Cloning ＆ sequence identification of Hsp27 gene and expression analysis of the protein on thermal stress in Lucilia cuprina

Hsp27, a highly conserved small molecular weight heat shock.protein, is widely k.own to be developmentally regulated and heat inducible. Its role in thermotolerance is also implicated. This study is a sequel of our earlier studies to understand the molecular organization of heat shock.genes/proteins and their role in development and thermal adaptation in a sheep pest, Lucilia cuprina （blowfly）, which exhibits unusually high adaptability to a variety of environmental stresses, including heat and chemicals. In this report our aim was to understand the evolutionary relationship of Lucilia hsp27 gene/protein with those of other species and its role in thermal adaptation. We sequence characterized the Lchsp27 gene （coding region） and analyzed its expression in various larval and adult tissues under normal as well as heat shock.conditions. The nucleotide sequence analysis of 678 bps long-coding region of Lchsp27 exhibited closest evolutionary proximity with Drosophila （90.09%）, which belongs to the same order, Diptera. Heat shock.caused significant en- hancement in the expression ofLchsp27 gene in all the larval and adult tissues examined, however, in a tissue specific manner. Significantly, in Malpighian tubules, while the heatinduced level ofhsp27 transcript （mRNA） appeared increased as compared to control, the protein level remained unaltered and nuclear localized. We infer that Lchsp27 may have significant role in the maintenance of cellular homeostasis, particularly, during summer months, when the fly remains exposed to high heat in its natural habitat.

Totally Anisotropic Cosmological Models with Bulk.Viscosity for Variable G and Λ

Einstein's field equations with variable gravitational and cosmological constants are considered in the presence of bulk.viscous fluid for the totally anisotropic Bianchi type II space-time in such a way as to preserve the energy momentum tensor. We have presented solutions of field equations which represent expanding, shearing and non-rotating cosmological models of the universe. The physical behaviours of the models are discussed .We observe that the results obtained match with recent observations of SNIa.

Colorectal cancer(CRC)is k.own to harbor considerable heterogeneity.1 Consequently,it could be hypothesized that similar-appearing tumors might exhibit substantial genetic differences while diverse-appearing tumors may have a similar genetic landscape.2 Due to these differences at the molecular level,they behave or respond differently to therapies as well.CRC progression is a multistep process and involves the accumulation of substantial genetic and epigenetic events in a stage-dependent manner.

A Clinical Data Analysis Based Diagnostic Systems for Heart Disease Prediction Using Ensemble Method

The correct diagnosis of heart disease can save lives,while the incorrect diagnosis can be lethal.The UCI machine learning heart disease dataset compares the results and analyses of various machine learning approaches,including deep learning.We used a dataset with 13 primary characteristics to carry out the research.Support vector machine and logistic regression algorithms are used to process the datasets,and the latter displays the highest accuracy in predicting coronary disease.Python programming is used to process the datasets.Multiple research initiatives have used machine learning to speed up the healthcare sector.We also used conventional machine learning approaches in our investigation to uncover the link. between the numerous features available in the dataset and then used them effectively in anticipation of heart infection risk..Using the accuracy and confusion matrix has resulted in some favorable outcomes.To get the best results,the dataset contains certain unnecessary features that are dealt with using isolation logistic regression and Support Vector Machine(SVM)classification.

Molecular subtypes of colorectal cancer: An emerging therapeutic opportunity for personalized medicine

Molecular subtypes-based therapies offer new potential framework.for desired and precise outcome in clinical settings.Current treatment strategies in colorectal cancer are largely‘one drug fit all’model for patients that display same pathological conditions.However,CRC is a very heterogenous set ofmalignancy that does not support for above criteria.Each subtype displays different pathological and genetic signatures.Based on these features,therapeutic stratification for individual patients may be designed,which may ultimately lead to improved therapeutic outcomes.In this comprehensive review,we have attempted to briefly outline major CRC pathways.A detailed overview of molecular subtypes and their clinical significance has been discussed.Present and futuremethods,governing CRC subtyping in the era of personalized therapywith a special emphasis on CMS subtypes of CRC has been reviewed.Together,discovery and validation of new CRC patient stratification methods,screening for novel therapeutic targets,and enhanced diagnosis of CRC may improve the treatment outcome.

SEIHCRD Model for COVID-19 Spread Scenarios,Disease Predictions and Estimates the Basic Reproduction Number,Case Fatality Rate,Hospital,and ICU Beds Requirement

We have proposed a new mathematical method,the SEIHCRD model,which has an excellent potential to predict the incidence of COVID-19 diseases.Our proposed SEIHCRD model is an extension of the SEIR model.Three-compartments have added death,hospitalized,and critical,which improves the basic understanding of disease spread and results.We have studiedCOVID-19 cases of six countries,where the impact of this disease in the highest are Brazil,India,Italy,Spain,the United k.ngdom,and the United States.After estimating model parameters based on available clinical data,the modelwill propagate and forecast dynamic evolution.Themodel calculates the Basic reproduction number over time using logistic regression and the Case fatality rate based on the selected countries’age-category scenario.Themodel calculates two types of Case fatality rate one is CFR daily,and the other is total CFR.The proposed model estimates the approximate time when the disease is at its peak.and the approximate time when death cases rarely occur and calculate how much hospital beds and ICU beds will be needed in the peak.days of infection.The SEIHCRD model outperforms the classic ARXmodel and the ARIMA model.RMSE,MAPE,andRsquaredmatrices are used to evaluate results and are graphically represented using Taylor and Target diagrams.The result shows RMSE has improved by 56%–74%,and MAPE has a 53%–89%improvement in prediction accuracy.

Synergism of co-delivered nanosized antioxidants displayed enhanced anticancer efficacy in human colon cancer cell lines

Combination of chemopreventive and/or therapeutic agents is the imminent smart approach to cope upwith cancer because it may act on multiple targets through different pathways. In the present study, wehave synthesized multiple chemopreventive and/or therapeutic agents (Curcumin, Quercetin andAspirin) loaded nanoparticles by simple cation-anion interaction among the amine groups of chitosan(CS) and phosphate groups of sodium hexametaphosphate (SHMP). These nanosized bioactive materials(CS-SHMP-CQA-NPs) were well characterized and found most effective in colon cancer cell line (HCT-116)compared to other cancer cell lines. Triplex chemopreventive and/or therapeutic agents-loaded NPs weresynergistically inducing apoptosis in HCT-116 cells compared to two-chemopreventive agents-loadedNPs as evident by an increase in sub-G1 cells (percent), and chromatin condensation along with thedecrease in mitochondrial membrane potential (MMP). Interestingly, ChoueTalalay analysis revealedthat CS-SHMP-CQA-NPs showed strong synergistic effect in its all doses. Thus, our study demonstratesthat nanoparticles based bioactive materials significantly inhibit the growth of HCT-116 cells and thuscould be a promising approach for colon cancer chemoprevention.

Pancreatic fluid collections(PFCs)result from injury to the pancreas from acute or chronic pancreatitis,surgery,or trauma.Management of these collections has evolved over the last 2 decades.The choice of interventions includes percu-taneous,endoscopic,minimally invasive surgery,or a combined approach.Endoscopic drainage is the drainage of PFCs by creating an artificial communi-cation between the collection and gastrointestinal lumen that is maintained by placing a stent across the fistulous tract.In this editorial,we endeavored to update the current status of endoscopic ultrasound-guided drainage of PFCs.

The control of Leishmania infection relies primarily on chemotherapy till date.Resistance to pentavalent antimonials,which have been the recommended drugs to treat cutaneous and visceral leishmaniasis,is now widespread in Indian subcontinents.New drug formulations lik. amphotericin B,its lipid formulations,and miltefosine have shown great efGcacy to treat leishmaniasis but their high cost and therapeutic complications limit their usefulness.In addition, irregular and inappropriate uses of these second line drugs in endemic regions lik. state of Bihar, India threaten resistance development in the parasite.In context to the limited drug options and unavailability of either preventive or prophylactic candidates,there is a pressing need to develop true antileishmanial drugs to reduce the disease burden of this debilitating endemic disease.Notwithstanding significant progress of leishmanial research during last few decades, identification and characterization of novel drugs and drug targets are far from satisfactory.This review will initially describe current drug regimens and later will provide an overview on few important biochemical and enzymatic machineries that could be utilized as putative drug targets for generation of true antileishmanial drugs.

Study of Non-Pharmacological Interventions on COVID-19 Spread

COVID-19 disease has emerged as one of the life threatening threat to the society.A novel beta coronavirus causes it.It began as unidentified pneumonia of unk.own etiology in Wuhan City,Hubei province in China emerged in December 2019.No vaccine has been produced till now.Mathematical models are used to study the impact of different measures used to decrease pandemic.Mathematical models have been designed to estimate the numbers of spreaders in different scenarios in the present manuscript.In the present manuscript,three different mathematical models have been proposed with different scenarios,such as screening,quarantine,andNPIs,to estimate the number of virus spreaders.The analysis shows that the numbers of COVID-19 patients will be more without screening the peoples coming from other countries.Since every people suffering fromCOVID-19 disease are spreaders.The screening and quarantine with NPIs have been implemented to study their impact on the spreaders.It has been found that NPI measures can reduce the number of spreaders.The NPI measures reduce the spread function’s growth and provide decision mak.rs more time to prepare with in dealing with the disease.
