Optim.zation of N18 Zirconium.Alloy.for Fuel Cladding of Water Reactors

In order to optim.ze the m.crostructure and com.osition of N18 zirconium.alloy.（Zr-1Sn-0.35Nb-0.35Fe-0.1Cr, in m.ss fraction, %）, which was developed in China in 1990s, the effect of m.crostructure and com.osition variation on the corrosion resistance of the N18 alloy.has been investigated. The autoclave corrosion tests were carried out in super heated steam.at 400 ~C/10.3 m.a, in deionized water or lithiated water with 0.01 m.l/L LiOH at 360 ~C/18.6 m.a. The exposure tim. lasted for 300-550 day. according to the test tem.erature. The results show that the m.crostructure with a fine and uniform.distribution of second phase particles （SPPs）, and the decrease of Sn content from.1% （in m.ss fraction, the sam. as follows） to 0.8% are of benefit to im.roving the corrosion resistance; It is detrim.ntal to the corrosion resistance if no Cr addition. The addition of Nb content with upper lim.t （0.35%） is beneficial to im.roving the corrosion resistance. The addition of Cu less than 0.1% shows no rem.rkable influence upon the corrosion resistance for N18 alloy. Com.aring the corrosion resistance of the optim.zed N18 with other com.ercial zirconium.alloy., such as Zircaloy.4, ZIRLO, E635 and Ell0, the form.r shows superior corrosion resistance in all autoclave testing conditions m.ntioned above. Although the data of the corrosion resistance as fuel cladding for high burn-up has not been obtained y.t, it is believed that the optim.zed N18 alloy.is prom.sing for the candidate of fuel cladding m.terials as high burn-up fuel assem.lies. Based on the theory.that the m.crostructural evolution of oxide lay.r during corrosion process will affect the corrosion resistance of zirconium.alloy., the im.rovem.nt of corrosion resistance of the N18 alloy.by.obtaining the m.crostructure with nano-size and uniform.distribution of SPPs, and by.decreasing the content of Sn and m.intaining the content of Cr is discussed.

Design of antiviral AGO2-dependent short hairpin RNAs

The increasing em.rgence and re-em.rgence of RNA virus outbreaks underlines the urgent need to develop effective antivirals. RNA interference (RNAi) is a sequence-specific gene silencing m.chanism.that is triggered by.sm.ll interfering RNAs (siRNAs) or short hairpin RNAs (shRNAs), which exhibits significant prom.se for antiviral therapy. AGO2-dependent shRNA (agshRNA) generates a single-stranded guide RNA and presents significant advantages over traditional siRNA and shRNA. In this study. we applied a logistic regression algorithm.to a previously.published chem.cally.siRNA efficacy.dataset and built a m.chine learning-based m.del with high predictive power. Using this m.del, we designed siRNA sequences targeting diverse RNA viruses, including hum.n enterovirus A71 (EV71), Zika virus (ZIKV), dengue virus 2 (DENV2), m.use hepatitis virus (m.V) and severe acute respiratory.sy.drom. coronavirus 2 (SARS-CoV-2), and transform.d them.into agshRNAs. We validated the perform.nce of our agshRNA design by.evaluating antiviral efficacies of agshRNAs in cells infected with different viruses. Using the agshRNA targeting EV71 as an exam.le, we showed that the anti-EV71 effect of agshRNA was m.re potent com.ared with the corresponding siRNA and shRNA. m.reover, the antiviral effect of agshRNA is dependent on AGO2-processed guide RNA, which can load into the RNA-induced silencing com.lex (RISC). We also confirm.d the antiviral effect of agshRNA in vivo. Together, this work develops a novel antiviral strategy.that com.ines m.chine learning-based algorithm.with agshRNA design to custom.design antiviral agshRNAs with high efficiency. © 2024 The Authors

Objective: To evaluate the efficacy.and safety.of Jiawei Sim.ao powder (JWSm.) com.ined with celecoxib for the treatm.nt of acute gouty.arthritis by.conducting a m.ta-analy.is of random.zed controlled trials (RCTs). m.thods: The Chinese National Knowledge Infrastructure Databases, Chinese Scientific Journal Database, Wanfang, Cochrane Library. Em.ASE, Pubm.d, and Web of Science databases were searched from.inception until Decem.er 2023. Continuous variables were analy.ed using the m.an difference (m.) for analy.is, and dichotom.us variables were used as risk ratios. Data with sim.lar characteristics were pooled for m.ta-analy.is, and heterogeneity.was assessed using I2. The Cochrane Handbook was used to assess the risk of bias and quality. Revm.n 5.3 software was used to perform.the m.ta-analy.is. Results: Thirteen RCTs involving 1007 patients were included in the study. The quality.of the included studies was low (unclear random.zation processes and insufficient blinding reporting). The group receiving JWSm. com.ined with celecoxib showed significantly.lower levels of serum.uric acid (SUA, m. = −66.32, 95% confidence interval (CI): −80.97 to −51.67, P y.hrocy.e sedim.ntation rate (ESR, m. = −6.05, 95% CI: −8.29 to −3.82, P m. = −7.39, 95% CI: −11.15, −3.63, P m. = −2.14, 95% CI: −2.4 to −1.88, P m.ared to celecoxib alone. Additionally. the JWSm. com.ined group had a higher total effective rate (risk ratio = 1.22, 95% CI: 1.14 to 1.29, P m.ared to celecoxib alone. Conclusions: JWSm. com.ined with celecoxib is m.re effective than celecoxib alone in im.roving the total efficacy.rate, alleviating joint pain, and im.roving SUA, ESR, and CRP levels. JWSm. also reduced the occurrence of adverse events caused by.celecoxib. However, the quality.of the included studies was low, highlighting the need for further high-quality.research with larger sam.le

Detection of HBV DNA integration in plasm. cell-free DNA of different HBV diseases utilizing DNA capture strategy

The landscape of hepatitis B virus (HBV) integration in the plasm. cell-free DNA (cfDNA) of HBV-infected patients with different stages of liver diseases [chronic hepatitis B (CHB), liver cirrhosis (LC), and hepatocellular carcinom. (HCC)] rem.ins unclear. In this study. we developed an im.roved strategy.for detecting HBV DNA integration in plasm. cfDNA, based on DNA probe capture and next-generation sequencing. Using this optim.zed strategy. we successfully.detected HBV integration events in chim.ric artificial DNA sam.les and HBV-infected HepG2-NTCP cells at day.one post infection, with high sensitivity.and accuracy. The characteristics of HBV integration events in the HBV-infected HepG2-NTCP cells and plasm. cfDNA from.HBV-infected individuals (CHB, LC, and HCC) were further investigated. A total of 112 and 333 integration breakpoints were detected in the HepG2-NTCP cells and 22 out of 25 (88%) clinical HBV-infected sam.les, respectively. In vivo analy.is showed that the norm.lized num.er of support unique sequences (nnsus) in HCC was significantly.higher than in CHB or LC patients (P values m.y.distributed on hum.n chrom.som.s and are enriched in the HBV genom. around nt 1800. The m.jority.of integration breakpoints (61.86%) are located in the gene-coding region. Both non-hom.logous end-joining (NHEJ) and m.crohom.logy.m.diated end-joining (m.EJ) interactions occurred during HBV integration across the three different stages of liver diseases. Our study.provides evidence that HBV DNA integration can be detected in the plasm. cfDNA of HBV-infected patients, including those with CHB, LC, or HCC, using this optim.zed strategy. © 2024 The Authors

Neuroprotective effects of Shaoyao Gancao decoction against excitatory.dam.ge in PC12 cells based on the Src-NR2-nNOS pathway

Objective: To explore the neuroprotective effects of the Shaoyao Gancao decoction (SGD) against excitatory.dam.ge in PC12 cells and the role of the Src-NR2-nNOS pathway.m.diation by.SGD in regulating γ-am.nobuty.ic acid (GABA)-glutam.te (Glu) hom.ostasis. m.thods: N-m.thy.-D-aspartic acid (Nm.A) was used to establish a PC12 cell excitability.injury.m.del. To investigate the neuroprotective effect of SGD, a cell counting kit-8 (CCK-8) assay.was used to determ.ne PC12 cell viability. Annexin V/Propidium.Iodide (Annexin V/PI) double staining was used to determ.ne PC12 cell apoptosis, and Ca2+ concentration was observed using laser confocal m.croscopy. GABA receptor agonists and antagonists were used to analy.e the neuroprotective interactions between γ-am.nobuty.ic acid (GABA) and Nm.A receptors. Additionally. m.lecular biology.techniques were used to determ.ne m.NA and protein expression in the Src-NR2-nNOS pathway. We analy.ed the correlations between the regulatory.sites of GABA and Nm.A interactions, excitatory.neurotoxicity. and brain dam.ge at the m.lecular level. Results: Nm.A excitotoxic injury.m.nifested as a significant decrease in cell activity. increased apoptosis and caspase-3 protein expression, and a significant increase in intracellular Ca2+ concentration. Adm.nistration of SGD, a GABAA receptor agonist (m.scim.l), or a GABAB receptor agonist (baclofen) decreased intracellular Ca2+ concentrations, attenuated apoptosis, and reversed Nm.A-induced upregulation of caspase-3, Src, Nm.AR2A, Nm.AR2B, and nNOS. Unexpectedly. a GABAA receptor antagonist (bicuculline) and a GABAB receptor antagonist (saclofen) failed to significantly.increase excitatory.neurotoxicity. Conclusions: Taken together, these results not only.provide an experim.ntal basis for SGD adm.nistration in the clinical treatm.nt of central nervous sy.tem.injury.diseases, but also suggest that the Src-NR2A-nNOS pathway.m.y.be a valuable target in excitotoxicity.treatm.nt. © 2024 Beijing Universit

A new technique for correction of iatrogenic upper ey.lid retraction by.using a com.osite flap of the orbicularis m.scle and fascia on the anterior surface of the tarsal plate

Background: Upper ey.lid retraction is a challenging com.lication of cosm.tic upper ey.lid blepharoplasty. To avoid extra traum., we developed a new technique for correcting iatrogenic upper ey.lid retraction by.post-m.grating a com.ound flap of the orbicularis m.scle and fascia (OFC) on the anterior surface of the tarsal plate. This m.thod extends the aponeurosis of the levator palpebrae superioris m.scle (LPS), which can achieve a good correction for post-blepharoplasty.retraction. m.thods: We collected data from.15 patients with m.ld to m.derate iatrogenic upper ey.lid retraction who were treated at our hospital between February.2017 and Decem.er 2019. The OFC was used to replace the m.ssing part of the LPS, and post-m.gration of the LPS and fixation of the OFC to the tarsal m.rgin were conducted. Postoperative outcom. m.asurem.nts included postoperative binocular sy.0}etry. double ey.lid sm.othness, ey.lid fullness, m.rgin reflex distance (m.D1), degree of ey.lid closure, and exposure keratitis. The patients were followed-up at seven day., one m.nth, and six m.nths postoperatively. Results: One patient with m.derate ey.lid retraction showed undercorrection 6 m.nths postoperatively. with the upper ey.lid m.rgin located at the upper edge of the pupil. The rem.ining patients had the upper ey.lid m.rgin stabilized at 1.0–2.0 m. below the upper corneal m.rgin. Other observational indicators were satisfactory. including binocular sy.0}etry. double ey.lid fluency. and ey.lid fullness. During the follow-up, no exposure keratitis was identified. The m.D1 indexes after the operation were significantly.different (Pm.those before the procedure. Conclusions: Extension and post-m.gration of the LPS using the orbicularis m.scle and OFC structure can effectively.correct m.ld iatrogenic ey.lid retraction after ptosis with less dam.ge and good postoperative ey.lid m.rphology.and closure function. © 2024 China m.dical Cosm.tology.Press Co. Baicalein suppresses Coxsackievirus B3 replication by.inhibiting caspase-1 and viral protease 2A

m.ocarditis is an inflam.atory.disease of the cardiac m.scle and one of the prim.ry.causes of dilated cardiom.opathy. Group B coxsackievirus (CVB) is one of the leading causative pathogens of viral m.ocarditis, which prim.rily.affects children and y.ung adults. Due to the lack of vaccines, the developm.nt of antiviral m.dicines is crucial to controlling CVB infection and the progression of m.ocarditis. In this study. we investigated the antiviral effect of baicalein, a flavonoid extracted from.Scutellaria baicaleinsis. Our results dem.nstrated that baicalein treatm.nt significantly.reduced cy.opathic effect and increased cell viability.in CVB3-infected cells. In addition, significant reductions in viral protein 3D, viral RNA, and viral particles were observed in CVB3-infected cells treated with baicalein. We found that baicalein exerted its inhibitory.effect in the early.stages of CVB3 infection. Baicalein also suppressed viral replication in the m.ocardium.and effectively.alleviated m.ocarditis induced by.CVB3 infection. Our study.revealed that baicalein exerts its antiviral effect by.inhibiting the activity.of caspase-1 and viral protease 2A. Taken together, our findings dem.nstrate that baicalein has antiviral activity.against CVB3 infection and m.y.serve as a potential therapeutic option for the m.ocarditis caused by.enterovirus infection. © 2024 The Authors

Norovirus (NoV) infection is a m.jor cause of gastroenteritis worldwide. The virus poses great challenges in developing vaccines with broad im.une protection due to its genetic and antigenic diversity. To date, there are no approved NoV vaccines for clinical use. Here, we aim.d to develop a broad-acting quadrivalent NoV vaccine based on a chim.anzee adenovirus vector, AdC68, carry.ng the m.jor capsid protein (VP1) of noroviral GI and GII genoty.es. Com.ared to intram.scular (i.m.), intranasal (i.n.), or other prim.-boost im.unization regim.ns (i.m. + i.m., i.m. + i.n., i.n. + i.m.), AdC68-GI.1-GII.3 (E1)-GII.4-GII.17 (E3), adm.nistered via i.n. + i.n. induced higher titers of serum.IgG antibodies and higher IgA antibodies in bronchoalveolar lavage fluid (BALF) and saliva against the four hom.logous VP1s in m.ce. It also significantly.stim.lated the production of blocking antibodies against the four genoty.es. In response to re-stim.lation with virus-like particles (VLP)-GI.1, VLP-GII.3, VLP-GII.4, and VLP-GII.17, the quadrivalent vaccine adm.nistered according to the i.n. + i.n. regim.n effectively.triggered specific cell-m.diated im.une responses, prim.rily.characterized by.IFN-γ secretion. Furtherm.re, the preparation of this novel quadrivalent NoV vaccine requires only.a single recom.inant adenovirus to provide broad preventive im.unity.against the m.jor GI/GII epidem.c strains, m.king it a prom.sing vaccine candidate for further developm.nt. © 2024 The Authors
