Characterization of the genes involved in nitrogen cycling in wastewater treatment plants using DNA microarray and most probable number-PCR

To improve nitrogen removal performance of wastewater treatment plants （WWTPs）, it is essential to understand the behavior of nitrogen cycling communities, which comprise various microorganisms. This study characterized the quantity and diversity of nitrogen cycling genes in various processes of municipal WWTPs by employing two molecular-based methods：most probable number-polymerase chain reaction （MPN-PCR） and DNA microarray. MPN-PCR analysis revealed that gene quantities were not statistically different among processes, suggesting that conventional actwated sludge processes （CAS） are similar to nitrogen removal processes in their ability to retain an adequate population of nitrogen cycling microorganisms. Furthermore, most processes in the WWTPs that were researched shared a pattern：the nitS and the bacterial amoA genes were more abundant than the nirK and archaeal amoA genes, respectivelv. DNA microarray analysis revealed that several kinds of nitrification and denitrification genes were detected in both CAS and anaerobic-oxic processes （AO）, whereas limited genes were detected in nitrogen removal processes. Results of this study suggest that CAS maintains a diverse community of nitrogen cycling microorganisms; moreover, the microbial communities in nitrogen removal processes may be specific.

Osmotic pressure of serum and cerebrospinal fluid in patients with suspected neurological conditions

Interstitial fluid movement in the brain parenchyma has been suggested to contribute to sustaining the metabolism in brain parenchyma and maintaining the function of neurons and glial *** pulsatile hydrostatic pressure gradient may be one of the driving forces of this bulk ***,osmotic pressure- related factors have not been studied until *** this prospective observational study,to elucidate the relationship between osmolality (mOsm/kg) in the serum and that in the cerebrospinal fluid (CSF),we simultaneously measured the serum and CSF osmolality of 179 subjects with suspected neurological *** osmolality was 283.6 ± 6.5 mOsm/kg and CSF osmolality was 289.5 ± 6.6 mOsm/*** the specific gravity of serum and CSF is known to be 1.024–1.028 and 1.004–1.007,respectively,the estimated average of osmolarity (mOsm/L) in the serum and CSF covered exactly the same range (i.e.,290.5–291.5 mOsm/L).There was strong correlation between CSF osmolality and serum osmolality,but the difference in osmolality between serum and CSF was not correlated with serum osmolality,serum electrolyte levels,protein levels,or quotient of *** conclusion,CSF osmolarity was suggested to be equal to serum *** is not one of the driving forces of this bulk *** factors such as hydrostatic pressure gradient should be used to explain the mechanism of bulk flow in the brain *** study was approved by the Institutional Review Board of the Tohoku University Hospital (approval *** No.2015-1-257) on July 29,2015.

A new method for analysis of lichen triterpenoids was established using high performance liquid chromatography with the combination of a differential refractive index detector(RID) and a photodiode array detector(PDA).It is proved that this method was convenient to detect and identify aromatic and aliphatic lichen substances;it enabled quantitative analysis of substances having no or less absorption of ultraviolet rays such as *** addition,they can be measured in high accuracy compared with the TLC method.

Safety and effectiveness of propofol sedation during and after outpatient colonoscopy

AIM:To study the safety and effectiveness of propofol sedation for outpatient ***:Propofol was given by bolus injection with an age-adjusted standard protocol consisting of 60 mg for patients < 70 years old,40 mg for patients age 70-89 years,and 20 mg for those ≥ 90 years,and additional injections of 20 mg propofol were given up to a maximum of 200 *** principal parameters were the occurrence of adverse events within 24 h after colonoscopy and overall satisfaction for this *** parameters included successful procedure,respiratory depression,and other ***:Consecutive patients were entered prospectively and all 2101 entered successfully completed outpatient *** mean dose of propofol used was 96.4 mg(range 40-200 mg).Younger patients required higher doses of propofol than older patients(20-40 years vs ≥ 61 years:115.3 ± 32 mg vs 89.7 ± 21 mg,P < 0.001).Transient supplemental oxygen supply was needed by five patients(0.2%);no other complications *** questionnaires were completed by 1820(87%) of 2101 patients and most rated their overall satisfaction as excellent(80%) or good(17%).The majority(65%) of patients drove home or to their office after their ***(99%) were willing to repeat the same *** incidents occurred within 24 h after ***:Propofol sedation using a dose < 200 mg proved both safe and practical for outpatient colonoscopy.

Development of a competitive enzyme-linked immunosorbent assay for therapeutic drug monitoring of afatinib

Afatinib is an oral tyrosine kinase inhibitor(TKI) approved for treating advanced non-small cell lung cancer. It is necessary to develop a simple quantification method for TKIs in order to facilitate therapeutic drug monitoring(TDM) in clinical settings. This study sought to develop a simple and sensitive competitive enzyme-linked immunosorbent assay(ELISA) to quantify afatinib in plasma for routine pharmacokinetic applications. An anti-afatinib antibody was obtained using(S)-N-4-(3-chloro-4-fluorophenyl)-7-(tetrahydrofuran-3-yloxy)-quinazoline-4,6-diamine(CTQD), which has the same substructure as afatinib, as a hapten. Enzyme labeling of afatinib with horseradish peroxidase was similarly performed using CTQD. A simple competitive ELISA for afatinib was developed based on the principle of direct competition between afatinib and the enzyme marker for the anti-afatinib antibody, which had been immobilized on the plastic surface of a microtiter plate. Plasma afatinib concentrations below the limit of quantification of 30 pg/mL were reproducibly measurable. Also, the values of plasma afatinib levels measured from 20 patients were comparable with those measured by high-performance liquid chromatography, and there was a strong correlation between the values determined by both methods(Y=0.976 X – 0.207, r=0.975). As indicated by its specificity and sensitivity, this newly developed ELISA for afatinib is an important tool for TDM and studies of the pharmacokinetics of afatinib.

Asian international grid connection and potentiality of DC superconducting power transmission

In 2011, a large scale earthquake and tsunami hit the northeastern coast of Japan, and nuclear plants were damaged to a large extent. Before the Tsunami, 54 nuclear plants were operated, however presently, only three nuclear plants are in operation. Therefore, Japan is suffering from high price of electricity and low supply reliability. In generation mix, nuclear plants generate only one percent and 87 percent of electricity is generated by fossil energy. This is not preferable for Japan from the viewpoints of energy security and stable electricity supply. Therefore, it is mandatory to increase sustainable energy and to decrease consumption of fossil fuel. International grid connection and the Global Energy Interconnection will be one of countermeasures against these problems existing in Japan. In this paper, international grid connection initiatives proposed in Asia are described and features and issues of electric power grids in Japan are discussed to implement the international grid connection. As the key technology for implementing the international grid connection, the present status of DC superconducting power transmission lines and power converters for long-distance power transmission lines are presented. In conclusion, conditions and legal frameworks for realizing the international grid connection are described.

Adhesion Force Measurements of Polymer Particles by Detachment Field Method

The adhesion force distributions of polymer particles to aluminum substrates were measured by the detachment field method. Polymer particles with conducting surface treatment were used for the measurements. Further the conventional detachment field method was modified to be applicable to the adhesion force measurements of a single particle. The adhesion force of the polymer particles increased with an increase in relative humidity. The surface roughness of the substrate influenced the adhesion forces of particles significantly. The influence of the CF4 plasma treatment of the polymer particles and thin layer coating of the substrate surface on the adhesion forces of the polymer particles was also studied, and factors affecting adhesion forces of polymer particles are discussed.

Expression of the tumor suppressor gene maspin and its significance in intraductal papillary mucinous neoplasms of the pancreas

BACKGROUND: Maspin is a member of the serpin family of protease inhibitors and is thought to inhibit carcinoma invasion, metastasis, and angiogenesis and induce apoptosis. We examined maspin expression immunohistochemically and assessed its significance in intraductal papillary mucinous neoplasm (IPMN) of the pancreas. METHODS: We examined 39 surgically resected specimens of IPMN that included 17 adenomas (IPMAs), 5 borderline tumors (IPMBs), 4 non-invasive carcinomas (non-invasive IPMCs), and 13 invasive carcinomas (invasive IPMCs). Immunostaining was performed according to the EnVision ChemMate method. The degree of maspin expression was scored and assessed according to the percentage and staining intensity of positive cells. RESULTS: Maspin expression was minimal in normal pancreatic duct epithelium, whereas in IPMNs, maspin was expressed in neoplasms of all stages. Maspin expression increased with increasing grade from IPMAs, IPMBs, to non-invasive IPMCs but decreased significantly in invasive IPMCs. No specific association between maspin expression and mucin type was found. Analysis of maspin expression with respect to clinicopathologic factors in cases of invasive IPMC revealed a greater extent of invasion in cases of low maspin expression and significantly fewer apoptotic cells in the ***: Maspin was expressed at high levels in IPMNs at various stages from adenoma to invasive carcinoma, and our results suggest that maspin may be involved in the occurrence and progression of IPMN. In addition, our data suggest that the apoptosis-inducing action of maspin suppresses invasion and progression of IPMN.
