Rethinking neurodegenerative diseases:neurometabolic concept linking lipid oxidation to diseases in the central nervous system

Currently,there is a lack of effective medicines capable of halting or reve rsing the progression of neurodegenerative disorde rs,including amyotrophic lateral sclerosis,Parkinson s disease,multiple sclerosis,or Alzheimer s *** the unmet medical need,it is necessary to reevaluate the existing para digms of how to to rget these *** considering neurodegenerative diseases from a systemic neurometabolic perspective,it becomes possible to explain the shared pathological *** innovative approach presented in this paper draws upon exte nsive research conducted by the authors and researchers *** this review,we highlight the importance of metabolic mitochondrial dysfunction in the context of neurodegenerative *** provide an overview of the risk factors associated with developing neurodegenerative disorders,including genetic,epigenetic,and environmental fa ***,we examine pathological mechanisms implicated in these diseases such as oxidative stress,accumulation of misfolded proteins,inflammation,demyelination,death of neurons,insulin resistance,dysbiosis,and neurotransmitter ***,we outline a proposal for the restoration of mitochondrial metabolism,a crucial aspect that may hold the key to facilitating curative therapeutic interventions for neurodegenerative disorders in forthcoming advancements.

Fitness components of Drosophila melanogaster developed on a standard laboratory diet or a typical natural food source

Drosophila melanogaster is often used as a model organism in evolution- ary biology and ecophysiology to study evolutionary processes and their physiological mechanisms. Diets used to feed Drosophila cultures differ between laboratories and are often nutritious and distinct from food sources in the natural habitat, Here we rear D. melanogaster on a standard diet used in our laboratory and a field diet composed of decomposing apples collected in the field. Flies developed on these two diet compositions are tested for heat, cold, desiccation, and starvation resistance as well as developmental time, dry body mass and fat percentage. The nutritional compositions of the standard and field diets were analyzed, and discussed in relation to the phenotypic observations. Results showed marked differences in phenotype of flies from the two types of diets. Flies reared on the field diet are more starvation resistant and they are smaller, leaner, and have lower heat resistance compared to flies reared on the standard diet. Sex specific effects of diet type are observed for several of the investigated traits and the strong sexual dimorphism usually observed in desiccation resistance in D. melanogaster disappeared when rearing the flies on the field diet. Based on our results we conclude that care should be taken in extrapolating results from one type of diet to another and especially from laboratory to field diets.

Novel immune modulatory vaccines targeting TGFβ

Immune modulatory vaccines (IMVs) are novel therapies designed to activate anti-regulatory T cells (anti-Tregs) [1]. Anti-Tregs recognize HLA-restricted antigens on the surface of regulatory immune cells derived from tumor microenvironment antigens (TMAs) [2], such as IDO [3], PD-L1 [4], and TGFβ [5, 6]. Having established that specific T cells recognize target cells in a TGFβ-dependent manner, we reported that such TGFβ-specific T cells are associated with clinical benefit and improved survival after combined treatment with radiotherapy (RT) and immune checkpoint inhibitors (ICIs) in patients with pancreatic cancer (PC) [7]. Additionally, we found that a TGFβ-based IMV impacted tumor growth in murine models of PC via immune modulation of the tumor microenvironment (TME) [8]. These data suggest that combining a TGFβ-based IMV with RT/ICI therapy will be beneficial for patients with PC.

Neural Network Based Feasible Region Approximation Model for Optimal Operation of Integrated Electricity and Heating System

This paper proposes a neural network based feasible region approximation model of a district heating system(DHS),and it is intended to be used for optimal operation of integrated electricity and heating system(IEHS)considering privacy *** this model,a neural network is trained to approximate the feasible region of the DHS operation and then is reformulated as a set of mixed-integer linear *** on the received approximation models of DHSs and detailed electricity system model,the electricity operator conducts centralized optimization,and then sends specific heating generation plans back to corresponding heating ***,subsequent optimization is formulated for each DHS to obtain detailed operation strategy based on received heating generation *** this scheme,optimization of the IEHS could be achieved and privacy protection requirement is satisfied since the feasible region approximation model does not contain detailed system *** studies conducted on a small-scale system demonstrate accuracy of the proposed strategy and a large-scale system verify its application possibility.

Ecological speciation in Darwin＇s finches： Parsing the effects of magic traits

Many recent studies of ecological speciation have focused on ＂magic trait＂ scenarios, in which divergent selection on viability traits leads inextricably to corresponding divergence in mechanisms, especially mate recognition systems, that facilitate assortative mating. Speciation however may also proceed via other scenarios, such as when populations experience directly se- lected or random divergence in mate recognition systems. The relative contributions of magic trait versus other scenarios for speciation remain virtually unexplored. The present study aims to test the relative contribution of the magic trait scenario in the divergence of populations of the medium ground finch Geospiza fortis of Santa Cruz Island, Galapagos. First, we assess differ- ences in G. fortis song between a northern population （Borrero Bay） and a southeastern population （El Garrapatero）, differences that we propose （along with other within-island geographic song variations） have arisen via scenarios that do not involve a magic trait scenario. Pairwise comparisons of raw and composite （PC） song parameters, as well as discriminant functions analyses, re- veal significant patterns of song divergence between sites. Second, we test the ability of territorial males at Borrero Bay to dis- criminate songs from the two sites. We find that G. fortis males can discriminate within-island song variants, responding more strongly to local than to ＂foreign＂ songs, along 3 raw and 1 composite response measures. Third, we compare these findings to prior data sets on song divergence and discrimination in Santa Cruz G. fortis. These comparisons suggest that song divergence and discrimination are shaped less strongly by geographic sources than by morphological （beak-related） sources. We thus argue that interpopulation song divergence and discrimination, fundamental elements of assortative mating in Darwin＇s finches, can be fos- tered in early stages of divergence under magic trait as well as alternative scenar

An Accelerated Method for Simulating Population Dynamics

We present an accelerated method for stochastically simulating the dynamics of heterogeneous cell *** algorithm combines a Monte Carlo approach for simulating the biochemical kinetics in single cells with a constant-number Monte Carlo method for simulating the reproductive fitness and the statistical characteristics of growing cell *** benchmark accuracy and performance,we compare simulation results with those generated from a previously validated population dynamics *** comparison demonstrates that the accelerated method accurately simulates population dynamics with significant reductions in runtime under commonly invoked steady-state and symmetric cell division *** the increasing complexity of cell population models,the method is an important addition to the arsenal of existing algorithms for simulating cellular and population dynamics that enables efficient,coarse-grained exploration of parameter space.

Understanding the effects of structured self-assessment in directed,self-regulated simulation-based training of mastoidectomy:A mixed methods study

Objective:Self-directed training represents a challenge in simulation-based training as low cognitive effort can occur when learners overrate their own level of *** study aims to explore the mechanisms underlying the positive effects of a structured self-assessment intervention during simulation-based training of ***:A prospective,educational cohort study of a novice training program consisting of directed,self-regulated learning with distributed practice(5x3 procedures)in a virtual reality temporal bone *** intervention consisted of structured self-assessment after each procedure using a rating form supported by small ***-structured telephone interviews upon completion of training were conducted with 13 out of 15 *** were analysed using directed content analysis and triangulated with quantitative data on secondary task reaction time for cognitive load estimation and participants’self-assessment ***:Six major themes were identified in the interviews:goal-directed behaviour,use of learning supports for scaffolding of the training,cognitive engagement,motivation from self-assessment,selfassessment bias,and feedback on self-assessment(validation).Participants seemed to self-regulate their learning by forming individual sub-goals and strategies within the overall goal of the *** scaffolded their learning through the available learning ***,structured self-assessment was reported to increase the participants’cognitive engagement,which was further supported by a quantitative increase in cognitive ***:Structured self-assessment in simulation-based surgical training of mastoidectomy seems to promote cognitive engagement and motivation in the learning task and to facilitate self-regulated learning.
