The relationship between DNA fragmentation and the intensity of morphologically abnormal human spermatozoa

Objective:To determine the relationship between teratozoospermia and sperm DNA fragmentation(SDf)in the human ***:This retrospective study included 100 normozoospermic men as a control cohort(abnormal forms>14%),210 patients with a high level of abnormal forms(≤4%)and 65 patients presenting with a moderate level of abnormal forms(>4%to≤14%)based on the World Health Organization *** morphology was assessed using bright field *** DNA fragmentation was assessed using the sperm chromatin dispersion ***-parametric analyses were conducted to examine the relationship between abnormal sperm morphology and sperm DNA fragmentation;receiver operating characteristic(ROC)analyses were conducted to assess sensitivity and specificity of this ***:A correlation analysis revealed that the higher the proportion of abnormal spermatozoa in the ejaculate,the higher the level of SDf(Spearman's Rho=-0.230;Pficant differences in the proportion of SDf were found when all cohorts were compared(Pficant differences were also retained when the different cohorts were compared *** analysis showed a moderate but significant predictive value for SDf to differentiate patients with different levels of ***:Although analysis of a more continuous range of values for teratozoospermia would help further clarify any causal relationship with SDf,there is clearly a synergistic or coincident affiliation between these variables that needs to be acknowledged by the clinician when interpreting the spermiogram.

Evaluation of Clinical Characteristics and Plasma Cortisol Concentrations in Individuals with COVID-19 and Post-COVID Syndrome

COVID-19 generates systematic alterations in humans both in active stages of infection and over time, called post-COVID syndrome. Cortisol is a hormone that is overexpressed in inflammation and cellular stress processes. Its main function is to return to physiological homeostasis, so its evaluation together with other clinical parameters can allow us to determine the degree of systemic affectation by COVID-19. Objective: To evaluate changes in clinical parameters and plasma cortisol concentrations in patients with active COVID-19 and post-COVID syndrome. Material and Methods: Healthy patients, in stages of mild infection, critical and with post-COVID syndrome, were recruited, obtaining, through clinical diagnoses and interviews, their main clinical characteristics, in addition to plasma, in which cortisol concentrations were determined using competitive ElISA. Results: The critical stage group had higher frequencies of comorbidities, clinical symptoms, as well as more altered laboratory parameters compared to the other subgroups. In the post-COVID syndrome group after the initial infection, most laboratory parameters recovered, however, several clinical symptoms remained latent over time. The determination of cortisol showed an increase in its concentration, being higher in patients in critical stage and with post-COVID syndrome. Conclusion: COVID-19 disease generates clinical alterations that trigger an increase in plasma cortisol. These alterations increase as the stages of infection become more severe and some of them remain altered in patients with post-COVID syndrome.

TGAIN: Geospatial Data Recovery Algorithm Based on GAIN-lSTM

Accurate geospatial data are essential for geographic information systems (GIS), environmental monitoring, and urban planning. The deep integration of the open Internet and geographic information technology has led to increasing challenges in the integrity and security of spatial data. In this paper, we consider abnormal spatial data as missing data and focus on abnormal spatial data recovery. Existing geospatial data recovery methods require complete datasets for training, resulting in time-consuming data recovery and lack of generalization. To address these issues, we propose a GAIN-lSTM-based geospatial data recovery method (TGAIN), which consists of two main works: (1) it uses a long-short-term recurrent neural network (lSTM) as a generator to analyze geospatial temporal data and capture its temporal correlation; (2) it constructs a complete TGAIN network using a cue-masked fusion matrix mechanism to obtain data that matches the original distribution of the input data. The experimental results on two publicly accessible datasets demonstrate that our proposed TGAIN approach surpasses four contemporary and traditional models in terms of mean absolute error (MAE), root mean square error (RMSE), mean square error (MSE), mean absolute percentage error (MAPE), coefficient of determination (R2) and average computational time across various data missing rates. Concurrently, TGAIN exhibits superior accuracy and robustness in data recovery compared to existing models, especially when dealing with a high rate of missing data. Our model is of great significance in improving the integrity of geospatial data and provides data support for practical applications such as urban traffic optimization prediction and personal mobility analysis. © 2024 The Authors. Precipitation transformation pathway and mechanical behavior of nanoprecipitation strengthened fe-Mn-Al-C-Ni austenitic low-density steel

Precipitation strengthening has been widely adopted in austenitic low-density steel owing to excellent hardened *** approach generally employs the coherentκ′carbides and non-coherent B2 *** the precipitation transformation pathway is decisive for further optimizing the microstructures under specific engineering ***,the detailed precipitation sequence of fe-28Mn-11Al-1C-5Ni(wt%)austenitic low-density steel as well as its influence on mechanical prop-erties during aging process is systematically *** results reveal that nano-sizedκ′carbides domains(2 nm)exist in the solution-treated *** aging at 500℃for 1 h,the cuboidalκ′carbides(15-20 nm)uniformly disperse in austenite ***,after aging at 700℃for 15 min,the coarsenκ′carbides(30-35 nm)inhomogeneously distribute and align preferentially along the〈100〉***,extending the aging time to 60 min,the needle-type B2 particles replace theκ′carbides due to the enrichment of Ni elements at the phase boundaries among the austenite andκ′*** aging at 900℃,κ′carbides entirely dissolve into the austenite matrix,and the intragranular B2 particles are the sole precipitates in the austenite matrix and follow the K-S orientation relationship with *** work hardening capability seriously deteriorates due to the shearing ofκ′carbides by gliding *** the intragranular B2 particles preserve excellent work hardening rate by dislocations bow-out *** present work is meaningful for guiding the design of new generation dual-nano precipitation austenitic lightweight steel.

The current spotlight of cancer therapeutics is shifting towards personalized medicine with the widespread use of monoclonal antibodies(mAbs).Despite their increasing potential,mAbs have an intrinsic limitation related to their inability to cross cell membranes and reach intracellular *** offers promising solutions to overcome this limitation,however,formulation challenges *** challenges are the limited loading capacity(often insufficient to achieve clinical dosing),the complex formulation methods,and the insufficient characterization of mAb-loaded ***,we present a new nanocarrier consisting of hyaluronic acid-based nanoassemblies(HANAs)specifically designed to entrap mAbs with a high efficiency and an outstanding loading capacity(50%,w/w).HANAs composed by an mAb,modified HA and phosphatidylcholine(PC)resulted in sizes of~100 nm and neutral surface *** modeling identified the principal factors governing the high affinity of mAbs with the amphiphilic HA and *** composition and structural configuration were analyzed using the orthogonal techniques cryogenic transmission electron microscopy(cryo-TEM),asymmetrical flow field-flow fractionation(Af4),and small-angle X-ray scattering(SAXS).These techniques provided evidence of the formation of core-shell nanostructures comprising an aqueous core surrounded by a bilayer consisting of phospholipids and amphiphilic *** vitro experiments in cancer cell lines and macrophages confirmed HANAs’low toxicity and ability to transport mAbs to the intracellular *** reproducibility of this assembling process at industrial-scale batch sizes and the long-term stability was *** conclusion,these results underscore the suitability of HANAs technology to load and deliver biologicals,which holds promise for future clinical translation.

Precipitation of multi-type nano-quasicrystals in a Mg-Zn-Y alloy

Mg-6Zn-1Y(at.%)ribbons with strengthening precipitates of multi-type nanoquasicrystals were prepared by melt-spinning followed by aging *** evolution of the rapidly solidified ribbons during isothermal aging was comprehensively studied using various electron microscopy *** new kinds of decagonal quasicrystals were formed in aged ribbons,in addition to precipitation of nanometer icosahedral ***-resolution observations reveal that both decagonal quasicrystals can be modeled by quasiperiodic tiling with decagonal clusters of 2.5 nm in diameter,but overlap of neighboring clusters in both decagonal quasicrystals is different from the Gummelt model observed in other quasicrystals.A shell composed of complex laves Mg-Zn domains was formed surrounding each decagonal quasicrystal precipitate upon prolonged *** addition,all kinds of nanoprecipitates exhibit excellent structure and size stability at 573 *** findings may have implications for not only fundamental studies about quasicrystals,but also microstructural manipulation of high-performance Mg alloys.

Metabolic collaboration between cells in the tumor microenvironment has a negligible effect on tumor growth

The tumor microenvironment is composed of a complex mixture of different cell types interacting under conditions of nutrient deprivation,but the metabolism therein is not fully understood due to difficulties in measuring metabolic fluxes and exchange of metabolites between different cell types in ***-scale metabolic modeling enables estimation of such exchange fluxes as well as an opportunity to gain insight into the metabolic behavior of individual cell ***,we estimated the availability of nutrients and oxygen within the tumor microenvironment using concentration measurements from blood together with a metabolite diffusion *** addition,we developed an approach to efficiently apply enzyme usage constraints in a comprehensive metabolic model of human *** combined modeling reproduced severe hypoxic conditions and the Warburg effect,and we found that limitations in enzymatic capacity contribute to cancer cells’preferential use of glutamine as a substrate to the citric acid ***,we investigated the common hypothesis that some stromal cells are exploited by cancer cells to produce metabolites useful for the cancer *** identified over 200 potential metabolites that could support collaboration between cancer cells and cancer-associated fibroblasts,but when limiting to metabolites previously identified to participate in such collaboration,no growth advantage was *** work highlights the importance of enzymatic capacity limitations for cell behaviors and exemplifies the utility of enzyme-constrained models for accurate prediction of metabolism in cells and tumor microenvironments.
