Specific probiotics alleviate allergic rhinitis during the birch pollen season

AIM:To investigate whether birch pollen allergy symptoms are linked with gut microbiota changes and whether probiotics have an effect on ***:Forty seven children with confirmed birch pollen allergy were randomized to receive either a probiotic combination of Lactobacillus acidophilus (***) NCFMTM (ATCC 700396) and Bifidobacterium lactis (***) Bl-04 (ATCC SD5219) or placebo in a double-blind manner for 4 mo,starting prior to onset of the birch pollen *** were recorded in a *** samples were taken for analysis of cytokines and *** samples were analysed for microbiota components,calprotectin and *** swabs were taken for analysis of ***:The pollen season induced a reduction in Bifidobacterium,Clostridium and Bacteroides which could not be prevented by the probiotic *** the intervention,significantly higher numbers of *** 11.2×107 ± 4.2×107 vs 0.1×107 ± 0.1×107 bacteria/g feces (P<0.0001) and *** NCFMTM 3.5×106 ± 1.3×106 vs 0.2×106 ± 0.1×106 bacteria/g feces (P<0.0001) were observed in the probiotic group compared to the placebo *** May,there was a tendency for fewer subjects,(76.2% vs 95.2%,P=0.078) to report runny nose,while during June,fewer subjects,11.1% vs 33.3%,reported nasal blocking in the probiotics group (P =0.101).Concomitantly,fewer subjects in the probiotic group had infiltration of eosinophils in the nasal mucosa compared to the placebo group,57.1% vs 95% (P=0.013).Eye symptoms tended to be slightly more frequent in the probiotic group,12.5 d [interquartile range (IQR) 6-18] vs 7.5 d (IQR 0-11.5) (P=0.066) during *** IgA was increased in the placebo group during the pollen season;this increase was prevented by the probiotics (P=0.028).CONCLUSION:Birch pollen allergy was shown to be associated with changes in fecal microbiota *** specific combination of probiotics used was shown to prevent the pollen-induced infiltration

The clinical challenges of homologous recombination proficiency in ovarian cancer: from intrinsic resistance to new treatment opportunities

Ovarian cancer is the most lethal gynecologic cancer. Optimal cytoreductive surgery followed by platinum-based chemotherapy with or without bevacizumab is the conventional therapeutic strategy. Since 2016, the pharmacological treatment of epithelial ovarian cancer has significantly changed following the introduction of the poly (ADP-ribose) polymerase inhibitors (PARPi). BRCA1/2 mutations and homologous recombination deficiency (HRD) have been established as predictive biomarkers of the benefit from platinum-based chemotherapy and PARPi. While in the absence of HRD (the so-called homologous recombination proficiency, HRp), patients derive minimal benefit from PARPi, the use of the antiangiogenic agent bevacizumab in first line did not result in different efficacy according to the presence of homologous recombination repair (HRR) genes mutations. No clinical trials have currently compared PARPi and bevacizumab as maintenance therapy in the HRp population. Different strategies are under investigation to overcome primary and acquired resistance to PARPi and to increase the sensitivity of HRp tumors to these agents. These tumors are characterized by frequent amplifications of Cyclin E and MYC, resulting in high replication stress. Different agents targeting DNA replication stress, such as ATR, WEE1 and CHK1 inhibitors, are currently being explored in preclinical models and clinical trials and have shown promising preliminary signs of activity. In this review, we will summarize the available evidence on the activity of PARPi in HRp tumors and the ongoing research to develop new treatment options in this hard-to-treat population.

El Soplao outcrop, an Early Cretaceous amber deposit recently discovered in northern Spain （Cantabria）, has been shown to be the largest site of amber with arthropod inclusions that has been found in Spain so far. Relevant data provided herein for biogeochemistry of the amber, palynology, taphonomy and arthropod bioinclusions complement those previously published. This set of data suggests at least two botanical sources for the amber of El Soplao deposit. The first （type A amber） strongly supports a source related to Cheirolepidiaceae, and the second （type B amber） shows non-specific conifer biomarkers. Comparison of molecular composition of type A amber with Frenelopsis leaves （Cheirolepidiaceae） strongly suggests a biochemical affinity and a common botanical origin. A preliminary palynological study indicates a regional high taxonomical diversity, mainly of pteridophyte spores and gymnosperm pollen grains. According to the preliminary palynological data, the region was inhabited by conifer forests adapted to a dry season under a subtropical climate. The abundant charcoalified wood associated with the amber in the same beds is evidence of paleofires that most likely promoted both the resin production and an intensive erosion of the litter, and subsequent great accumulation of amber plus plant cuticles. In addition, for the first time in the fossil record, charcoalified plant fibers as bioinclnsions in amber are reported. Other relevant taphonomic data are the exceptional presence of serpulids and bryozoans on the surfaces of some amber pieces indicating both a long exposure on marine or brackish-water and a mixed assemblage of amber. Lastly, new findings of insect bioiuclusions, some of them uncommon in the fossil record or showing remarkable adaptations, are reported. In conclusion, a documented scenario for the origin of the El Soplao amber outcrop is provided.

Impairment of pre-mRNA splicing in liver disease: Mechanisms and consequences

Pre-mRNA splicing is an essential step in the process of gene expression in eukaryotes and consists of the removal ofintrons and the linking of exons to generate mature mRNAs. This is a highly regulated mechanism that allows the alternative usage of exons, the retention ofintronic sequences and the generation of exonic sequences of variable length. Most human genes undergo splicing events, and disruptions of this process have been associated with a variety of diseases, including cancer. Hepatocellular carcinoma (HCC) is a molecularly heterogeneous type of tumor that usually develops in a cirrhotic liver. Alterations in pre-mRNA splicing of some genes have been observed in liver cancer, and although still scarce, the available data suggest that splicing defects may have a role in hepatocarcinogenesis. Here we briefly review the general mechanisms that regulatepre-mRNA splicing, and discuss some examples that illustrate how this process is impaired in liver tumorigenesis, and may contribute to HCC development. We believe that a more thorough examination of pre-mRNA splicing is still needed to accurately draw the molecular portrait of liver cancer. This will surely contribute to a better understanding of the disease and to the development of new effective therapies.

Lack of functional link in the tadpole morphology induced by predators

Most studies of predator-induced plasticity have focused on documenting how prey species respond to predators by modifying phenotypic traits and how traits correlate with fitness. We have previously shown that tadpoles exposed to the dragonfly responded strongly by showing morphological changes, less activity, and better survival than non-exposed tadpoles. Here, we tested whether there is a functional link between morphological plasticity and increased survival in the presence of predators. Tadpoles that experienced predation risk were smaller, less developed, and much less active than tadpoles without this experience. Burst speed did not correlate significantly with morphological changes and predator-induced deeper tails did not act as a lure to divert predator strikes away from the head. Although we have previously found that tadpoles with predator-induced morphology survive better under a direct predator threat, our results on the functional link between morphology and fitness are not conclusive. Our results suggest that in tadpoles (1) burst speed is not important to evade predators, (2) those exposed to predators reduce their activity, and (3) morphological changes do not divert predator attacks away from areas that compromise tadpole survivalEE. Our results show that morphological changes in tadpoles do not explain burst speed or lure attraction, although there was a clear reduction of activity, which itself reduces predation. We propose that changes in tadpole activity could be further analyzed from another perspective, with morphological change as an indirect product of behavior mediated by physiological mechanisms.
