Our previous study published in Cellular&Molecular Immunology(Harling et al.1)described aberrant immunological features in the pathogenesis of tuberculosis.Overall,we provided evidence supporting an impact chain regarding aberrant high interleukin(IL)-6 and IL-10 cytokine expression and respective receptor signaling affecting T-cell functions in acute tuberculosis.Constitutive STAT3 phosphorylation,high expression of its key regulator suppressor of cytokine signaling(SOCS)3 and spontaneous IL-6/IL-10 secretion characterized CD4^(+)T cells from tuberculosis patients.

T-cells critically contribute to protection against Mycobacterium tuberculosis infection,and impaired T-cell responses can lead to disease progression.Pro-inflammatory and immunosuppressive cytokines affect T-cells,and fine-tuned regulation of cytokine signaling via the Jak/STAT signaling pathways is crucial for appropriate T-cell function.Constitutive STAT3 phosphorylation as a consequence of aberrant cytokine signaling has been described to occur in pathognomonic T-cell responses in inflammatory and autoimmune diseases.We characterized blood samples from tuberculosis patients(n=28)and healthy contacts(n=28)from Ghana for M.tuberculosis-specific T-cell responses,constitutive cytokine production,and SOCS3 and pSTAT3 expression.Lentiviral modulation of primary CD4+T-cells was performed to determine the effects of SOCS3 on T-cell functions.T-cells from tuberculosis patients expressed higher levels of IL-10 and IL-6 and lower levels of T helper type(TH)17 cytokines after M.tuberculosis-specific stimulation compared to healthy contacts.In addition,tuberculosis patients had higher IL-10 and IL-6 levels in the supernatants of non-stimulated immune cells and plasma samples compared to healthy contacts.Notably,aberrant cytokine expression was accompanied by high constitutive pSTAT3 levels and SOCS3 expression in T-cells.Multivariate analysis identified an IL-6/IL-10 co-expression-based principal component in tuberculosis patients that correlated with high pSTAT3 levels.SOCS3 contributed to a regulatory component,and tuberculosis patients with high SOCS3 expression showed decreased TH1 cytokine expression and impaired IL-2-induced STAT5 phosphorylation.SOCS3 over-expression in primary CD4+T-cells confirmed the SOCS3 inhibitory function on IL-2-induced STAT5 phosphorylation.We conclude that constitutive pSTAT3 and high SOCS3 expression are influential factors that indicate impaired T-cell functions in tuberculosis patients.

Background:Exercise training promotes brain plasticity and is associated with protection against cognitive impairment and Alzheimer’s disease(AD).These beneficial effects may be partly mediated by blood-borne factors.Here we used an in vitro model of AD to investigate effects of blood plasma from exercise-trained donors on neuronal viability,and an in vivo rat model of AD to test whether such plasma impacts cognitive function,amyloid pathology,and neurogenesis.Methods:Mouse hippocampal neuronal cells were exposed to AD-like stress using amyloid-βand treated with plasma collected from human male donors 3 h after a single bout of high-intensity exercise.For in vivo studies,blood was collected from exercise-trained young male Wistar rats(high-intensity intervals 5 days/week for 6 weeks).Transgenic AD rats(McGill-R-Thyl-APP)were inj ected 5 times/fortnight for 6 weeks at2 months or 5 months of age with either(a)plasma from the exercise-trained rats,(b)plasma from sedentary rats,or(c)saline.Cognitive function,amyloid plaque pathology,and neurogenesis were assessed.The plasma used for the treatment was analyzed for 23 cytokines.Results:Plasma from exercised donors enhanced cell viability by 44.1%(p=0.032)and reduced atrophy by 50.0%(p<0.001)in amyloid-β-treated cells.In vivo exercised plasma treatment did not alter cognitive function or amyloid plaque pathology but did increase hippocampal neurogenesis by~3 fold,regardless of pathological stage,when compared to saline-treated rats.Concentrations of 7 cytokines were significantly reduced in exercised plasma compared to sedentary plasma.Conclusion:Our proof-of-concept study demonstrates that plasma from exercise-trained donors can protect neuronal cells in culture and promote adult hippocampal neurogenesis in the AD rat brain.This effect may be partly due to reduced pro-inflammatory signaling molecules in exercised plasma.

There are various publications on the 90th birthday of Gottfried Konecny.His involvement in the International Society for Photogrammetry and Remote Sensing(ISPRS)is particularly described by others.Here,his forward-oriented impulses in research and development at the former Institute for Photogrammetry and Engineering Survey of the(Technical)University of Hannover,today Institute for Photogrammetry and Geoinformation,Leibniz University Hannover,and some of his activities supporting developing and other countries by given shortterm courses are highlighted.Gottfried Konecny was always one of the first in pushing new technologies.With the analytical plotter AP/C-3,he introduced analytical photogrammetry in Germany.Based on his experience with this not really operational version,he induced the company Zeiss to develop the first really operational analytical plotter Planicomp,which resulted in the end of analog photogrammetric devices and their limitations.At the invitation of Gilbert Hobrough,a mainly hardware-based image correlator was developed for the AP/C-3 in Hannover which anticipated many of today’s applications.Gottfried Konecny initiated remote sensing in Germany.Based on his proposal with the Metric Camera Flight on Spacelab,the then highest resolution civil stereoscopic space images available were generated.Despite limited computer performance at his institute,a digital stereo plotter was developed,using hardware components from the cooperating Swedish company Context Vision,long before digital stereo workstations with increased and affordable computer capacity were possible.Also,in the area of GIS,he too,pushed development in collaboration with companies and administrations.Shortly before the end of his time as head of the institute,and more so after he became emeritus professor,he started a series of educational workshops,particularly in developing countries,but also other countries,to support the development of photogrammetry,remote sensing and GIS.He promoted the use of space imagery for mapping to imp

This paper considers the problem of supply-demand imbalances in Mobility-on-Demand(MoD)services.These imbalances occur due to uneven stochastic travel demand and can be mitigated by proactively rebalancing empty vehicles to areas where the demand is high.To achieve this,we propose a method that takes into account uncertainties of predicted travel demand while minimizing pick-up time and rebalance mileage for autonomous MoD ride-hailing.More precisely,first travel demand is predicted using Gaussian Process Regression(GPR)which provides uncertainty bounds on the prediction.We then formulate a stochastic model predictive control(MPC)for the autonomous ride-hailing service and integrate the demand predictions with uncertainty bounds.In order to guarantee constraint satisfaction in the optimization under estimated stochastic demand prediction,we employ a probabilistic constraining method with user-defined confidence interval,using Chance Constrained MPC(CCMPC).The benefits of the proposed method are twofold.First,travel demand uncertainty prediction from data can naturally be embedded into the MoD optimization framework,allowing us to keep the imbalance at each station below a certain threshold with a user-defined probability.Second,CCMPC can be relaxed into a Mixed-Integer-Linear-Program(MILP)and the MILP can be solved as a corresponding Linear-Program,which always admits an integral solution.Our transportation simulations show that by tuning the confidence bound on the chance constraint,close to optimal oracle performance can be achieved,with a median customer wait time reduction of 4%compared to using only the mean prediction of the GPR.

In the context of climate change,especially for the temperate continental climate,new potential crop species are emerging,originating from the tropics.One of them is quinoa(Chenopodium quinoa Willd.)with multiple benefits for seed and leaf production.Quinoa is native to South America,grown mainly for seeds,with a high ecological plasticity.Little is known about the potential of using quinoa plants as a leafy vegetable for food diversification.In this study,the nutritional and antinutritional content of quinoa leaves was evaluated in three cultivars(Titicaca,Puno,Vikinga),considering different densities and times of sowing.Puno cultivar had a higher total content of carbohydrates,lipids,proteins and dietary fibers,and lower mineral contents in leaves.Low levels of antinutrient compounds were found in Vikinga leaves.Regarding the time of the crop establishment,the highest content of primary metabolic compounds(carbohydrates,lipids,proteins,dietary fibers)was achieved by April 17,the sowing date.Crop densities of 7.7,3.2,and 1.6 mil.plants/ha did not significantly influence the content of compounds with antinutritive role,such as oxalates,saponins or trypsin inhibitors.The content of mineral elements such as:Fe,Zn,Na and K were significantly influenced by the cultivar,compared to Mg and Ca whose values were insignificant regardless of the treatment.

We study N-cluster correlation functions in four-and five-dimensional(4D)and 5D)bond percolation by extensive Monte Carlo simulation.We reformulate the transfer Monte Carlo algorithm for percolation[Phys.Reu.E 72,016126(2005)]using the disjoint-set data structure,and simulate a cylindrical geometry Ld^-1×∞,with the linear size up to L=512 for 4D and 128 for 5D.We determine with 1 high precision all possible N-cluster exponents,for N=2 and 3,and the universal amplitude for a logarithmic correlation function.From the symmetric correlator with N=2,we obtain the correlation-length critical exponent as 1/v=1.4610(12)for 4D and 1/v=1.737(2)for 5D,significantly improving over the existing results.Estimates for the other exponents and the universal logarithmic amplitude have not been reported before to our knowledge.Our work demonstrates the validity of logarithmic conformal field theory and adds to the growing knowledge for high-dimensional percolation.
