In nature,plants acquire nutrients from soils to sustain growth,and at the same time,they need to avoid the uptake of toxic compounds and/or possess tolerance systems to cope with them.This is particularly challenging when the toxic compound and the nutrient are chemically similar,as in the case of phosphate and arsenate.In this study,we demonstrated that regulatory elements of the phosphate starvation response(PSR)coordinate the arsenate detoxification machinery in the cell.We showed that arsenate repression of the phosphate transporter PHT1;1 is associated with the degradation of the PSR master regulator PHR1.Once arsenic is sequestered into the vacuole,PHR1 stability is restored and PHT1;1 expression is recovered.Furthermore,we identified an arsenite responsive SKP1-like protein and a PHR1 interactor F-box(PHIF1)as constituents of the SCF complex responsible for PHR1 degradation.We found that arsenite,the form to which arsenate is reduced for compartmentalization in vacuoles,represses PHT1;1 expression,providing a highly selective signal versus phosphate to control PHT1;1 expression in response to arsenate.Collectively,our results provide molecular insights into a sensing mechanism that regulates arsenate/phosphate uptake depending on the plant’s detoxification capacity.

Dear Editor,Hereditary prostate cancer(PC)comprises 5%-10%of all PC cases.The increased risk of PC in menwith a family his-tory of the disease is well known and is commonly caused by germline mutations,leading to clinical guidelines men-tioning various genes for identifying high-risk individuals.However,the complex inheritance patterns involvingmul-tiple single nucleotide polymorphisms(SNPs)make it a genetically heterogeneous disease,with genetic testing still in its early stages.

Importance of complementary data to histopathological image analysis of oral leukoplakia and carcinoma using deep neural networks

Background Oral cancer is one of the most common types of cancer in men causing mortality if not diagnosedearly.In recent years,computer-aided diagnosis(CAD)using artificial intelligence techniques,in particular,deepneural networks have been investigated and several approaches have been proposed to deal with the automateddetection of various pathologies using digital images.Recent studies indicate that the fusion of images with thepatient’s clinical information is important for the final clinical diagnosis.As such dataset does not yet exist fororal cancer,as far as the authors are aware,a new dataset was collected consisting of histopathological images,demographic and clinical data.This study evaluated the importance of complementary data to histopathologicalimage analysis of oral leukoplakia and carcinoma for CAD.Methods A new dataset(NDB-UFES)was collected from 2011 to 2021 consisting of histopathological imagesand information.The 237 samples were curated and analyzed by oral pathologists generating the gold standardfor classification.State-of-the-art image fusion architectures and complementary data(Concatenation,MutualAttention,MetaBlock and MetaNet)using the latest deep learning backbones were investigated for 4 distincttasks to identify oral squamous cell carcinoma,leukoplakia with dysplasia and leukoplakia without dysplasia.We evaluate them using balanced accuracy,precision,recall and area under the ROC curve metrics.Results Experimental results indicate that the best models present balanced accuracy of 83.24%using images,demographic and clinical information with MetaBlock fusion and ResNetV2 backbone.It represents an improvement in performance of 30.68%(19.54 pp)in the task to differentiate samples diagnosed with oral squamous cellcarcinoma and leukoplakia with or without dysplasia.Conclusion This study indicates that cured demographic and clinical data may positively influence the performance of artificial intelligence models in automated classification of oral cancer.

Breeding effects on the genotype×environment interaction for yield of durum wheat grown after the Green Revolution:The case of Spain

This study evaluates the changes caused by breeding in the genotype by environment(G×E)interaction of the durum wheat varieties most widely cultivated in Spain after the Green Revolution.A set of 12 cultivars was tested in 27 environments,which are understood as the combination of different sites,years,and treatments(water regime and planting dates),representative of the durum wheat growing conditions in Spain with average grain yields(GY)ranging between 2.8 and 9.1 Mg ha^-1.The most important environmental factors affecting the G×E interaction for yield were themaximum and the mean temperature during the entire crop cycle.An improvement in genetic yield was observed in warm environments and under optimal water conditions that resemble those where the germplasm originated(essentially as advanced lines)before its release in Spain.Therefore,the adaptation of semi-dwarf durum in Spain has shown a tendency to specific adaptation rather than large-scale adaptation.Two different patterns of selection have been reported due the G×E interaction and changes in the ranking of genotypes:in the high yielding environments(GY>5 Mg ha^-1),plants favor increased water uptake,with higher levels of transpiration and more open stomata(more negative values of carbon isotope composition,δ^13 C,and higher canopy temperature depression,CTD),whereas,in low yielding environments(GY<5 Mg ha^-1)plants close their stomata and favor greater water use efficiency(less negativeδ^13 C values and lower CTD values).

Propranolol hydrochloride can be considered a persistent and bioaccumulative pharmaceutical in the environment.This drug and its by-products are potentially toxic and have adverse effects,since these compounds have been associated with endocrine-disrupting effects,reproductive deficiencies,embryo abnormalities and pericardial oedema.TiO_(2)–La 0.05%–carboxymethyl-β-cyclodextrin(CMCD)nanoparticles were successfully prepared by a simple two-step method,which consists of sonification and functionalization.The characterization analyses reveal that lanthanum is dispersed on the semiconductor surface,probably forming Ti–O–La bonds,which can induce oxygen vacancies and surface defects that effectively restrain the recombination of photogenerated electron/holes pairs.The efficiency of TiO_(2)–La 0.05%–CMCD samples in degradation of propranolol under UV-light irradiation is higher than that of pristine TiO_(2) within 20 min reaction,probably due to complex formation between theβ-blocker and the oligosaccharide,which allows us to propose a photocatalytic mechanism based on the formation of intermediates and competition of these compounds to the radicals and CMCD cavities.

Background:The damage inflicted by the coronavirus diseases 2019(COVID-19)pandemic upon humanity is and will continue to be considerable.Unprecedented progress made in global health over the past 20 years has reverted and economic growth has already evaporated,giving rise to a global recession,the likes of which we may not have experienced since the Second World War.Our aim is to draw the attention of the neglected tropical disease(NTD)community towards some of the major emerging economic opportunities which are quickly appearing on the horizon as a result of COVID-19.Main text:This scoping review relied on a literature search comprised of a sample of articles,statements,and press releases on initiatives aimed at mitigating the impact of COVID-19,while supporting economic recovery.Of note,the donor scenario and economic development agendas are highly dynamic and expected to change rapidly as the COVID-19 pandemic unfolds,as are donor and lender priorities.Con elusions:The NTD community,particularly in low-and middle-income countries(LMICs),will need to work quickly,diligently,and in close collaboration with decision-makers and key stakeholders,across sectors at national and international level to secure its position.Doing so might enhance the odds of grasping potential opportunities to access some of the massive resources that are now available in the form of contributions from corporate foundations,trust funds,loans,debt relieve schemes,and other financial mechanisms,as part of the ongoing and future economic development agendas and public health priorities driven by the COVID-19 pandemic.This paper should serve as a starting point for the NTD community to seek much needed financial support in order to sustain and revitalize control and elimination efforts pertaining to NTDs in LMICs.

Presacral venous bleeding during mobilization in rectal cancer

AIM To analyze the anatomy of sacral venous plexus flow, the causes of injuries and the methods for controlling presacral hemorrhage during surgery for rectal cancer.METHODS A review of the databases MEDLINE? and Embase? was conducted, and relevant scientific articles published between January 1960 and June 2016 were examined. The anatomy of the sacrum and its venous plexus, as well as the factors that influence bleeding, the causes of this complication, and its surgical management were defined.RESULTS This is a review of 58 published articles on presacral venous plexus injury during the mobilization of the rectum and on techniques used to treat presacral venous bleeding. Due to the lack of cases published in the literature, there is no consensus on which is the best technique to use if there is presacral bleeding during mobilization in surgery for rectal cancer. This review may provide a tool to help surgeons make decisions regarding how to resolve this serious complication.CONCLUSION A series of alternative treatments are described; however, a conventional systematic review in which optimal treatment is identified could not be performed because few cases were analyzed in most publications.

As stated in the Declaration of the Rights of the Child, children have the right to go to school for free, to play, and to have the same opportunity to develop and to learn to be responsible and useful[1]. Young children are curious by nature and they have passion for learning [2]. An environment that encourages scientific thinking is what gives young children the time, space and materials to exercise their curiosity [3]. The aim of this paper is to present a methodology for the development of educational environments based on digital technology, internet and free programs available on the web, for the use and application of structural bioinformatics protein and DNA, for early stimulation in basic education at rural and indigenous communities in state of Oaxaca, conducted in On Line Learning & Training “OLL & T”. The course is divided into 4 sections: 1)Introduction to the topic of protein and DNA structure;2)Search in protein data bank;3)Download RasTop program and use it as bioinformatics tool;4)Emulation of building a viral molecule. Workshops were given to children in elementary, middle and high school at the Universidad Del Papaloapan during the National Week of Science and Technology, every year in Oaxaca since 2007 to 2012 and in Monterrey 2010 at the Universidad Autonoma de Nuevo León. Conclusions: positive impact was achieved in basic education in the area of natural sciences by analyzing the molecular basis of protein and deoxyribonucleic acid, theoretically and practically, as a support for teachers.

A method for microbial decontamination of Acanthamoeba cultures using the peritoneal cavity of mice

Objective: To evaluate whether the inoculation of contaminated cultures in the peritoneal cavity of mice could implement decontamination of Acanthamoeba cultures.Methods: Suspensions of Acanthamoeba, Acanthamoeba polyphaga ATCC 30461, or Acanthamoeba spp. isolated from soil(Un B13 strain) were inoculated in the peritoneal cavity of Swiss mice(n = 24). After 1, 6, 12, or 24 h of exposure the peritoneal cavity was washed and assessed for the presence of bacteria, fungi, and Acanthamoeba.Results: After 1 h of intraperitoneal inoculation at least 97% of the bacteria and 96% of the fungi(P < 0.05) and 99% of the bacteria(P < 0.05) were successfully eliminated from the ATCC 30461 strain and from the soil isolate Un B13 strain, respectively. This method also allowed the recovery of most trophozoites and cysts from both Acanthamoeba cultures at the end of 24 h.Conclusions: Our data demonstrated that this technique has great potential for decontamination of Acanthamoeba cultures in a short period of time.
