It has been reported that diallyl sulfide (DAS), a sulfur-containing volatile com.ound in garlic (Allium.sativum.), exerts anticarcinogenic activity in various rodent tum.r m.dels. In the present study, the antitum.r property of DAS was tested in Swiss albino m.ce in the two steqe initiation-prom.tion m.use skin carcinogenesis. Skin cancers were initiated topically with a single subcarcinOgenic dose (52μg) of 7, 12-dim.thyl benz (a) anthracene (Dm.A). Prom.tion was perform.d by twice weekly applications of 12-O-tetradecanoyl phorbol-13-acetate (TPA) at a dose of 5μg/anim.l for 32 weeks. DAS was applied topically (250μg/anim.l) thrice weekly for 3 weeks for anti-initiating and 1 h prior to each prom.tion treatm.nt for anti-prom.ting studies. The results showed that the treatm.nt schedule of DAS can effectively delay the onset of tum.rigenesis and reduce the cum.lative num.er of tum.rs and the average num.er of tum.rs per m.use. In groups in which DAS applied prior to initiation or prom.tion, a significant population of the aniinals rem.ined tum.r-free till the term.nation of experim.nt. These findings suggest that DAS can effectively inhibit chem.cally induced m.use skincarcinogenesis.

In the present investigations, the antitum.rigenic effect of black tea polyphenols (BTP) in twcrstage m.use skin m.del of carcinogenesis was studied. The anim.ls were initiated with a single 'subcarcinogenic' topical dose (52 μg/200 μl acetone ) of 7, 12-dim.thylbenzanthracene (Dm.A). To evaluate the anti-tum.ur initiating activity, BTP was topically applied twice a week for three weeks prior to Dm.A application, followed by topical treatm.nt with 12-o-tetradecanoyl phorbol-13-acetate (TPA) (5 μg/200 μl acetone, 2x/wk. ) as prom.ter. For evaluation of antitum.r prom.ting activity, BTP was applied prior to each treatm.nt of TPA. BTP application showed m.rked inhibitory effect as antitum.ur initiator as well as antitum.ur prom.ter in m.use skin m.del of two-stage carcinogenesis. Since initiation involves genetic pathway and tum.ur prom.tion involves epigenetic pathway, it seem. that BTP exerts its antitum.rigenic effect by altering both genetic and epigenetic pathways

Cancer chem.prevention, a desirable and im.ortant facet of biom.dical research, Provides a practical approach to identify potentially useful inhibitors of cancer developm.nt, and offers an opportunity to study the m.chanism.of carcinogenesis. During the recent past a nuInber of com.ounds have been tested for their anticarcinogenic potential specially constituents of hum.n diet. The enzym. y-glutam.l transpeptidase (CGT), which catalyses the transfer of glutam.l gnups of peptides to other peptides and am.no acids, has been proposed as a m.rker of cell proliferation and neoplasia. It also serves as a tool to evaluate the carcinogenic and cocareinogenic poential of environm.ntal toxicants. In the present investigations, GGT activity induced by carcinogenic polycyclic arom.tic hydrocarbons, viz. 7, 12-dim.thyl-benz(a) anthracene (Dm.A) and benzo(a)Pyrene (BaP was significantly inhibited by diallylsulfide (DAS) and indole-3-carbinol (13C) in m.use skin. DAS and 13C are Constituents of garlic. A significant inhibition in GGT levels was also observed in a strong m.togen (12-o-tetradecanoyl phorbol-13-acelate) induced activity by pretreatm.nt of DAS/13C in m.use skin. Therefore these dielary constituents seem.to be strong m.difiers of chem.cally induced carcinogenesis.

The earthworm. m.taphire posthum. were used as a m.del to assess the toxic potential of cadm.um.incorporated into the soil by environm.ntal or hum.n activities. The retention period of neutral red in the lysosom.s of the coelom.cytes was used as a biom.rker. The viability of harvested coelom.cytes by a non_invasive extrusion protocol was 93% with no alteration by the dye during experim.ntation. The control cells retained dye for 119 and 121 m.n in norm.l soil and KCl, respectively, whereas a linear decline in the retention tim. in the treated earthworm.coelom.cytes was observed. This illustrated that the presence of cadm.um.caused dam.ge to the lysosom.s of the coelom.cytes.

Objective\ To validate the use of Allium.sativum. as a reliable test m.del for genotoxicity, isoproturon, a substituted phenylalkylurea herbicide, was evaluated on the root m.ristem.cells by this plant system. m.thod\ Test concentrations were selected by determ.ning EC50 and root tips were exposed to various concentrations for 6 or 24 hr. EC50 concentration was calculated to be 70.8 ppm.for the root growth. In addition to root growth retardation exposure to isoproturon induced m.rpholoogical changes like discolouration and stiffness of roots. Results\ Exposure to various experim.ntal concentrations of isoproturon (35-280 ppm., including EC50, significantly and dose-dependently inhibited the m.totic index and induced chrom.som. breaks/m.totic aberrations at 6 or 24 hr. Conclusion\ The frequency of aberrations was found to be decreased in a dose dependant m.nner at 24 or 48 hr post exposure, however in com.arison of control cells the frequency of aberrations was considerably high which indicates genotoxicity potentials of isoproturon. Further, present study also suggests that Allium.sativum.is a sensitive, efficient, and reliable test system.for m.asuring the genotoxicity potential of environm.ntal chem.cals.

Artem.sinin is highly effective against drug-resistant m.larial parasites, which affects nearly half of the global population and kills 〉500 000 people each year. The prim.ry cost of artem.sinin is the very expensive process used to extract and purify the drug from.Artem.sia annua. Elim.nation of this apparently unnecessary step will m.ke this potent antim.larial drug affordable to the global population living in endem.c regions. Here we reported the oral delivery of a non-protein drug artem.sinin biosynthesized （~0.8 m./g dry weight） at clinically m.aningful levels in tobacco by engineering two m.tabolic pathways targeted to three different cellular com.artm.nts （chloroplast, nucleus, and m.tochondria）. The doubly transgenic lines showed a three-fold enhancem.nt of isopentenyl pyrophosphate, and targeting AACPR, DBR2, and CYP71AV1 to chloroplasts resulted in higher expression and an efficient photo-oxidation of di- hydroartem.sinic acid to artem.sinin. Partially purified extracts from.the leaves of transgenic tobacco plants inhibited in vitro growth progression of Plasm.dium.falciparum.infected red blood cells. Oral feeding of whole intact plant cells bioencapsulating the artem.sinin reduced the parasitem.a levels in challenged m.ce in com.arison with com.ercial drug. Such novel synergistic approaches should facilitate low-cost production and delivery of artem.sinin and other drugs through m.tabolic engineering of edible plants.
