Our previous studies have reported that activation of the NLRP3(NOD-,LRR-and pyrin domain-containing protein 3)-inflammasome complex in ethanol-treated astrocytes and chronic alcohol-fed mice could be associated with neuroinflammation and brain *** stem cell-derived extracellular vesicles(MSC-EVs)have been shown to restore the neuroinflammatory response,along with myelin and synaptic structural alterations in the prefrontal cortex,and alleviate cognitive and memory dysfunctions induced by binge-like ethanol treatment in adolescent *** the therapeutic role of the molecules contained in mesenchymal stem cell-derived extracellular vesicles,the present study analyzed whether the administration of mesenchymal stem cell-derived extracellular vesicles isolated from adipose tissue,which inhibited the activation of the NLRP3 inflammasome,was capable of reducing hippocampal neuroinflammation in adolescent mice treated with binge *** demonstrated that the administration of mesenchymal stem cell-derived extracellular vesicles ameliorated the activation of the hippocampal NLRP3 inflammasome complex and other NLRs inflammasomes(e.g.,pyrin domain-containing 1,caspase recruitment domain-containing 4,and absent in melanoma 2,as well as the alterations in inflammatory genes(interleukin-1β,interleukin-18,inducible nitric oxide synthase,nuclear factor-kappa B,monocyte chemoattractant protein-1,and C–X3–C motif chemokine ligand 1)and miRNAs(miR-21a-5p,miR-146a-5p,and miR-141-5p)induced by binge-like ethanol treatment in adolescent *** analysis further revealed the involvement of miR-21a-5p and miR-146a-5p with inflammatory target genes and NOD-like receptor signaling *** together,these findings provide novel evidence of the therapeutic potential of MSC-derived EVs to ameliorate the hippocampal neuroinflammatory response associated with NLRP3 inflammasome activation induced by binge drinking in adolescence.

Many historic buildings in old urban centers in Eastern Canada are made of stone masonry reputed to be highly vulnerable to seismic *** risk assessment of stone masonry buildings is therefore the first step in the risk mitigation process to provide adequate planning for retrofit and preservation of historical urban *** paper focuses on development of analytical displacement-based fragility curves reflecting the characteristics of existing stone masonry buildings in Eastern *** old historic center of Quebec City has been selected as a typical study *** standard fragility analysis combines the inelastic spectral displacement,a structure-dependent earthquake intensity measure,and the building damage state correlated to the induced building *** proposed procedure consists of a three-step development process：（1） mechanics-based capacity model,（2） displacement-based damage model and（3） seismic demand *** damage estimation for a uniform hazard scenario of 2% in 50 years probability of exceedance indicates that slight to moderate damage is the most probable damage experienced by these stone masonry *** is also made with fragility curves implicit in the seismic risk assessment tools Hazus and *** shows the highest probability of the occurrence of no to slight damage,whereas the highest probability of extensive and complete damage is predicted with *** comparison shows the importance of the development of fragility curves specific to the generic construction characteristics in the study area and emphasizes the need for critical use of regional risk assessment tools and generated results.

Drainage of central nervous system(CNS) antigens to the brain-draining cervical lymph nodes(CLN) is likely crucial in the initiation and control of autoimmune responses during multiple sclerosis(MS). We demonstrate neuronal antigens within CLN of MS patients. In monkeys and mice with experimental autoimmune encephalomyelitis(EAE) and in mouse models with non-inflammatory CNS damage, the type and extent of CNS damage was associated with the frequencies of CNS antigens within the cervical lymph nodes. In addition, CNS antigens drained to the spinal-cord-draining lumbar lymph nodes. In human MS CLN, neuronal antigens were present in pro-inflammatory antigen-presenting cells(APC), whereas the majority of myelincontaining cells were anti-inflammatory. This may reflect a different origin of the cells or different drainage mechanisms. Indeed, neuronal antigen-containing cells in human CLN did not express the lymph node homing receptor CCR7, whereas myelin antigen-containing cells in situ and in vitro did. Nevertheless, CLN from EAE-affected CCR7-deficient mice contained equal amounts of myelin and neuronal antigens as wild-type mice. We conclude that the type and frequencies of CNS antigens within the CLN are determined by the type and extent of CNS damage. Furthermore, the presence of myelin and neuronal antigens in functionally distinct APC populations within MS CLN suggests that differential immune responses can be evoked.

A case study of the role of climate, humans, and ecological setting in Holocene fire history of northwestern Europe

We present the major results from studies of fire history over the last 11000 years(Holocene) in southern Sweden, on the basis of palaeoecological analyses of peat sequences from three small peat bogs. The main objective is to emphasize the value of multiple, continuous sedimentary records of macroscopic charcoal(macro-C) for the reconstruction of local to regional past changes in fire regimes, the importance of multi-proxy studies, and the advantage of model-based estimates of plant cover from pollen data to assess the role of tree composition and human impact in fire history. The chronologies at the three study sites are based on a large number of 14 C dates from terrestrial plant remains and age-depth models are achieved using Bayesian statistics. Fire history is inferred from continuous records of macro-C and microscopic charcoal counts on pollen slides. The Landscape Reconstruction Algorithm(LRA) for pollen-based quantitative reconstruction of local vegetation cover is applied on the three pollen records for plant cover reconstruction over the entire Holocene. The results are as follows:(1) the long-term trends in fire regimes are similar between sites, i.e., frequent fires during the early Holocene until ca. 9 ka BP, low fire frequency during the mid-Holocene, and higher fire frequency from ca. 2.5 ka BP;(2) this broad trend agrees with the overall fire history of northwestern and western Europe north of the Mediterranean area, and is due to climate forcing in the early and mid-Holocene, and to anthropogenic land-use in the late Holocene;(3) the LRA estimates of plant cover at the three sites demonstrate that the relative abundance of pine played a primordial role in the early and mid-Holocene fire history; and(4) the between-site differences in the charcoal records and inferred fire history are due to local factors(i.e., relative abundance of pine, geomorphological setting, and anthropogenic land-use) and taphonomy of charcoal deposition in the small peat bog

Beside its poor prognosis and its late diagnosis, pancreatic cancer remains one of the most painful malignancies. Optimal management of pain in this cancer represents a real challenge for the oncologist whose objective is to ensure a better quality of life to his patients. We aimed in this paper to review all the treatment modalities incriminated in the management of pain in pancreatic cancer going from painkillers, chemotherapy, radiation therapy and interventional techniques to agents under investigation and alternative medicine. Although specific guidelines and recommendations for pain management in pancreatic cancer are still absent, we present all the possible pain treatments, with a progression from medical multimodal treatment to radiotherapy and chemotherapy then interventional techniques in case of resistance. In addition, alternative methods such as acupuncture and hypnosis can be added at any stage and seems to contribute to pain relief.

Liquid chromatography （LC） is considered by many as a mature technique. Nonetheless, LC technology continues to evolve driven by the need for high-throughput and high-resolution analyses. Over the past several years, small particle size packing materials have been introduced by several column manufacturers to enable fast and efficient LC separations. Several examples of pharmaceutical analyses, including impurity profiling of taxanes and atracurium besylate, in-process testing of peptides in injectable dosage form, using sub-2 um column technology are presented in this paper, demonstrating some of the capabilities and limitations of the technology.

The objective of this paper is to present a new method for designing absorbing or non-reflective boundary conditions (ABC) or (NRBC), illustrated by the case study of the modelling of a solid body in water, specifically the capillary gravity waves generated by its motion at the surface. The study analyses the flow of an inviscid, barotropic, and compressible fluid around the stationary solid body. The dynamic behaviour of the fluid is analysed using a two-dimensional coupled Neumann-Kelvin model extended with capillarity and inertia terms. For computational purposes, it is necessary to truncate the unbounded spatial domain with artificial boundaries and then introduce appropriate absorbing boundary conditions. The propagation of short wavelength waves in a convective fluid medium with significant differences in properties between the interior and the surface of the fluid presents a number of difficulties in the design of these conditions. The results are illustrated numerically and commented upon.

Olfactory ensheathing glia promote axonal regeneration in the mammalian central nervous system,including retinal ganglion cell axonal growth through the injured optic ***,it is unknown whether olfactory ensheathing glia also have neuroprotective *** ensheathing glia express brain-derived neurotrophic factor,one of the best neuroprotectants for axotomized retinal ganglion ***,we aimed to investigate the neuroprotective capacity of olfactory ensheating glia after optic nerve *** ensheathing glia cells from an established rat immortalized clonal cell line,TEG3,were intravitreally injected in intact and axotomized retinas in syngeneic and allogeneic mode with or without microglial inhibition or immunosuppressive *** and gene expression analyses were *** bulb-derived primary olfactory ensheathing glia and TEG3 express major histocompatibility complex classⅡ*** and syngenically transplanted TEG3 cells survived in the vitreous for up to 21 days,forming an *** axotomized retinas,only the allogeneic TEG3 transplant rescued retinal ganglion cells at 7 days but not at 21 *** these retinas,microglial anatomical activation was higher than after optic nerve crush *** intact retinas,both transplants activated microglial cells and caused retinal ganglion cell death at 21 days,a loss that was higher after allotransplantation,triggered by pyroptosis and partially rescued by microglial inhibition or ***,neuroprotection of axotomized retinal ganglion cells did not improve with these *** different neuroprotective properties,different toxic effects,and different responses to microglial inhibitory treatments of olfactory ensheathing glia in the retina depending on the type of transplant highlight the importance of thorough preclinical studies to explore these variables.
