The immune-stromal cell interactions play a key role in health and diseases. In periodontitis, the most prevalent infectious disease in humans, immune cells accumulate in the oral mucosa and promote bone destruction by inducing receptor activator of nuclear factor-κB ligand (RANKL) expression in osteogenic cells such as osteoblasts and periodontal ligament cells. However, the detailed mechanism underlying immune–bone cell interactions in periodontitis is not fully understood. Here, we performed single-cell RNAsequencing analysis on mouse periodontal lesions and showed that neutrophil–osteogenic cell crosstalk is involved in periodontitis-induced bone loss. The periodontal lesions displayed marked infiltration of neutrophils, and in silico analyses suggested that the neutrophils interacted with osteogenic cells through cytokine production. Among the cytokines expressed in the periodontal neutrophils, oncostatin M (OSM) potently induced RANKL expression in the primary osteoblasts, and deletion of the OSM receptor in osteogenic cells significantly ameliorated periodontitis-induced bone loss. Epigenomic data analyses identified the OSM-regulated RANKL enhancer region in osteogenic cells, and mice lacking this enhancer showed decreased periodontal bone loss while maintaining physiological bone metabolism. These findings shed light on the role of neutrophils in bone regulation during bacterial infection, highlighting the novel mechanism underlying osteoimmune crosstalk.

Using a novel hemostatic peptide solution to prevent bleeding after endoscopic submucosal dissection of a gastric tumor

BACKGROUND Endoscopic mucosal dissection has become the standard treatment for early gastric ***,post-endoscopic submucosal dissection(ESD)ulcer occurs in 4.4%of *** study hypothesized whether applying PuraStat,a novel hemostatic peptide solution,prevents post-ESD *** To investigate the preventive potential of PuraStat,a hemostatic formulation,against bleeding in post-ESD gastric *** Between May 2022 and March 2023,101 patients(Group P)underwent ESD for gastric diseases at our hospital and received PuraStat(2 mL)for post-ESD *** retrospectively compared this group with a control group(Group C)com-prising 297 patients who underwent ESD for gastric diseases at our hospital between April 2017 and March 2021.P values<0.05 on two-sided tests indicated *** Post-ESD bleeding occurred in 6(5.9%)(95%CI:2.8-12.4)and 20(6.7%)(95%CI:4.4-10.2)patients in Groups P and C,respectively,with no significant between-group *** relative risk was 1.01(95%CI:0.95-1.07).The lesser curvature or anterior wall was the bleeding site in all 6 patients who experienced postoperative bleeding in Group *** multivariate analysis,the odds ratios for resection diameter≥50 mm and oral anticoagulant use were 6.63(95%CI:2.52-14.47;P=0.0001)and 4.04(1.26-0.69;P=0.0164),*** adjusted odds ratio of post-ESD bleeding and PuraStat was 1.28(95%CI:0.28-2.15).CONCLUSION PuraStat application is not associated with post-ESD ***,the study suggests that gravitational forces may affect the effectiveness of applied PuraStat.

A Case of Cerebrospinal Drainage for Paraplegia Complicated by Acute Aortic Dissection (Stanford B) Followed by TEVAR in the Subacute Phase

The patient is 50-year-old man. He was admitted to our hospital with a strong back pain and diagnosed as an acute type B aortic dissection. On the second day of hospitalization, he developed symptoms of paraplegia, and we considered TEVAR, but we were concerned that TEVAR intervention in the acute phase might worsen the dissection, so we first placed a cerebrospinal drainage (CSFD) device, which resulted in improvement of his symptoms. Thereafter, although his lower limb mobility was fine, he underwent thoracic stent graft aortic repair (TEVAR) in the subacute phase due to worsening ULP. The patient had a good postoperative course and was discharged home unassisted. The initial placement of CSFD was effective in reducing the incidence of paraplegia as a complication of TEVAR and in bringing the timing of TEVAR intervention from the acute phase to the subacute phase.

We introduced a dual electron accepting layer composed of tetrafluoro-tetracyanoquinodimethane （F4-TCNQ） and MoO3 for thermoelectric devices based on a pentacene layer. We found that the power factor is enhanced by placing an F4-TCNQ layer directly in contact with the pentacene layer and it is also enhanced by placing a MoO3 layer between the F4-TCNQ layer and the Au electrode. By examining the contact resistance using a field effect transistor and a hole-only diode, we confirmed that the hole injection is improved due to the reduction of contact resistance at the interface between the MoO3 layer and the Au electrode.

Biomimetic Riblets Inspired by Sharkskin Denticles： Digitizing, Modeling and Flow Simulation

While sharkskin surface roughness in terms of denticle morphology has been hypothesized but remains yet controversial to be capable of achieving turbulent flow control and drag reduction, sharkskin-inspired ＂riblets＂ have been reported to be an effective biomimetic design. Here we address an integrated study of biomimetic riblets inspired by sharkskin denticles by combining 3D digitizing and mod- eling of＂fresh＂ denticles and computational fluid dynamic modeling of turbulent flows on a rough surface with staggered denticles and hound-tooth-patterned grooves. Realistic microstructures of denticles in five shark species of Galapagos, great white, whitetip reef, blacktip reef, and hammerhead sharks were first measured and digitized in three fold：（1） 2D imaging of lubricated sharkskin in a wet state by means of a ＂nano-suit＂ technique with a Field-Emission Scanning Electron Microscope （FE-SEM）; （2） 3D structures of sharkskin denticles with a micro-focus X-ray CT; and （3） single denticles of the five shark species in a 3D manner with 3D-CAD. The denticles at mid-body location in the five species were observed to have a structure of five non-uniform-ridges （herein termed ＂non-uniform grooves＂） with Angles Of Inclination （AOI） ranging over 20° - 32°. Hydrodynamics associated with the unique five-ridge denticles were then in- vestigated through modeling turbulent flow past a denticle-staggered skin surface. We further constructed a biomimetic riblet model inspired by the non-uniform grooves and investigated the hydrodynamic effects of height-to-spacing ratios of mid-ridge and side-ridges. Our results indicate that the morphological non-uniformity in sharkskin denticles likely plays a critical role in passively controlling local turbulent flow and point to the potential of denticle-inspired biomimetic riblets for turbulent-flow control in aquatic vehicles as well as other fluid machinery.

Aberrant gene methylation in the peritoneal fluid is a risk factor predicting peritoneal recurrence in gastric cancer

AIM:To investigate whether gene methylation in the peritoneal fluid (PF) predicts peritoneal recurrence in gastric cancer ***: The gene methylation of CHFR (checkpoint with forkhead and ring finger domains), p16, RUNX3 (runt-related transcription factor 3), E-cadherin, hMLH1 (mutL homolog 1), ABCG2 (ATP-binding cassette, sub-family G, member 2) and BNIP3 (BCL2/adenovirus E1B 19 kDa interacting protein 3) were analyzed in 80 specimens of PF by quantitative methylation-specific polymerase chain reaction (PCR). Eighty patients were divided into 3 groups; Group A (n=35):the depth of cancer invasion was less than the muscularis propria; Group B (n=31): the depth of cancer invasion was beyond the muscularis propria. Both group A and B were diagnosed as no cancer cells in peritoneal cytology and histology; Group C (n=14): disseminated nodule was histologically diagnosed or cancer cells were cytologically defi ned in the peritoneal ***: The positive rates of methylation in CHFR, E-cadherin and BNIP3 were significantly different among the 3 groups and increased in order of group A, B and C (0%,0% and 21% in CHFR, P<0.05; 20%, 45% and 50% in E-cadherin, P<0.05;26%,35% and 71% in BNIP3, P<0.05). In addition, the multigene methylation rate among CHFR, E-cadherin and BNIP3 was correlated with group A, B and C (9%,19% and 57%, P<0.001). Moreover, the prognosis was analyzed in group B, excluding 3 patients who underwent a non-curative resection. Two of the 5 patients with multigene methylation showed peritoneal recurrence after surgery, while those without or with a single gene methylation did not experience recurrence (P<0.05).CONCLUSION: This study suggested that gene methylation in the PF could detect occult neoplastic cells in the peritoneum and might be a risk factor for peritoneal metastasis.

革命不是请客吃饭——做生意更加不是。因此在当年,日本LESSTHANHUMAN品牌的掌门人甲贺润先生会出价买下一个还在设计学院的年轻人一系列的设计作品,并把它们正儿八经地作为该品牌的主力款式进行大规模的推广宣传,就是一件让人不容易想明白的事。但后来,看见这些精致又成熟的作品,人们则会猜中其中缘由。这可不是一般意义上交给老师获得高分就万事大吉的课外作业,而令甲贺润青睐的作品主人masatsugu,也迅速地用随后获得的一页订单创建起了自己的牌子——2005年,年轻的SOLIDBLUE就这样诞生了。在此之前,这个叫masatsugu的年轻人只是在从设计学院毕业后,短短地在实习了一阵——这叫人不得不佩服初生牛犊的热情和闯劲。而面对记者,masatsugu不太愿意多谈自己的过往成绩和个人履历,按照他自己的说法,自己现在仍然也还是个新人。所以在对话过程中,我们对眼镜设计领域的探讨远远要大于对他个人本身的探讨。

The leaf extract of Mallotus japonicus and its major active constituent, rutin,suppressed on melanin production in murine B16F1 melanoma

Objective: To find anti-melanogenesis materials used in whitening ***: The ethanolic leaf extract of Mallotus japonicus(M. japonicus) having an anti-melanogenesis activity was separated by a sephadex LH-20 chromatography. Each fraction was measured for its tyrosinase inhibitory activity together with its polyphenol content using the Folin–Ciocalteu method. The anti-melanogenesis activity of the active fractions was determined by the melanin content in the murine B16F1 melanoma. The active fractions were put together due to similar constituents, and then separated by high performance liquid chromatography using a C-18 ODS column. The major antimelanogenesis compound was identified using1 H and13C-NMR and liquid chromatography-mass ***: The ethanolic leaf extract of M. japonicus showed an anti-tyrosinase activity with a high polyphenol content, resulting in suppression of melanin production in the B16F1 melanoma. The extract was separated and the active compound was identical as rutin based on the1 H,13C-NMR and liquid chromatography-mass spectrometry analysis data. In addition, the rutin treatment with cells reduced the melanin content in a concentration dependent manner without any cell toxicity. The leaf extract of M. japonicus containing rutin would be useful in whitening cosmetics for protection from UV-light exposure to be limiting the accumulation of melanin in ***: The leaf extract of M. japonicus and/or rutin isolated from the extract as a key whitening agent would be useful as a whitening cosmetic material for protecting against disorder skin due to melanin accumulation.
