Targeting biosignatures of hyperglycemia and oxidative stress in diabetes comorbid depressive rats: effectiveness of hydroethanolic extract of the whole plant of Ludwigia octovalvis

Background:The prime objective of the current research was to evaluate the whole plant hydroalcoholic extract of Ludwigia octovalvis(HLO)against hyperglycemia,and oxidative stress biomarkers in rats induced with diabetes comorbid depression,diabetes comorbid depression(streptozotocin-nicotinamide+electric footshocks).Methods:2,2-Diphenyl-1-picrylhydrazyl assay of HLO versus ascorbic acid was *** of 200 and 400 mg/kg body weight/day HLO doses versus 25 mg/kg body weight/day metformin was studied through insulin,glucose,superoxide dismutase,lipid peroxidation,catalase,and behavioral assessment(forced swim and open field tests).Results:IC50 values of HLO and ascorbic acid were 33.52 and 27.86μg/mL *** the HLO doses showed intended results with respect to oxidative stress biomarkers in diabetes comorbid depression rats in comparison to *** field test showed better results for HLO in diabetes comorbid depression ***,hypoglycemic effects,and forced swim test performance of metformin was slightly higher than the 400 mg dose,followed by the 200 mg dose of *** gallate,gallic acid,β-sitosterol,and quercetin in HLO might resulted in attenuating diabetic as well as depression ***:Inhibition of glucosidase and lipase activity,and AMP-activated protein kinase phosphorylation might be the possible biochemical changes occurred in HLO treated rats.

Onsite mine fire generates large volumes of heat-affected coal in Jharia coalfields,*** utilization of such heat-affected coal in thermal utilities is not feasible as such coal does not have the desirable volatile matter required for *** the present work,experimental studies have been carried out to investigate the possible utilization of such heat-affected coal in thermal utilities by blending with other ***-affected coal(31%ash and 5300 kcal/kg GCV)collected from Jharia coalfield were blended with thermal coal(28%ash and 5650 kcal/kg GCV)in different ratios of 90:10,80:20,70:30 and 60:40 to identify the desirable blend ratio for burning of blended coal in thermal *** characteristics of all the coals were carried out using *** combustion parameters such as ignition temperature,peak temperature,burnout temperature,ignition index,burnout index,combustion performance index,rate and heat intensity index of the combustion process and activation energy were evaluated to analyse the combustion *** and theoretical analysis shows the blend ratio of 90:10 can be used in place of only thermal coal in utilities to reduce the fuel cost.

Electric discharge phenomenon in dielectric and electrolyte medium

Electric discharge is a common tool nowadays for machining of materials. It may be through a liquid medium or through air. Any metals, hard alloys, and nonmetals can be machined using the energy of electric discharge. In electric discharge machining (EDM), the discharge occurs between two electrodes through a liquid medium and it is applicable only for electrically con ducting materials and alloys. In electrochemical discharge machining (ECDM), the medium is an aqueous electrolyte and it is of two types. In the first type, the discharge occurs between two electrodes. One of the electrodes is the workpiece, and the other is the tool. In the second type, the discharge occurs between one electrode and an electrolyte. It is used for electrically nonconducting materials and the discharge energy is utilized maintaining the nonconducting workpiece in proximity of the discharge. All these electrical discharges are transient phenomena and do not result in a stable discharge in the form of arc. The output parameters depend on the discharge energy that requires precise control to maintain the accuracy of the machining. For micromachining, the control of the discharge is paramount both in terms of energy and pattern. Using various shaped tools, machining media with additives, different types of applied potentials, and supporting mechanical motions are some of the attempts made to improve the machining output. Optimization of these parameters for machining particular materials (or alloys) is a popular field of research. The present work is directed toward the investigation of discharge initiation and development by analyzing the cell current and discharge voltage relationship for both EDM and ECDM. Objective: The current study evaluated various new colchicine analogs for their anticancer activity and to study the primary mechanism of apoptosis and in vivo antitumor activity of the analogs with selective anticancer properties and minimal toxicity to normal ***: Sulforhodamine B(SRB) assay was used to screen various colchicine analogs for their in vitro cytotoxicity. The effect of N-[(7S)-1,2,3-trimethoxy-9-oxo-10-(pyrrolidine-1-yl)5,6,7,9-tetrahydrobenzo[a] heptalene-7-yl] acetamide(ⅢM-067) on clonogenicity, apoptotic induction, and invasiveness of A549 cells was determined using a clonogenic assay, scratch assay, and staining with 4’,6-diamidino-2-phenylindole(DAPI) and annexin V/propidium iodide. Mitochondrial membrane potential(MMP) and reactive oxygen species(ROS) levels were observed using fluorescence microscopy. Western blot analysis was used to quantify expression of proteins involved in apoptosis, cell cycle, and phosphatidylinositol 3-kinase(PI3K)/protein kinase B(AKT)/mammalian target of rapamycin(mTOR) signaling. Pharmacokinetic and in vivo efficacy studies against Ehrlich ascites carcinoma(EAC) and Ehrlich solid tumor models were conducted using Swiss albino ***: ⅢM-067 showed potent cytotoxicity and better selectivity than all other colchicine analogs screened in this study. The selective activity of ⅢM-067 toward A549 cells was higher among other cancer cell lines,with a selectivity index(SI) value of 2.28. ⅢM-067 demonstrated concentration-and time-dependent cytotoxicity against A549 cells with half-maximal inhibitory concentration values of 0.207, 0.150 and0.106 μmol/L at 24, 48 and 72 h, respectively. It also had reduced toxicity to normal cells(SI > 1) than the parent compound colchicine(SI = 1). ⅢM-067 reduced the clonogenic ability of A549 cells in a dose-dependent manner. ⅢM-067 enhanced ROS production from 24.6% at 0.05 μmol/L to 82.1% at 0.4 μmol/L and substantially decreased the MMP(100% in control to 5.6% at 0.4 μmol

In recent years,iron(Fe)based degradable metal is explored as an alternative to permanent fracture fixation *** the present work,copper(Cu)is added in Fe-Mn system to enhance the degradation rate and antimicrobial ***-Mn-x Cu(x=0.9,5 and 10 wt.%)alloys are prepared by the meltingcasting-forging *** analysis confirms austenite phase stabilization due to the presence of Mn and *** predicted by Thermo-Calc calculations,Cu rich phase precipitations are noticed along the austenite grain *** behaviours of Cu added Fe-Mn alloys are investigated through static immersion and electrochemical polarization where enhanced degradation is found for higher Cu added *** challenged against *** bacteria,the Fe-Mn-Cu alloy media extract shows a significant bactericidal effect compare to the base *** vitro cytocompatibility studies,as determined using MG63 and MC3T3-E1 cell lines,indicate increased cell density as a function of time for all the *** implanted in rabbit femur,the newly developed alloy does not show any kind of tissue necrosis around the *** osteogenesis and higher new bone formation are observed with Fe-Mn-10 Cu alloy as evident from micro-computed tomography(μ-CT)and fluorochrome labelling.

In-Situ Hydrothermal Synthesis of Bi–Bi2O2CO3 Heterojunction Photocatalyst with Enhanced Visible Light Photocatalytic Activity

Bismuth containing nanomaterials recently received increasing attention with respect to environmental applications because of their low cost, high stability and nontoxicity. In this work, Bi–Bi_2O_2CO_3 heterojunctions were fabricated by in-situ decoration of Bi nanoparticles on Bi_2O_2CO_3 nanosheets via a simple hydrothermal synthesis approach. X-ray diffraction(XRD), scanning electron microscopy(SEM), transmission electron microscopy(TEM) and high-resolution TEM(HRTEM) were used to confirm the morphology of the nanosheet-like heterostructure of the Bi–Bi_2O_2CO_3 composite. Detailed ultrafast electronic spectroscopy reveals that the in-situ decoration of Bi nanoparticles on Bi_2O_2CO_3 nanosheets exhibit a dramatically enhanced electron-hole pair separation rate, which results in an extraordinarily high photocatalytic activity for the degradation of a model organic dye, methylene blue(MB) under visible light illumination. Cycling experiments revealed a good photochemical stability of the Bi–Bi_2O_2CO_3 heterojunction under repeated irradiation. Photocurrent measurements further indicated that the heterojunction incredibly enhanced the charge generation and suppressed the charge recombination of photogenerated electron-hole pairs.

Effect of weathering on physico-chemical properties and combustion behavior of an Indian thermal coal

An open air stockpile of conical shape was formed with 3.5 m base diameter and 5 m height using 500 tons of -200 mm size ROM thermal coal on a concrete floor. Coal sample from the stockpile were collected at 30 days interval for analysis of various chemical properties like;proximate analysis, ultimate analysis, gross calorific value (GCV) and various combustion related properties such as ignition temperature, peak temperature, burnout temperature, maximum combustion rate, ignition index, burnout index, combustion performance index and combustion rate intensity index. Experimental results show that, due to weathering of coal fixed carbon decreased from 35.6% to 19.9%, elemental carb on (C) decreased from 46.6% to 28.6%, hydrogen (H) decreased from 3.3% to 2.9% and GCV decreased by up to 55% of original value during 330 days of storage of coal in an open atmosphere. Ash con tent of coal increased form 29.2% to 46.6% due to loss of combustibles. Sulfur (S) of coal was found to get increased from 0.33% to 1.08% during storage. The activation energy of coal combustion increased from 22 kJ/mol to 54 kJ/mol. Variation in combustion parameters signifies that weathering has significant negative impact on coal combustion properties as coal become difficult to ignite.

Relationship between proximate analysis parameters and combustion behaviour of high ash Indian coal

This work presents the analysis of combustion characteristics of high ash Indian coal(28%-40%)collected from different mines of Singaurali coalfield,*** the coal samples were characterized by proximate and gross calorific value *** performance of the coals were characterised using thermo-gravimetric analysis to identify the burning profile of individual *** combustion kinetic parameters such as ignition temperature,peak temperature and burnout temperature,ignition index and burnout index,combustion performance index plus rate and intensity index of combustion process,activation energy were determined to analyse the combustion behaviour of *** all these combustion properties were compared with the volatile matter,ash,fixed carbon and fuel ratio of each *** analysis shows that with increase in ash content,combustion performance initially increases and later ***,coal with(25±1.75)%volatile matter,20%-35%ash and fuel ratio 1.4-1.5 were found to be optimum for coal combustion.

Development of a Polyclonal Antibody-based AC-ELISA and Its Comparison with PCR for Diagnosis of Canine Parvovirus Infection

A polyclonal antibody-based antigen-capture ELISA (AC-ELISA) has been developed for detection of Canine parvovirus (CPV) antigens in faecal samples of dogs. The assay uses rabbit anti-CPV polyclonal antibody as the capture antibody, guinea pig anti-CPV polyclonal antibody as tracing antibody and anti-guinea pig HRPO conjugate as the detection system. The optimum dilution of the capture antibody and the tracing antibody capable of detecting the CPV-2 antigens was found to be 1:1 600 and 1:400, respectively, in the check-board titration. In this study, a total of 152 samples (129 faecal samples and 23 cell culture supernatant) were tested both by AC-ELISA and by polymerase chain reaction (PCR). Of the samples tested, 69 and 78 samples were found positive by AC-ELISA and PCR, respectively. The AC-ELISA had relative sensitivity, relative specificity and accuracy of 88.4%, 100.0% and 91.4% respectively. The analytical sensitivity of AC-ELISA was estimated to be 102.8 TCID50/mL whereas PCR sensitivity was 100.8 TCID50/mL. The AC-ELISA is a simple, quick and reliable method for screening large numbers of faecal samples of dogs suspected of CPV infection.
