Viral hepatitis is still a public health problem affecting several million people around the *** are polymorphonuclear cells that have a critical role in antibacterial ***,the role of neutrophils in viral infection is not fully *** using a mouse model of lymphocytic choriomeningitis virus infection-induced viral hepatitis,we observed increased neutrophil recruitment in the liver accompanied by enhanced CD8+T-cell *** neutrophils expressed high levels of immunomodulatory cytokines,such as C-X-C chemokine ligand 2,arginase-1,inducible nitric oxide synthase and interleukin(IL)-10,demonstrating immunosuppressive *** of neutrophils in vivo by a neutralizing antibody resulted in the exacerbation of liver injury and the promotion of T-cell responses at the immune contraction ***-33 significantly induced neutrophil recruitment in the liver and attenuated liver injury by limiting effector T-cell ***,we found that IL-33 promoted the expression of arginase-1 in neutrophils through the type 2 innate lymphoid cell(ILC2)-derived ***,IL-13 increased the inhibitory effect of neutrophils on CD8+T-cell proliferation in vitro,partially through ***,we found that IL-13 induced arginase-1 expression,depending on signal transducer and activator of transcription factor 6(STAT6)***,IL-33 induced immunosuppressive neutrophils via an ILC2/IL-13/STAT6 ***,our findings shed new light on the mechanisms associated with IL-33-triggered neutrophils in the liver and suggest potential targets for therapeutic investigation in viral hepatitis.

目的证明亚马孙利什曼原虫前鞭毛体和无鞭毛体的基因表达水平。方法用RNA分离试剂盒,分别提取3种不同来源的无鞭毛体(由小鼠模型皮损组织获得的无鞭毛体、由前鞭毛体培养转化而来的无鞭毛体,以及来自J774.G8巨噬细胞株的无鞭毛体)的总RNA,以及前鞭毛体总RNA,然后用SuperScripⅡ逆转录聚合酶将其逆转录为cD-NA,再经PCR扩增无鞭毛体特异核酸酶(P-4)和前鞭毛体特异膜糖蛋白(GP-46)的特异片段,经1.5%琼脂糖凝胶电泳分析。结果3种不同来源的无鞭毛体均观察到P-4特异性条带(273bp),且密度相似,但在前鞭毛体中未观察到;在前鞭毛体中观察到高表达的GP-46特异性条带(325bp),但在3种无鞭毛体中弱表达。结论由前鞭毛体转化的无鞭毛体能高水平表达亚马孙利什曼原虫无鞭毛体P-4特异基因,可为其生物化学及免疫学研究提供无鞭毛体来源。

Type 1 interferon-induced IL-7 maintains CD8＋ T-cell responses and homeostasis by suppressing PD-1 expression in viral hepatitis

Type 1 interferon （IFN-I） promotes antigen-presenting cell maturation and was recently shown to induce hepatic IL-7 production during infection. Herein, we further explored the underlying mechanisms used by I FN-I to orchestrate antiviral immune responses in the liver. Acute viral hepatitis was induced by i.v. injection of adenovirus （Ad） in IFN-a receptor knockout （IFNAR-/-） and control mice. To disrupt signaling, monoclonal antibodies （mAbs） against IL-7 receptor alpha （IL-7Re） or PD-L1 were i.p. injected. We found that CD8＋ T cells in IFNAR-/- mice were less effective than those in control mice. The reduced T-cell function was accompanied by increased levels of PD-1 expression, apoptosis and decreased IFN-7 production. The lack of IFN-I signaling also impaired the expression of accessory molecules in both intrahepatic dendritic cell （DCs） and hepatocytes. PD-L1 was comparably and highly expressed on hepatocytes in both IFNAR-/- and control mice. Injection of PD-Ll-specific mAb in IFNAR-/- mice reversed the compromised immune responses in the liver. Further investigation showed that hepatic IL-7 elevation was less pronounced in IFNAR-/- mice compared to the controls. A treatment with recombinant IL-7 suppressed PD-1 expression on CD8＋ T cells in vitro. Accordingly, blocking IL-7R signaling in vivo resulted in increased PD-1 expression on CD8＋ T cells in Ad-infected mice. Collectively, the results suggest that IFN-I-induced hepatic IL-7 production maintains antiviral CD8＋ T-cell responses and homeostasis by suppressing PD-1 expression in acute viral hepatitis.

目的建立亚马逊利什曼原虫无鞭毛体体外培养方法,并用间接免疫荧光法分析其抗原特异性。方法小鼠皮损组织处获得的无鞭毛体(Al)、由前鞭毛体转化来的无鞭毛体(Ap)、J774.G8巨噬细胞来源的无鞭毛体(Aj)置于改良后的Schneider’sDrosophila培养基,pH4.6、33℃条件下培养,取培养物光镜下观察并用间接免疫荧光法分析。结果三种不同来源无鞭毛体体外培养物光镜检查形态相似,均与纯培养获得的无鞭毛体一样。与无鞭毛体特异性蛋白P-8和P-4特异性单克隆抗体进行间接免疫荧光分析显示,三种不同来源的无鞭毛体均出现阳性,且浓度与部位相似。而前鞭毛体与抗P-8单克隆抗体(mAb)染色呈弱阳性反应,与抗P-4单克隆抗体呈阴性反应。相反,GP-46/M-2McAb与前鞭毛体染色呈强阳性反应,与无鞭毛体仅呈弱阳性反应。结论在33℃条件下,用改良的Schneider’sDrosophila培养基进行无鞭毛体体外培养获得成功。用阶段特异性单克隆抗体,进行间接免疫荧光分析法可鉴定亚马逊利什曼原虫的无鞭毛体。

Extraction of rare earth elements from neodymium(NdFeB) magnet scrap using magnesium halides

The demand for neodymium(NdFeB) permanent magnets for electric vehicles and eco-friendly generators is ***,NdFeB magnets contain rare earth elements(REEs),which are limited in *** this study,we performed an exchange reaction between magnesium halides(fluoride and chloride) and waste NdFeB scrap and then compared the characteristics of the extracted halides *** compositions of the ternary Mg fluoride(LiF:NaF:MgF_(2)=50:40:10 in mole ratio) and chloride(LiCl:NaCl:MgCl_(2)=10:50:40 in mole ratio) salts were thermodynamically determined for achieving low eutectic *** reactions between the NdFeB scrap powder(1-2 mm) and Mg halide salts were carried out at 1073 and 873 K for the fluoride and chloride systems,respectively,in an argon *** the reaction,we separated Nd halide from the residual salt and evaluated the Ndextraction *** phase formation of the salt was analyzed using X-ray diffraction(XRD),and the extraction rate of Nd was analyzed using inductively coupled plasma optical emission spectroscopy(ICPOES).Nd was extracted in the form of Nd halide(NdF_(3) or NdCl_(3)),and the extraction rates in the fluoride and chloride systems are 98.64% and 84.59%,***,the fluoride system is more effective than the chloride system for Nd *** study provides a comprehensive comparative analysis of the effectiveness of fluo ride and chlo ride systems in extracting REEs from NdFeB magnet *** study findings can be used to develop an effective method for recycling magnet scraps.

Flow Structure and Heat Transfer Between Two Disks Rotating Independently

In the present paper, fluid flow and convective heat transfer between two co-axial disks rotating independently are dealt with mainly based on the author's recent research on that topic. Three rotational modes, i.e. co-rotation, rotor-stator, and counter-rotation, are considered. Theory of rotating non-isothermal fluids with the presence of disk rotation and thermal effects is addressed. Rotational buoyancy effects on the flow structure development are highlighted. Results of flow visualization and heat transfer measurements are discussed to explore the thermal flow mechanisms involved in the two-disk flows at various rotational and geometric conditions. Potential issues open to the future investigation are also proposed.

Dislocation characteristics and dynamic recrystallization in hot deformed AM30 and AZ31 alloys

Zn addition to Mg alloys activates non-basal slip or twinning with solute softening *** the other hand,the effects of the Zn solute on the macroscopic dislocation behavior and dynamic recrystallization are not completely ***,it is unclear ifslip can be affected by changes in the c/a ratio of solute *** study was conducted to understand the solute strengthening of Zn addition and its effects on the dislocation characteristics and dynamic recrystallization.A hot torsion test was performed on both AM30 and AZ31 alloys up to a high strain to investigate the Zn solute effect on the dislocation characteristics and dynamic *** dislocation components of the hot torsioned alloys were evaluated by X-ray line profile analysis and electron backscatter *** results showed that the Zn solutes slightly accelerate strain accumulation at the initial stages of hot deformation,which accelerated recrystallization at high *** dislocation characteristics were changed dynamically by Zn addition:fortified-type slip,dislocation arrangement and strain anisotropy *** most important point was that the dislocation characteristics were changed dramatically at the critical strain for recrystallization and high strain *** addition of Zn also acted greatly in these strain *** indicates that the rapid formation of-type slip at the serrated grain boundaries occurs at the initiation of dynamic recrystallization and the increase in the grain triple junction because grain refinement has a great influence on the dislocation characteristics at high strain.

Control of seismic and operational vibrations of rotating machines using semi-active mounts

A dual isolation problem for rotating machines consists of isolation of housing structures from the machine vibrations and protection of machines during an earthquake to maintain their functionality. Desirable characteristics of machine mounts for the above two purposes can differ significantly due to difference in nature of the excitation and performance criteria in the two situations. In this paper, relevant response quantities are identified that may be used to quantify performance and simplified models of rotating machines are presented using which these relevant response quantities may be calculated. Using random vibration approach with a stationary excitation, it is shown that significant improvement in seismic performance is achievable by proper mount design. Results of shaking table experiments performed with a realistic setup using a centrifugal pump are presented. It is concluded that a solution to this dual isolation problem lies in a semi-active mount capable switching its properties from ‘operation-optimum’ to ‘seismic-optimum’ at the onset of a seismic event.

Increasing penetration of distributed energy resources(DERs)introduced by different stakeholders,poses an immense challenge to power network *** traditional direct control of local DERs has the risk of violating preferences and privacies of stakeholders.A promising solution for supplydemand coordination is to utilize a transactive energy(TE)based energy management method to indirectly coordinate the local DERs,which enables the distribution-level energy providers,consumers,and prosumers to trade energy with each other through a transactive energy system(TES)trading *** paper provides a comprehensive review of a TES and presents a detailed classification from different perspectives,including TES participants,structure,commodity,clearing method,and solution *** presented detailed component-scale classification can be used as a reference for future TES ***,two additional market tools,i.e.,penalty mechanism and loss allocation mechanism,are discussed as future focus areas,which can be seen as necessary complements to a TES for ensuring feasibility and fairness of energy trading.
