AIM:To evaluate the association between Helicobacter pylori(***) infection and MLH1 and MGMT methylation and its relationship with microsatellite instability(MSI).METHODS:The methylation status of the MLH1 and MGMT promoter region was analysed by methylation specific methylation-polymerase chain reaction(MSPPCR) in gastric biopsy samples from uninfected or ***-infected children(n = 50),from adults with chronic gastritis(n = 97) and from adults with gastric cancer(n = 92).MLH1 and MGMT mRNA expression were measured by real-time PCR and normalised to a constitutive gene(β actin).MSI analysis was performed by screening MSI markers at 4 loci(Bat-25,Bat-26,D17S250 and D2S123) with PCR;PCR products were analysed by single strand conformation polymorphism followed by silver *** analyses were performed with either the χ 2 test with Yates continuity correction or Fisher's exact test,and statistical significance for expression analysis was assessed using an unpaired Student's ***:Methylation was not detected in the promoter regions of MLH1 and MGMT in gastric biopsy samples from children,regardless of *** infection *** MGMT promoter was methylated in 51% of chronic gastritis adult patients and was associated with *** infection(P ved methylation of the MLH1 promoter(39%) and increased MSI levels(68%) in samples from gastric cancer patients in comparison to samples from ***-infected adult chronic gastritis patients(P vely).The frequency of promoter methylation for both genes was higher in gastric cancer sam

Could cancer and infection be adverse effects of mesenchymal stromal cell therapy?

Multipotent mesenchymal stromal cells [also referred to as mesenchymal stem cells(MSCs)] are a heterogeneous subset of stromal cells. They can be isolated from bone marrow and many other types of tissue. MSCs are currently being tested for therapeutic purposes(i.e., improving hematopoietic stem cell engraftment, managing inflammatory diseases and regenerating damaged organs). Their tropism for tumors and inflamed sites and their context-dependent potential for producing trophic and immunomodulatory factors raises the question as to whether MSCs promote cancer and/or infection. Thisarticle reviews the effect of MSCs on tumor establishment, growth and metastasis and also susceptibility to infection and its progression. Data published to date shows a paradoxical effect regarding MSCs, which seems to depend on isolation and expansion, cells source and dose and the route and timing of administration. Cancer and infection may thus be adverse or therapeutic effects arising form MSC administration.

We present major and trace element compositions of mineral concentrates comprising garnet xenocrysts,ilmenite,phlogopite,spinel,zircon,and uncommon minerals(titanite,calzirtite,anatase,baddeleyite and pyrochlore)of a newly discovered Late Cretaceous kimberlite(U-Pb zircon age 90.0±1.3 Ma;2σ)named Osvaldo Franca 1,located in the Alto Paranaíba Igneous Province(APIP),southeastern *** grains are lherzolitic(Lherz-1,Lherz-2 and Lherz-3),harzburgitic(Harz-3)and wehrlitic(Wehr-2).The pyrope xenocrysts cover a wide mantle column in the subcratonic lithosphere(66–143 km;20–43 kbar)at relatively low temperatures(811–875°C).The shallowest part of this mantle is represented by Lherz-1 pyropes(20–32 kbar),which have low-Cr(Cr_(2)O_(3)=1.74–6.89 wt.%)and fractionated middle to heavy rare earth elements(MREE-HREE)*** deepest samples are represented by Lherz-2,Lherz-3,Harz-3,and Wehr-2 pyropes(36–43 kbar).They contain high-Cr contents(Cr_(2)O_(3)=7.36–11.19 wt.%)and are characterized by sinusoidal(Lherz-2 and Wehr-2)and spoon-like(Lherz-3 and Harz-3)REE *** to their REE and trace elements,pyrope xenocrysts have enriched nature(e.g.,Ce and Yb ***_(2)O_(3)),indicating that the cratonic lithosphere has been affected by a silicate melt with subalkaline/tholeiite composition due to their low Zr,Ti and Y *** minerals with typical kimberlitic signatures,such as ilmenite and zircon,the exotic compositions of phlogopite and ulvöspinel suggest an enriched component in the magma *** formation of rare mineral phases with strong enrichment of light-REE(LREE)and high field strength elements(HFSE)is attributed to the late-stage kimberlitic *** propose a tectonic model where a thermal anomaly,represented by the low-velocity seismic anomaly observed in P-wave seismic tomography images,supplied heat to activate the alkaline magmatism from a metasomatized cratonic mantle source during the late-stages of Gondwana fragmentation and conse

Cost-effectiveness of enhanced liver fibrosis test to assess liver fibrosis in chronic hepatitis C virus and alcoholic liver disease patients

AIM To assess liver fibrosis(LF) in hepatitis C virus(HCv) and alcoholic liver disease(ALD), estimate health outcomes and costs of new noninvasive testing strategies METHODS A Markov model was developed to simulate LF progression in HCv and ALD for a cohort of 40-yearold men with abnormal levels of transaminases. Three different testing alternatives were studied: a single liver biopsy; annual Enhanced liver fibrosis(ELF?) followed by liver stiffness measurement(LSM) imaging as a confirmation test if the ELF test is positive; and annual ELF test without LSM. The analysis was performed from the perspective of a university hospital in *** data were obtained from published literature. Costs were sourced from administrative databases of the hospital. A proposed Global Boundary Stratotype Section and Point for the base of the Middle Ordovician Series： The Niquivil section, Precordillera of San Juan, Argentina

The Niquivil section, Argentine Precordillera, fulfills most of the requirements for a Global Stratotype Section and Point （GSSP） for the base of the Middle Ordovician Series. It has excellent conodont biostratigraphy in a continuous succession of uniform lithology, is readily accessible, and the critical interval is not tectonically affected. The position of the Lower/Middle Ordovician Series boundary in the Central Precordillera is placed at the middle part of the San Juan Formation, an open-platform carbonate unit. The Niquivil section is proposed as a GSSP for the base of the Middle Ordovician, which is marked by the FAD of the conodont Cooperignathus aranda （Cooper）. It occurs in level NCA, at the upper part of the Oepikodus evae Zone （overlapping uppermost records of O. evae in association with other guide species）; i.e., 100.15 m above the base of the reference section. The auxiliary Pena Sombria section from northern Precordillera records graptolites of the lower （but not lowest） Middle Ordovician in the Isograptus victoriae maximus Zone, few meters above the proposed boundary biohorizon. The range of C. aranda can be correlated with the I. victoriae lunatus Zone up to the lower part of the Undulograptus austrodentatus Zone in diverse sections of the world. C. aranda presents cosmopolitan distribution and occurs in virtually all environments. The proposed GSSP preserves a carbon-isotope record that provides an auxiliary marker of potential global application. Radiometric dating of K-bentonite samples, interbedded with carbonate strata yielding lower records of C. aranda, were recovered from the auxiliary Talacasto section in Central Precordillera （469.8 ±2.3 Ma, U-Pb SHRIMP dating）.
