Synthetic host-guest pairs as novel bioorthogonal tools for pre-targeting

Due to its simplicity, high efficiency, and chemo-selectivity, bioorthogonal chemistry has shown a great application potential in pre-targeting.Currently, four bioorthogonal pairs as targeting tools, including (strept)avidin/biotin, antibody/antigen, oligonucleotide hybridization and IEDDA tools, have been developed and applied in targeted delivery.Nevertheless, all of these tools still suffer from some limitations, such as difficult modification, biochemical fragility and larger molecular weight for biological association tools, as well as chemical instability for IEDDA tools.Synthetic host-guest pairs with relatively small molecular sizes not only possess strong chemical stability, but also have the features of fast conjugation rate, tunable binding affinity , easy modification, and high chemo-selectivity.Consequently, they can be used as a novel non-covalent bioorthogonal tool for pre-targeting.In order to further promote the development of host-guest pairs as novel bioorthogonal tools for pre-targeted delivery, we firstly calculate their conversion rate to make researcher aware of their unique advantages;next, we summarize the recent research progress in this area.The future perspectives and limitations of these unique tools will be discussed.This review will provide a systemic overview of the development of synthetic host-guest pairs as novel bioorthogonal tools for pre-targeting, and may serve as a “go for” resort for researchers who are interested in searching for new synthetic tools to improve pre-targeting.

Realizing high initial Coulombic efficiency in manganese-based layered oxide cathodes for sodium-ion batteries via P2/O'3 biphasic structure optimization

Abstract Mn-based layered oxides are among the most promising cathode materials for sodium-ion batteries owing to the advantages of abundance,environmenta friendliness,low cost and high specific capacity.P2 and O'3 are two representative structures of Mn-based layered oxides.However,the P2 structure containing insufficien Na generally exhibits low initial charge capacity,while O'3structure with sufficient Na delivers high initial charge capacity but poor cycle stability.This study prepared a multitude of NaxMnO2（x=0.7,0.8,0.9） cathode materials with varying P2/O'3 ratios and further investigated their electrochemical performances.The optimized Na0.8MnO2comprising 69.9 wt%O'3 and 30.1 wt% P2 phase,exhibited relatively balanced specific capacity,Coulombic efficiency and cycle stability.Specifically,it achieved a high specific capacity of 128.9 mah·g-1with an initia Coulombic efficiency of 98.2%in half-cell configuration The Na0.8MnO2// hard carbon full cell also achieved a high specific capacity of 126.7 mah·g-1with an initia Coulombic efficiency of 98.9%.Moreover,the capacity fading mechanism was revealed by combining in-situ and ex-situ X-ray diffraction.The findings of this study provide theoretical guidance for further modification design of Mnbased layered cathodes.

Diagnosis and treatment experience of atypical hepatic cystic echinococcosis type 1 at a tertiary center in China

BACKGROUND Some hydatid cysts of cystic echinococcosis type 1(CE1)lack well-defined cyst walls or distinctive endocysts,making them difficult to differentiate from simple hepatic cysts.AIM To investigate the diagnostic methods for atypical hepatic CE1 and the clinical efficacy of laparoscopic surgeries.METHODS The clinical data of 93 patients who had a history of visiting endemic areas of CE and were diagnosed with cystic liver lesions for the first time at the People's Hospital of Xinjiang Uygur Autonomous Region(China)from January 2018 to September 2023 were retrospectively analyzed.Clinical diagnoses were made based on findings from serum immunoglobulin tests for echinococcosis,routine abdominal ultrasound,high-frequency ultrasound,abdominal computed tomography(CT)scan,and laparoscopy.Subsequent to the treatments,these patients underwent reexaminations at the outpatient clinic until October 2023.The evaluations included the diagnostic precision of diverse examinations,the efficacy of surgical approaches,and the incidence of CE recurrence.RESULTS All 93 patients were diagnosed with simple hepatic cysts by conventional abdominal ultrasound and abdominal CT scan.Among them,16 patients were preoperatively diagnosed with atypical CE1,and 77 were diagnosed with simple hepatic cysts by high-frequency ultrasound.All the 16 patients preoperatively diagnosed with atypical CE1 underwent laparoscopy,of whom 14 patients were intraoperatively confirmed to have CE1,which was consistent with the postoperative pathological diagnosis,one patient was diagnosed with a mesothelial cyst of the liver,and the other was diagnosed with a hepatic cyst combined with local infection.Among the 77 patients who were preoperatively diagnosed with simple hepatic cysts,4 received aspiration sclerotherapy of hepatic cysts,and 19 received laparoscopic fenestration.These patients were intraoperatively diagnosed with simple hepatic cysts.During the followup period,none of the 14 patients with CE1 experienced recurrence or implantation of hydatid sc

Span-based joint entity and relation extraction augmented with sequence tagging mechanism

Span-based joint extraction simultaneously conducts named entity recognition(NER) and relation extraction(RE) in a text span form. However, since previous span-based models rely on span-level classifications, they cannot benefit from token-level label information, which has been proven advantageous for the task. In this paper, we propose a sequence tagging augmented span-based network(STSN), a spanbased joint model that can make use of token-level label information. In STSN, we construct a core neural architecture by deep stacking multiple attention layers, each of which consists of three basic attention units.On the one hand, the core architecture enables our model to learn token-level label information via the sequence tagging mechanism and then uses the information in the span-based joint extraction; on the other hand, it establishes a bi-directional information interaction between NER and RE. Experimental results on three benchmark datasets show that STSN consistently outperforms the strongest baselines in terms of F1,creating new state-of-the-art results.

Braille Character Segmentation Algorithm Based on Gaussian Diffusion

Optical braille recognition methods typically employ existing target detection models or segmentation modelsfor the direct detection and recognition of braille characters in original braille images. However, these methodsneed improvement in accuracy and generalizability, especially in densely dotted braille image environments. Thispaper presents a two-stage braille recognition framework. The first stage is a braille dot detection algorithmbased on Gaussian diffusion, targeting Gaussian heatmaps generated by the convex dots in braille images. Thisis applied to the detection of convex dots in double-sided braille, achieving high accuracy in determining thecentral coordinates of the braille convex dots. The second stage involves constructing a braille grid using traditionalpost-processing algorithms to recognize braille character information. Experimental results demonstrate that thisframework exhibits strong robustness and effectiveness in detecting braille dots and recognizing braille charactersin complex double-sided braille image datasets. The framework achieved an F1 score of 99.89% for Braille dotdetection and 99.78% for Braille character recognition. Compared to the highest accuracy in existing methods,these represent improvements of 0.08% and 0.02%, respectively.

BACKGROUND According to clinical data,a significant percentage of patients experience pain after surgery,highlighting the importance of alleviating postoperative pain.The current approach involves intravenous self-control analgesia,often utilizing opioid analgesics such as morphine,sufentanil,and fentanyl.Surgery for colo-rectal cancer typically involves general anesthesia.Therefore,optimizing anes-thetic management and postoperative analgesic programs can effectively reduce perioperative stress and enhance postoperative recovery.The study aims to analyze the impact of different anesthesia modalities with multimodal analgesia on patients'postoperative pain.AIM To explore the effects of different anesthesia methods coupled with multi-mode analgesia on postoperative pain in patients with colorectal cancer.METHODS Following the inclusion criteria and exclusion criteria,a total of 126 patients with colorectal cancer admitted to our hospital from January 2020 to December 2022 were included,of which 63 received general anesthesia coupled with multi-mode labor pain and were set as the control group,and 63 received general anesthesia associated with epidural anesthesia coupled with multi-mode labor pain and were set as the research group.After data collection,the effects of postoperative analgesia,sedation,and recovery were compared.RESULTS Compared to the control group,the research group had shorter recovery times for orientation,extubation,eye-opening,and spontaneous respiration(Pmany scores at 6 h and 12 h,and improved cognitive function at 24 h,48 h,and 72 h(P<0.05).Additionally,interleukin-6 and interleukin-10 levels were significantly reduced at various time points in the research group compared to the control group(P<0.05).Levels of CD3+,CD4+,and CD4+/CD8+were also lower in the research group at multiple time points(P<0.05).CONCLUSION For patients with colorectal cancer,general anesthesia coupled with epidural anesthes

Research on denoising of joint detection signal of water quality with multi-parameter based on IEEMD

An improved ensemble empirical mode decomposition(IEEMD) is suggested to process water quality spectral signals in order to address the issue that noise interference makes it difficult to extract and evaluate water quality spectral signals.This algorithm effectively solves the problems of modal mixing,poor reconstruction accuracy in the empirical mode decomposition(EMD),and a large amount of calculation in the ensemble empirical mode decomposition(EEMD).Based on EEMD,IEEMD firstly preprocesses the original water quality spectral signals,then performs Savitzky-Golay(S-G) smoothing on the decomposed effective intrinsic mode function(IMF) components,and finally reconstructs them to obtain the denoised signals.Water sample data at different concentrations can be accurately analyzed based on the noise-reduced spectral signals.In this paper,three water quality parameters are used as research objects:benzene(C_(6)H_(6)),benzo(b)fluoranthene(C_(20)H_(12)),and chemical oxygen demand(COD).The original water quality multi-parameter(C_(6)H_(6),C_(20)H_(12),COD) spectral signals were subjected to denoising based on the IEEMD and the water quality multi-parameter joint detection technology.The signal-to-noise ratio(SNR) and the correlation coefficient(R^(2)) of the fitted curves obtained from the processing of the IEEMD were compared and analyzed with those obtained from the processing of the EMD and the EEMD.The experimental results show that the SNR of the spectral signals and the R^(2) of the fitting curve in three water quality parameters have been significantly improved.Therefore,the IEEMD effectively improves the phenomenon of modal mixing,reduces the amount of calculation,improves the reconstruction accuracy,and provides an important guarantee for the effective extraction of multi-parameter spectral signals of water quality.

Enaminones,which possesses both the nucleophilic enamine as well as electrophilic enone structures,are well known versatile building blocks in organic synthesis.Meanwhile,visible light-mediated reactions have emerged as useful synthetic strategy with enhanced sustainability.Around the last decade,various photochemical transformations of enaminones have been developed to construct cyclic or acyclic compounds.In this review,we describe the recent advances in visible light-mediated chemical transformations of enaminones.Detailed discussion on the reaction mechanism of the related reactions is given to provide guide to the reader.Finally,a summary on the existing challenges and the future outlook towards the development of practical photocatalytic reactions of enaminones is also presented.

A Dynamic Covalent Bonding-based Nanoplatform for Intracellular Co-Delivery of Protein Drugs and Chemotherapeutics with Enhanced Anti-Cancer Effect

Efficient intracellular delivery of protein drugs is critical for protein therapy.The combination of protein drugs with chemotherapeutics represents a promising strategy in enhancing anti-cancer effect.However,co-delivery systems for efficient delivery of these two kinds of drugs are still lacking because of their different properties.Herein,we show a well-designed delivery system based on dynamic covalent bond for efficient intracellular co-delivery of ribonuclease A(RNase A)and doxorubicin(DOX).Two polymers,PEG-b-P(Asp-co-AspDA)and PAE-b-P(Asp-co-AspPBA),and two 2-acetylphenylboronic acid(2-APBA)-functionalized drugs,2-APBA-RNase A and 2-APBA-DOX,self-assemble into mixed-shell nanoparticles(RNase A/DOX@MNPs)via dynamic phenylboronic acid(PBA)-catechol bond between PBA and dopamine(DA)moieties.The PBA-catechol bond endows the nanoparticles with high stability and excellent stimulus-responsive drug release behavior.Under the slight acidic environment at tumor tissue,RNase A/DOX@MNPs are positively charged,promoting their endocytosis.Upon cellular uptake into endosome,further protonation of PAE chains leads to the rupture of endosomes because of the proton sponge effect and the cleavage of PBA-catechol bond promotes the release of two drugs.In cytoplasm,the high level of GSH removed the modification of 2-APBA on drugs.The restored RNase A and DOX show a synergistic and enhanced antic-cancer effect.This system may be a promising platform for intracellular co-delivery of protein drugs and chemotherapeutics.

Effect of Guanxin Danshen Dripping Pills on Coronary Heart Disease Comorbid with Depression or Anxiety: The ADECODE-Real World Study

Objective: To evaluate the efficacy of Guanxin Danshen Dripping Pill(GXDSDP) in treating anxiety and depression in patients with coronary heart disease(CHD). Methods: A total of 1,428 patients diagnosed with CHD screened for anxiety, depression, and quality of life(QOL) at baseline received 0.4 g of GXDSDP treatment 3 times per day and returned for monthly reassessment. Patients were recruited after stable treatment for CHD and received assessment of General Anxiety Disorder-7(GAD-7), Patient Health Questionnaire-9(PHQ-9), and Seattle Angina Questionnaire(SAQ) for evaluating anxiety, depression, and QOL.Patients were followed up 3 times, once every 4 weeks, during outpatient visits for 12 weeks. Results: At the third follow-up(F3), the anxiety symptom of 63.79%(673/1,055) of the patients improved to sub-clinical level, and the GAD-7 score improved significantly(8.11 vs. 3.87, Pmay be a potential treatment option for CHD patients comorbid with anxiety or depression.(Registration No. ChiCTR2100051523)
