In vitro and in vivo evaluation of.the antiangiogenic activities of.Trigonella f.enum.graecum.extracts

Objective: To assess the antiangiogcnic activity of.***: Dif.erent f.actions of.f.nugreek crude extracts were prepared and their antiangiogenic properties were assessed using the ex vivo rat aortic ring assay and in vivo chicken em.ryo chorioallantoic m.m.rane(CAm. assay. They were investigated f.r their direct cytotoxic activity in the m.f. cells using the m.T ***: The ethanol extract showed 100% inhibition of.blood vessel outgrowth f.om.prim.ry tissue explants in the rat aortic ring assay at a concentration of.100μg/m. while the other extracts did not show signif.cant antiangiogenic activity. The ethanol extract was theref.re investigated at varying concentrations and exhibited a signif.cant dose dependent ef.ect. The CAm.assay coincided with the results of.the aortic ring assay as ethanol extract showed a signif.cant inhibition of.f.rm.tion of.new blood vessels. The extracts only showed anti-prolif.rative activity at the highest concentration of.400μg/m. towards m.f. breast cancer cell lines in the m.T ***: f.ndings of.the both assays conf.rm.d that the ethanol extract inhibited vascularization signif.cantly. f.rther studies on the ethanol extract would be benef.cial in isolating the active ingredient responsible f.r the inhibition.

The ternary or quaternary layered com.ounds called m.B phases are f.equently m.ntioned recently together with the well-known m.X ***,m.B phases are generally ref.rred to layered transition m.tal borides,while m.X phases are layered transition m.tal carbides and nitrides with dif.erent types of.crystal structure although they share the com.on nano-lam.nated structure *** order to prove that m.B phases can share the sam. type of.crystal structure with m.X phases and extend the com.osition window of.m.X phases f.om.carbides and nitrides to borides,two new m.B phase com.ounds Zr_(2)SeB and Hf.(2)SeB with the Cr_(2)AlC-type m.X phase(211 phase)crystal structure were discovered by a com.ination of.f.rst-principles calculations and experim.ntal verif.cation in this ***-principles calculations predicted the stability and lattice param.ters of.the two new m.B phase com.ounds Zr_(2)SeB and Hf.(2)*** they were successf.lly synthesized by using a therm.l explosion m.thod in a spark plasm. sintering(SPS)*** crystal structures of.Zr_(2)SeB and Hf.(2)SeB were determ.ned by a com.ination of.the X-ray dif.raction(XRD),scanning electron m.croscopy(SEm.,and high-resolution transm.ssion electron m.croscopy(HRTEm..The lattice param.ters of.Zr_(2)SeB and Hf.(2)SeB are a=3.64398Å,c=12.63223Åand a=3.52280Å,c=12.47804Å,*** the atom.c positions are m.at 4f.1/3,2/3,0.60288[Zr]or 0.59889[Hf.),Se at 2c(1/3,2/3,1/4),and B at 2a(0,0,0).And the atom.c stacking sequences f.llow those of.the Cr_(2)AlC-type m.X *** work opens up the com.osition window f.r the m.B phases and m.X phases and will trigger the interests of.m.terial scientists and physicists to explore new com.ounds and properties in this new f.m.ly of.m.terials.

In vivo subsurf.ce m.rphological and f.nctional cellular and subcellular im.ging of.the gastrointestinal tract with conf.cal m.ni-m.croscopy

AIm. To evaluate a newly developed hand-held conf.cal probe f.r in vivo m.croscopic im.ging of.the com.lete gastrointestinal tract in rodents. m.THODS： A novel rigid conf.cal probe （diam.ter 7 m.） was designed with optical f.atures sim.lar to the f.exible endom.croscopy system.f.r use in hum.ns using a 488 nm.single line laser f.r f.uorophore excitation, Light em.ssion was detected at 505 to 750 nm. The f.eld of.view was 475 μm.× 475 μm. Optical slice thickness was 7 μm.with a lateral resolution of.0.7 μm. Subsurf.ce serial im.ges at dif.erent depths （surf.ce to 250 μm. were generated in real tim. at 1024 × 1024 pixels （0.8 f.am.s/s） by placing the probe onto the tissue in gentle, stable contact. Tissue specim.ns were sam.led f.r histopathological correlation.RESULTS： The esophagus, stom.ch, sm.ll and large intestine and m.so, liver, pancreas and gall bladder were visualised in vivo at high resolution in n = 48 m.ce. Real tim. m.croscopic im.ging with the conf.cal m.nim.croscopy probe was easy to achieve. The dif.erent staining protocols （f.uorescein, acrif.avine, f.TC-labelled dextran and L. esculentum.lectin） each highlighted specif.c aspects of.the tissue, and in vivo im.ging correlated excellently with conventional histology. In vivo blood f.ow m.nitoring added a f.nctional quality to m.rphologic im.ging.CONCLUSION： Conf.cal m.croscopy is f.asible in vivo allowing the visualisation of.the com.lete GI tract at high resolution even of.subsurf.ce tissue structures. The new conf.cal probe design evaluated in this study is com.atible with laparoscopy and signif.cantly expands the f.eld of.possible applications to intra-abdom.nal organs. It allows im.ediate testing of.new in vivo staining and application options and theref.re perm.ts rapid transf.r f.om.anim.l studies to clinical use in patients.

The notion of.a f.zzy retract was introduced by Rodabaugh (1981). The notion of.a f.zzy pairwise retract was introduced in 2001. Som. weak f.rm. and som. strong f.rm. of.α-continuous m.ppings were introduced in 1988 and 1997. The authors extend som. of.these f.rm. to the L-f.zzy bitopological setting and construct various α-f.zzy pairwise retracts. The concept of.weakly induced spaces in the case L = [0,1] was introduced by martin (1980). Liu and Luo (1987) generalized this notion to the case that L is an arbitrary f.lattice and introduced the notion of.induced L-f.s. Several results are obtained, especially, f.r L-valued pairwise stratif.cation spaces.

Regenerative Surgery f.r the Rehabilitation of.a Patient af.er Surgery and Radiation Therapy f.r Head and Neck Cancer: A Case Report

Oral cancer is usually treated com.ining surgery, radiation therapy and chem.therapy, f.llowing ef.ective therapeutic protocols. Severe m.xillary and m.ndibular bone atrophy resulting af.er therapies are usually treated with autologous bone graf.ing procedures even though these techniques of.en present a signif.cant risk of.postoperative com.lications and disadvantages. We describe the case of.a 59-year-old wom.n presenting severe bone def.ct af.er being treated with surgery and radiotherapy f.r recurrent oral verrucous squam.us cell carcinom.. We perf.rm.d bone regeneration using Platelet-Derived Growth f.ctors (PDGf. in com.ination with autologous bone chips. Our procedure of.bone regeneration allowed the placem.nt of.dental im.lants and the achievem.nt of.a good aesthetic and f.nctional result. Regenerative surgery m.y enable the regeneration of.substantial bone def.cts. m.reover, PDGf.application decreases the risk of.im.lant f.ilure in irradiated patients.

Background: Adipocyte and osteoblast derive f.om.the sam. m.senchim.l progenitor. Age-related decrease in bone m.ss is accom.anied by an increase in m.rrow adipose tissue. Vitam.n D3 (VD3) inhibits adipogenesis in 3T3-L1 preadipocytes. Recently it has been dem.nstrated that alendronate (ALN) inhibits adipogenesis while prom.ting osteoblast dif.erentiation of.m.senchim.l stem.cells. Aim.of.the Study: To evaluate the role of.ALN on adipocyte dif.erentiation in vitro and the potential synergic role of.VD3 co-treatm.nt. Procedures: m.rine 3T3-L1 and 3T3-f.42A preadipocytes were routinely dif.erentiated in presence of.ALN and VD3 10-9 - 10-7 m.f.r 7 days and then stained with Oil Red O. The ef.ect of.these treatm.nts on m.NA expression of.the m.in m.lecular m.rkers of.adipocyte dif.erentiation (PPARγ and C/EBPα) and VD Receptor (VDR) were analyzed through RT-PCR. Results: Both ALN and VD3 showed a m.rked anti-adipogenic ef.ect on 3T3-L1 cells. Co-incubation of.ALN 10-8 m.and VD3 10-9 m.displayed no synergic ef.ect on inhibition of.adipogenesis. PPARγ m.NA expression was signif.cantly reduced by ALN and VD3. m.NA expression of.C/EBPα was reduced only by VD3 treatm.nt. An increase in VDR m.NA expression of.3T3-L1 cells was observed with both ALN and VD3. On the contrary, 3T3-f.42A cells, which are in a m.re advanced adipogenic dif.erentiation stage com.ared to 3T3-L1, did not express detectable levels of.VDR. Interestingly, adipose dif.erentiation of.3T3-f.42A was not af.ected by ALN nor VD3. These results suggest that VDR m.y represent the m.lecular target of.the anti-adipogenic ef.ect of.ALN. Conclusion: VDR plays a critical role in m.diating the anti-adipogenic ef.ect of.ALN. f.rther studies to clarif. this m.chanism.are warranted.

f.brous dysplasia(f.)is a rare,disabling skeletal disease f.r which there are no established *** evidence supports inhibiting the osteoclastogenic f.ctor receptor activator of.nuclear kappa-B ligand(RANKL)as a potential treatm.nt *** this study,we investigated the m.chanism. underlying RANKL inhibition in f. tissue and its likely indirect ef.ects on osteoprogenitors by evaluating hum.n f. tissue pre-and post-treatm.nt in a phase 2 clinical trial of.denosum.b(NCT03571191)and in m.rine in vivo and ex vivo preclinical *** analysis of.hum.n and m.use tissue dem.nstrated increased osteogenic m.turation,reduced cellularity,and reduced expression of.the pathogenic Gαs variant in f. lesions af.er RANKL *** sequencing of.hum.n and m.use tissue supported these *** interaction between osteoclasts and m.tant osteoprogenitors was f.rther assessed in an ex vivo lesion m.del,which indicated that the prolif.ration of.abnorm.l f. osteoprogenitors was dependent on *** results f.om.this study dem.nstrated that,in addition to its expected antiosteoclastic ef.ect,denosum.b reduces f. lesion activity by decreasing f. cell prolif.ration and increasing osteogenic m.turation,leading to increased bone f.rm.tion within *** f.ndings highlight the unappreciated role of.cellular crosstalk between osteoclasts and preosteoblasts/osteoblasts as a driver of.f. pathology and dem.nstrate a novel m.chanism.of.action of.denosum.b in the treatm.nt of.bone disease.
