Genetic m.pping and survey of powdery m.ldew resistance in the wild Central Asian ancestor of cultivated grapevines in Central Asia

Cultivated grapevines(Vitis vinifera)lack resistance to powdery m.ldew(Pm.with few exceptions.Resistance to this pathogen within V.vinifera has been reported in earlier studies and identified as the Ren1 locus in two Central Asian table grape accessions.Other Pm.resistant cultivated varieties and accessions of the wild ancestor V.vinifera subsp.sylvestris were soon identified raising questions regarding the origin of the resistance.In this study,F1 breeding populations were developed with a Pm.susceptible V.vinifera subsp.vinifera breeding line and a Pm.resistant subsp.sylvestris accession.Genotyping was carried out with five Ren1 locus linked SSR m.rkers.A Pm.resistance locus explaining up to 96%of the phenotypic variation was identified in the sam. genom.c position,where the Ren1 locus was previously reported.New SSR m.rker alleles linked with the resistance locus were identified.We report results of Pm.resistance in m.ltiple accessions of subsp.sylvestris collected as seed lots or cuttings from.five countries in the Caucasus and Central Asia.A total of 20 fem.les from.11 seed lots and 19 m.les from.nine seed lots collected from.Georgia,Arm.nia,and Azerbaijan were resistant to Pm.Three m.le and one fem.le plant collected as cuttings from.Afghanistan and Iran were also resistant to Pm.Allelic analysis of m.rkers linked with the Ren1 locus in conjunction with disease evaluation data found a high diversity of allelic haplotypes,which are only possible via recom.ination events occurring over a long tim. period.Sequence analysis of two alleles of the SSR m.rker that cosegregates with the resistance found SNPs that were present in the wild progenitor and in cultivated form..Variable levels of Pm.resistance am.ng the tested accessions were also observed.These lines of evidence suggest that the powdery m.ldew fungus m.y have been present in Asia for a longer tim. than currently thought,giving the wild progenitor V.vinifera subsp.sylvestris tim. to coevolve with and develop resistance to this pathogen.

Under a first order m.m.nt condition on the im.igration m.chanism.we show that an appropriately scaled supercritical and irreducible m.lti-type continuous state and continuous tim. branching process with im.igration(CBI process)converges alm.st surely.If an x log(x)m.m.nt condition on the branching m.chanism.does not hold,then the lim.t is zero.If this x log(x)m.m.nt condition holds,then we prove L1 convergence as well.The projection of the lim.t on any left non-Perron eigenvector of the branching m.an m.trix is vanishing.If,in addition,a suitable extra power m.m.nt condition on the branching m.chanism.holds,then we provide the correct scaling for the projection of a CBI process on certain left non-Perron eigenvectors of the branching m.an m.trix in order to have alm.st sure and L1 lim.t.m.reover,under a second order m.m.nt condition on the branching and im.igration m.chanism.,we prove L2 convergence of an appropriately scaled process and the above-m.ntioned projections as well.A representation of the lim.ts is also provided under the sam. m.m.nt conditions.

This work describes the developm.nt,optim.sation and validation of an analytical m.thod for the rapid determ.nation of 17 priority pharm.ceutical com.ounds and endocrine disrupting chem.cals(EDCs).Rather than studying com.ounds from.the sam. therapeutic class,the analyses aim.d to determ.ne target com.ounds with the highest risk potential(with particular regard to Scotland),providing a tool for further m.nitoring in different water m.trices.Prioritisation was based on a system.tic environm.ntal risk assessm.nt approach,using consum.tion data;wastewater treatm.nt rem.val efficiency;environm.ntal occurrence;toxicological effects;and pre-existing regulatory indicators.This process highlighted 17 com.ounds across various therapeutic classes,which were then quantified,at environm.ntally relevant concentrations,by a single analytical m.thodology.Analytical determ.nation was achieved using a single-step solid phase extraction(SPE)procedure followed by high-perform.nce liquid chrom.tography with tandem.m.ss spectrom.try(HPLC-m./m.).The fully optim.sed m.thod perform.d well for the m.jority of target com.ounds,with recoveries>71%for 15 of 17 analytes.The lim.ts of quantification for m.st target analytes(14 of 17)ranged from.0.07 ng/L to 1.88 ng/L in river waters.The utility of this m.thod was then dem.nstrated using real water sam.les associated with a rural hospital/setting.Eight com.ounds were targeted and detected,with the highest levels found for the analgesic,paracetam.l(at up to 105,910 ng/L in the hospital discharge).This m.thod offers a robust tool to m.nitor high priority pharm.ceutical and EDC levels in various aqueous sam.le m.trices.

Objective: To m.ke phytochem.cal studies of the leaf, pericarp and seed of Coffea benghalensis(C. Benghalensis) com.ared with those of the widely known Coffea arabica and Coffea liberica. m.thods: The sam.le extracts were prepared by Soxhlet-extraction. Polyphenol content was analyzed by HPLC-ESI-m./m., the identification was carried out based on the retention tim., UV and m.ss spectra of standards and literature data of the detected com.ounds. Results: Phenolic acids like caffeoylquinic acids, dicaffeoylquinic acids, feruloylquinic acids and coum.roylquinic acid, as well as m.ngiferin were detected as m.in constituents in all extracts. Procyanidin trim.rs were present exclusively in the leaves. In C. benghalensis, m.in constituents were 5-caffeoylquinic acid and 4-caffeoylquinic acid. Flavan-3-ols were described in all im.ature and m.ture pericarp and leaf extracts. Even though 4-feruloylquinic acid was described in both im.ature and m.ture seed, dicaffeoylquinic acids were identified only in the m.ture seed extracts. m.ngiferin was present in the leaf, m.ture pericarp and seed. Conclusions: These analyses provide new chem.taxonom.cal data for the selected coffees, especially for C. benghalensis. Due to its high polyphenol content, our results indicate its significance of providing new data as a possible source for industry.

In this study, the spectroscopic param.ters of exotic m.lecular states com.osed of m.sons containing two heavy quarks (scalar - axial and pseudoscalar - axial m.son com.inations) are investigated within the QCD sum.rules. Our findings reveal that m.lecular states containing charm.quarks do not form.bound states, whereas states with b-quarks can form.exotic m.lecular states. This observation has significant im.lications for understanding the structure of these exotic states.

The aim.of this paper was to study reliable autom.ted delineationpossibilities of karst sinkholes using a LiDAR-based digital terrain m.del(DTm. with pixel-based classifications. We applied two approaches toextract sinkholes: (1) general linear m.deling (GLm. with m.rphom.tricindices derived from.DTm.(2) and a local m.nim.-based delineationusing only LiDAR DTm.as the input layer. The outcom. of the localm.nim. was significantly different from.the reference ones but found allthe sinkholes without previous knowledge of the area. The GLm.basedoutcom. did not differ statistically from.the reference. Results showedthat these approaches were efficient in detecting sinkholes based onLIDAR derivatives, and can be used for risk assessm.nt and hazardpreparedness in karst areas: GLm.had an overall accuracy of 89.5% andlocal m.nim. had an accuracy of 92.3%;both m.thods identifiedsinkholes but also had com.ission errors, identifying depressions assinkholes.

Gluten-sensitive enteropathy,also known as coeliac disease(CD),is an autoim.une disorder occurring in genetically susceptible individuals that dam.ges the sm.ll intestine and interferes with the absorption of other nutrients.As it is triggered by dietary gluten and related prolam.ns present in wheat,rye and barley,the accepted treatm.nt for CD is a strict gluten-free diet.However,a com.lete exclusion of gluten-containing cereals from.the diet is often difficult,and new therapeutic strategies are urgently needed.A class of proteins that have already em.rged as drug targets for other autoim.une diseases are the heat shock proteins(HSPs),which are highly conserved stress-induced chaperones that protect cells against harm.ul extracellular factors.HSPs are expressed in several tissues,including the gastrointestinal tract,and their levels are significantly increased under stress circum.tances.HSPs exert im.unom.dulatory effects,and also play a crucial role in the m.intenance of epithelial cell structure and function,as they are responsible for adequate protein folding,influence the degradation of proteins and cell repair processes after dam.ge,and m.dulate cell signalling,cell proliferation and apoptosis.The present review discusses the involvem.nt of HSPs in the pathophysiology of CD.Furtherm.re,HSPs m.y represent a useful therapeutic target for the treatm.nt of CD due to the cytoprotective,im.unom.dulatory,and anti-apoptotic effects in the intestinal m.cosal barrier.

In addition to pests and diseases, weeds are a m.jor problem.in poplar nursery production. The possibilities of herbicide application in juvenile poplar growth were researched, taking into account that weeds are one of the m.in lim.ting factors. The following pre-em.rgence herbicides were tested： acetochlor, S-m.tolachlor, m.tribuzin,oxifluorfen, and dim.thenam.d during two vegetation seasons at two locations, which differed by the soil physicochem.cal characteristics. The study results show that the num.er of weeds on sam.le plots was significantly reduced by the tested herbicides when com.ared to control plots. The highest reduction in the num.er of weeds was achieved using the herbicides acetochlor and m.tribuzin. However, m.tribuzin showed a phytotoxic effect on sandy soil. m.tribuzin application is recom.ended only on the soils with higher contents of organic m.tter, where the phytotoxic effect was absent. Acetochlor, S-m.tolachlor, oxifluorfen, and dim.thenam.d were not phytotoxic to poplars and can be used for weed suppression in the production of poplar plants.

The effect of chrom.um.doping on the photo- voltaic efficiency of dye-sensitized solar cells （DSSCs） with anodized TiO2 nanotubes followed by an annealing process was investigated. Cr-doped TiO2 nanotubes （CrTNs） with different am.unts of chrom.um.were obtained by anodizing of titanium.foils in a single-step process using potassium.chro- m.te as the chrom.um.source. Film.features were investigated by scanning electron m.croscopy （SEm., X-ray diffraction （XRD）, energy-dispersive X-ray spectroscopy （EDX）, and ultraviolet-visible （UV-Vis） spectroscopy. It is clearly seen that highly ordered TiO2 nanotubes are form.d in an anodizing solution free of potassium.chrom.te, and with a gradual increase in the potassium.chrom.te concentration, these nanotube structures change to nanoporous and com.act film. without porosity. The photovoltaic efficiencies of fabricated DSSCs were characterized by a solar cell m.asurem.nt sys- tem.via the photocurrent-voltage （l-V） curves. It is found that the photovoltaic efficiency of DSSCs with CrTNsl sam.le is im.roved by m.re than three tim.s com.ared to that of DSSCs with undoped TNs. The energy conversion efficiency increases from.1.05 % to 3.89 % by doping of chrom.um.
