Amyotrophic lateral sclerosis (ALS) is a fatal motor neuron disease typically leading to death within 5 years from symptom onset. ALS familial forms are associated with mutations in several genes, including Superoxide Dismutase 1 (SOD1) and Fused in Sarcoma (FUS). Although genes linked to ALS participate in disparate biological processes, ALS genetic variants largely trigger shared pathogenic events such as oxidative stress, protein aggregation and defects in RNA processing. Moreover, ALS patients show systemic hypermetabolism that leads to increased energy expenditure at rest and thus weight loss during the disease course.1 ALS hypermetabolic phenotype and weight loss have been extensively characterized in mice bearing the G93A substitution in SOD1 protein (SOD1-G93A), which exhibit skeletal-muscle metabolic reprogramming before disease onset.

Dietary phenolic compounds （PCs） have been receiving intercst lor their presumcd roles ill disease prevention. However, there is a lack of studies on the underlying molecular mechanisnls. In this regard, in vitro metabolomic approaches are suitable for the investigation o1 the molecular changes in response to PC exposure. Up to date, the biological effects of PCs have only been cxalnined for PCs from rosemary （Rosmarinus officinalis）, olive oil, and reSVCl-atrol using cell-based mctabolomic approach, although transcriptomic and/or proteomic studies have also been conducted in the same in vi[ro cell experiment in some cases. Our integral analysis of the reviewed studies suggest that PCs may be involved not only in basic cellular processes or macro-and micro-nutrient meta- bolism, but also in specific metabolic pathways that have been thoroughly investigated. These modulated pathways could have a clinical impact on neurodegenerativc diseases, type 2 diabetes, cancer, and cardiovascular diseases. In conclusion, the in vitro metabolomic approaches provide additional information of the molecular mechanisms involved in disease risk reduction of dietary PCs. In order to elucidate the mechanisms of action of PCs, more metabolomic cell-based studies are nccdcd and testing the physiological conjugated forms of PCs in these cell systems could be of special interest.

Fatigue is considered one of the most frequent and debilitating symptoms in primary biliary cholangitis(PBC),affecting over 50%of PBC *** in five patients with PBC suffer from severe fatigue,which significantly impairs quality of *** is made up of a central and a peripheral component,whose pathophysiology is still greatly *** fatigue is characterised by a lack of self-motivation and can manifest both in physical and mental activities(lack of intention).Peripheral fatigue includes neuromuscular dysfunction and muscle weakness(lack of ability).Peripheral fatigue could be explained by an excessive deviation from aerobic to anaerobic metabolism leading to excessive lactic acid accumulation and therefore accelerated decline in muscle function and prolonged recovery *** opposed to itching,and with the exception of endstage liver disease,fatigue is not related to disease *** objective of this review is to outline current understanding regarding the pathophysiology of fatigue,the role of comorbidities and contributing factors,the main tools for fatigue assessment,the failed therapeutic options,and future treatment perspectives for this disabling *** fatigue is an extremely common and debilitating symptom and there is still no licensed therapy for fatigue in PBC patients,further research is warranted to understand its causative mechanisms and to find an effective treatment.

The Sergipano Orogenic System(SOS)in southern Borborema Province(NE Brazil)hosts voluminous Neoproterozoic plutonism related to the Brasiliano/Pan-African *** paper presents comprehensive whole-rock geochemical data,titanite U-Pb ages,and the first combined zircon U-Pb and Lu-Hf isotope results for plutonic rocks from the MacururéDomain to constrain their sources,petrogenesis,and tectonic *** magmatic episodes are recognized and record the evolutionary stages of the orogen.(i)Early-collisional magmatism(643–628 Ma)comprises gabbros and diorites with minor tonalites characterized by well-developed tectonic foliation and evidence of solid-state *** rocks are magnesian,high-K calc-alkaline,LILE-and LREE-enriched and provide subchondriticε_(Hf)(t)values(6.5 to4.7)and Orosirian Hf-T^(C)_(DM)model ages(1.83–1.94 Ga).Such features indicate derivation from a lithospheric mantle source metasomatized by incorporating crustal components through subduction processes prior to magma generation,possibly related to the Rhyacian Orogeny(2.20–1.96 Ga).Extensive mixing/mingling between basaltic and crust-derived magmas took place at lower crustal depths,producing coeval hybrid diorites and quartz-diorites.(ii)Syn-collisional magmatism(630–624 Ma)encompasses biotite-and muscovite-bearing granodiorites and monzogranites,preserving their structures parallel to the schistosity of the country *** rocks are leucocratic,weakly metaluminous to peraluminous,and contain abundant surmicaceous *** features and geochemical composition suggest an origin related to the partial melting of graywacke protoliths with a subordinate igneous component.(iii)Late-collisional magmatism(625–600 Ma)includes undeformed and isotropic monzonites and granodiorites,which truncate the regional *** rocks are consistently metaluminous and magnesian,showing affinities with the high-K calc-alkaline to shoshonite *** element modeling with su

Objective： To examine the effect of metanol extract of Petiveria alliacea （PM） on airway inflammation in a murine model of chronic asthma. Methods： Two-month-old male BALB/c mice （n=6-8/group） were sensitized on days 0 and 14 by intraperitoneal injection of 20μg ovalbumin （OVA）. On day 25, the mice received an airway challenge with OVA （3%, w/v, in phosphate buffered saline）. PM was administered orally by oral gavage to mice at doses of 100, 200 and 400 mg/kg body weight once daily from days 18 to 23. Control mice were orally administered phosphate buffered saline （PBS） to induce a model of asthma. At the end of the test, respiratory reactivity was assayed, the total cell number, interleukin-4 （IL-4）, IL-5, IL-13, tumor necrosis factor-alpha （TNF-α） and reactive oxygen species （ROS） in the bronchoalveolar lavage fluid （BALF） were determined and the levels of serum IgE, intercellular cell adhesion molecule 1 （ICAM-1） and eotoxin were measured. In addition, lung tissue was used to qualify the IL-4, IL-5, IL-13, TNF-α and transforming growth factor beta 1 （TGF-β1）. Histologic examination was performed to observe inflammatory cellular infiltration. Results： The administration of PM in comparison with the OVA-only treated group significantly attenuated the infiltration of eosinophUs and other inflammatory cells （P〈0.01）. Airway resistance （RI） in the OVA-only induced group was significantly higher than that of the PBS control group （P〈0.01） when methacholine was added. TNF-α, IgE, TGF-β1 and cytokine levels IL-4, IL-5, IL-β in the BALF decreased compared to control mice （P〈0.01 or P〈0.05）. PM treatment also inhibited the production of chemokines, eotaxin and ICAM-1 in BALF （P〈0.01）, which improved lung function. Histopathological examination revealed that the sensitized treated PM groups had significant lower in inflammatory scores similar to dexamethasone treatments and the untreated group. Phenology allows organisms to overcome seasonally variable conditions through life-cycle adjustment. Changes in phenology can drastically modify the evolutionary tra- jectory of a population, while a shift in the reproductive time may cause allochronic differentiation. The hypothesis of heritable reproductive time was experimentally tested, by studying a unique population of the pine processionary moth Thaumetopoeapityocampa （Den. ＆ Schiff.） which has a shifted phenology, and however co-occurs with the typical population following the classical life cycle. When populations of both types were reared under controlled conditions, the reproductive time was maintained asynchronous, as ob- served in the field. The shifted population was manipulated in the laboratory to reproduce later than usual, yet the offspring emerged in the next year at the expected dates thus ＂com- ing back＂ to the usual cycle. Hybrids from crosses performed between the 2 populations showed an intermediate phenology. From the emergence times of parents and offspring, a high heritability of the reproductive time （h = 0.76） was observed. The offspring ob- tained from each type of cross was genetically characterized using microsatellite markers. Bayesian clustering analysis confirmed that hybrids can he successfully identified and separated from the parental genetic classes by genotyping. Findings support the hypothesis that, for this particular population, incipient allochronic speciation is due to a heritable shift in the reproductive time that further causes assortative mating and might eventually cause ecological adaptation/maladaptation in response to environmental changes.

The diet composition of the Lycian salamander Lyciasalamandra luschani basoglui across both age and sex groups was studied. Specimens were collected from a small island in the Southeast Aegean Sea. The dominant prey group of juveniles consisted of Collembola, while Coleoptera dominated the diet of males and females. The number and size of prey items consumed by males and females of L. luschani basoglui were quite similar, while juveniles ate fewer items of much smaller size. The numerical abundance along with the four measures of the size of prey permitted discrimination among males, females and juveniles. Although discrimination between adults and juveniles is expected due to dissimilarity in body size, discrimination between males and females remains inexplicable despite their similarity in body size. Future studies should be focused on understanding how and why prey choice differs between sexes in L. luschani basoglui.
