Background： The tools of synthetic biology have enabled researchers to explore multiple scientific phenomena by directly engineering signaling pathways within living cells and artificial protoeells. Here, we explored the potential for engineered living cells themselves to assemble signafing pathways for non-living protocells. This analysis serves as a preliminary investigation into a potential origin of processes that may be utilized by complex living systems. Specifically, we suggest that if living cells can be engineered to direct the assembly of genetic signaling pathways from genetic biomaterials in their environment, then insight can be gained into how naturally occurring living systems might behave similarly. Methods： To this end, we have modeled and simulated a system consisting of engineered cells that control the assembly of DNA monomers on microparticle scaffolds. These DNA monomers encode genetic circuits, and therefore, these microparticles can then be encapsulated with minimal transcription and translation systems to direct protocell phenotype. The modeled system relies on multiple previously established synthetic systems and then links these together to demonstrate system feasibility. Results： In this specific model, engineered cells are induced to synthesize biotin, which competes with biotinylated, circuit-encoding DNA monomers for an avidinized-microparticle scaffold. We demonstrate that multiple synthetic motifs can be controlled in this way and can be tuned by manipulating parameters such as inducer and DNA concentrations. conclusions： We expect that this system will provide insight into the origin of living systems as well as serve as a tool for engineering living cells that assemble complex biomaterials in their environment.

Growth, Optical, Mechanical, Dielectric and Theoretical Studies on Potassium Pentaborate Tetrahydrate (KB_5O_8·4H_2O) Single crystal by Modified Sankaranarayanan-Ramasamy Method

A nonlinear optical single crystal of potassium pentaborate tetrahydrate （KB5O8·4H2O） has been grown from aqueous solution by using unidirectional crystal growth method of Sankaranarayanan-Ramasamy （SR） with a due modification in the growth assembly. Potassium pentaborate crystal of 60 mm length and 10 mm diameter has been grown along （100） plane with a growth rate of 3 mm per day within a period of 20 days. The grown crystal was subjected to single crystal X-ray diffraction analysis to confirm that the crystal belongs to the orthorhombic system. Some fundamental data such as valance electron plasma energy, Penn gap, Fermi energy and electronic polarizability of the grown crystal were calculated. The presence of borate in the grown crystal was confirmed by Fourier transform infrared （FTIR） spectroscopy. The optical transmission property of the grown crystal was analyzed using ultra violet （UV） visible spectral analysis. Surface morphology of the growth plane was observed using scanning electron microscopy （SEM）. The mechanical strength of the crystals was found out using Vickers microhardness test along the growth axis. Frequency dependent dielectric constant of the grown crystal was studied for various temperatures along （100） plane.

Health communication Framing： The contribution of Weiner＇s Attribution Theory

Message framing has been shown to influence the persuasiveness of health communications. In this conceptual paper, the authors suggest that message framing can be extended to include causal attributions, and Weiner＇s Attribution Theory （Weiner＇s AT） is an appropriate theoretical framework for the study of message framing and health communications. Weiner＇s AT proposes a specific motivational sequence that may explain cognitive, emotional, and behavioral responses to message frames. Propositions regarding relationships between message frames, causal attributions, and emotional and behavioral responses are presented to guide future research. The implications of this attribution-based message-framing process for future research are discussed

GABAA Receptor Modulation by compounds Isolated from <i>Salvia triloba</i>L.

Salvia triloba, traditionally known as Greek sage, has been used to enhance memory, as a sedative and to treat headaches. Pharmacological evaluation of purified extracts and isolated compounds of S. triloba were carried out on functional assays using two-electrode voltage clamp methods on recombinant GABA receptors expressed in Xenopus laevis oocytes. Bio-assay guided fractionation led to seven compounds being isolated from S. triloba: ursolic acid, carnosol, oleanolic acid, salvigenin, rosmanol, cirsimaritin and hispidulin. The purified extracts of S. triloba inhibited 54% of the current produced by 300 μM GABA at α1β2γ2L GABAA receptors. Ursolic acid, carnosol, oleanolic acid and rosmanol also acted as negative allosteric modulators. The flavonoids salvigenin, cirsimaritin and hispidulin acted as positive modulators when applied in the presence of low concentrations of GABA but in the presence of high concentrations of GABA acted as negative modulators, demonstrating a biphasic action. These results are consistent with the concept that Salvia triloba may have cognition enhancing properties. In most cases these activities are likely to be occurring via different modulatory sites on GABAA receptor complexes. It may be that the combination of these activities permits cognition enhancement whilst offering protection from convulsant activity.

Parental Awareness and Perception of Their children’s Body Size

Objective: To investigate parents’ misperception of their active children’s size. Subjects: One hundred and forty male or female parents and their children from a youth soccer league participated. Actual child Body Mass Index (BMI) percentiles and BMI categories were compared to the results of a Body Size Estimation Task and lifestyles questionnaire results. Results: Parents underestimated the body size of their children and placed them in a lower BMI category than the children actually belonged in. As was the case in other studies children in the unhealthy or at risk to become overweight category where seen as a healthy weight. Unlike other studies, many healthy weight children were seen as underweight. conclusions: An explanation of body size misperception and underestimation of body size may be “change blindness”. The growing prevalence of obesity in children may be better addressed by focusing on the parents’ apparent lack of concern about excess childhood weight and the parents’ identification of excess childhood weight as “just normal” rather than seeing excess weight as a potential problem.

Fishways at hydroelectric dams are unnatural environments that typically present fish with channels of uniform depth,variable water velocity,and lack of *** retention and overall passage success may be improved by providing rest and sheltering areas inside *** examined adult Pacific lamprey Entosphenus tridentatus use of two specially designed fishway refuges at Bonneville Dam on the columbia River(northwestern USA).These relatively small boxes(1.1 m×0.4 m×0.2 m)provided low-velocity,dark refuge from predation for Pacific lamprey,a largely nocturnal *** boxes were equipped with antennas to detect entry of lamprey tagged with passive integrated transponder(PIT)*** PIT tagged and released 3,247 lamprey,including 599 that were double-tagged with a PIT and a radio transmitter,over three study years(2012-2014).In each year,PIT-tagged lamprey detected at nearby fishway exits had used a refuge:12%in 2012,28%in 2013,and 36%in *** residence time of PIT-tagged lamprey in the refuges for each year was 20.3 h,16.6 h,and 13.1 *** entrance into refuges peaked at 0300-0500 PDT,and they typically exited at around 2000 PDT,suggesting that refuges primarily functioned as shelter from *** of refuge use increased with a morphological indicator of sexual maturity(distance between dorsal fins).In the radio-tagged group,refuge users were equally likely to pass Bonneville Dam as ***,refuge users were less likely than non-users to be detected at sites upstream from the dam,perhaps owing to their maturation status or longer mean passage time through our study area(2.0 vs.0.6 d for non-users).Refuges show promise for improving fish retention in fishways,particularly for nocturnal and/or small-bodied species that seek shelter from light or predation.

Objective:To evaluate antioxidant,cytotoxic,and anti-venom capacity of crude bark extracts of Alstonia parvifolia ***:Gas chromatography-mass spectrometry(Gc-MS)and energy dispersive X-ray analyses were accomplished to characterize the chemical constituents of Alstonia *** characterization was evaluated using an inhibitory phospholipase A_(2)(PLA_(2))assay,DPPH,and cytotoxicity *** the constituents listed in the Gc-MS analyses,molecular docking was conducted to inspect the binding energies between the chosen compounds and selected PLA_(2) ***:Gc-MS analyses showed that the Alstonia parvifolia crude extract consisted predominantly of acetylmarinobufogenin(14.89%),γ-sitosterol(10.44%),3-O-methyl-D-glucose(5.88%),3,5-dimethoxy-4-hydroxyphenylacetic acid(5.30%),(2α,5α)-17-methoxyaspidofractinin-3-one(AFM)(4.08%),and 2,3,5,6,7,8,9-heptahydro-1-phenyl-5-(p-chlorophenylimino)-1H-benzo[e][1,4]thiazepine(HPT)(1.37%).The principal elemental components of Alstonia parvifolia were ca(4.012%)and K(1.496%),as exhibited by energy dispersive X-ray *** parvifolia showed significant free radical scavenging ability(Ic_(50):0.287 mg/mL)and was non-cytotoxic to normal HDFn cells(Ic_(50)>100μg/mL).Moreover,Alstonia parvifolia was favorably cytotoxic to McF-7(Ic_(50):4.42µg/mL),followed by H69PR,HT-29,and THP-1,with Ic_(50) values of 4.94,5.07,and 6.27µg/mL,*** parvifolia also displayed notable inhibition against PLA_(2) activity of Naja philippinensis Taylor venom with Ic_(50) of(15.2±1.8)μg/*** and cluster analyses projected negative binding energies from AFM(−6.36 to−9.68 kcal/mol),HPT(−7.38 to−9.77 kcal/mol),and acetylmarinobufogenin(−7.22 to−9.59 kcal/mol).These calculations were for the particular interactions of Alstonia parvifolia constituents to PLA_(2) homologues where the utmost affinity was detected in HPT owing to the dipole interactions with amino acid ***:The bark e
