Effects of natural mineral-rich water consumption on the expression of sirtuin 1 and angiogenic factors in the erectile tissue of rats with fructose-induced metabolic syndrome

Consuming a high-fructose diet induces metabolic syndrome （MS）-Iike features, including endothelial dysfunction. Erectile dysfunction is an early manifestation of endothelial dysfunction and systemic vascular disease. Because mineral deficiency intensifies the deleterious effects of fructose consumption and mineral ingestion is protective against MS, we aimed to characterize the effects of 8weeks of natural mineral-rich water consumption on the structural organization and expression of vascular growth factors and receptors on the corpus cavernosum （CC） in 10% fructose-fed Sprague-Dawley rats （FRUCT）. Differences were not observed in the organization of the CC either on the expression of vascular endothelial growth factor （VEGF） or the components of the angiopoietins/Tie2 system. However, opposing expression patterns were observed for VEGF receptors （an increase and a decrease for VEGFR1 and VEGFR2, respectively） in FRUCT animals, with these patterns being strengthened by mineral-rich water ingestion. Mineral-rich water ingestion （FRUCTMIN） increased the proportion of smooth muscle cells compared with FRUCT rats and induced an upregulatory tendency of sirtuin I expression compared with the control and FRUCT groups. Western blot results were consistent with the dual immunofluorescence evaluation. Plasma oxidized low-density lipoprotein and plasma testosterone levels were similar among the experimental groups, although a tendency for an increase in the former was observed in the FRUCTMIN group. The mineral-rich water-treated rats presented changes similar to those observed in rats treated with MS-protective polyphenol-rich beverages or subjected to energy restriction, which led us to hypothesize that the effects of mineral-rich water consumption may be more vast than those directly observed in this study.

Aims The Inclusive Niche Hypothesis has not been validated for plants using ecophysiological *** few experiments have measured growth and competition but not the physiological response of plants.A metallophyte plant that hyperaccumulates aluminium(Al),Plantago almogravensis,showed a defined spa-tial distribution by occurring mostly on vegetation gaps associ-ated with metalliferous areas(geochemical islands).This case was used to determine,in situ,whether the Inclusive Niche Hypothesis was suitable to explain the extent of the species real-ized *** The vegetation associated with *** geochemical islands in the SW coast of Portugal was *** biotic(neighbouring plants)and abiotic(edaphic)components of the niche were cor-related with parameters of the plant’s ecological and physiological performances(plant density and cover;leaf C and N concentration and isotopic composition;growth).The results were obtained using image analysis,abundance and morphological measures,isotopic signatures and chemical *** Findings The species showed better physiological performance where its ecological performance was lower due to trade-offs with environ-mental *** species’realized niche was mostly limited by shrub competition and soil *** limits contribute to explain the rarity status of the species:the species has a poor capacity to compete but,due to an enhanced Al-tolerance and Al-hyperaccumulator trait,has the ability to find refuge in geochem-ical islands that are too harsh for most other *** work,based on ecophysiological field studies,provides support for the Inclusive Niche Hypothesis relating to plant species.

Characterization and Biological Activities of Protein-Bound Polysaccharides Produced by Cultures of Pleurotus ostreatus

Several species of mushrooms, as Pleurotus ostreatus, have been valued as edible and medicinal resources. These mushrooms may be an important source of polysaccharides with medicinal properties as antioxidant, antitumoral, antimicrobial and immunological properties. The aim of this work was to produce and to evaluate the biological properties of protein-bound polysaccharide complexes, extra intracellular （E-PPS and I-PPS）, extracted from P. ostreatus cultures, using agricultural sunflower wastes as carbon source. Three main compounds in the E-PPS and four main compounds in the I-PPS were identified by SEC-UV-RI-HPLC. These complexes of P. ostreatus present no toxicity in Artemia salina cultures, after 24 h of incubation. Antioxidant properties of the complexes were evaluated by radical scavenging activity using DPPH method and lipid peroxidation inhibition capacity, determined by erytbsocytes hemolysis. Additionally, E-PPS and I-PPS extracts revealed capacity to mimetize superoxide dismutase and catalase enzymatic activities. The hepatoprotector effect of E-PPS extracts in Wistar rats was evaluated by AST, ALT, ALP and y-GT activities, showing capacity to reduce the liver damage induced by ethanol-administration. This hepatoprotective effect is equivalent to that observed by silymarin, a standard drug. Our results suggests that the extracts of E-PPS and I-PPS produced by P. ostreatus cultures, using agricultural sunflower wastes as main carbon source, can be used as an important source of bioactive compounds with potential medicinal value.

AIM: To study the benef icial effects of triterpene α,β-amyrin and the underlying mechanisms in an experimental pancreatitis model. METHODS: Acute pancreatitis was induced in five groups of rats (n = 8) by L-arginine (2 × 2.5 g/kg, intraperitoneal, 1 h apart) and 1 h later, they received a single oral dose of α,β-amyrin (10, 30 and 100 mg/kg),methylprednisolone (30 mg/kg) and vehicle (3% Tween 80). A saline (0.9% NaCl) treated group served as a normal control. Efficacy was assessed at 24 h by determination of serum levels of amylase, lipase and proinflammatory cytokines [tumor necrosis factor (TNF)-α and interleukin (IL)-6], pancreatic myeloperoxidase (MPO) activity, lipid peroxidation [thiobarbituric acid reactive substances (TBARS)], nitrate/nitrite levels, and the wet weight/body weight ratio. Tissue histology and the immunoreactivity for TNF-α and inducible nitric oxide synthetase (iNOS) were performed. RESULTS: α,β-amyrin and methylprednisolone treatments significantly (P < 0.05) attenuated the L-arginine-induced increases in pancreatic wet weight/body weight ratio, and decreased the serum levels of amylase and lipase, and TNF-α and IL-6, as compared to the vehicle control. Also, pancreatic levels of MPO activity, TBARS, and nitrate/nitrite were signifi cantly lower. Histological f indings and TNF-α and iNOS immunostaining further confirmed the amelioration of pancreatic injury by α,β-amyrin. CONCLUSION: α,β-amyrin has the potential to combat acute pancreatitis by acting as an anti-in? ammatory and antioxidant agent.

Pilomatricoma or calcifying epithelioma of Malherbe is a rare benign tumor of the skin and/or the subcutaneous tissue originating from the hair matrix. It is usually seen in children and adolescents, however a smaller second peak of onset is seen in the elderly. The head, neck, or upper extremities are the most common anatomical sites of pilomatricomas, being less frequently seen on the trunk and lower extremities, and very rarely have they been diagnosed in the breast. Its malignant counterpart, pilomatrix carcinoma, is rare and found more often in men. Due to its incapacity of regression, treatment consists of surgical excision. We present the case of a man with a pilomatrixoma of the breast, presenting as ACR BI-RADS 4, and we review the literature regarding pilomatrixomas.

Cyanobacterium Spirulina platensis is commercially produced as a nutrient source for food,animal feed and pharmaceutical industries,and is also explored in other applications in areas such as material sciences,materials engineering and for the production of biofuels and *** to the increasing interest in the use of this microalga,a complete characterization was intended,as to provide data to the insufficient *** this work,various techniques were used for thermal(thermogravimetric analysis(TGA)/derivative thermogravimetry(DTG),differential scanning calorimetry(DSC)),structural(scanning electron microscopy(SEM)and wide-angle X-ray diffraction(XRD))and chemical(atomic absorption spectroscopy(AAS),attenuated total reflection Fourier transform infrared spectroscopy(ATR-FTIR),energy dispersive X-ray spectroscopy(EDX))characterization of Spirulina cultivated in *** have shown that in addition to the high quantity of protein(over 50%),Brazilian Spirulina is a source for carbohydrates(33%)and also has good thermal stability up to 200°*** pigment protein Phycocyanin could be identified by FTIR and ultraviolet-visible(UV-vis)*** show favorable properties of Spirulina as a source for new materials and biomass.
