Evaluation of cotton germplasm for morphological and biochemical host plant UPdates resistance traits against sucking insect pests complex

Background:Sucking insect pests cause severe damage to cotton crop *** development of insect resistant cotton cultivars is one of the most effective measures in curtailing the yield *** the role of morphological and biochemical host plant resista nee(HPR)traits in plant defense,12 cotton genotypes/varieties were evaluated for leaf area,leaf glanding,total soluble sugars,total soluble proteins,total phenolics,tannin and total flavonoids against fluctuating populations of whitefly,thrips and jassid under field ***:The population of these insects fluctuated during the growing seas on and remained above threshold level(whitefly>5,thrips>(8-10)f or jassid>1 per leaf)during late June and early *** and negative association of whitefly(r=-0.825)and jassid(r=-0.929)with seed cotton yield was *** population of insects were the highest in Glandless-1 followed by NIA-82 and ***-Kiran followed by NI AB-878 and Sadori were the most resistant,with the mean population of 1.41,1.60,1.66(whitefly);2.24,232,2.53(thrips)and 037,0.31,036(jassid),*** resistant variety NIAB-Kiran showed less soluble sugars(8.54 mg.g^(-1)),soluble proteins(27.11 mg.g^(-1))and more phenolic(36.56 mg.g^(-1))and flavonoids(13.10mg.g^(-1))as compared with the susceptible check ***,all insect populations were positively correlated with total soluble sugars and *** populations exhibited negative response to leaf gossypol glands,total phenolics,tannins and *** thrips and jassid populations had a significant and negative correlation with these four biochemical HPR ***:The ide ntified resistant resources and HPR traits can be deployed against sucking in sect pests'complex in future breeding programs of developing insect resistant cotton varieties.

Pleural Fluid Alkaline Phosphate Levels to Differentiate between Tuberculosis and Malignant Pleural Effusion a Tertiary Care Experience

Introduction: Pleural effusion (PF) is a common clinical presentation in several diseases. Tuberculosis is one of the most frequent causes of exudative pleural effusions in immunocompetent patients. Tuberculosis is the leading cause of morbidity and mortality from an infectious disease in developing countries. Pakistan is ranked fifth in the world in terms of tuberculosis high-burden countries. Various pleural fluid parameters have been used to identify the cause of pleural effusion. It has been discovered that tuberculous pleural effusions had a greater alkaline phosphatase (ALP) concentration than transudative effusions. This study used pleural fluid alkaline phosphatase levels to distinguish between tuberculous pleural effusion and malignant pleural effusion because there is little information from tuberculosis-high burden nations like Pakistan. Study Design: A descriptive cross-sectional study conducted at the Jinnah Postgraduate Medical Center in Karachi between October 2016 and October 2017. Material and Methods: The study comprised all patients who were admitted to the department of chest medicine at Jinnah post graduate medical centre (JPMC) of either gender between the ages of 18 and 70 who had exudative lymphocytic pleural effusions lasting two weeks or more included in the study. Non probability consecutive sampling was used to collect data. Patients who have tonsillitis, pharyngitis, pneumonia, asthma, Chronic obstructive pulmonary disease (COPD), or a history of hemoptysis, Bleeding disorders like, platelet function disorder, thrombocytopenia, Liver cirrhosis and Pregnant women were excluded. Parents’ informed consent was obtained after being informed of the study’s protocol, hazards, and advantages. Each patient had their level of pleural fluid alkaline phosphate (PALP) assessed. Faithful segregation of mitotic chromosomes requires bi-orientation of sister chromatids, which relies on the sensing of correct attachments between spindle microtubules and kinetochores. Although the mechanisms underlying PLK1 activation have been extensively studied, the regulatory mechanisms that couple PLK1 activity to accurate chromosome segregation are not well understood. In particular, PLK1 is implicated in stabilizing kinetochore–microtubule attachments, but how kinetochore PLK1 activity is regulated to avoid hyperstabilized kinetochore–microtubules in mitosis remains elusive. Here, we show that kinetochore PLK1 kinase activity is modulated by SET7/9 via lysine methylation during early mitosis. The SET7/9-elicited dimethylation occurs at the Lys191 of PLK1, which tunes down its activity by limiting ATP utilization. Overexpression of the non-methylatable PLK1 mutant or chemical inhibition of SET7/9 methyltransferase activity resulted in mitotic arrest due to destabilized kinetochore–microtubule attachments. These data suggest that kinetochore PLK1 is essential for stable kinetochore–microtubule attachments and methylation by SET7/9 promotes dynamic kinetochore–microtubule attachments for accurate error correction. Our findings define a novel homeostatic regulation at the kinetochore that integrates protein phosphorylation and methylation with accurate chromosome segregation for maintenance of genomic stability.

In this research,green synthesized magnesium oxide nanoparticles(MgO NPs)from lemon fruit extracts and their fungicidal potential was evaluated against Alternaria dauci infection on carrot(Daucus carota L.)under greenhouse *** scanning and transmission electron microscopy(SEM and TEM)and ultra-violet(UV)visible spectroscopy were used to validate and characterize MgO *** crystalline nature of MgONPs was determined using selected area electron diffraction(SAED).MgO NPs triggered substantial antifungal activity against *** when exposed to 50 and 100 mg L^(–1)concentrations but the higher antifungal potential was noticed in 100 mg L^(–1)under invitro *** fungal inoculated plants,a marked decrease in growth,photosynthetic pigments,and an increase in phenol,proline contents,and defense-related enzymes of carrot were seen over control(distilled water).However,foliar application of MgO NPs at 50 and 100 mg L^(–1)resulted in significant improvement of plant growth,photosynthetic pigments,phenol and proline contents,and defense enzymes activity of carrots with and without *** *** of MgO NPs at 100 mg L^(–1)had more plant length(17.11%),shoot dry weight(34.38%),plant fresh weight(20.46%),and root dry weight(49.09%)in carrots when challenged with *** over inoculated *** leaf blight indices and percent disease severity were also reduced in *** inoculated plants when sprayed with MgO *** non-bonding interactions of Alternaria genus protein with nanoparticles were studied using molecular docking.

Phytochemical investigations of the ethanolic extract of leaves of Clutia lanceolata(Family:Euphorbiaceae)resulted in the isolation of four compounds viz.3,4-dihydroxy-2-methylbenzoic acid(1),2,20-dihydroxy-1,10-binaphthyl(2),1,3,8-trihydroxy-6-methylanthracene-9,10-dione(3)and 5-hydroxy-1,7-bis(4-hydroxy-3-methoxyphenyl)hepta-1,4,6-trien-3-one(4).Although all the isolated compounds were known but this was the first report from this plant *** structures were established on the basis of chemical and physical evidences *** analysis,FT-IR,1 H-NMR,13CNMR and mass spectral *** of compound 2 and 4 was further authenticated by single-crystal X-ray analysis and density functional theory *** isolated compounds(1–4)were screened for AChE enzyme inhibition assay in which compound 3 and 4 were found to be more potent AChE *** docking study of potent AChE inhibitor was performed to find the probable binding mode of the compounds into the active site of ***,the isolated compounds were also screened for in vivo cytotoxicity by brine shrimp lethality assay.

Intervarietal Variation in Proximate Composition and Antioxidant Potential of Dry Peas （Pisum sativum L）

The present research work was carried out to investigate the proximate composition and antioxidant capacities of newly invented varieties of dry peas. The proximate composition in terms of moisture （7.00-8.50%）, ash （3.85-4.97%）, fiber （3.8-12.3%） and protein （17.3-18.8%） contents varied significantly with respect to various cultivars. Total phenolic contents （TPC）, total flavonoids contents （TFC）, percentage inhibition of peroxidation of linoleic acid system, reducing power and DPPH free radical scavenging capacity of different dry peas extracts were found in range of 0.651-0.684 g/100g of GAE, 0.021-0.041 g/100g of CE, 0.713-0.895, 40.051-84.608%, 18.097-24.591 mg/mL, respectively. The amount of TFC, and percent inhibition of per oxidation in linoleic system of different dry peas extracts varied significantly among various cultivars and solvents. From the results of present investigations it is reasonable to say that 80% methanolic extracts of dry peas have exhibited varying degree of antioxidant activity.
