The HDAC3 enzymatic activity regulates skeletal muscle fuel metabolism

Histone deacetylase 3 (HDAC3) is a major HDAC, whose enzymatic activity is targeted by small molecule inhibitors for treating a variety of conditions. However, its enzymatic activity is largely dispensable for its function in embryonic development and hepatic lipid ***3 plays a pivotal role in regulating muscle fuel metabolism and contractile function. Here, we address whether these muscular functions of HDAC3 require its enzymatic activity. By mutating the NCoR/SMRT corepressors in a knock - in mouse model named NS-DADm, we ablated the enzymatic activity of HDAC3 without affecting its protein levels. Compared to the control mice, skeletal muscles from NS-DADm mice showed lower force generation, enhanced fatigue resistance, enhanced fatty acid oxidation, reduced glucose uptake during exercise, upregulated expression of metabolic genes involved in branchedchain amino acids catabolism, and reduced muscle mass during aging, without changes in the muscle fiber-type composition or mitochondrial protein content. These muscular phenotypes are similar to those observed in the HDAC3-depleted skeletal muscles, which demonstrates that, unlike that in the liver or embryonic development, the metabolic function of HDAC3 in skeletal muscles requires its enzymatic activity. These results suggest that drugs specifically targeting HDAC3 enzyme activity could be developed and tested to modulate muscle energy metabolism and exercise performance.

Objective:To evaluate the correlation between ALT,AST and IVIG-resistance Kawasaki disease by using ***:we searched PubMed,the Cochrane Library,EMBASE,Wanfang database,VIP,CNKI database for study reports which describe the correlation between alt,AST and IVIG-resistance Kawasaki disease,literature retrieval time from 2002.1 to 2016.1,then extract the data of studys which meet the requirements,at last Cochrane Collaboration review manger software(Revman 5.2) was used,meta-analysis and systematic review was ***:a total of 20 articles were included in the final meta-analysis,19 articles were about ALT and 15 articles were about *** meta-analysis Results showed that ALT(overall MD=43.71,95%CI 31.19-56.23,P< 0.01) AND AST(overall MD=44.69,95%CI 28.59-60.80,P < 0.01) are associated with the occurrence of IVIGresistance Kawasaki *** The serum levels of ALT and AST were higher in the children of IVIGresistance than the children of IVIG-sensitive in kawasaki *** result is based on the clinical research already published,but due to the definitions of IVIG-resistance kawasaki disease are not unified,part of the studys design are not standardized,relevant researches exist publication bias problem,still need more multicenter,large sample clinical research to further observe the relevance.

Objectives:Although many significant physiological changes have been discovered during the maturation process of the heart,the genetic mechanisms involved in the changes between immature and mature myocardium have only been partially ***,high-throughput microarray screening and improved bioinformatics were used to characterize the gene expression profile changed between the human fetal and adult ***:An Arraystar Human Microarray was applied to define gene expression signatures of the fetal hearts(13-17 weeks of gestation,n = 4) and normal adult hearts(30-40 years old,n = 4).Gene ontology analysis,pathway analysis,gene set enrichment analyses and signal transduction network were performed to predict the function of the differentially expressed *** differentially expressed mRNAs were selected and confirmed by quantificational real-time polymerase chain reaction(qRT-PCR).Results:A total of 5547 coding RNAs were found to be significantly differentially expressed(fold change ≥ 2.0,P < 0.05 and false discovery rate < 0.05) between fetal and adult hearts,among which 3,220 mRNAs were downregulated and 2,327 were up-regulated in the adult group compared with the fetal *** ontology analysis revealed that the down-regulated genes were mainly involved in "cell cycle".KEGG pathway analysis and gene set enrichment analyses identified that "cell cycle pathway" was the most enriched pathway involved in the downregulated *** signal transduction network suggested that epidermal growth factor receptor(EFGR) and insulin-like growth factor 1 receptor(IGF1 R) play a central role in the regulation of heart ***,IGF1 R and fibroblast growth factor receptor 2(FGFR2) were identified to be the core genes in the signal transduction network related to cell *** qRT-PCR Results suggested that the expression tendency was concordant with the microarray ***:Taken together,our data identified the transcriptional regul
