained ***-human primates with anxious temperament(the macaque equivalent of behavioural inhibition)have increased BNST activity to ambiguous social situations,and activity of the BNST in anxious temperament is significantly ***,the BNST is sexually dimorphic and continues to develop into adulthood,paralleling the development of anxiety disorders in ***,these findings suggest that further investigation of the BNST in behavioural inhibition is necessary and may lead to new avenues for the prevention and treatment of anxiety disorders.

This paper presents a comprehensive experimental study on the effect of extreme waves on a LNG *** LNG carrier model was equipped with a variety of sensors to measure motions,green water height on deck as well as local and global *** in transient wave packets provided the general performance in waves in terms of response amplitude operators and were accompanied by tests in regular waves with two different wave *** tests allowed detailed insights into the nonlinear behavior of the vertical wave bending moment in steep waves showing that green water on deck can contribute to a decrease of vertical wave bending ***,systematic model tests in irregular waves were performed to provide the basis for statistical *** is shown that the generalized extreme value distribution model is suitable for the estimation of the extreme peak values of motions and ***,model tests in tailored extreme wave sequences were conducted comparing the results with the statistical *** this purpose,analytical breather solutions of the nonlinear Schrödinger equation were applied to generate tailored extreme waves of certain critical wave lengths in terms of ship *** these design extreme waves,the LGN carrier was also investigated in the model scale reproduction of the real-world Draupner *** comparing the motions,vertical wave bending moment,green water column and slamming pressures it is concluded that the breather solutions are a powerful and efficient tool for the generation of design extreme waves of certain critical wave lengths for wave/structure investigations on different subjects.

Liquidity risk integration in portfolio choice： The bid efficient frontier

In this paper, a tractable solution is proposed to integrate, to a certain extent, market liquidity risk in the portfolio selection process. It is shown how an investor may take advantage of this additional risk source within the standard mean-variance optimization framework, by in certain circumstances overcoming the pitfalls of illiquidity and in others seizing a liquidity premium. Bid prices appear effective to capture liquidity risk. The efficient frontier conceived with bid prices consists of mean-variance optimal allocations that cover more liquid stocks (large caps) under stressed market conditions and less liquid stocks (small caps) under normal conditions.

Efficiency for Sustainability—Smart Energy Management in Social Housing

An average energy consumption distribution in household at the worldwide level illustrates that more than three quarters of total consumption is contributed to room heating and almost 12% to water heating for all the living necessities. Although a slow fall of domestic energy consumption has occurred in the recent 20 years (from 1990 to 2011) with a regular decrease between ?1.2% and ?1.4%/year per dwelling as a result of a decrease income corresponding to the economic crisis in 2008, whereas energy prices for households has increase since 2004, the energy cost of paying especially for space heating and domestic hot water (DHW) supplying is still going up. At the EU level, the building sector including residential, commercial and other service buildings is regarded as the key to greater energy efficiency, because according to statistics the final energy consumption for building sector has taken the largest proportion about 40% until 2020, which is apparently higher than the share in transportation sector by 32% and industry sector by 24%. In the scope of the ongoing research and investigation on energy efficiency in residential fields and its impact on environment and climate, how to investigate smart energy management methods for the promotion of sustainable consumption and green living patterns has been already paid much attention, however it has to be studied further and thoroughly, especially among energy consumer groups whose energy costs have no or just little relevance or dependence on major income source, which leads to a lack of energy saving awareness by users. Energy consumers living in social housing buildings represent this kind of energy consumer group which receive the governmental relief fund as their family income in a great measure, they have different culture, educational and age backgrounds. This paper presents firstly some research results based on authors’ practical experiences on the projects about energy efficiency in social housing buildings

Rheumatoid arthritis(RA) is an autoimmune disease affecting 1% of the world population and is characterized by chronic inflammation of the joints sometimes accompanied by extra-articular manifestations. K/Bx N mice, originally described in 1996 as a model of polyarthritis, exhibit knee joint alterations. The aim of this study was to describe temporomandibular joint(TMJ)inflammation and damage in these mice. We used relevant imaging modalities, such as micro-magnetic resonance imaging(μMRI)and micro-computed tomography(μCT), as well as histology and immunofluorescence techniques to detect TMJ alterations in this mouse model. Histology and immunofluorescence for Col-I, Col-II, and aggrecan showed cartilage damage in the TMJ of K/Bx N animals, which was also evidenced by μCT but was less pronounced than that seen in the knee joints. μMRI observations suggested an increased volume of the upper articular cavity, an indicator of an inflammatory process. Fibroblast-like synoviocytes(FLSs)isolated from the TMJ of K/Bx N mice secreted inflammatory cytokines(IL-6 and IL-1β) and expressed degradative mediators such as matrix metalloproteinases(MMPs). K/Bx N mice represent an attractive model for describing and investigating spontaneous damage to the TMJ, a painful disorder in humans with an etiology that is still poorly understood.
