The hair follicle is a biological oscillator that alternates growth,regression,and rest phases driven by the sequential activation of the proliferation differentiation programs of resident stem cell populations.The activation of hair follicle stem cell niches and subsequent entry into the growing phase is mainly regulated by Wnt/β-catenin signalling,while regression and resting phases are mainly regulated by Tgf-β/Bmp/Smad activity.A major question still unresolved is the nature of the molecular switch that dictates the coordinated transition between both signalling pathways.Here we have focused on the role of Endoglin(Eng),a key co-receptor for members of the Tgf-β/Bmp family of growth factors.Using an Eng haploinsufficient mouse model,we report that Eng is required to maintain a correct follicle cycling pattern and for an adequate stimulation of hair follicle stem cell niches.We further report thatβ-catenin binds to the Eng promoter depending on Bmp signalling.Moreover,we show thatβ-catenin interacts with Smad4 in a Bmp/Eng-dependent context and both proteins act synergistically to activate Eng promoter transcription.These observations point to the existence of a growth/rest switching mechanism in the hair follicle that is based on an Eng-dependent feedback cross-talk between Wnt/β-catenin and Bmp/Smad signals.

Dose-response associations,physical activity intensity and mortality risk:A narrative review

Physical activity is consistently associated with reduced mortality,decreased risk for non-communicable diseases,and improved mental health in observational studies.Randomized controlled trials and observational Mendelian randomization studies support causal links between physical activity and health outcomes.However,the scarcity of evidence from randomized controlled trials,along with their inherent challenges like exposure contrasts,healthy volunteer biases,loss to follow-up,and limited real-world dose-response data,warrants a comprehensive approach.This review advocates synthesizing insights from diverse study designs to better understand the causal relationship between physical activity,mortality risk,and other health outcomes.Additionally,it summarizes recent research since the publication of current physical activity recommendations.Novel observational studies utilizing device-measured physical activity underscore the importance of every minute of activity and suggest that all intensity levels confer health benefits,with vigorous-intensity potentially requiring lower volumes for substantial benefits.Future guidelines,informed by device-measured physical activity studies,may offer refined age-specific recommendations,emphasize vigorous-intensity physical activity,and include daily step counts as a simple,easily assessable metric using commercial wearables.

Trombe walls with porous medium insertion and their influence on thermal comfort in flats in Cusco,Peru

The Trombe wall is a passive indirect heating system which should be used in Cusco,Peru to improve thermal conditions against the typical frosts and low temperatures during winter in the high Andean regions.Due to this problem,the use of a modified Trombe wall with insertion of porous medium is proposed to improve thermal comfort in flat buildings in Cusco.This research aims to analyse and compare the performance of dwellings without Trombe wall,with traditional Trombe wall,and with Trombe wall with glass and plastic pellets insertion in thermal comfort improvement.Autodesk■CFD was used to simulate and analyse the system.The simulation was performed with three prototype flats(55 m^(2),75 m^(2) and 95 m^(2))for six months of the year.From the results obtained,the level of thermal comfort in the traditional scenarios is low with an average PMV of-1.86,in the scenarios with Trombe wall is good and in the scenarios with Trombe wall with insertion of porous medium is slightly better than the previous one,reaching an average PMV of+0.10 and a temperature of 21.90℃.The study carried out is important because it represents an efficient eco-sustainable heating alternative that improves the thermal comfort sensation in houses during the coldest months of the year.

Avian responses to an extreme weather event:The case of the‘Filomena’snowstorm in Madrid(central Spain)

Cold spells and severe weather may have detrimental effects in wild birds,but owing to the stochastic nature of these events,it is not always possible to measure their impacts.in January 2021,a strong snowstorm known as‘Filomena’affected Spain,mainly the region of Madrid(Central Spain),which was fully covered with snow and suffered freezing temperatures for a fortnight.Using bird surveys conducted in the winter-spring of 2020 and 2021(before and after Filomena)at same locations,we addressed the possible impact of the cold spell in three widespread gamebirds:Wood Pigeon(Columba palumbus),Eurasian Magpie(Pica pica)and Red-legged Partridge(Alectoris rufa).No significant abundance differences were found between years for none of the species and hunting pressure was only significantly reduced for Partridges,hence this species may have been negatively affected by Filomena.Results suggest that in response to Filomena,Pigeons conducted partial migration in search of better conditions,while Magpies aggregated in certain areas of Madrid and Partridges endured the harsh conditions within their home ranges.Our results highlight the importance to alleviate negative impacts of extreme weather events in wild birds through management actions,especially for sedentary species.

How to fill the biodiversity data gap:is it better to invest in fieldwork or curation?

Data gaps and biases are two important issues that affect the quality of biodiversity information and downstream results.Understanding how best to fill existing gaps and account for biases is necessary to improve our current information most effectively.Two current main approaches for obtaining and improving data include(1)curation of biological collections,and(2)fieldwork.However,the comparative effectiveness of these approaches in improving biodiversity data remains little explored.We used the Flora de Bogota project to study the magnitude of change in species richness,spatial coverage,and sample coverage of plant records based on curation versus fieldwork.The process of curation resulted in a decrease in species richness(synonym and error removal),but it significantly increased the number of records per species.Fieldwork contributed to a slight increase in species richness,via accumulation of new records.Additionally,curation led to increases in spatial coverage,species observed by locality,the number of plant records by species,and localities by species compared to fieldwork.Overall,curationwas more efficient in producing new information compared to fieldwork,mainly because of the large number of records available in herbaria.We recommend intensive curatorial work as the first step in increasing biodiversity data quality and quantity,to identify bias and gaps at the regional scale that can then be targeted with fieldwork.The stepwise strategy would enable fieldwork to be planned more costeffectively given the limited resources for biodiversity exploration and characterization.

Purpose:This study aimed to quantify the dose-response association and the minimal effective dose of leisure-time physical activity(PA)to prevent mortality and cardiovascular disease in adults with type 2 diabetes.Methods:Cross-country comparison of 2 prospective cohort studies including 14,913 and 17,457 population-based adults with type 2 diabetes from the UK and China.Baseline leisure-time PA was self-reported and categorized by metabolic equivalent hours per week(MET-h/week)according to World Health Organization recommendations:none,below recommendation(>0-7.49 MET-h/week);at recommended level(7.5-14.9 MET-h/week);above recommendation(>15 MET-h/week).Mortality and cardiovascular disease data were obtained from national registries.Results:During a median follow-up of 12.4 and 9.7 years,in the UK and China cohorts,repectively,higher levels of leisure-time PA were inversely associated with all-cause(1571 and 2351 events)and cardiovascular mortality(392 and 1060 events),mostly consistent with a linear dose-response relationship.PA below,at,and above recommendations,compared with no activity,yielded all-cause mortality hazard ratios of0.94(95%confidence interval(95%Ci):0.79-1.12),0.90(95%Ci:0.74-1.10),and 0.85(95%Ci:0.70-1.02)in British adults and 0.87(95%Ci:0.68-1.10),0.88(95%Ci:0.74-1.03),and 0.77(95%Ci:0.70-0.85)in Chinese adults.Associations with cardiovascular mortality were more pronounced in British adults(0.80(95%Ci:0.58-1.11),0.75(95%Ci:0.52-1.09),and 0.69(95%Ci:0.48-0.97))but less pronounced in Chinese adults(1.06(95%Ci:0.76-1.47),1.01(95%Ci:0.80-1.28),and 0.79(95%Ci:0.69-0.92)).PA at recommended levels was not associated with lower rates of major adverse cardiovascular events(2345 and 4458 events).Conclusion:Leisure-time PA at the recommended levels was not convincingly associated with lower mortality and had no association with risk of major adverse cardiovascular events in British or Chinese adults with type 2 diabetes.Leisure-time PA above current recommendations may be needed to prevent cardiovascul

Trypanosoma cruzi is the etiologic agent of Chagas disease.This flagellated protozoan is transmitted to humans as well as different species of domestic and wild animals via vectors from the Reduviidae family(known as"kissing bugs").Despite the fact that hundreds of species of wild mammals are part of the reservoir system,the morphologi-cal changes and clinical manifestations resulting from the pathogenesis of the infection have been largely neglected.The aim of this review is to systematically compile the available information regarding clinicopathological altera-tions in wild mammals due to natural infection by T.cruzi.information was obtained from six online bibliographic data search platforms,resulting in the identification of 29 publications that met the inclusion criteria.Mortality was the most common clinical manifestation,cardiac damage was the main finding at necropsy,and lymphoplas-macytic inflammation was the most frequent microscopic injury.Thus,regardless of its role as a reservoir,T.cruzi has the potential to affect the health status of wild mammals,a situation that highlights the need for further research to analyze,measure,and compare its effects at both the individual and population levels.

Purpose:The present work examines the associations of dietary habits,sedentarism,physical activity(PA)levels and sleep habits,with thyroid function in young euthyroid adults.Methods:A total of 105 young euthyroid adults participated in this cross-sectional study.Thyroid function was determined in fasting conditions(>6 h).Dietary habits were measured by a food frequency questionnaire and three non-consecutive 24 h recalls,and different dietary intake and patterns were then estimated.The time spent in sedentary,PA levels and sleep habits were objectively measured using a wrist-worn accelerometer.Results:Energy and carbohydrate intake were positively associated with thyroid stimulating hormone(TSH)(β=0.222;R^(2)=0.102;P=0.022 andβ=0.425;R^(2)=0.129;P=0.007,respectively)whereas fat intake was negatively associated with TSH(β=-0.428;R^(2)=0.137;P=0.004).Energy intake was also positively associated with free triiodothyronine(β=0.277;R^(2)=0.137;P=0.004).Further,adherence to the Mediterranean diet was negatively related to TSH and free thyroxine(FT4)(β=-0.221;R^(2)=0.113;P=0.020 andβ=-0.268;R^(2)=0.071;P=0.007,respectively).Vigorous-intensity and overall PA were negatively associated with FT4(β=-0.227;R^(2)=0.052;P=0.022 andβ=-0.204;R^(2)=0.042;P=0.041,respectively).in contrast,no associations were found between sleep parameters and thyroid function.Conclusions:Lifestyle factors such as dietary intake and PA levels seems to be related to thyroid function even in young euthyroid adults.

A nonlinear relapse model with disaggregated contact rates:Analysis of a forward-backward bifurcation

Throughout the progress of epidemic scenarios,individuals in different health classes are expected to have different average daily contact behavior.This contact heterogeneity has been studied in recent adaptive models and allows us to capture the inherent differences across health statuses better.Diseases with reinfection bring out more complex scenarios and offer an important application to consider contact disaggregation.Therefore,we developed a nonlinear differential equation model to explore the dynamics of relapse phenomena and contact differences across health statuses.Our incidence rate function is formulated,taking inspiration from recent adaptive algorithms.it incorporates contact behavior for individuals in each health class.We use constant contact rates at each health status for our analytical results and prove conditions for different forward-backward bifurcation scenarios.The relationship between the different contact rates heavily in-fluences these conditions.Numerical examples highlight the effect of temporarily recov-ered individuals and initial conditions on infected population persistence.

Long non-coding RNAs(lncRNAs)are non-coding RNAs longer than 200 nucleotides with dynamic regulatory functions.They interact with a wide range of molecules such as DNA,RNA,and proteins to modulate diverse cellular functions through several mechanisms and,if deregulated,they can lead to cancer development and progression.Recently,it has been described that incRNAs are susceptible to form gene fusions with mRNAs or other incRNAs,breaking the paradigm of gene fusions consisting mainly of protein-coding genes.However,their biological significance in the tumor phenotype is still uncertain.Therefore,their recent identification opens a new line of research to study their biological role in tumor-igenesis,and their potential as biomarkers with clinical relevance or as therapeutic targets.The present study aimed to review the incRNA fusions identified so far and to know which of them have been associated with a potential function.We address the current challenges to deepen their study as well as the reasons why they represent a future therapeutic window incancer.
