Co-delivery of PArP and PI3K inhibitors by nanoscale metal-organic frameworks for enhanced tumor chemoradiation

Due to their complementary activity,the use of DNA damage repair(DDr)inhibitors during radiotherapy(rT)has yielded promising ***,this approach is often hindered by toxicity and poor in vivo stability of the DDr in *** scale metal-organic frameworks(nMOFs)represent an emerging class of crystalline materials which exhibit advantageous properties over traditional nanomaterials and demonstrate great potential in ***,a unique,synergistic treatment strategy for enhancing the therapeutic efficacy of rT vianMOF-mediated drug delivery and rT enhancement was evaluated.A nMOF containing the high-Zelement Hf and the ligand 1,4-dicarboxybenzene(Hf-BDC)was synthesized and loaded with the two DDr inhibitor drugs,talazoparib and buparlisib(TB@Hf-BDC-PEG).TB@Hf-BDC-PEGaugmented rT by increasing reactive oxygen species generation and thus DNA damage of which repair was inhibited *** enhancement was demonstrated in vivo where the combination of concurrent radiation with in travenous TB@Hf-BDC-PEG administration resulted in improved tumor control and increased ***,no apparent toxicity was observed with nMOF treatment,supporting its potential as an attractive candidate over traditional nano *** work provides the first report of nMOFs employed to enhance thetherapeutic response to rT through DDr inhibitor delivery and physical radiation dose enhancement.

receptor-like kinases(rLKs)are plasma membrane(PM)proteins that monitor the extracellular environment and regulate various processes such as biotic *** motif(LysM)rLKs are a specific class of rLKs involved in the recognition of chitin-derived molecules produced by microorganisms(Buendia et al.,2018).Notably,the LysM-rLK heterocomplex LjNFr1/LjNFr5 recognizes lipochitooligosaccharides(LCOs,also called Nod factors)leading to root nodule symbiosis(rNS)in Lotus japonicus,whereas the heterocomplexes AtLYK5/AtCErK1 and OsCEBIP/OsCErK1 recognize chitooligosaccharides(COs)leading to defense responses in Arabidopsis thaliana and rice,respectively(Buendia et al.,2018).These receptors function as *** ligand binding activates the intracellular kinase activity of LjNFr1,AtCErK1,and OsCErK1,which mediate phosphorylation-dependent signaling *** mechanisms are essential to tightly control rLK homeostasis and signaling,allowing cells to respond efficiently while preventing constitutive *** PM-resident pool of rLKs must be maintained for rapid response,while rLK endocytosis and ultimately degradation in lytic vacuoles is critical for both signaling and downregulation of the *** addition to its role in proteasome-dependent degradation,ubiquitination,a post-translational protein modification,has emerged as a key regulator of PM proteins by inducing their internalization and degradation in lytic vacuoles(romero-Barrios and Vert,2018).

Thermal characterization of bio-based phase changing materials in decorative wood-based panels for thermal energy storage

Decorative wood panels containing pouches of bio-based phase changing materials(PCMs) were prepared. Three different PCM mixtures were used: a blend of capric and lauric acids as well as two commercial products, Puretemp?20 and Puretemp?23(Puretemp). The panels consist of engraved Medium Density Fiberboard(MDF) filled with a plastic pouch filled with PCM. High density fiberboard(HDF) was used on top of the panels to enclose the PCM pouches. PCM mixtures were first tested by differential scanning calorimetry(DSC). Phase change temperature and total heat storage of the panels were measured for both fusion and solidification with a Dynamic Heat-Flow Meter Apparatus(DHFMA). DSC and DHFMA results were compared, allowing a better understanding of results gathered from these two techniques. DSC calibration has been revealed important when assessing PCMs. The panels present a phase change temperature and a latent heat storage suitable for buildings applications. The panel made with Puretemp?23 presented the highest energy, with 57.1 J g^(-1). Thermal cycling was conducted on the panels to investigate thermal reliability, which revealed small modifications of thermal properties for two products. For all cases, latent heat was found stable. Hygro-mechanical behavior of the panels was also evaluated as these where designed to be esthetic decorative panels. This study exposes the potential of a new type of wood-based panels loaded with PCM for thermal energy storage and brings overall knowledge about PCM products thermal characterization.

Microscopic imaging of the brain continues to reveal details of its structure, connectivity, and function. To further improve our understanding of the emergent properties and functions of neural circuits, we need to directly visualize the relationship between brain structure, neuron activity, and neurochemistry. Advances in the chemical and optical properties of nanomaterials, and developments in deep-tissue microscopy, may help to overcome the current challenges of in-vivo brain imaging, particularly when imaging the brain through optically-dense brain tissue, skull, and scalp. Developments in nanomaterials may enable the implementation of tunable chemical functionality for neurochemical targeting and sensing, and fluorescence stability for long-term imaging. In this review, we summarize the current methods used for brain microscopy and describe the diverse classes of nanomaterials recently offered as contrast agents and functional probes for microscopic optical imaging of the brain.

Identifying the dominant transport mechanism in single nanoscale pores and 3D nanoporous media

Gas transport mechanisms can be categorized into viscous flow and mass diffusion,both of which may coexist in a porous media with multiscale pore *** determine the dominant transport mechanism and its contribution to gas transport capacity,the gas viscous flow and mass diffusion processes are analyzed in single nanoscale pores via a theoretical method,and are simulated in 3D nanoporous media via pore-scale lattice Boltzmann *** apparent permeability from the viscous flow and apparent diffusivity from the mass diffusion are estimated.A dimensionless parameter,i.e.,the diffusion-flow ratio,is proposed to evaluate the dominant transport mechanism,which is a function of the apparent permeability,apparent diffusivity,bulk dynamic viscosity,and working *** results show that the apparent permeability increases by approximately two orders of magnitude when the average Knudsen number(Kn_(avg))of the nanoporous media or Knudsen number(Kn)of single nanoscale pores increases from 0.1 to *** the same conditions,the increment in the apparent diffusivity is only approximately one order of *** Knrent permeability has a lower bound(i.e.,absolute permeability).When Kn>10,the apparent diffusivity has an upper bound(i.e.,Knudsen diffusivity).The dominant transport mechanism in single nanoscale pores is the viscous flow for 0.01re the maximum diffusion-flow ratio is less than *** nanoporous media,the dominant transport relies heavily on Kn_(avg) and the structural *** nanoporous media with the pore throat diameter of 3 nm,Kn_(avg)=0.2 is the critical point,above which the mass diffusion is dominant;otherwise,the viscous flow is *** Kn_(avg) increases to 3.4,the mass diffusion is overwhelming,with the maximum diffusion-flow ratio reaching ~4.

Insight into the Origin of Iron Ore Based on Elemental Contents of Magnetite and Whole-rock Geochemistry:A Case of the Bipindi Banded Iron Formations,Nyong Complex,SW Cameroon

The Bipindi iron ore district is located in the central section of the Nyong Complex at the northwestern margin of the Congo Craton in Southwest *** iron district contains numerous iron mineralization hosted by the Mewongo,Bibole,Kouambo,and Zambi banded iron formations(BIFs).These BIFs contain magnetite as the main iron ore mineral associated with pyrite,and gangue minerals are quartz with minor chlorite and *** origin of iron ore from these BIFs was investigated using a combination of in-situ magnetite and whole-rock *** studied BIF ore samples have a narrow range of TFe between 30.90 wt.%and 43.20 wt.%,indicating a low-grade *** geochemical signatures of magnetite such as low contents of base metals(e.g.,Cu,Co,V,and Zn)and low Co/Zn ratiosrothermal *** with the geochemical features of these BIFs,e.g.,high Fe/Ti and Fe/Al ratios(mean>600 and>75,respectively),we suggest that magnetite was derived from a mixture of seawater and~0.1%low-temperature hydrothermal fluids in an oxidizing ***,low-temperature hydrothermal and later metamorphic fluids were necessary for the transformation of the protolith Nyong Complex BIFs to iron ore.

The Bafoussam-Mamfe region(BMr)belongs to the Western Cameroon Highland margin,an amalgam of transient landscape that evolved from the interactions between tectonics,lithology and climate *** area is tectonically active and shows differential tectonic uplift,contrasting relief,variations in erosion rates,in river incision,and in channel *** order to appraise the BMr tectono-geomorphic evolution,it is essential to understand relationships between internal and external processes and their geomorphic expression at a river basin *** this purpose,we use drainage texture(Dt),stream frequency(Fs),drainage density(Dd),asymmetry factor(AF),the transverse topographic symmetry factor(TTSF),the mean of mountain front sinuosity per unit(Smf),concavity factor(Cf),hypsometry integral(HI)and streamlength gradient index(SL-index)based on 30 m Digital Elevation Models(DEM)and field *** results reveal considerable variations in geology,relief,topography and geomorphic indices values differentiated into three geomorphic units;the Mamfe unit developed on the Mamfe Cretaceous basin,the Nguti and Bafoussam high plateau units build up on the Precambrian granito-gneissic basement overlaid by a volcanic *** stepped(longitudinal and normalized)river profiles and complex hypsometric curves show the lithostructural control of the hydrographic ***(from~1.21 to~4.59),Fs(from~0.26 to~1.91),Dd(from~0.66 to~1.71)geomorphic indice values indicate a part of lithologic control on this morphology while the AF(from~1.16to~24.91),TTSF(from~0.20 to~0.71),Smf(U2=1.11 and U3=1.19),Cf(from~-20.91 to~78.93),HI(from~0.12 to~0.52)and SL-index designate the active tectonics as the main factor that had controlled the morphology of this same *** results pinpoint the combined post-Pan-African to recent tectonic features,lithologic heterogeneities and the differential erosion processes as main factors that have controlled the BMr *** results highlight how
