The organism Caenorhabditis elegans is a performant model system for studying human biological processes and diseases,but until now all phenome data are produced as population-averaged *** of individual responses to drug treatments would however be more ***,a new strategy to track different phenotypic traits of individual *** nematodes throughout their full life-cycle-i.e.,embryonic and post-embryonic development,until adulthood onset,differently from life-span-is *** an automated fashion,single worms were synchronized,isolated,and cultured from egg to adulthood in a microfluidic device,where their identity was preserved during their whole *** phenotypes were monitored and quantified for each animal,resulting in high-content phenome ***,the method was validated by analyzing the response of *** to doxycycline,an antibiotic fairly well-known to prolong the development and activate mitochondrial stress-response pathways in different ***,the obtained extensive single-worm phenome not only confirmed the dramatic doxycycline effect on the worm developmental delay,but more importantly revealed subtle yet severe treatment-dependent phenotypes that are representative of minority subgroups and would have otherwise stayed hidden in an averaged *** heterogeneous response started during the embryonic development,which makes essential having a dedicated chip that allows including this early developmental stage in the drug *** approach would therefore allow elucidating pharmaceutical or therapeutic responses that so far were still being overlooked.

Bioplastics were first introduced as environmentally friendly materials,with properties similar to those of conventional plastics.A bioplastic is defined as biodegradable if it can be decomposed into carbon dioxide under aerobic degradation,or methane and CO_(2)under anaerobic conditions,inorganic compounds,and new cellular biomass,by the action of naturally occurring *** definition however does not provide any information on the environmental conditions,timescale and extent at which decomposition processes should *** regard to the aquatic environment,recognized standards have been established to assess the ability of plastics to undergo biodegradation;however,these standards fail to provide clear targets to be met to allow labelling of a bioplastic as ***,these standards grant the user an extensive leeway in the choice of process *** these reasons,the comparison of results deriving from different studies is *** authors analysed and discussed the degree of biodegradability of a series of biodegradable bioplastics in aquatic environments(both fresh and salt water)using the results obtained in the laboratory and from on-site testing in the context of different research *** Oxygen Demand(BOD),CO_(2)evolution,surface erosion and weight loss were the main parameters used by researchers to describe the percentage of *** results showed a large variability both in weight loss and BOD,even when evaluating the same type of *** confirms the need for a reference range of values to be established with regard to parameters applied in defining the biodegradability of bioplastics.

Prolonged exposure of colon cancer cells to the epidermal growth factor receptor inhibitor gefitinib(Iressa^(TM))and to the antiangiogenic agent ZD6474:Cytotoxic and biomolecular effects

AIM： To analyze the biological effects of prolonged in vitro exposure of HT-29 and LoVo colon cancer cell lines to gefitinib （Iressa^TM）, an inhibitor of epidermal growth factor receptor （EGFR） activity, and ZD6474, an inhibitor of both KDR and EGFR activities. METHODS： Cells were treated with each drug for up to 2 wk using either a continuous or an intermittent （4 d of drug exposure followed by 3 d of washout each week） schedule. RESULTS： In both cell types, prolonged exposure （up to 14 d） to gefitinib or ZD6474 produced a similar inhibition of cell growth that was persistent and independent of the treatment schedule. The effects on cell growth were associated with a pronounced inhibition of p-EGFR and/ or p-KDR expression. Treatment with gefitinib or ZD6474 also inhibited the expression of EGFR downstream signal molecules, p-Erkl/2 and p-Akt, although the magnitude of these effects varied between treatments and cell lines. Furthermore, expression of the drug resistance-related protein ABCG2 was shown to significantly increase after 14 d of continuous exposure to the two drugs. CONCLUSION： We conclude that long-term exposure of colon cancer cells to gefitinib and ZD6474 does not modify their cytotoxic effects but it might have an effect on sensitivity to classical cytotoxic drugs.

Fertilization is required for optimum plant growth, particularly in unfertile soils, while optimizing nutrient use effciency is an alternative to reduce inorganic fertilizer needs and reduce environmental problems caused by nutrient leaching. This study investigated soil properties and cowpea yield responses to biochars(BCs) made from different feedstocks, baby corn peel biochar(BC1), branches of mango tree biochar(BC2), and rice husk biochar(BC3), applied in combination with nitrogen-phosphorus-potassium(NPK) *** experiment was conducted in a greenhouse, using an acid sandy soil(Arenosol) that was submitted for 70 d to the following eight treatments: i) control;ii) full dose of NPK(a commercial compound fertilizer(12-24-12 of N-P2O5-K2O)+ urea(46% N));iii) BC1+ half dose of NPK;iv) BC1 + full dose of NPK;v) BC2 + half dose of NPK;vi) BC2 + full dose of NPK;vii) BC3 + half dose of NPK;and viii) BC3 + full dose of NPK. All biochars were applied at a rate of 0.9%(weight/weight), and each type of biochar was combined with half and full doses of NPK fertilizers. Soil pH increased significantly(P < 0.05) in treatments with BC1 and BC2,while cation exchange capacity(CEC) and available P were higher in the treatments with BC1;BC1 and BC2 also induced higher activity of enzymes related to the P cycle and higher cowpea yield. Similar soil properties and cowpea yield parameters were obtained with the full and half doses of NPK fertilizers for each type of biochar used. In conclusion, biochars in the combination with NPK fertilizers improved soil chemistry and enzymatic activities, allowing reduced fertilizer application and food production costs in the acid soil studied.

Phosphorus(P)deficiency is one of the main constraints on crop production in Arenosols(acid sandy soil).The high cost of P fertilizers may represent an insurmountable obstacle in many poor countries,leaving the exploitation of their own calcareous and phosphate rocks as the only low-cost and long-term *** is suggested to have positive effects on soil properties;however,there is no published research on the synergistic effects of biochar and rocky materials in modifying soil *** aim of this study was to investigate the chemical and biochemical responses of an acid Arenosol treated with phosphate rock(PR),calcareous rock(CR),and biochar(BC),and the implications for corn yield.A soil from Marracuene District,Mozambique was used,where corn was grown for 90 d with the soil treated with:no addition(control),water-soluble zinc phosphite fertilizer(WSP),PR,WSP+CR,WSP+BC,WSP+CR+BC,PR+BC,and PR+CR+*** was produced by pyrolysis of babycorn peels for 4 h at 450?C and applied at 11 g *** soil pHH2 Oincreased from about 4.54 in the control to 7.38 in the PR+CR+BC *** oxidizable organic carbon,cation exchange capacity,and available P were higher in the treatments containing BC than in the *** treatments containing CR and/or BC led to the highest activities of alkaline phosphomonoesterase,phosphodiesterase,andα-glucosidase,which increased P availability and gave the greatest biomass and *** suggest that biochar provides additional soluble P and supplies adsorption sites for phosphate,preventing its evolution to unavailable ***,PR applied together with BC contributed to an 840%yield increase compared to the *** treatments containing WSP and BC facilitated phosphite oxidation to phosphate and increased crop yield.

Evolutionary consequences of deception： Complexity and informational content of colony signature are favored by social parasitism

Nestmate recognition codes show remarkable chemical complexity, involving multiple biochemical pathways. This complexity provides the opportunity to evaluate the ecological and social conditions that favor the evolution of complex signaling. We investigated how the chemical signatures of three populations of the social paper wasp Polistes biglumis differed in terms of concentration of hydrocarbons, proportions of branched hydrocarbons and overall variation. We tested whether the variation in chemical signatures among populations could be explained by the prevalence of social parasites or whether this was just an effect of local abiotic conditions which influenced the composition of the hydrocarbon cuticular layer. We studied the chemical signa- ture in three populations in which obligate social parasites differed in the selection pressures they imposed on host populations. Within each population, we restricted our analyses to non-parasitized hosts, to avoid potential short-term effects of parasite pres- ence on the host chemical signatures. We found that host colonies in parasitized populations had more diverse profiles than the parasite-free population. Moreover, the overall concentration of hydrocarbons and the relative proportion of branched hydrocar- bons were larger in the parasitized populations, relative to the non-parasitized one. This is to our knowledge the first evidence in favour of the hypothesis that different traits in the host chemical signatures as a whole undergo evolutionary changes resulting from directional or balancing selection imposed by social parasites. We conclude that obligate social parasites act as ＇engines of diversity＇ on host chemical signatures and operate in favor of a geographic mosaic of diverging communication codes

Ticagrelor therapy and atrioventricular block:Do we need to worry?

Ticagrelor is a potent,direct P2Y12 antagonist with rapid onset of action and intense platelet inhibition,indicated in patients with acute coronary syndromes(ACS).This drug is usually well tolerated,but some patients experience serious adverse effects:Major bleeding;gastrointestinal disturbances;dyspnoea;ventricular pauses > 3 *** the unexpected high incidence of bradyarrhythmias,a PLATO substudy monitored this side effect,showing that ticagrelor was associated with an increase in the rate of sinus bradycardia and sinus arrest compared to *** side effect was usually transient,asymptomatic and not associated with higher incidence of severe atrioventricular(AV) block or pacemaker needs.A panel of experts from Food and Drug Administration did not consider bradyarrhythmias a serious problem in clinical practice and,accordingly,current labeling of the drug does not give any precaution or contraindication regarding this ***,recently some articles have described ACS patients with high-degree,life-threatening,AV block requiring drug discontinuation and,in some cases,pacemaker *** this paper,we describe and discuss five published case reports of severe AV block following ticagrelor therapy and two other cases managed in our *** analysis of literature suggests that,although rarely,ticagrelor can be associated with lifethreatening AV *** and careful monitoring are required especially in patients with already compromised conduction system and/or treated with AV blocking *** studies,with long-term rhythm monitoring,would help to define the outcome of patients at higher risk of developing this complication.

Leather is one of the most popular products across globe and holds a significant place in the economy,while the pollution,associated to traditional leather industry,is far away on the"green chemistry"*** this sense,polyurethanes,which exhibit tunable chemical structures by selecting suitable precursors,can fit specific requirements,and the developments of green strategies make them important candidates for leather *** mini review briefly outlines the recent development of conventional(petrol-based)and sustainable polyurethanes in the leather industry,including their design and properties,in applications such as synthetic leather and surface-finishing(coatings/binders).Finally,outlooks of the future tendency,including more environmental-friendly strategies,bio-sourced/recycled materials and development of high-value multifunctional leather materials,are also here proposed.

Acoustic scattering cross sections of smart furtive obstacles are studied and discussed.A smart furtive obstacle is an obstacle that,when hit by an incoming field,avoids detection through the use of a pressure current acting on its boundary.A highly parallelizable algorithm for computing the acoustic scattering cross section of smart obstacles is *** a case study,this algorithm is applied to the(acoustic)scattering cross section of a"smart"(furtive)simplified version of the NASA space shuttle when hit by incoming time-harmonic plane waves,the wavelengths of which are small compared to the characteristic dimensions of the *** solution to this numerically challenging scattering problem requires the solution of systems of linear equations with many unknowns and *** to the sparsity of these systems of equations,they can be stored and solved using affordable computing resources.A cross section analysis of the simplified NASA space shuttle highlights three findings:i)the smart furtive obstacle reduces the magnitude of its cross section compared to the cross section of a corresponding"passive"obstacle;ii)several wave propagation directions fail to satisfactorily respond to the smart strategy of the obstacle;iii)satisfactory furtive effects along all directions may only be obtained by using a pressure current of considerable *** experiments and virtual reality applications can be found at the website:http://***/ceri/zirilli/w7.
