Preserving microbial diversity has become a strategic undertaking. Thus, ex situ microalgal culture conservation results in strategic and functional resource in both biodiversity protection and application domains. Cryopreservation of microalgae has been practiced since the 1960s and is now considered the optimal preservation strategy. Furthermore, the overall monitoring during growth of cultures after freezing/thawing protocols was hardly investigated and there is poor evaluation related to preserve especially the photosystem apparatus. The present study focuses on Stichococcus bacillaris as case study for short-term cryopreservation at −80 °C storage. Various freezing pretreatments using cryoprotective agents, and two thawing methods were compared introducing a novel variable to evaluate viability recovery and assessing growth kinetics of cultures immediately after thawing and after a series batch cultivation. Photosynthetic rate and pigments assessment were proposed to evaluate hidden metabolic cell damage. Results underline cryoprotective agents can increase the kinetic recovery of preserved cells in terms of reduction of lag phase during batch cultivation tests: the use of dimethyl sulfoxide and glycerol granted a growth comparable to unpreserved cells when sudden thawing occurs after 24 hours of storage, but recovery after preservation is less sensitive to cryoprotective agents when gradual thawing and 1 month of storage is considered. However, cells are always able to restore their physiological pathways even without agents, so their kinetic effect has been proved and quantified. Interestingly, both the photosynthetic efficiency and the ratio between total chlorophyll and carotenoids are comparable (0.75 Fv/Fm, 2.2 ± 0.25 g/g) to unpreserved cells and they are unsensitive to chosen agents, but the ratio between chlorophyll a and chlorophyll b was clearly altered (up to 10 times), suggesting that photoactive pigments relative proportions

Genetic association of interleukin-6 polymorphism (-174 G/C) with chronic liver diseases and hepatocellular carcinoma

Interleukin-6 (IL-6) is a pleiotropic cytokine which is expressed in many inflammatory cells in response to different types of stimuli, regulating a number of biological processes. The IL-6 gene is polymorphic in both the 5' and 3' flanking regions and more than 150 single nucleotide polymorphisms have been identified so far. Genetic polymorphisms of IL-6 may affect the outcomes of several diseases, where the presence of high levels of circulating IL-6 have been correlated to the stage and/or the progression of the disease itself. The -174 G/C polymorphism is a frequent polymorphism, that is located in the upstream regulatory region of the IL-6 gene and affects IL-6 production. However, the data in the literature on the genetic association between the -174 G/C polymorphism and some specific liver diseases characterized by different etiologies are still controversial. In particular, most of the studies are quite unanimous in describing a correlation between the presence of the high-producer genotype and a worse evolution of the chronic liver disease. This is valid for patients with hepatitis C virus (HCV)-related chronic hepatitis and liver cirrhosis and hepatocellu-lar carcinoma (HCC) whatever the etiology. Studies in hepatitis B virus-related chronic liver diseases are not conclusive, while specific populations like non alcoholic fatty liver disease/non-alcoholic steatohepatitis, autoimmune and human immunodeficiency virus/HCV coinfected patients show a higher prevalence of the lowproducer genotype, probably due to the complexity of these clinical pictures. In this direction, a systematic revision of these data should shed more light on the role of this polymorphism in chronic liver diseases and HCC.

Employing fuzzy logic in the diagnosis of a clinical case

Fuzzy logic is a logical calculus which operates with many truth values (while classical logic works with the two values of true and false). Since fuzzy logic considers the truth of scientific statements like something softened, it is fruitfully applied to the study of biological phenomena, biology is indeed considered the field of complexity, uncertainty and vagueness. In this paper fuzzy logic is successfully applied to the clinical diagnosis of a patient who suffers from different diseases bound by a complex causal chain. In this work it is presented a mathematical foundation of fuzzy logic (with connectives and inference rules) and then the application of fuzzy reasoning to the study of a clinical case. Probabilistic logic is widely considered the unique logical calculus useful in clinical diagnosis, thus the usefulness of fuzzy logic and its relation with probabilistic logic is here explored. The presentation of the case is supplied with all the features necessary to affect a clinical diagnosis: physical exam, anamnesis and tests.

Prediction of the seismic behavior of an underground railway station and a tunnel in Napoli(Italy)

Purpose We sought to describe and examine the interrelationships between energy intake,body composition,and estimated energy *** Using self-reported hourly food intake and formula-based energy expenditure(EE)protocols,19 female profes-sional cheerleaders(mean age 25.4 years)were assessed to obtain energy balance(EB)for a typical training *** intake(EI)was predicted using the USDA Food Composition Database SR27,and EE was predicted using the Harris-Benedict equation plus a MET-based relative intensity activity *** composition was predicted using a multi-current,8-mode segmental bioelectrical impedance analysis *** and daily EB was calculated from EI and EE *** Subjects reported a 24 h EI significantly below(P<0.001)the unadjusted predicted energy requirement(1482 kcal vs.2199 kcal,respectively),resulting in an average negative net EB of−720 *** intake was significantly below the minimum recommended level(3.1 g/kg vs.6 g/kg,P<0.001)while protein and fat intakes met the recommended *** fat intake(g/kg)was significantly associated with a higher EI kcal/kg(r=0.726;P<0.001),which was sig-nificantly associated(r=−0.55;P=0.01)with a lower body fat percent(BF%).Using the median of BF%(20.9)as the cut point,participants with fewer hours in a negative EB had lower BF%(P=0.043)and those with lower BF%spent more time in an EB of±300 kcal(P=0.013).Conclusions These athletes reported low energy intakes that resulted in large EB deficits and/or more hours in a negative EB,which could be counterproductive for achieving a lean body composition overtime.

clinical course and prognostic factors of hepatorenal syndrome:a retrospective single-center cohort study

AIM: To investigate clinical and biochemical features of hepatorenal syndrome(HRS), to assess short and long- term survival evaluating potential predictors of early mortality. METHODS: Sixty-two patients with liver cirrhosis and renal failure, defined as a serum creatinine value > 1.5 mg/dL on at least two measurements within 48 h, admitted to our tertiary referral Unit from 2001 to 201, were retrospectively reviewed. Among them, 33 patients(53.2%) fulfilled the revised criteria of the International Ascites Club for the diagnosis of HRS. Twenty-eight patients were treated with combinations of terlipressin and albumin, two with dopamine and al- bumin, and three with albumin alone. No patients were suitable for liver transplantation. Complete response was defined as normalization of creatinine levels to less than 1.5 mg/dL, partial response as a decrease of at least 50% but not to less than 1.5 mg/dL, no response as no reduction in creatinine or a decrease of less 50% compared to pre-treatment values. All of the patients were followed up for at least 1 year until January 2013. RESULTS: HRS type 1 was diagnosed in 15 patients(45.5%). Hepatitis C virus infection was the primary etiology(69.6%), followed by alcohol(15.2%), and cryptogenesis(15.2%). Complete response to therapy was obtained in only 3 cases(9.1%) and partial re- sponse in 7 patients(21.2%). Median survival was 30 d(range: 10-274) without significant differences be- tween type 1 and type 2 HRS. By univariate analysis, Child-Pugh class C(P = 0.009), presence of hepatocel- lular carcinoma(P = 0.04), low serum sodium(P = 0.02), high bilirubin values(P = 0.009) and high Model for End-stage Liver Disease(MELD) score(P = 0.03) were predictive factors of 30-d mortality. By multivari- ate analysis, only serum sodium 27(OR = 18.72; P = 0.01) were independently associated with a survival of less than one month. CONCLUSION: HRS still has a poor prognosis, even when va

旅游业与跨文化沟通的密切联系对旅游企业和顾客的交流有重大影响,尤其当服务或者旅游产品的形象不符合游客的价值观与需求时。因此,设计旅游产品的时候,除了考虑市场的因素外,还要重视跨文化背景下顾客的文化特征。关于跨文化和服务的研究比较丰富,因此本论文基于前人的研究成果以歌诗达邮轮公司的针对中国市场和意大利市场推出的旅游宣传册为研究对象通过霍夫斯泰德的文化维度理论进行旅游产品的形象设计进行研究。通过宣传册的内容分析,即供应、内容分类与顺序安排、词语、图像以及页面结构分析,得到的研究结果显示设计旅游宣传册的过程中,最有影响的文化维度为“男性化／女性化”与“回避不确定性”。此文化维度在提供信息的精密度和丰富度以及强调的内容类型上表现明显。研究结果指出在不同市场上,旅游产品的形象设计,即使要重视市场调查与潮流,也要参考目标市场的文化特色做出一定改进。

A Quantum Neurological Model of Perception-Cognition and Awareness in Ambiguous Figures and the Case of the Dalmatian Dog

The present paper has mainly the aim to formulate a quantum neurological model of brain dynamics during perception and cognition of ambiguous figures here including the case of the Dalmatian dog. First of all, we summarize our previous experimental and theoretical studies, evidencing as the basic conceptual foundations of quantum mechanics enter in a decisive manner in explaining the basic functions of brain dynamics at perceptive and cognitive level and at the level of our consciousness. This is done in detail in Sections 2 and 3 where we discuss also the role of the so called quantum interference effect and we include here the results of an our recent experiment confirming the role of quantum theory and of its quantum interference effect also at perceptive-cognitive and awareness levels. In Section 4, following our previous study, we consider the so called Mach-Zehnder interferometer. It is a physical device that is employed in quantum mechanical experiments in order to analyze such quantum interference effect. Soon after the discussion of this device, we proceed with the elaboration of a detailed quantum neurological model in the sense that we consider the brain regions identified in V1;V2/V3/V4 and A20 and we obtain that they may behave just as the beam splitters involved in the Mach-Zehnder quantum mechanical interferometer. In this manner, we formulate a model of V1;V2/V3/V4, A20 brain dynamics as a Mach-Zehnder interferometer and such model delineates a proposed neurological mechanism explaining perception-cognition and awareness in humans under the input of ambiguous figures and in the particular case of the Dalmatian dog.

Changes in human epidermal growth factor receptor 2 status between primary breast/gastric carcinomas and synchronous metastatic lymph nodes: how can we explain them?

Aim:Previous studies demonstrated discordant expression of human epidermal growth-factor receptor 2(HER2)between primary cancer and their recurrence/*** study further evaluated HER2 status between primary gastric and breast invasive carcinomas and paired metastatic disease to lymph ***:This study collected formalin-fi xed paraffi n-embedded representative tissue blocks from 62 gastric and 65 breast primary carcinomas as well as synchronous metastatic lymph nodes(male:female=39:88;age ranged between 44 and 95 years with mean age of 69.32 years)for immunohistochemical staining of HER2 expression(DAKO HercepTest^(TM)kit).If immunohistochemical HER2 score reached to 2+,HER2 amplifi cation was then assessed using fl uorescence in situ hybridization(PharmDx^(TM)kit DAKO).Results:The discordant HER2 pooled rate,regardless either negative or positive conversion,was 9.67%in primary gastric carcinoma and corresponding nodal metastasis,while the changes in HER2 expression were revealed in 4.61%of mammary and lymph node neoplastic samples.A high-level concordance in HER2 expression between primary carcinoma and synchronous metastatic lymph nodes was confi rmed in both types of cancer;the observed event of discordant HER2 status should be ascribed to intra-tumor heterogeneity,mostly appreciable in gastric ***:In any case,the shift from positive to negative HER2 expression suggests that trastuzumab could be the targeted treatment choice whereas the opposite shift should be evaluated by a simultaneous HER2 determination in both primary and metastatic lymph nodes.
