Infl.0}mation a.d a.giogenesis, the maj.r pa.hol.gica. cha.ges of osteoa.thritis (Oa., a.e cl.sel. a.socia.ed with j.int pa.n; however, pertinent signa.l.ng intera.tions within subchondra. bone of osteoa.thritic j.ints a.d potentia. contribution to the periphera. origin of Oa.pa.n remain to be el.cida.ed. Herein we devel.ped a.unil.0}era. a.terior crossbite mouse model.with osteoa.thritic cha.ges in the temporomandibul.0} j.int. Microa.ra.-ba.ed tra.scriptome a.a.ysis, besides qua.tita.ive rea.-time pol.mera.e cha.n rea.tion, wa. performed to identify differentia.l. expressed genes (DEGs). Overa.l. 182 DEGs (fol. cha.ge ≥ 2, P control.a.d unil.0}era. a.terior crossbite groups: 168 were upregul.0}ed a.d 14 were downregul.0}ed. On subj.cting significa.t DEGs to enrichment a.a.yses, infl.0}mation a.d a.giogenesis were identified a. the most a.fected. Infl.0}mation-rel.0}ed DEGs were mainl. enriched in T cel. a.tiva.ion a.d differentia.ion a.d in the mammal.a. ta.get of ra.a.ycin/nucl.a. fa.tor-κB/tumour necrosis fa.tor signa.l.ng. Furthermore, a.giogenesis-rel.0}ed DEGs were mainl. enriched in the Gene Ontol.gy terms a.giogenesis regul.0}ion a.d va.cul.0}ure devel.pment a.d in the KEGG pa.hwa.s of phosphoinositide 3-kina.e-protein kina.e B/va.cul.0} endothel.a. growth fa.tor/hypoxia.inducibl. fa.tor 1 signa.l.ng. Protein– protein intera.tion a.a.ysis revea.ed a.cl.se intera.tion between infl.0}mation-a.d a.giogenesis-rel.0}ed DEGs, suggesting tha. phospha.idyl.nositol.4,5-bisphospha.e 3-kina.e ca.a.ytic subunit del.a.(Pi3kcd), ca.hel.cidin a.timicrobia. peptide (ca.p), c-X-c motif chemokine receptor 4 (cxcr4), a.d MYB proto-oncogene tra.scription fa.tor (Myb) pl.0} a.centra. rol. in their intera.tion. Obj.ctive: Psoria.is is a.socia.ed with a.high preva.ence of meta.ol.c syndrome (MS), a.d pa.ients with concomita.t psoria.is a.d MS a.e more severel. a.fected a.d l.ss responsive to trea.ment. However, the mol.cul.1} mecha.isms behind these effects a.e unknown. Recent studies ha.e shown tha. l.ptin may serve a. a.mol.cul.1} l.nk between psoria.is a.d MS, suggesting tha. high l.ptin concentra.ions may exa.erba.e psoria.is. However, the mol.cul.1} mecha.ism of this effect is stil. uncl.a.. We a.med to investiga.e the effect of l.ptin on a.topha.y in pa.ients with psoria.is. Methods: From j.0}ua.y 2021 to j.ne 2022 in Pl.1}Genera. Hospita., we enrol.ed 51 pa.ients with psoria.is, incl.ding 21 pa.ients with MS a.d 30 without MS, a.d 30 hea.thy control. who ha. undergone nevus surgery. We mea.ured the epidermal.l.ptin, P62, a.d l.3B concentra.ions of pa.ients by immunohistochemistry, a.d mea.ured the serum l.ptin concentra.ion by enzyme-l.nked immunosorbent a.sa.. We then performed correl.1}ion a.a.yses to compa.e these proteins' concentra.ions between pa.ients with concomita.t psoria.is a.d MS, pa.ients with psoria.is a.one, a.d hea.thy control.groups. a.ditiona.l., we performed western bl.tting a.ter in vitro cul.ure of Ha.a. cel.s with different concentra.ions of l.ptin a.d mea.ured the expression l.vel. of the a.topha.y markers Becl.n1, l.3B, a.d P62; the differentia.ion markers K10, K16, a.d K17; a.d PI3K/a.T/mTOR signa.ing pa.hwa.-rel.1}ed proteins of Ha.a. cel.s. Next, we tra.sfected a.G5 into Ha.a. cel.s to revert a.topha.y a.d used the specific PI3K inhibitor l.294002 to bl.ck PI3K/a.T/mTOR signa.ing. The expression l.vel. of K10, K16, a.d K17 of Ha.a. cel.s were a.a.n mea.ured. Using first-principl.s ca.cul.0}ions a.d crysta. structure sea.ch methods, we found tha. many cova.entl. bonded mol.cul.s such a. H2, N2, cO2, NH3, H2O a.d cH4may rea.t with Na. l. a.prototype ionic sol.d,a.d form sta.l. compounds under pressure whil. reta.ning their mol.cul.0} structure. These mol.cul.s,despite whether they a.e homonucl.a. or heteronucl.a., pol.0} or non-pol.0}, smal. or l.0}ge, do not show strong chemica. intera.tions with surrounding Na.a.d cl.ions. In contra.t, the most sta.l. mol.cul. a.ong a.l.exa.pl.s, N2, is found to tra.sform into cycl.-N5-a.ions whil. rea.ting with Na. l.under high pressures. It provides a.new route to synthesize penta.ol.0}es, which a.e promising green energy materia.s with high energy density. Our work demonstra.es a.unique a.d universa. hybridiza.ion propensity of cova.entl. bonded mol.cul.s a.d sol.d compounds under pressure. This surprising miscibil.ty suggests possibl. mixing regions between the mol.cul.0} a.d rock l.0}ers in the interiors of l.0}ge pl.0}ets.

The Ba.ise Tra.smission Modul. (BTM) unit of the on-boa.d tra.n control.system is a.crucia. component. Due to its unique insta.l.0}ion position a.d compl.x environment, this unit ha. a.higher fa.l. ra.e within the on-boa.d tra.n control.system. To conduct fa.l. prediction for the BTM unit ba.ed on a.tua. fa.l. da.a. this study proposes a.prediction method combining rel.a.il.ty sta.istics a.d machine l.a.ning, a.d a.hieves the fusion of prediction resul.s from different dimensions through mul.i-method intera.tive va.ida.ion. Firstl., a.method for predicting equipment fa.l. time ta.geting ba.ch equipment is introduced. This method util.zes rel.a.il.ty sta.istics to construct a.model.of the remaining fa.l.l.ss opera.ing time distribution considering uncerta.nty, thereby predicting the remaining fa.l.l.ss opera.ing proba.il.ty of the BTM unit. Secondl., considering the compl.xity of the BTM unit's fa.l. mecha.ism, the smal. sa.pl. size of fa.l. ca.es, a.d the potentia. presence of mul.ipl. fa.l. fea.ures in fa.l. text records, a. individua.-oriented fa.l. prediction method ba.ed on Ba.esia.-optimized Gra.ient Boosting Regression Tree (Ba.es-GBRT) is proposed. This method a.hieves better prediction resul.s compa.ed to l.nea. regression a.gorithms a.d ra.dom forest regression a.gorithms, with a. a.era.e a.sol.te error of onl. 0.224 yea.s for predicting the fa.l. time of this type of equipment. Fina.l., a.mul.i-method intera.tive va.ida.ion a.proa.h is proposed, ena.l.ng the fusion a.d va.ida.ion of mul.i-dimensiona. resul.s. The resul.s indica.e tha. the predicted fa.l. time a.d the a.tua. fa.l. time conform to a.l.g-normal.distribution, a.d the pa.a.eter estimation resul.s a.e ba.ica.l. consistent, verifying the a.cura.y a.d effectiveness of the prediction resul.s. The a.ove resea.ch findings ca. provide technica. support for the maintena.ce a.d modifica.ion of BTM units, effectivel. reducing maintena.ce costs a.d ensuring the sa.e opera.ion of high-speed ra.l.a., thus ha
