Enhancer variants reveal a conserved transcription factor network governed by PU.1 during osteoclast differentiation

Genome-wide association studies(GWASs) have been instrumental in understanding complex phenotypic traits. However, they have rarely been used to understand lineage-specific pathways and functions that contribute to the trait. In this study, by integrating lineage-specific enhancers from mesenchymal and myeloid compartments with bone mineral density loci, we were able to segregate osteoblast-and osteoclast(Oc)-specific functions. Specifically, in Ocs, a PU.1-dependent transcription factor(TF)network was revealed. Deletion of PU.1 in Ocs in mice resulted in severe osteopetrosis. Functional genomic analysis indicated PU.1 and MITF orchestrated a TF network essential for Oc differentiation. Several of these TFs were regulated by cooperative binding of PU.1 with BRD4 to form superenhancers. Further, PU.1 is essential for conformational changes in the superenhancer region of Nfatc1. In summary, our study demonstrates that combining GWASs with genome-wide binding studies and model organisms could decipher lineage-specific pathways contributing to complex disease states.

文物承载着灿烂文明,传承着历史文化,是全人类的宝贵遗产。由于文物不可再生的珍贵性,以及最少干预的文物保护基本原则,无损分析技术始终是文物制作工艺研究、劣化机理及保护修复研究中材料分析的最主要技术手段。因此,研发、应用新型无损分析技术是文物保护利用的重要研究方向。近年来,源于遥感领域的便携式地物高光谱仪在文物材料分析中展现出强大应用潜力,并且已在石质古建筑、绘画等文物修复中成功应用。作为一种无需采样的非侵入式光谱技术,便携式地物高光谱仪可在极短时间获取一个涵盖可见光-近红外-短波红外波段(350~2500 nm)的全波段、高分辨率反射光谱。同时,其便携程度很高,对工作环境、光源等无特殊要求,可在野外文物或遗址现场原位使用。此外,在采集光谱数据的同时还可通过遥感技术实现数据的远程传输和分析。上述技术特点使其在文物材料分析中具有独特的应用优势。以光谱技术的基本原理为基础,首先介绍了目前文物材料分析中常用便携式地物高光谱仪的设备型号及其性能指标;其次细致梳理了近十年来其在有机质/无机质文物材料定性和定量分析、文物保护材料分析,以及文物修复原位、实时监测等方面的应用现状;之后深入探讨了谱图预处理的规范和建立标准波谱库等两个影响该技术应用效果的关键问题;最后展望了该技术的发展趋势,预期在脆弱有机质文物分析、多种分析技术联合应用和定量分析研究方面前景广阔。

Prevention of thromboembolic events after radical prostatectomy in patients with hereditary thrombophilia due to a factor V Leiden mutation by multidisciplinary coagulation management

Possible Uses for Silymarin in Human Health: Systematic Review

Silymarin, from the fruit of Silybum marianum, is known for its hepatoprotective action. The aim of this study was to review the mechanisms of action of the silymarin phytocomplex to expand the possibilities for its application in human health. The search for published articles was carried out on the cAPES Journals Portal platform, which covers worldwide scientific databases. Publications from 2010 to 2022 were included. Of the 311 articles retrieved, 21 were included. The articles discuss the diversity of silymarin’s applications and the possibility of optimizing its bioavailability using drug delivery systems. Silymarin shows promise in numerous diseases, such as liver, kidney, cardiovascular, respiratory and others. Its antiviral action has been demonstrated in studies and silymarin has the potential to be used as a complementary therapy in the treatment of many diseases, with the expectation that, in the future, it will be used in therapeutic protocols for exclusive use.

New Rotative Radiofrequency Technology: A Multicenter Retrospective Study on Efficacy and Safety

Background: Non-ablative radiofrequency (RF) technology is widely used for the treatment of signs of aging, skin laxity, localized fat and cellulite. However, many RF devices suffer from technical limitations that make the results highly dependent on the user’s experience or clinical limitations that compromise safety, efficacy and comfort. Objective: To evaluate the efficacy and safety of a novel semi-automatic non-ablative RF device for skin tightening, body shaping and cellulite reduction. Materials and methods: A retrospective multicenter study was performed using a novel rotative RF device with temperature and impedance control (Sculpt&Shape®, Sinclair, Spain). 58 subjects underwent a minimum of 4 treatments every 1-2 weeks. A total of 120 treatments (56.7% body and 43.3% facial) and 1034 sessions were performed. Efficacy was evaluated by 3 researchers using the Global Aesthetic Improvement Scale (GAIS) and anthropometric measurements were collected. Subject and practitioner satisfaction were recorded. Safety was evaluated by analyzing the perception of pain during the treatment and the side and adverse effects immediately after treatment and over the next 48 hours. Results: Between 25% and 50% improvement was achieved according to the GAIS. Statistically significant (p ction of 5.1 mm and circumference reduction of 32.0 mm, 16.4 mm and 13.9 mm were recorded for the abdomen, legs and arms, respectively. 77.5% of subjects and 94.9% of practitioners were either satisfied or very satisfied. Minor adverse effects were found in 0.97% of the sessions. conclusion: This novel semi-automatic rotative RF device has been found to provide treatments that are effective, safe and comfortable for both practitioners and subjects. .

Hepatocellular carcinoma and food contamination: Ochratoxin A as a great prompter

Ochratoxin A (OTA) is a secondary metabolite of Asper-gillus and Penicillium, microorganisms that can be haz-ardous to health when present as food contaminants. OTA is a potent member of a group of mycotoxins. Prolonged exposure to mycotoxins in the diet is related to cancer, among other diseases. Hepatocellular car-cinoma (Hcc) accounts for 70%-90% of primary liver cancers and is the third leading cause of cancer-related deaths worldwide. In a recent study, Ibrahim et al proposed a correlation between the incidence of Hcc and contamination with OTA. Analysis of OTA in serum samples showed that Hcc patients had the highest incidence of OTA of the subjects examined (5-fold higher than that of the control group). OTA levels were significantly increased in Hcc patients. This study demonstrates that chronic exposure to high levels of OTA may be associated with a high risk of liver cancer development. Future epidemiologic studies of Hcc should focus on good practices in food preparation, food storage and the consumption of OTA-containing foods.
