Signaling interactions among neurons impact cell fitness and death in Alzheimer's disease

The pathology of Alzheimer’s disease involves a long preclinical period,where the characteristic clinical symptoms of the changes in the brain are undetectable.During the preclinical period,homeostatic mechanisms may help prevent widespread cell death.Evidence has pointed towards selective cell death of diseased neurons playing a potentially protective role.As the disease progresses,dysregulation of signaling pathways that govern cell death contributes to neurodegeneration.Aberrant activation of the c-Jun N-terminal kinase pathway has been established in human and animal models of Alzheimer’s disease caused by amyloid-beta 42-or tau-mediated neurodegeneration.Clonal mosaic studies in Drosophila that examine amyloid-beta 42 in a subset of neurons suggest complex interplay between amyloid-beta 42-expressing and wild-type cells.This review examines the role of c-Jun N-terminal kinase signaling in the context of cell competition and short-range signaling interactions between amyloid-beta 42-expressing and wild-type neurons.Cell competition is a conserved phenomenon regulating tissue integrity by assessing the fitness of cells relative to their neighbors and eliminating suboptimal cells.Somatic clones of amyloid-beta 42 that juxtapose genetically distinct neuronal cell populations show promise for studying neurodegeneration.Generating genetic mosaics with labeled clones of amyloid-beta 42-or tau-expressing and wild-type neurons will allow us to understand how short-range signaling alterations trigger cell death in neurons and thereby contribute to the progression of Alzheimer’s disease.These approaches have the potential to uncover biomarkers for early Alzheimer’s disease detection and new therapeutic targets for intervention.

Desulfurization of dibenzothiophene(DBT) by a novel strain Lysinibacillus sphaericus DMT-7 isolated from diesel contaminated soil

A new bacterial strain DMT-7 capable of selectively desulfurizing dibenzothiophene（DBT） was isolated from diesel contaminated soil.The DMT-7 was characterized and identified as Lysinibacillus sphaericus DMT-7（NCBI GenBank Accession No.GQ496620） using 16S rDNA gene sequence analysis.The desulfurized product of DBT,2-hydroxybiphenyl（2HBP）,was identified and confirmed by high performance liquid chromatography analysis and gas chromatography-mass spectroscopy analysis respectively.The desulfurization kinetics revealed that DMT-7 started desulfurization of DBT into 2HBP after the lag phase of 24 hr,exponentially increasing the accumulation of 2HBP up to 15 days leading to approximately 60% desulfurization of the DBT.However,further growth resulted into DBT degradation.The induced culture of DMT-7 showed shorter lag phase of 6 hr and early onset of stationary phase within 10 days for desulfurization as compared to that of non-induced culture clearly indicating the inducibility of the desulfurization pathway of DMT-7.In addition,Lysinibacillus sphaericus DMT-7 also possess the ability to utilize broad range of substrates as sole source of sulfur such as benzothiophene,3,4-benzo DBT,4,6-dimethyl DBT,and 4,6-dibutyl DBT.Therefore,Lysinibacillus sphaericus DMT-7 could serve as model system for efficient biodesulfurization of diesel and petrol.

Enhanced Deep Learning for Detecting Suspicious Fall Event in Video Data

Suspicious fall events are particularly significant hazards for the safety of patients and elders.Recently,suspicious fall event detection has become a robust research case in real-time monitoring.This paper aims to detect suspicious fall events during video monitoring of multiple people in different moving back-grounds in an indoor environment;it is further proposed to use a deep learning method known as Long Short Term Memory(LSTM)by introducing visual atten-tion-guided mechanism along with a bi-directional LSTM model.This method contributes essential information on the temporal and spatial locations of‘suspi-cious fall’events in learning the video frame in both forward and backward direc-tions.The effective“You only look once V4”(YOLO V4)–a real-time people detection system illustrates the detection of people in videos,followed by a track-ing module to get their trajectories.Convolutional Neural Network(CNN)fea-tures are extracted for each person tracked through bounding boxes.Subsequently,a visual attention-guided Bi-directional LSTM model is proposed for the final suspicious fall event detection.The proposed method is demonstrated using two different datasets to illustrate the efficiency.The proposed method is evaluated by comparing it with other state-of-the-art methods,showing that it achieves 96.9%accuracy,good performance,and robustness.Hence,it is accep-table to monitor and detect suspicious fall events.

Curcumin and Apigenin – novel and promising therapeutics against chronic neuroinflammation in Alzheimer's disease

Alzheimer’s disease is a progressive neurodegenerative disorder, characterized by deposition of amyloid beta, neurofibrillary tangles, astrogliosis and microgliosis, leading to neuronal dysfunction and loss in the brain. Current treatments for Alzheimer’s disease primarily focus on enhancement of cholinergic transmission. However, these treatments are only symptomatic, and no disease-modifying drug is available for Alzheimer’s disease patients. This review will provide an overview of the proven antioxidant, anti-inflammatory, anti-amyloidogenic, neuroprotective, and cognition-enhancing effects of curcumin and apigenin and discuss the potential of these compounds for Alzheimer’s disease prevention and treatment. We suggest that these compounds might delay the onset of Alzheimer’s disease or slow down its progression, and they should enter clinical trials as soon as possible.

Probing and understanding interaction of Eu(Ⅲ)withγ-alumina in presenceof malonic acid

Radionuclide migration in aquatic environment is influenced by its sorption onto colloids/mineral oxides and the presence of organic complexing anions.With a view to understand the sorption of trivalent actinides by mineral oxides in presence of organic acid,in the present study,Eu(Ⅲ),malonic acid(MA)andγ-alumina are considered as representatives of trivalent actinides,low molecular weight natural occurring organic acid and aluminol sites,respectively.The influence of MA on sorption of Eu(Ⅲ)byγ-alumina was elucidated by batch sorption,spectroscopic techniques and surface complexation modeling,for the first time.Attenuated Total Reflection-Fourier Transform Infrared spectroscopic studies of MA sorbed onγ-alumina revealed the presence of two inner-sphere surface complexes.Batch sorption for binary(alumina-Eu(Ⅲ))and ternary(alumina-Eu(Ⅲ)-MA)systems were investigated as a function of p H,Eu(Ⅲ)concentration and sequential addition of Eu(Ⅲ)/MA.The p H edge for Eu(Ⅲ)sorption shifts to higher p H with increasing Eu(Ⅲ)concentration.In ternary systems,Eu(Ⅲ)sorption is significantly enhanced at p H<4.5.Eu(Ⅲ)speciation onγ-alumina is independent of addition sequence of Eu(Ⅲ)/MA.Time resolved fluorescence spectroscopy of Eu(Ⅲ)sorbed onγ-alumina exhibited two surface species,≡XOEu^2+and(≡YO)2 Eu^(+).The enhancement in I 616/I 592 and lifetime for ternary systems,as compared to binary system,at low p H,indicates the participation of Eu-MA complexes in the formation of surface species in ternary systems.The diffuse layer model has been employed to successfully model the experimental sorption profiles of binary and ternary systems,using code FITEQL 4.0,by considering the surface species identified by spectroscopic techniques.

Reliability of Sawai's classification for dental cervical abrasions:A pilot study

AIM To test the reliability of the Sawai’s classification for dental cervical abrasions.METHODSIntraoral photographs of 70 teeth from 23 patients with tooth abrasions were taken by the first examiner MS.The teeth were marked and the photos were maintained in a soft copy sequentially.Two other examiners FA and SC were trained in the use of the classification and any clarifications needed were provided at the beginning of the study.Each examiner was then given the soft copy of the complied photographs and was asked to classify the dental cervical abrasion according to their understanding of the Sawai’s classification.They were given sheets to write their responses for every marked tooth.All the examiners were blinded to each other’s observations which were then tested for inter-rater agreement among the three examiners.RESULTSThe 70 teeth with tooth abrasions from 23 patients were examined by 3 investigators(MS,FA and SC)to test the reliability of the Sawai’s classification system for tooth abrasion.Each examiner marked their responses in separate sheets which were blinded to each other.The kappa statistics were performed for inter-rater agreement among the three examiners.The level of agreement was evaluated according to the six-level nomenclature given by Landis and Koch.ICC and 95%CI between two examiners,i.e.,the inter-rater agreement among 1^(st) examiner(MS) and 2^(nd) examiner(FA) was 0.89.The inter-rater agreement among 1^(st) examiner(MS) and 3^(rd) examiner(SC) was 0.89.And the inter-rater agreement among 2^(nd) examiner(FA) and 3^(rd) examiner(SC) was 0.83.All the three comparisons show an almost perfect agreement between them.CONCLUSIONThere is an almost perfect agreement between multiple observers for classifying dental cervical abrasions using Sawai’s classification.Hence,this classification is reliable.

Approximate Data Aggregation in Sensor Equipped IoT Networks

As Internet-of-Things(IoT) networks provide efficient ways to transfer data, they are used widely in data sensing applications. These applications can further include wireless sensor networks. One of the critical problems in sensor-equipped IoT networks is to design energy efficient data aggregation algorithms that address the issues of maximum value and distinct set query. In this paper, we propose an algorithm based on uniform sampling and Bernoulli sampling to address these issues. We have provided logical proofs to show that the proposed algorithms return accurate results with a given probability. Simulation results show that these algorithms have high performance compared with a simple distributed algorithm in terms of energy consumption.

Integrated approach of heavy metal pollution indices and complexity quantification using chemometric models in the Sirsa Basin, Nalagarh valley, Himachal Pradesh, India

Chemometric techniques and pollution assessment indices were applied to determine the source and intensity of pollution in the Sirsa River, Himachal Pradesh,India. Results show EC, Cr, Fe, Mn, and Ni were above the permissible limit as per the Bureau of Indian Standards.The heavy metal pollution index(HPI) and contamination index(Cd) provided contrasting outcome and poor correlation was observed. A heavy metal evaluation index(HEI)method was developed using a multiple of the mean and correlation coefficient values to provide an alternative pollution classification. The criteria of HEI adopted for reclassification of HPI and Cdproduced comparable results; 40 % samples were labeled as low contamination,50 % as medium contamination, and 10 % as high contamination for all indices. Principal component analysis along with cluster analysis was used to identify the main factors responsible for degradation of water quality,namely discharge of industrial effluent, river bed mining,agricultural runoff, and minor natural or geogenic input.The methods and chemometric study proposed here can be used as effective tools to gather information about water quality and water resource management.

The CXCR3alt-CXCL11 axis in bladder cancer: potential for prediction of neoadjuvant chemotherapy response

Muscle-invasive bladder cancer(MIBC)constitutes-25%of newly diagnosed bladder cancer cases.Patients with MIBC are optimally treated with platinum-based neoadjuvant chemotherapy(NAC)prior to radical cystectomy,which improves overall survival compared to surgery alone.While the goal of NAC is to downstage the primary tumor and afford early systemic treatment of micrometastasis,more than 60%of patients receiving NAC will not demonstrate significant local responses.Without accurate biomarkers predicting those that will benefit from NAC,a large cohort of patients will not only be exposed to the side effects of chemotherapy but also unnecessarily be subjected to delayed curative surgical intervention.This dilemma in the current management of patients with this aggressive disease highlights the importance of a recent discovery by Vollmer and colleagues,who reported that the intratumoral CXCR3alt-CXCL11 chemokine axis may be associated with the response to NAC in MIBC.

Severe acute respiratory syndrome coronavirus-2(SARS-CoV-2)has affected millions of people across the world engendering an unprecedented pandemic.Coronavirus disease(COVID)-19 can present asymptomatic or in the form of the acute respiratory syndrome,viral pneumonia,or sepsis.Due to the novelty of the disease,the endocrine manifestations are not fully understood.It becomes indispensable to address the underlying endocrine disruptions contributing to the severe form of illness and thereby increasing the mortality.We discuss here the SARS-CoV-2 virus and endocrine reverberations based on the research with structurally similar SARS-COV-1.SARS-CoV-2 enters the body via its attachment to the angiotensin-converting enzyme 2(ACE2)receptors.Apart from lungs,ACE2 expression on various organs can lead to endocrine perturbations.In COVID-19 infection,pre-existing endocrine disorders warrant cautious management and may require replacement therapy.COVID-19 and its repercussions on hormones are discussed extensively in this review.
