Detection of blaNDM-1 and Genetic Relatedness in Clinical Isolates of Escherichia coli Producing Extended Spectrum β-Lactamase from Tertiary Care Centres in South India

Background: Extended spectrum β-lactamases (ESBL) producing E. coli co-producing other β-lactamases and exhibiting co-resistance to different antibiotic classes continue to emerge as a threat to clinical field. This study aimed to analyze the co-production of New Delhi metallo-β-lactamase-1 (blaNDM-1) in ESBL producing plasmid-bearing clinical isolates collected from two tertiary care centres in Kerala, South India, and to understand their genetic relatedness. Methods: Antibiotic resistance phenotypes of 44 clinical isolates were determined by disc-diffusion method. Plasmid-bearing isolates, detected by the alkaline-lysis method, which also tested positive for ESBL production, were screened for the presence of blaNDM-1 by polymerase chain reaction. Plasmid, random amplified polymorphic DNA profiles and blaNDM-1 sequence-based phylogenetic tree were analyzed to understand the genotypic similarities among the isolates. Results: Beta-lactam antibiotics, quinolones, cephalosporins, used in this study, and AZM were found to be ineffective against the isolates as significantly high number of isolates were resistant to these antibiotics (P < 0.01). Plasmid bearing isolates constituted 57% (n = 25), all of which were found to be ESBL producers. blaNDM-1 amplicons were noticed in four (16%) isolates and these DNA sequences showed homology between them and with similar sequences reported from other countries like Japan and Korea. Plasmid and RAPD profiles demonstrated that most of the isolates, including those harbouring blaNDM-1 shared genetic similarities as well as an apparent geographical distinctiveness. Conclusion: The predominance of ESBL production and the occurrence of blaNDM-1 in plasmid-bearing isolates observed in our study corroborate the worldwide drug-resistance scenario. This study thus warrants the need for constant surveillance in the face of sparse information available in Kerala State on the emerging drug resistance in clinical bacteria.

The present study focused on MexCD-OprJ efflux pump and its regulatory gene nfxB in multidrug resistant (MDR) clinical isolates of Pseudomonas aeruginosa collected from Kerala, South India. Semi-quantitative reverse transcription-PCR technique was employed to detect hyperexpression of the efflux pump gene, mexD. Amplicons from nfxB gene of isolates hyperexpressing the efflux pump were sequenced for mutational and phylogenetic analysis. Among 29 isolates of MDR P. aeruginosa, increased mexD transcription was detected in 10.3% of the isolates when compared with P. aeruginosa reference strain, PAO (MTCC-3541). Various synonymous and non-synonymous mutations in nfxB regulatory gene sequences were detected. Notably, mutations detected in the strains designate Pa6 and Pa7 have been found to be novel and are hitherto unreported in GenBank data base. The genetic divergence and homogeneity of the nfxB regulatory gene sequences of mexCD-oprJ operon were clearly apparent in the phylogram generated employing similar sequences retrieved from the public database.

The role of serratiopeptidase in the resolution of inflammation

Inflammation remains a key event during most of the diseases and physiological imbalance. Acute inflammation is an essential physiological event by immune system for a protective measure to remove cause of inflammation and failure of resolution lead to chronic inflammation. Over a period of time, a number of drugs mostly chemical have been deployed to combat acute and chronic inflammation. Recently, enzyme based anti-inflammatory drugs became popular over conventional chemical based drugs. Serratiopeptidase, a proteolytic enzyme from trypsin family, possesses tremendous scope in combating inflammation. Serine protease possesses a higher affinity for cyclooxygenase(COX-Ⅰ and COX-Ⅱ), a key enzyme associated with production of different inflammatory mediators including interleukins(IL),prostaglandins(PGs) and thromboxane(TXs) etc. Currently, arthritis, sinusitis, bronchitis,fibrocystic breast disease, and carpal tunnel syndrome, etc. are the leading inflammatory disorders that affected the entire the globe. In order to conquer inflammation, both acute and chronic world, physician mostly relies on conventional drugs. The most common drugs to combat acute inflammation are Nonsteroidal anti-inflammatory drugs(NSAIDs) alone and or in combination with other drugs. However, during chronic inflammation, NSAIDs are often used with steroidal drugs such as autoimmune disorders. These drugs possess several limitations such as side effects, ADR, etc. In order to overcome these limitations and complications, enzyme based drugs(anti-inflammatory) emerged, and aim for a new high since the last decade. Serine protease, the largest proteolytic family has been reported for several therapeutic applications, including anti-inflammatory. Serratiopeptidase is a leading enzyme which has a very long history in medical as an effective anti-inflammatory drug. Glucose 6 phosphate dehydrogenase(G6PD)is a key and rate limiting enzyme in the pentose phosphate pathway(PPP).The physiological significance of enzyme is providing reduced energy to specific cells like erythrocyte by maintaining co-enzyme nicotinamide adenine dinucleotide phosphate(NADPH).There are preponderance research findings that demonstrate the enzyme(G6PD)role in the energy balance,and it is associated with bloodrelated diseases and disorders,primarily the anemia resulted from G6PD *** Xlinked genetic deficiency of G6PD and associated non-immune hemolytic anemia have been studied widely across the *** advancement in biology,more precisely neuroscience has revealed that G6PD is centrally involved in many neurological and neurodegenerative *** neuroprotective role of the enzyme(G6PD)has also been established,as well as the potential of G6PD in oxidative damage and the Reactive Oxygen Species(ROS)produced in cerebral *** G6PD deficiency remains a global health issue,however,a paradigm shift in research focusing the potential of the enzyme in neurological and neurodegenerative disorders will surely open a new avenue in diagnostics and enzyme ***,in this study,more emphasis was made on exploring the role of G6PD in neurological and inflammatory disorders as well as non-immune hemolytic anemia,thus providing diagnostic and therapeutic opportunities.

Intelligent Multilevel Node Authentication in Mobile Computing Using Clone Node

Nodes in a mobile computing system are vulnerable to clone attacks due to their *** such attacks,an adversary accesses a few network nodes,generates replication,then inserts this replication into the network,potentially resulting in numerous internal network *** existing techniques use a central base station,which introduces several difficulties into the system due to the network’s reliance on a single point,while other ways generate more overhead while jeopardising network *** this research,an intelligent double hashing-based clone node identification scheme was used,which reduces communication and memory costs while performing the clone detection *** approach works in two stages:in the first,the network is deployed using an intelligent double hashing procedure to avoid any network collisions and then in the second,the clone node identification procedure searches for any clone node in the *** first phase verifies the node prior to network deployment,and then,whenever a node wants to interact,it executes the second level of ***-to-end delay,which is bound to increase owing to the injection of clone nodes,and packet loss,which is reduced by the double hashing technique,were used to evaluate the performance of the aforementioned approach.

Bearing ratio and leachate analysis of pond ash stabilized with lime and lime sludge

Recycling of industrial waste is one of the effective ways to overcome their disposal problem. Ash produced by thermal power plants and lime sludge produced by paper mills require huge disposal land and may create environmental problems such as dusting and leaching of harmful heavy metals. Stabilization of the ash can improve its engineering properties and address the environmental problems. This paper reports the laboratory test results of a Class F pond ash stabilized with lime(2%, 4%, 6% and 8% by weight)alone and in combination with lime sludge(5%, 10% and 15% by weight). The X-ray diffraction(XRD) and scanning electron micrograph(SEM) tests were also performed to identify the possible formation of crystalline phases after stabilization. The effects of lime sludge on the unsoaked and soaked bearing ratios of pond ash with different lime contents, after 7 d, 28 d and 45 d of curing, were observed. Test results indicated that the bearing ratio increased considerably up to a 4% lime content which can be taken as the optimum lime content. Further increase in lime content increased bearing ratio gradually but at a slower rate. The effect of lime sludge was more pronounced at the optimum lime content,particularly at a low curing period. Lime sludge improved the bearing ratio in soaked condition significantly. Leachate analysis of stabilized ash was performed using toxicity characteristic leaching procedure(TCLP-1311) method. The concentrations of toxic elements Zn, Cu, Cd, Ni and Cr in the stabilized mixes were lower than those in the unstabilized waste. The results indicated that the pond ash-lime-lime sludge mixes have potential application as road subbase material.

In this paper,a nonlinear mathematical model is proposed and analyzed to study the effects of population pressure augmented industrialization on the survival of competing species dependent on *** is assumed that the growths of competing species are logistic and carrying capacities increase with increase in the density of resource ***,it is assumed that the resource biomass too is growing logistically in the envi-ronment and its carrying capacity decreases with the increase in densities of competing species and *** growth rate of population pressure is assumed to be proportional to the densities of competing *** of all equilibria and con-ditions which influence the permanence of the system are carried out using theory of differential *** simulations are performed to accomplish our analytical *** is shown that the equilibrium density of resource biomass decreases as(i)the growth rate coefficient of population pressure increases(ii)the growth rate coeffi-cient of industrialization due to population pressure increases and(iii)the growth rate coefficient of industrialization due to resource biomass *** is found that the competitive outcome alters with increase in the growth rate coefficient of population *** in the equilibrium densities of competing species is also noted with increase in the growth rate coefficient of industrialization due to resource biomass.

The present paper demonstrates the applicability of finite element method of weighted residuals to study the effects of precipitation scavenging through raindrops on the steady-state dispersion of air pollutants in the atmosphere under realistic variable wind and diffusivity ***,the washout of pollutants by rain droplets is considered and the whole analysis for the pollutant concentration is carried out in two phases *** phase and droplet *** model solutions are obtained by using local basis and asymmetric quadratic weighting functions which can provide a promising alternative to the standard Galerkin formulation for problems with advective and diffusive *** results of the model reveal that the precipitation scavenging by rain is quite effective in cleaning the polluted *** the need of pollutant concentration in rain drops regarding acid precipitation(or acid rain),the concentration of the absorbed pollutant in the droplet phase are also ***,the present study provides an understanding on the concentration distribution of pollutant in gaseous and droplet phases,under the effects of variable wind and diffusivity profiles.

Background The internet is an integral part of everyone’s *** going adolescents are highly vulnerable to the misuse of the *** To estimate the pooled prevalence of internet addiction(IA)among college students in *** Literature databases(PubMed,Web of Science,Scopus,EMBASE,PsycINFO and Google Scholar)were searched for studies assessing IA using the Young Internet Addiction Test(Y-IAT)among adolescents from India,published in the English language up to December *** included studies from 2010 to 2020 as this is the marked era of momentum in wireless internet connectivity in *** methodological quality of each study was scored,and data were extracted from the published *** prevalence was estimated using the fixed-effects *** bias was evaluated using Egger’s test and visual inspection of the symmetry in funnel *** Fifty studies conducted in 19 states of India estimated the prevalence of IA and the overall prevalence of IA as 19.9%(95%CI:19.3%to 20.5%)and 40.7%(95%CI:38.7%to 42.8%)based on the Y-IAT cut-off scores of 50 and 40,*** estimated prevalence of severe IA was significantly higher in the Y-IAT cut-off points of 70 than 80(12.7%(95%CI:11.2%to 14.3%)vs 4.6%(95%CI:4.1%to 5.2%)).The sampling method and quality of included studies had a significant effect on the estimation of prevalence in which studies using non-probability sampling and low risk of bias(total quality score≥7)reported lower *** overall quality of evidence was rated as‘moderate’based on the Grading of Recommendations Assessment,Development and Evaluation *** Our nationally representative data suggest that about 20%to 40%of college students in India are at risk for *** is a need for further research in the reconsideration of Y-IAT cut-off points among Indian college students.
