Impact of chronic exposure to bevacizumab on EpCAM-based detection of circulating tumor cells

Background： Circulating tumor cells （CTCs） are often undetected through the immunomagnetic epithelial cell adhesion molecule （EpCAM）-based CellSearch~ System in breast and colorectal cancer （CRC） patients treated with bevacizumab （BEV）, where low CTC numbers have been reported even in patients with evidence of progression of disease. To date, the reasons for this discrepancy have not been clarified. This study was carried out to investigate the molecular and phenotypic changes in CRC cells after chronic exposure to BEV in vitro. Methods： The human CRC cell line WiDr was exposed to a clinically relevant dose of BEV for 3 months in vitro. The expression of epithelial and mesenchymal markers and EpCAM isoforms was determined by western blotting and immunofluorescence. To evaluate the impact of EpCAM variant isoforms expression on CTC enumeration by CellSearch, untreated and treated colon cancer cells were spiked into 7.5 mL of blood from a healthy donor and enumerated by CellSearch. Results： Chronic exposure of CRC cell line to BEV induced decreased expression of EpCAM 40 kDa isoform and increased expression EpCAM 42 kDa isoform, together with a decreased expression of cytokeratins （CK）, while no evidence of epithelial to mesenchymal transition （EMT） in treated cells was observed. The recovery rate of cells through CellSearch was gradually reduced in course of treatment with BEV, being 84% , 70% and 40% at l, 2 and 3 months, respectively. Conclusions： We hypothesize that BEV may prevent CellSearch from capturing CTCs through altering EpCAM isoforms.

Chemical composition of Rosmarinus officinalis essential oil and antioxidant action against gastric damage induced by absolute ethanol in the rat

Objective: To evaluate the antioxidant activity of the essential oil obtained from Rosmarinus officinalis(R. officinalis) in ethanol-induced gastric ulcer model in ***: The antioxidant properties of the essential oil obtained from R. officinalis were evaluated against gastric injury induced by absolute ethanol. Gastric tissues were prepared to enzymatic assays. The levels of glutathione, lipid peroxides, and the activities of glutathione peroxidase, superoxide dismutase and myeloperoxidase were ***: Ethanol produced severe hemorrhagic lesions in the stomach with ulcerative lesion of(140.2 ± 37.2) mm2. In animals pretreated with essential oil of R. officinalis(50 mg/kg, p.o.), a significant inhibition of mucosal injury of(21.2 ± 7.1) mm2(84%inhibition) was observed. The essential oil of R. officinalis protected the gastric mucosa probably by modulating the activities of the enzymes(superoxide dismutase and glutathione peroxidase) and increasing or maintaining the levels of glutathione. In addition,lipid peroxides levels were reduced. The essential oil of R. officinalis was analyzed by gas chromatography–mass spectrometer and the main constituents were cineole(28.5%),camphor(27.7%) and alpha-pinene(21.3%).Conclusions: We suggest that the monoterpenes present in the essential oil obtained from R. officinalis may be among the active principles responsible for the antioxidant activity shown by essential oil of R. officinalis.

Is It Workplace Stress a Trigger for Alcohol and Drug Abuse?

Those workers most vulnerable to pressure tend to suffer from scarce social and personal resources with which to respond adaptively to stress. In this case, the effects of psychoactive substances may exceed the stressed worker’s positive expectations. Thus, the aim is to analyze the scientific evidence on the relationship between drug abuse and workplace stress, based on an integrative review of the literature. Data were collected in February 2016 from the databases of the Virtual Health Library and PubMed. The final sample of 16 articles was divided into two categories: alcohol and drugs abuse in professions with high degree of psychosocial hazards and risks, and alcohol and drugs abuse for workplace stress in other professions. A relationship between precarious conditions, the nature of the work and its influence on drug abuse could be seen. However, other variables may strengthen psychoactive drug use as a coping strategy for stress.

Validation of a Marker Model for Gait Analysis with Wearable Exoskeletons

The aim of the present study was to develop and validate a new marker model for optoelectronic systems adapted to wearable devices, in order to have an analysis tool for kinematic gait evaluation of reproduced patterns by exoskeletons. The marker model has a total of 36 retro-reflective markers attached bilaterally to anatomical landmarks during the static measures （without exoskeleton） and 28 markers at the dynamics measures （with exoskeleton）. The main difference between others kinematic models and the described adapted model was the placement of the three markers in the back thigh and the other three in the back calf, what allowed removing the hip, thigh, knee, tibia and ankle markers. The proposed adapted marker model could be an effective tool to validate the joint movement and velocities of those wearable exoskeletons that at present have been developing.

Bcl-2 down modulation in WEHI-3B/CTRES cells resistant to Cholera Toxin(CT)-induced apoptosis

The very different effects of Cholera Toxin (CT) on cell growth and proliferation may depend on the type of ganglio-side receptors in cell membranes and different signal transduction mechanisms triggered,but other functions relatedto the drug resistance mechanisms can not be *** effect of CT treatment on the "in vitro" clonogenicity,thePopulation Doubling Time (PDT),apoptosis,PKA activation and Bax and Bcl-2 expression was evaluated in WEHI-3Bcell line and its CT-resistant subclone (WEHI-3B/CTRES).In WEHI-3B parental cells the dramatic accumulation ofcAMP induced by CT correlated well with PKA activation,increased PDT value,inhibition of clonogenicity and apop-tosis.H-89 treatment inhibited PKA activation by CT but did not protect the cells from apoptosis and growth *** WEHI-3B/CTRES no significant CT-dependent accumulation of cAMP occurred with any increase of PKA activityand *** CT resistant cells (WEHI-3B/CTRES),Bcl-2 expression was down regulated by both CT or drug treatment(eg.,ciprofloxacin,CPX) although these cells were protected from CT-dependent apoptosis but not from *** from other cell models described,down regulation of Bcl-2 is proved to be independent on cAMPaccumulation and PKA *** observations support the implication of cAMP dependent kinase (PKA) in theinhibition of WEHI-3B cells growth and suggest that,in WEHI-3B/CTRES,Bcl-2 expression could be modulated byCT in the absence of cAMP *** in consideration of many contradictory data reported in literature,ourcell models(of one sensitive parental cell strain and two clones with different uncrossed specific resistance to CT andCPX)provides a new and interesting tool for better investigating the relationship between the CT signal transductionmechanisms and Bcl-2 expression and function.

BACKGROUND Obesity and diabetes are associated with high levels of oxidative *** Romanian patients with obesity and(or)diabetes,this association has not been sufficiently *** To evaluate oxidative stress in obese and(or)diabetic subjects and to investigate the possible correlations between oxidative stress and anthropometric/biochemical *** Oxidative stress was evaluated from a single drop of capillary *** oxygen species(ROS)were evaluated using the free oxygen radical test(FORT).The free oxygen radical defence(FORD)assay was used to measure antioxidant *** FORT levels were higher in obese subjects(3.04±0.36 mmol/L H2O2)vs controls(2.03±0.14 mmol/L H2O2)(Pdex(BMI)(r=0.48,P=0.0000),waist circumference(WC)(r=0.31,P=0.0018),fasting plasma glucose(FPG)(r=0.31,P=0.0017),total cholesterol(TC)(r=0.27,P=0.0068)and uric acid(r=0.36,P=0.0001).FORD values correlated negatively with BMI(r=-0.43,P=0.00001),WC(r=-0.28,P=0.0049),FPG(r=-0.25,P=0.0130),TC(r=-0.23,P=0.0198)and uric acid(r=-0.35,P=0.0002).In obese diabetic subjects,FORT values correlated positively with BMI(r=0.49,P=0.0034)and TC(r=0.54,P=0.0217).FORD values were negatively associated with BMI(r=-0.54,P=0.0217)and TC(r=-0.58,P=0.0121).CONCLUSION Oxidative stress levels,as measured by the FORT and FORD assays,were higher in obese subjects vs *** levels were elevated in diabetic obese patients vs obese non-diabetic patients and controls.

This study aimed to identify the chemical composition of essential oil from fruits(EOAF)and the hexanic crude extract from aerial parts(At-Hex)of atemoya(Annona cherimola x Annona squamosa),a hybrid belonging to the Annonaceae *** and antimicrobial activity was also *** was obtained by hydrodistillation using a Clevenger apparatus,and their composition was analyzed by gas chromatography-mass spectrometry(GC-MS)*** was tested against human tumor cell lines HCT-116(colon carcinoma),SF-295(glioblastoma),OVCAR-8(ovarian carcinoma)and HL60(leukemia)using 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide(MTT)assay,while antimicrobial activity was conducted by bioauthography method against eleven microorganisms ***-four compounds were identified in the EOAF and twenty-nine in *** monoterpenes linalool(25.70%),α-pinene(10.38%),β-pinene(9.12%),transocimene(7.43%),and the sesquiterpene bicyclogermacrene(12.58%)were the major constituents of EOAF,whereas the sesquiterpene spathulenol(13.91%)was the main compound of ***-Hex showed a high cytotoxicity against SF-295(glioblastoma).These findings show an important chemotaxonomic contribution for the Annonaceae family,mainly for the Annona *** proved to be a promising source of substances with potential cytotoxic activity.
