目的:筛选与结直肠癌肝转移以奥沙利铂为基础的新辅助化疗患者疗效相关的特征性肠道菌群及粪便代谢物,分析肠道菌群与粪便代谢物之间的关系,评估相关标志物在结直肠癌新辅助化疗中的疗效预测价值。方法:病例对照研究。选取2021年10月—2022年7月在山东大学齐鲁医院就诊的34例结直肠癌肝转移患者为研究对象,根据疗效评价标准,分为化疗有效组(20例)和化疗无效组(14例),患者粪便样本进行宏基因组测序和非靶向代谢组学分析,获得与结直肠癌肝转移化疗疗效相关的菌群及代谢物特征,采用Logistic回归构建预测模型筛选可预测化疗效果的菌群及代谢标志物,绘制受试者工作特征(ROC)曲线以及生存分析曲线评价相关菌种和代谢物对新辅助化疗疗效的预测效果。结果:化疗有效组与无效组患者的肠道菌群的α和β多样性差异无统计学意义(P均>0.05)。在菌种方面,与化疗无效组比较,化疗有效组有5种菌种相对丰度上调,10种菌种相对丰度下调,差异有统计学意义(P均<0.05),其中唾液普雷沃菌能有效预测化疗效果(ROC曲线下面积为0.750,P=0.007),敏感度为80.0%,特异度为71.4%,且其丰度高水平患者(17例)总生存率低于丰度低水平患者(17例)(χ^(2)=5.239,P=0.022)。在代谢物方面,与化疗无效组比较,化疗有效组有20种代谢物上调,4种代谢物下调,差异有统计学意义(P均<0.05),其中苏氨酸和前列腺素F2α-1-乙醇酰胺可区分化疗有效与无效患者(AUC=0.743、0.707,P均<0.05),并且两者相对丰度高水平患者(17例)总生存率高于低水平患者(17例)(χ^(2)=4.748、5.407,P均<0.05)。通过筛选分析获得唾液普雷沃菌和前列腺素F2α-1-乙醇酰胺的Logistic回归模型,ROC曲线结果显示,该模型对患者的疗效具有较好的预测价值(AUC=0.836,P<0.05),敏感度为90.0%,特异度为78.6%,且模型预测生存概率高水平患者(17例)总生存率高于低水平患者(17例)(χ^(2)=9.260,P=0.002)。结论:唾液普雷沃菌和前列腺素F2α-1-乙醇酰胺可作为结直肠癌肝转移新辅助化疗效果的预测标志物,对患者预后判断具有较好的临床参考价值。

目的探讨微原纤维相关蛋白3(microfibril-associated protein 3,MFAP3)在胶质母细胞瘤(glioblastoma,GBM)中的表达与临床意义,以及对于胶质瘤干细胞(glioma stem cells,GSCs)恶性生物学行为的影响。方法利用TCGA数据和CGGA数据分析GBM中MFAP3表达情况及其与患者预后相关性;使用Western blotting检测胶质瘤干细胞系中MFAP3蛋白表达水平;探究敲减及过表达MFAP3对胶质瘤干细胞自我更新能力和表型的影响。结果MFAP3在GBM中高表达,且与患者不良预后相关。间充质型GSCs中MFAP3的表达水平高于前神经元型GSCs。MFAP3敲减后,GSCs自我更新能力减弱,间充质表型标志物CD44、YKL40蛋白水平均降低,过表达与之相反。结论MFAP3对GBM恶性生物学进展具有重要调控作用,有望成为新的GBM临床诊断标志物和治疗干预靶点。

目的评估自噬相关蛋白5(autophagy-related protein 5,ATG5)在结肠癌中的表达及其与临床病理特征的关系,分析ATG5对结肠癌细胞迁移及侵袭能力的影响。方法运用在线分析工具cProsite分析ATG5 mRNA以及ATG5蛋白在结肠癌组织及癌旁正常组织中的表达水平。使用Kaplan-Meier Plotter分析ATG5表达水平对结肠癌患者预后的影响。免疫组织化学法检测100例结肠癌组织中ATG5表达,χ^(2)和Fisher's检验分析ATG5表达水平与临床病理特征的相关性。转染质粒或小干扰过表达或敲低ATG5后,Western blotting检测转染效率及自噬相关标记物微管相关蛋白轻链3(microtubule-associated protein light chain 3,LC3)的变化,Transwell实验评估过表达或敲低ATG5后结肠癌细胞的迁移及侵袭能力。使用LinkedOmics数据库分析结肠癌中与ATG5相关的差异表达基因,并进行基因本体(gene ontology,GO)分析和京都基因与基因组百科全书(kyoto encyclopedia of genes and genomes,KEGG)分析。结果结肠癌组织中ATG5 mRNA及蛋白的表达水平均低于癌旁正常组织(P<0.001)。低表达ATG5的结肠癌患者无复发生存期明显短于高表达ATG5的结肠癌患者(P<0.001)。免疫组化结果表明,结肠癌组织中ATG5的表达水平与淋巴结转移相关,低表达ATG5的结肠癌患者更容易发生淋巴结转移(P=0.027)。Western blotting及Transwell实验显示在SW1116细胞中过表达ATG5后,LC3Ⅱ/LC3Ⅰ增加,细胞的迁移侵袭能力减弱(P_(ATG5)=0.001;P_(LC3Ⅱ/LC3Ⅰ)=0.04;P_(迁移)<0.001,P_(侵袭)<0.001);反之,在DLD1细胞中敲低ATG5后,LC3Ⅱ/LC3Ⅰ降低,细胞的迁移侵袭能力增加(P_(ATG5#1)=0.021,P_(ATG5#2)<0.001;P_(LC3Ⅱ/LC3Ⅰ#1)=0.013,P_(LC3Ⅱ/LC3Ⅰ#2)=0.02;P迁移<0.001,P侵袭<0.001)。ATG5相关的差异表达基因富集分析结果显示,结肠癌中ATG5可能通过影响DNA损伤反应、染色质重排及Notch信号通路等途径影响结肠癌的转移。结论ATG5在结肠癌中低表达,并与患者淋巴结转移及预后显著相关,结肠癌细胞中ATG5能够增加细胞自噬水平并抑制细胞侵袭转移,提示ATG5及其调控的自噬过程可能成为结肠癌临床治疗的新靶点。

Improvement of Microstructure and Mechanical Properties of Rapid Cooling Friction Stir-welded A1050 Pure Aluminum

Two-mm thick A1050 pure aluminum plates were successfully joined by conventional and rapid cooling friction stir welding(FSW), respectively. The microstructure and mechanical properties of the welded joints were investigated by electron backscatter diffraction characterization, Vickers hardness measurements, and tensile testing. The results showed that liquid CO_(2) coolant significantly reduced the peak temperature and increased the cooling rate, so the rapidly cooled FSW joint exhibited fine grains with a large number of dislocations. The grain refinement mechanism of the FSW A1050 pure aluminum joint was primarily attributed to the combined effects of continuous dynamic recrystallization, grain subdivision, and geometric dynamic recrystallization. Compared with conventional FSW, the yield strength, ultimate tensile strength, and fracture elongation of rapidly cooled FSW joint were significantly enhanced, and the welding efficiency was increased from 80% to 93%. The enhanced mechanical properties and improved synergy of strength and ductility were obtained due to the increased dislocation density and remarkable grain refinement. The wear of the tool can produce several WC particles retained in the joint, and the contribution of second phase strengthening to the enhanced strength should not be ignored.

Objective and Impact Statement:We describe an electroenzymatic mediator(EM)sensor based on an electroenzymatic assembly peak separation strategy,which can efficiently realize the simultaneous detection of 3 typical cardiovascular disease(CVD)metabolites in 5μl of plasma under one *** work has substantial implications toward improving the efficiency of chronic CVD ***:Monitoring CVD of metabolites is strongly associated with disease *** and time-consuming detection in hospitals is unfavorable for chronic CVD ***:The EM was flexibly designed by the cross-linking of electron mediators and enzymes,and 3 EM layers with different characteristics were assembled on one *** were transferred under tunable potential;3 metabolites were quantitatively detected by 3 peak currents that correlated with metabolite ***:In this study,the EM sensor showed high sensitivity for the simultaneous detection of 3 metabolites with a lower limit of 0.01 *** linear correlation between the sensor and clinical was greater than 0.980 for 242 patients,and the consistency of risk assessment was 94.6%.Conclusion:Metabolites could be expanded by the EM,and the sensor could be a promising candidate as a home healthcare tool for CVD risk assessment.

The frequency-dependent electrical properties and strain self-sensing behaviour of ultra-high performance concrete(UHPC)as cement-based stress/strain self-sensing(CBSS)smart materials were investigated in the frequency range from 100 Hz to 300 *** using the electrical parameters of the equivalent electric circuit model,the quantitative relations of capacitance and conductance of CBSS with the measurement frequency were *** capacitance and the conductance exhibit power-law type dependence on the measurement *** calculated capacitance values at frequencies beyond 2 kHz and conductance values are consistent with the experimental *** sweep-frequency test and the fixed-frequency test were performed to examine effects of the excitation frequencies on strain self-sensing properties of *** fractional change in capacitance(FCC)and resistance(FCR)of CBSS are frequency-dependent in the frequency range from 100 Hz to the f_(B),but frequency-independent in the frequency range from the f_(B)to 300 *** f_(A)and the f_(B)are 1.7-4.0 kHz and 11-78 kHz depending on the fiber dosages,*** and FCR reach their maximum at the f_(A)and 100 Hz,*** responses of capacitance and resistance of CBSS to strain show good repeatability during cyclic *** the fiber dosage increases,capacitance-based sensitivity to strain increases initially and then decreases at the f_(A),and resistance-based sensitivity to strain of CBSS increases with increasing fiber contents.

In this paper,we aim to design a practical low complexity low-density parity-check(LDPC)coded scheme to build a secure open channel and protect information from *** this end,we first propose a punctured LDPC coded scheme,where the information bits in a codeword are punctured and only the parity check bits are transmitted to the *** further propose a notion of check node type distribution and derive multi-edge type extrinsic information transfer functions to estimate the security performance,instead of the well-known weak metric bit error *** optimize the check node type distribution in terms of the signal-to-noise ratio(SNR)gap and modify the progressive edge growth algorithm to design finite-length *** results show that our proposed scheme can achieve a lower computational complexity and a smaller security gap,compared to the existing scrambling and puncturing schemes.

Super-resolution reconstruction in medical imaging has become more demanding due to the necessity of obtaining high-quality images with minimal radiation dose,such as in low-field magnetic resonance imaging(MRI).However,image super-resolution reconstruction remains a difficult task because of the complexity and high textual requirements for diagnosis *** this paper,we offer a deep learning based strategy for reconstructing medical images from low resolutions utilizing Transformer and generative adversarial networks(T-GANs).The integrated system can extract more precise texture information and focus more on important locations through global image matching after successfully inserting Transformer into the generative adversarial network for picture ***,we weighted the combination of content loss,adversarial loss,and adversarial feature loss as the final multi-task loss function during the training of our proposed model *** comparison to established measures like peak signal-to-noise ratio(PSNR)and structural similarity index measure(SSIM),our suggested T-GAN achieves optimal performance and recovers more texture features in super-resolution reconstruction of MRI scanned images of the knees and belly.

Forty-five years after the Apollo and Luna missions, China’s Chang’e-5 (CE-5) mission collected ∼1.73 kg of new lunar materials from one ofthe youngest basalt units on the Moon. The CE-5 lunar samples provide opportunities to address some key scientific questions related to theMoon, including the discovery of high-pressure silica polymorphs (seifertite and stishovite) and a new lunar mineral, changesite-(Y). Seifertitewas found to be coexist with stishovite in a silica fragment from CE-5 lunar regolith. This is the first confirmed seifertite in returned lunarsamples. Seifertite has two space group symmetries (Pnc2 and Pbcn) and formed from an α-cristobalite-like phase during “cold” compressionduring a shock event. The aftershock heating process changes some seifertite to stishovite. Thus, this silica fragment records different stagesof an impact process, and the peak shock pressure is estimated to be ∼11 to 40 GPa, which is much lower than the pressure condition forcoexistence of seifertite and stishovite on the phase diagram. Changesite-(Y), with ideal formula (Ca_(8)Y)◻Fe^(2+)(PO_(4))_(7) (where ◻ denotes avacancy) is the first new lunar mineral to be discovered in CE-5 regolith samples. This newly identified phosphate mineral is in the form ofcolumnar crystals and was found in CE-5 basalt fragments. It contains high concentrations of Y and rare earth elements (REE), reaching upto ∼14 wt. % (Y,REE)2O3. The occurrence of changesite-(Y) marks the late-stage fractional crystallization processes of CE-5 basalts combinedwith silicate liquid immiscibility. These new findings demonstrate the significance of studies on high-pressure minerals in lunar materials andthe special nature of lunar magmatic evolution.
