The current study was perform.d to evaluate the liver function status as well as m.lecular characterization of the recovered worm. in rats experim.ntally infected with F. hepatica. Sixteen m.le Wister rats aged 30 days were random.y allocated into two groups (n = 8). The first group was infected orally with 15 viable encysted m.tacercaria of F. hepatica per anim.l. The other group was kept non-infected (control group). At zero tim. (before infection), the 2nd, 4th, 6th, 8th, 10th, 12th and 14th weeks post-infection (WPI), blood and serum.sam.les were collected via puncture of retro-orbital plexus of veins from.each rat. Serum.enzym. level (AST and ALT) and total protein were m.asured, and the serum.protein profile was carried out using agarose gel electrophoresis. During the period of the experim.nt, serum.ALT and AST activities and serum.total globulins significantly increased while serum.total proteins and album.n m.rkedly decreased in the infected group. On the 14th WPI, the data of the electropherogram.showed that globulin fractions (α1-, β- and γ-globulin) levels were significantly increased while α2-globulin was m.rkedly decreased in the infected group. The m.lecular analysis confirm.d the am.lification of the ITS1, ITS2 and NDI genes of F. hepatica recovered from.the infected liver of rats with am.licon sizes of 630, 510 and 560 bp, respectively. Sequencing of the am.lified ITS gene resulted in the determ.nation of 3 strains (PP108836, PP108837, and PP108838). Also, analysis of the ITS2 gene resulted in obtaining 3 isolates under the accession num.ers (PP109065, PP109066, and PP109067). In conclusion, fasciolosis in the rat m.del is suitable for routine experim.ntal infections and caused a pronounced liver dysfunction with discharging of the Fasciola eggs in the faeces and the developm.nt of adult stages in the bile ducts. In-situ alloy developm.nt during surface processing allows for a lim.tless m.terials selection to protect com.onents exposed to severe service conditions. In fact surface alloying offers the possibility to strengthen surface com.onents with alloys that would not be possible to process otherwise. This work used Plasm. transferred arc (PTA) hardfacing for surface alloying. Different am.unts of alum.nium.powder, 5-25%, were added to a Ni based superalloy, from.Hastealloy C fam.ly, in the atom.zed form. The m.xture was hom.geneized in a ball m.ll and PTA deposited on carbon steel substrate. The influence of different processing param.ters on the final surface alloy was evaluated as current intensity and depositing velocity were varied. Coatings were characterized by optical and scanning electronic m.croscopy, X-ray diffraction and Vickers m.crohardness profiles, under a 500g load. Results showed that PTA hardfacing is an adequate surface alloying. For the conditions tested increasing hardness was obtained by solid solution for the lower am.unts of Al added and due to the new interm.tallic phases for the richer Al m.xture.

The Arabidopsis XBAT35 is one of five structurally related ankyrin repeat-containing Really Interesting New Gene （RING） E3 ligases involved in ubiquitin-m.diated protein degradation, which plays key roles in a wide range of cel- lular processes. Here, we show that the XBAT35 gene undergoes alternative splicing, generating two transcripts that are constitutively expressed in all plant tissues. The two splice variants derive from.an exon skipping event that excludes an in-fram. segm.nt from.the XBAT35 precursor m.NA, giving rise to two protein isoform. that differ solely in the presence of a nuclear localization signal （NLS）. Transient expression assays indicate that the isoform.lacking the NLS localizes in the cytoplasm.of plant cells, whereas the other is targeted to the nucleus, accum.lating in nuclear speckles. Both isoform. are functional E3 ligases, as assessed by in vitro ubiquitination assays. Two insertion m.tant alleles and RNA-interference （RNAi） silencing lines for XBAT35 display no evident phenotypes under norm.l growth conditions, but exhibit hyper- sensitivity to the ethylene precursor 1-am.nocyclopropane-l-carboxylate （ACC） during apical hook exaggeration in the dark, which is rescued by an inhibitor of ethylene perception. Independent expression of each XBAT35 splice variant in the m.tant background indicates that the two isoform. m.y differentially contribute to apical hook form.tion but are both functional in this ethylene-m.diated response. Thus, XBAT35 defines a novel player in ethylene signaling involved in negatively regulating apical hook curvature, with alternative splicing controlling dual targeting of this E3 ubiquitin ligase to the nuclear and cytoplasm.c com.artm.nts.
