The Zika virus(ZIKV)and dengue virus(DENV)flaviviruses exhibit similar replicative processes but have distinct clinical outcomes.A systematic understanding of virus–host protein–protein interaction networks can reveal cellular pathways critical to viral replication and disease *** we employed three independent systems biology approaches toward this ***,protein array analysis of direct interactions between individual ZIKV/DENV viral proteins and20,240 human proteins revealed multiple conserved cellular pathways and protein complexes,including proteasome ***,an RNAi screen of 10,415 druggable genes identified the host proteins required for ZIKV infection and uncovered that proteasome proteins were crucial in this ***,high-throughput screening of 6016 bioactive compounds for ZIKV inhibition yielded 134 effective compounds,including six proteasome inhibitors that suppress both ZIKV and DENV *** analyses of these orthogonal datasets pinpoint proteasomes as critical host machinery for ZIKV/DENV *** study provides multi-omics datasets for further studies of flavivirus–host interactions,disease pathogenesis,and new drug targets.

Modified vaccinia virus Ankara(MVA) is an approved smallpox vaccine and a promising vaccine vector for other pathogens as well as for cancer therapeutics with more than 200 current or completed clinical *** was derived by passaging the parental Ankara vac-cine virus hundreds of times in chick embryo fibroblasts during which it lost the ability to rep-licate in human and most other mammalian *** this replication deficiency is an important safety feature,the genetic basis of the host restriction is not ***,an unbiased human genome-wide RNAi screen in human A549 cells revealed that the zinc-finger antiviral protein(ZAP),previously shown to inhibit certain RNA viruses,is a host restriction factor for MVA,a DNA *** studies demonstrated enhanced MVA replication in several human cell lines following knockdown of ***,CRISPR-Cas9 knockout of ZAP in human A549 cells increased MVA replication and spread by more than one log but had no effect on a non-attenuated strain of vaccinia *** intact viral C16 protein,which had been disrupted in MVA,antagonized ZAP by binding and sequester-ing the protein in cytoplasmic punctate *** aimed at exploring the mecha-nism by which ZAP restricts MVA replication in the absence of C16 showed that knockout of ZAP had no discernible effect on viral DNA or individual mRNA or protein species as deter-mined by droplet digital polymerase chain reaction,deep RNA sequencing and mass spec-trometry,***,inactivation of ZAP reduced the number of aberrant,dense,spherical particles that typically form in MVA-infected human cells,suggesting that ZAP has a novel role in interfering with a late step in the assembly of infectious MVA virions in the absence of the C16 protein.

Comparative efficacy of hyperbaric bupivacaine vs hyperbaric ropivacaine in spinal anesthesia for cesarean section:A metaanalysis

Frozen shoulder-A prospective randomized clinical trial

AIM To compare the results of arthroscopic capsular release with intra-articular steroid injections in patients of frozen *** Fifty-six patients with frozen shoulder were randomised to one of two treatment groups: Group 1, complete 360 degree arthroscopic capsular release and group 2, intra-articular corticosteroid injection(40 mg methyl prednisolone acetate). Both groups were put on active and passive range of motion exercises following the intervention. The outcome parameters were visual analogue scale(VAS) score for pain, range of motion and Constant score which were measured at baseline, 4, 8, 12, 16 and 20 wk after *** All the parameters improved in both the groups. The mean VAS score improved significantly more in the group 1 as compared to group 2 at 8 wk. This greater improvement was maintained at 20 wk with P value of 0.007 at 8 wk, 0.006 at 12 wk, 0.006 at 16 wk and 0.019 at 20 wk. The Constant score showed a more significant improvement in group 1 compared to group 2 at 4 wk, which was again maintained at 20 wk with P value of 0.01 at 4, 8, 12 and 16 wk. The gain in abduction movement was statistically significantly more in arthroscopy group with P value of 0.001 at 4, 8, 12, 16 wk and 0.005 at 20 wk. The gain in external rotation was statistically significantly more in arthroscopy group with P value of 0.007 at 4 wk, 0.001 at 8, 12, and 16 wk and 0.003 at 20 wk. There was no statistically significant difference in extension and internal rotation between the two groups at any time. CONCLUSION Arthroscopic capsular release provides subjective and objective improvement earlier than intra-articular steroid injection.

Hot deformation behavior and processing maps of Ti-15Al-12Nb alloy

The isothermal hot compression tests of Ti-15Al-12Nb alloy under wide range of strain rates(0.01-10.00 s^(-1)and deformation temperatures(950,1000,1050,and 1100℃)were carried out using Gleeble-3500 thermo-simulation machine.A constitutive equation represented as a function of temperature,strain rate and true strain was developed,and the hot deformation appar-ent activation energy is calculated to be about 453 kJ·mol^(-1).By employing dynamic material model(DMM),the processing maps of Ti-15Al-12Nb alloy at various strains were *** efficiency of about 57%for power dissipation is obtained at high temperature and low strain *** to the high power dissipation efficiency and excellent processing ability in dynamic recrystallization(DRX)zone for metal material,the optimum processing conditions are selected as the temperature range of 1050-1100℃and the strain rate range of 0.01-0.10 s^(-1).Using transmission electron microscopy(TEM)studies,it is found that the dislocation density is directly associated with the value of processing *** is observed that when the processing effi-ciency is about 22%,the dislocation density is reasonably *** flow instability region occurs at strain rate of 10.00 s^(-1)with cracks,which should be avoided during hot processing to obtain the required mechanical properties.

Antioxidant and modulatory role of Chlorophytum borivilianum against arsenic induced testicular impairment

Arsenic has a suppressive influence on spermatogenesis and induces impairment in male reproductive system due to oxidative stress. The present study was aimed to test the arsenic induced toxicity and protection by Chlorophytum borivilianum. The effect of sodium arsenite （4 mg/（kg body weight （bw）.day）） via double distilled water without or with C. borivilianum （800 mg/（kg ***）） was evaluated in Swiss albino mice for 30 days. The radical scavenging activity of the aqueous C. borivilianum root extract was measured using DPPH （1,1-diphenyl-2-picryl hydrayzyl） radical. Qualitative assessment of various cell types in the testis, sperm count and motility, testicular activity of lipid peroxidation （LPO）, reduced glutathione （GSH）, acid and alkaline phosphatase, cholesterol and serum testosterone were monitored. Arsenic treatment showed a significant increase in LPO, acid and alkaline phosphatase, cholesterol and decrease in sperm count, sperm motility, GSH and serum testosterone. Combined treatment showed significant decrease in LPO, acid and alkaline phosphatase, cholesterol and elevation in sperm count, sperm motility, GSH and serum testosterone. Testicular histopathology showed that C. borivilianum had reduced degeneration of germ cell in the seminiferous tubules and loss of sperms induced by arsenic intoxication. The results thus led us to conclude that administration of C. borivilianum root extract is found to be protective against arsenic induced toxicity.

Sustainable intensification of China's agroecosystems by conservation agriculture

Agriculture in China started about 8000 BCE with cultivation of millet and rice. Thus, China's modern agriculture is rooted in a long evolutionary history of agricultural practices. China's population of 211 million in 1700 increased to 295 million in 1800, 400 million in 1900, 1266 million in 2000, and is 1409 million in 2017. Thus, China has to feed 18.5% of the world population on merely 7% of the world's arable land and with even scarcer water resources. Yet, between 1950 and 2017, the grain production in China increased at a faster rate than that of the population. Total grain production (million ton or Tg) was 113 in 1949, 305 in 1970, 462 in 2000, and 603 in 2015. The high agricultural productivity is attributed to high consumption of fertilizers (58.8 Tg in 2015), pesticides (1.81 Tg in 2014), and irrigation (65.7 Mha in 2014). However, the high production is also associated with the severe problems of soil degradation (erosion), water contamination and air pollution. It is estimated that ~40% of China's arable land is degraded to some degree. Thus, there is a strong need for a paradigm shift of sustaining high productivity while restoring quality of natural resources of soil, water and air. In this context, conversion to a systembased conservation agriculture (CA) may be an important strategy of sustainable intensification of agroecosystems for advancing and sustaining high production while restoring soil health, purifying water and air and improving the environment. The CA has been practiced in China since 1990s and was adopted on ~8.0 Mha in 2015. The available literature shows that conversion to CA increases soil organic C (SOC) concentration and stocks mostly in the surface layer while also producing an equivalent agronomic yield. Therefore, a widespread adoption of CA in China would necessitate a deeper understanding of its ecological underpinnings. Bulk arrival retrial G-queue with impatient customers and multi-servicessubject to server breakdowns has been analyzed. The system allows the arrival oftwo types of customers: positive customers and negative customers in the *** negative customers make the server fail if they find the server in busy state,whereas positive customers are served if the server is idle otherwise they join thevirtual pool of customers called orbit. The customers from the retrial orbit try theirchance again for the service. The customers have the option of obtaining more than oneservice. Moreover, the customers are impatient and may renege from the system withprobability (1−r ). The server is sent for repair as soon as it breakdowns;after repair,the service process starts again. Also, the server has the provision to initiate the servicewhen there areN customers accumulated in the system. Using supplementary variablestechnique and generating functions, various performance measures like reliabilityindices and long run probabilities have been obtained.
