The goal of asset management is to identify and track the maintenance and replacement of assets that have reached their useful life. For that reason, gathering data and collecting information is a critical step when developing an asset management plan. Such data gathering includes physical and operational properties of the assets as well as collecting and tracking important events during the lifespan of the asset (i.e., pipe breaks, replacement year, maintenance performed, etc.). Critical factors in the asset management plan may be overlooked when there is no data or poor quality data. However, many utilities lack the resources for examining buried infrastructure and lack good quality work order data, so other methods of data collection are needed. The concept for this paper was to develop a means to acquire data on the assets for a condition assessment to identify pipes that were most likely to break and those with the highest consequences for same. Three utilities were used as examples. It was found that for buried infrastructure, much more information was known than anticipated but the actual predictions relied on only a few factors related to pipe type. However, there is a need to track the consequences, in this case breaks, which would indicate a failure. The latter would be useful for predicting future maintenance needs and the most at-risk assets, but is often missing in utility systems as many utilities do not adequately track breaks sufficiently. In this case two utilities were analyzed and predication on a third was developed.

Background:Leber’s Hereditary Optic Neuropathy(LHON;MIM 535000)is one of the most commonly inherited optic neuropathies and it results in significant visual morbidity among young adults with a peak age of onset between the ages of 15–*** worldwide incidence of LHON is approximately 1 in 31,000.95%of LHON patients will have one of 3 primary mitochondrial mutations,G3460A(A52T of ND1),G11778A(R340H of ND4)and T14484C(M64V of ND6).There is incomplete penetrance and a marked gender bias in the development of visual morbidity with approximately 50%of male carriers and 10%of female carriers developing optic *** recovery can occur but is dependent on the mutation present with the highest level of visual recovery seen in patients who have the T14484C *** 3 primary mutations are typically identified by individual end-point PCR-restriction fragment length polymorphism(RFLP)or individual targeted bi-directional Sanger sequencing *** purpose of this study was to design a simple multiplex PCR-RFLP that could detect these 3 primary LHON mutations in one ***:PCR primers were designed to incorporate a MaeIII restriction site in the presence of 3460A and 14484C mutations with the 11778A mutation naturally incorporating a MaeIII site.A multiplex PCR-RFLP assay was developed to detect the 3 common mutations in a single *** LHON controls based on the mitochondrial genome harbouring the 3 common mutations were synthesized and cloned into plasmids to act as reliable assay *** from previously tested patients and the synthetic LHON controls were subjected to the multiplex PCR-RFLP *** RFLP products were detected by agarose gel ***:The novel PCR-RFLP assay accurately detects the 3 primary mutations both in patient DNA and in synthesized DNA control samples with a simple visual mutation detection *** synthesized DNA was demonstrated to be a robust control for the detection of LHON Mutations

Biomineralization of Uranium: A Simulated Experiment and Its Significance

A simulated experimental reduction of and the synthesis of uraninite by a sulfate-reducing bacteria, Desulfovibrio desulfuricans DSM 642, are first reported. The simulated physicochemical experimental conditions were: 35°C, pH=7.0-7.4, corresponding to the environments of formation of the sandstone-hosted interlayer oxidation-zone type uranium deposits in Xinjiang, NW China. Uraninite was formed on the surface of the host bacteria after a one-week's incubation. Therefore, sulfate-reducing bacteria, which existed extensively in Jurassic sandstone-producing environments, might have participated in the biomineralization of this uranium deposit. There is an important difference in the order- disorder of the crystalline structure between the uraninite produced by Desulfovibrio desulfuricans and naturally occurring uraninite. Long time and slow precipitation and growth of uraninite in the geological environment might have resulted in larger uraninite crystals, with uraninite nanocrystals arranged in order, whereas the experimentally produced uraninite is composed of unordered uraninite nanocrystals which, in contrast, result from the short time span of formation and rapid precipitation and growth of uraninite. The discovery has important implications for understanding genetic significance in mineralogy, and also indicates that in-situ bioremediation of U-contaminated environments and use of biotechnology in the treatment of radioactive liquid waste is being contemplated.

Using -24‰ and -14‰ as the endpoints of stable carbon isotopic composition of total organic carbon (δ 13CTOC) of surface soil under pure C3 and C4 vegetation, and surface soil δ 13CTOC data from eastern China, Australia and the Great Plains of North America, we estimate the relative abundance of C3/C4 plants (i.e., the ratio of C3 or C4 biomass to local primary production) in modern vegetation for each region. The relative abundance of modern C3/C4 vegetation from each region is compared to the corresponding climatic parameters (mean annual temperature and precipitation) to explore the relationship between relative C4 abundance and climate. The results indicate that temperature controls the growth of C4 plants. However, even where temperature is high enough for the growth of C4 plants, they will only dominate the landscape when precipitation declines as temperatures increase. Our results are consistent with those of other investigations of the geographic distribution of modern C4 plant species. Therefore, our results provide an important reference for interpretation of past C3/C4 relative abundance records in these three regions.

Reducing risk in long deep tunnels by using TBM and drill-and-blast methods in the same project-the hybrid solution

There are many examples of TBM tunnels through mountains, or in mountainous terrain, which have suffered the ultimate fate of abandonment, due to insufficient pre-investigation. Depth-of-drilling limitations are inevitable when depths approach or even exceed l or 2 km. Uncertainties about the geology, hydro-geology, rock stresses and rock strengths go hand-in-hand with deep or ultra-deep tunnels. Unfortunately, unexpected conditions tend to have a much bigger impact on TBM projects than on drill-and-blast projects. There are two obvious reasons. Firstly the circular excavation maximizes the tangential stress, making the relation to rock strength a higher source of potential risk. Secondly, the TBM may have been progressing fast enough to make probe-drilling seem to be unnecessary. If the stress-to-strength ratio becomes too high, or if faulted rock with high water pressure is unexpectedly encountered, the ＂unexpected events＂ may have a remarkable delaying effect on TBM. A simple equation explains this phenomenon, via the adverse local Q-value that links directly to utilization. One may witness dramatic reductions in utilization, meaning ultra-steep deceleration-of-the-TBM gradients in a log-log plot of advance rate versus time. Some delays can be avoided or reduced with new TBM designs, where belief in the need for probe-drilling and sometimes also pre-injection, have been fully appreciated. Drill-and-blast tunneling, inevitably involving numerous ＂probe-holes＂ prior to each advance, should be used instead, if investigations have been too limited. TBM should be used where there is lower cover and where more is known about the rock and structural conditions. The advantages of the superior speed of TBM may then be fully realized. Choosing TBM because a tunnel is very long increases risk due to the law of deceleration with increased length, especially if there is limited pre-investigation because of tunnel depth.
