DIW1 encoding a cladeⅠpp2C phosphatase negatively regulates drought tolerance by de-phosphorylating TaSnRK1.1 in wheat

Drought seriously impacts wheat production(Triticum aestivum L.),while the exploitation and utilization of genes for drought tolerance are insufficient.Leaf wilting is a direct reflection of drought tolerance in plants.Clade A pp2Cs are abscisic acid(ABA)co-receptors playing vital roles in the ABA signaling pathway,regulating drought response.However,the roles of other clade pp2Cs in drought tolerance,especially in wheat,remain largely unknown.Here,we identified a gain-of-function drought-induced wilting 1(DIW1)gene from the wheat Aikang 58 mutant library by map-based cloning,which encodes a cladeⅠprotein phosphatase 2C(Tapp2C158)with enhanced protein phosphatase activity.phenotypic analysis of overexpression and CRISpR/Cas9 mutant lines demonstrated that DIW1/Tapp2C158 is a negative regulator responsible for drought resistance.We found that Tapp2C158 directly interacts with TaSnRK1.1 and de-phosphorylates it,thus inactivating the TaSnRK1.1–Ta AREB3 pathway.Tapp2C158 protein phosphatase activity is negatively correlated with ABA signaling.Association analysis suggested that C-terminal variation of Tapp2C158 changing protein phosphatase activity is highly correlated with the canopy temperature,and seedling survival rate under drought stress.Our data suggest that the favorable allele with lower phosphatase activity of Tapp2C158 has been positively selected in Chinese breeding history.This work benefits us in understanding the molecular mechanism of wheat drought tolerance,and provides elite genetic resources and molecular markers for improving wheat drought tolerance.

Identifying mechanisms and pathways involved in gene–environment interplay and phenotypic plasticity is a long-standing challenge.It is highly desirable to establish an integrated framework with an environmental dimension for complex trait dissection and prediction.A critical step is to identify an environmental index that is both biologically relevant and estimable for new environments.With extensive field-observed complex traits,environmental profiles,and genome-wide single nucleotide polymorphisms for three major crops(maize,wheat,and oat),we demonstrated that identifying such an environmental index(i.e.,a combination of environmental parameter and growth window)enables genome-wide association studies and genomic selection of complex traits to be conducted with an explicit environmental dimension.Interestingly,genes identified for two reaction-norm parameters(i.e.,intercept and slope)derived from flowering time values along the environmental index were less colocalized for a diverse maize panel than for wheat and oat breeding panels,agreeing with the different diversity levels and genetic constitutions of the panels.In addition,we showcased the usefulness of this framework for systematically forecasting the performance of diverse germplasm panels in new environments.This general framework and the companion CERIS-JGRA analytical package should facilitate biologically informed dissection of complex traits,enhanced performance prediction in breeding for future climates,and coordinated efforts to enrich our understanding of mechanisms underlying phenotypic variation.

The vagus nerve(VN)plays an important role in regulating physiological conditions in the gastrointestinal(GI)tract by communicating via the parasympathetic pathway to the enteric nervous system(ENS).However,the lack of knowledge in the neurophysiology of the VN and GI tract limits the development of advanced treatments for autonomic dysfunctions related to the VN.To better understand the complicated underlying mechanisms of the VN-GI tract neurophysiology,it is necessary to use an advanced device enabled by microfabrication technologies.Among several candidates including intraneural probe array and extraneural cuff electrodes,microchannel electrode array devices can be used to interface with smaller numbers of nerve fibers by securing them in the separate channel structures.previous microchannel electrode array devices to interface teased nerve structures are relatively bulky with thickness around 200µm.The thick design can potentially harm the delicate tissue structures,including the nerve itself.In this paper,we present a flexible thin film based microchannel electrode array device(thickness:11.5µm)that can interface with one of the subdiaphragmatic nerve branches of the VN in a rat.We demonstrated recording evoked compound action potentials(ECAp)from a transected nerve ending that has multiple nerve fibers.Moreover,our analysis confirmed that the signals are from C-fibers that are critical in regulating autonomic neurophysiology in the GI tract.

Neuronal necroptosis-an emerging form of regulated cell death associated with neuroinflammatory signaling:Alzheimer’s disease(AD)is characterized by the presence of extracellular amyloid-β(Aβ)plaques and intracellular tau neurofibrillary tangles as well as progressive neuronal loss.Recent evidence has suggested that prolonged neuroinflammation with increased levels of cytokines,arising from neuronal injury,innate immune responses from glial cells,and peripheral inflammation,leads to neuronal death and AD progression.

Alterations of SARS-CoV-2 Evolutionary Dynamics by pharmaceutical Factors

The outbreak of SARS-CoV-2(severe acute respiratory syndrome coronavirus 2)has been influenced by the human response to the virus.These responses have undoubtedly impacted the evolutionary dynamics of the virus in ways distinct from a scenario lacking a widespread response.Two important pharmaceutical interventions,vaccination and the utilization of medications,particularly molnupiravir,known to have mutagenic properties,were the focus of this article.The impact of molnupiravir on human health was evaluated through 3 mechanisms:viral resistance,mutagenesis of SARS-CoV-2,and mutagenesis occurring in patients undergoing treatmentwithmolnupiravir.Thesemechanisms,as well as the impact of vaccination,have inadvertently given rise to unforeseen challenges in the management of the COVID-19 crisis.Taking a systems view in future pandemic responses,and taking into account the evolution of the pandemic virus,may be critical to ending the pandemic at an earlier date.

Evaluation of a software positioning tool to support SMEs in adoption of big data analytics

Big data analytics has been widely adopted by large companies to achieve measurable benefits including increased profitability,customer demand forecasting,cheaper development of products,and improved stock control.Small and medium sized enterprises(SMEs)are the backbone of the global economy,comprising of 90%of businesses worldwide.However,only 10%SMEs have adopted big data analytics despite the competitive advantage they could achieve.previous research has analysed the barriers to adoption and a strategic framework has been developed to help SMEs adopt big data analytics.The framework was converted into a scoring tool which has been applied to multiple case studies of SMEs in the UK.This paper documents the process of evaluating the framework based on the structured feedback from a focus group composed of experienced practitioners.The results of the evaluation are presented with a discussion on the results,and the paper concludes with recommendations to improve the scoring tool based on the proposed framework.The research demonstrates that this positioning tool is beneficial for SMEs to achieve competitive advantages by increasing the application of business intelligence and big data analytics.

Runx2 regulates peripheral nerve regeneration to promote Schwann cell migration and re-myelination

Runx2 is a major regulator of osteoblast differentiation and function;however,the role of Runx2 in peripheral nerve repair is unclea r.Here,we analyzed Runx2expression following injury and found that it was specifically up-regulated in Schwann cells.Furthermore,using Schwann cell-specific Runx2 knocko ut mice,we studied peripheral nerve development and regeneration and found that multiple steps in the regeneration process following sciatic nerve injury were Runx2-dependent.Changes observed in Runx2 knoc kout mice include increased prolife ration of Schwann cells,impaired Schwann cell migration and axonal regrowth,reduced re-myelination of axo ns,and a block in macrophage clearance in the late stage of regeneration.Taken together,our findings indicate that Runx2 is a key regulator of Schwann cell plasticity,and therefore peripheral nerve repair.Thus,our study shows that Runx2 plays a major role in Schwann cell migration,re-myelination,and peripheral nerve functional recovery following injury.

Designing a medical ethics curriculum in ophthalmology

Ethical principles form a bedrock to medical practice in any specialty,guiding physicians to appropriate attitudes and behaviors.A formal ethics curriculum can be difficult to generate de novo in an ophthalmology training program.A number of barriers exist in most ophthalmology departments:trainees may think ethics is of secondary importance compared to core basic and clinical science topics;most ophthalmology faculty have no formal degree in medical ethics;there is limited didactic time with competing academic,clinical,and surgical priorities;work-hours regulations may limit the time available to deliver“para-professional”lectures;and there is a belief that the medical ethics lectures during medical school is a sufficient amount of coursework to last through a physician’s career with no need for continuing professional development.The four pillars of medical ethics are beneficence,non-maleficence,autonomy,and justice.In addition,morals,ethics,and professionalism are important aspects of sound medical practice.A curriculum specific to medical ethics in ophthalmology can be developed in any of our sub-specialties and include lectures,curated readings,case rounds,and clinic wrap-up sessions.Ethical considerations are part of everyday clinical practice,and a structured ethics curriculum can be incorporated into ophthalmology training programs.The concept of backward design can be used to structure the curriculum,starting with the expected outcome,then designing authentic assessments,and finally putting together a learning plan that has students actively involved in ethical discussions.This paper will provide a guide to developing an ethics curriculum for an ophthalmology training program utilizing the concept of backwards design and guide the reader through the process of developing expected learning outcomes,authentic assessments,and a unified learning plan.

production of proinflammatory cytokines in the central nervous system is a key process in the neuroinflammatory response to trauma,infection,and neurodegenerative diseases(Kumar,2019).These intercellular signaling molecules play multiple roles in the immune response in the central nervous system including the orchestration of the sickness response to innate immune perturbations in the brain(Dantzer et al.,2008).
