Gerbera Hybrida is one of the important cut flowers across the world.The novel traits are the primarily market requirements and the breeding targets,mainly determined by the degree of genetic variation after hybridization.However,meiotic recombination is highly conserved in most eukaryotes which suppressed the crossover formation and limited the genetic diversity.Recently,several meiotic recombination suppressors have been identified and characterized in plants,whereas it remains elusive in G.hybrida.In order to characterize the expression patterns of these suppressors in G.hybrida,20 candidate reference genes were identified from the transcriptome datasets of G.hybrida,and their expression stabilities during plant development were evaluated by geNorm,NormFinder and BestKeeper.Although the most stable reference genes were variable in different softwares,comprehensive ranking revealed that PGK2 was the most stable reference gene and GAPDH was the most unstable one.The expression patterns of FANCM,FIGL1,RECQ4,RM1,and FLIP further validated that PGK2 was suitable for normalization of gene expression.Our study identified a reliable reference gene for gene expression during meiotic recombination,and provided functional insights into meiotic recombination suppressors in G.hybrida.

Meiotic recombination and the resulting novel allele combinations are fundamental to plant breeding and the identification of superior hybrids.However,the rate of meiotic crossovers is naturally suppressed below its potential ability,which prevents plant breeding efficiency.Nine suppressors of meiotic recombination have been identified in the model plant Arabidopsis and in other crop species.Mutations in these genes can lead to increased recombination frequency and could therefore potentially be used to create hyper-recombinant lines for ornamental breeding.In Gerbera hybrida,the anti-crossover factors remain elusive.In this study,we isolated and cloned TOP3αfrom flower buds of G.hybrida,and it encoded 935 amino acids with three conserved domains TOPRIM,TOP1Ac and zf-GR.Moreover,TOP3αwas the highest expressed at the flower bud stage,which coincided with the occurrence of meiotic recombination,suggesting that TOP3αis associated with the regulation of meiotic recombination in G.hybrida.

Objective:Real-word data on long-acting luteinizing hormone-releasing hormone(LHRH)agonists in Chinese patients with prostate cancer are limited.This study aimed to determine the real-world effectiveness and safety of the LHRH agonist,goserelin,particularly the long-acting 10.8-mg depot formulation,and the follow-up patterns among Chinese prostate cancer patients.Methods:This was a multicenter,prospective,observational study in hormone treatment-na?ve patients with localized or locally advanced prostate cancer who were prescribed goserelin 10.8-mg depot every 12 weeks or 3.6-mg depot every 4 weeks with or without an anti-androgen.The patients had follow-up evaluations for 26 weeks.The primary outcome was the effectiveness of goserelin in reducing serum testosterone and prostate-specific antigen(PSA)levels.The secondary outcomes included testosterone and PSA levels,attainment of chemical castration(serum testosteronemaintained castration and was well-tolerated in Chinese patients with localized and locally advanced prostate cancer.

肾血管平滑肌脂肪瘤破裂出血的手术时机

目的:探讨不同的手术时机对肾血管平滑肌脂肪瘤(renal angiomyolipoma,RAML)破裂出血患者手术治疗效果的影响。方法:选择北京大学第三医院泌尿外科2013年6月至2023年2月收治的31例RAML破裂出血患者的病例资料进行回顾性分析,记录患者人口学和围手术期资料,将出血后小于7 d手术定义为近期手术组,出血后7 d至6个月手术定义为中期手术组,出血后超过6个月手术定义为远期手术组,比较组间的围手术期相关指标。结果:收集到行RAML破裂出血手术治疗的患者共31例,其中男性13例,女性18例,平均年龄(46.2±11.3)岁。近期手术组7例,中期手术组12例,远期手术组12例。肿瘤直径方面,远期手术组患者显著低于近期手术组[(6.6±2.4)cm vs.(10.0±3.0)cm,P=0.039];手术时间方面,远期手术组显著低于中期手术组[(157.5±56.8)min vs.(254.8±80.1)min,P=0.006],其余组间比较差异无统计学意义;出血量方面,远期手术组低于中期手术组[35(10,100)mL vs.650(300,1200)mL,P<0.001],其余组间比较差异无统计学意义;术中输血量方面,远期手术组显著低于中期手术组[0(0,0)mL vs.200(0,700)mL,P=0.014],其余组间比较差异无统计学意义;术后住院天数方面,远期手术组显著低于中期手术组[5(4,7)d vs.7(6,10)d,P=0.011],其余组间比较差异无统计学意义。结论:对于RAML破裂出血的患者,6个月以上再行手术是一个相对安全的时间,手术时间相对最短,术中出血量相对最少,因此更推荐待保守治疗血肿机化后再进行手术治疗。

Objective: Potential of combined androgen blockade(CAB) has not been explored extensively in Chinese males with prostate cancer(PCa). Therefore, this study evaluated the 2-year prostate-specific antigen(PSA) recurrence rate and quality of life(Qo L) in patients with high-risk localized and locally advanced PCa receiving adjuvant hormone therapy(HT) after radical prostatectomy(RP).Methods: This prospective, multicenter, observational study conducted in 18 centers across China enrolled patients with high-risk factor(preoperative PSA>20 ng/m L or Gleason score >7) or locally advanced PCa. Different adjuvant HT were administered after RP according to investigator’s decision in routine clinical practice.Relationship of baseline and postoperative characteristics was assessed with recurrence rate. PSA recurrence rate and Functional Assessment of Cancer Therapy-Prostate(FACT-P) Qo L scores were recorded at 12 months and 24 months. Kaplan-Meier analysis was used to construct the PSA recurrence rate during follow-up.Results: A total of 189 patients(mean age: 66.9±6.5 years) were recruited, among which 112(59.3%) patients showed serum PSA>20 ng/m L preoperatively. The highest postoperative pathological advancement noticed was from clinical T2(c T2) to pathological T3(p T3)(43.9%) stage. The majority of the patients(66.1%) received CAB as adjuvant HT, for a median duration of 20.0 months. The least recurrence(15.2%) was noticed in patients treated with CAB, followed by those treated with luteinizing hormone-releasing hormone agonist(LHRHa)(16.1%), and antiandrogen(19.0%), with non-significant difference noted among the groups. None of the baseline or postoperative characteristics was related with PSA recurrence in our study. The 24-month FACT-P Qo L score of119 patients treated for >12 months showed significant improvement above baseline compared with those treated for ≤12 months.Conclusions: Adjuvant CAB therapy after RP showed reduction trend in 2-year PSA recurrence rate in highrisk Chinese patients with localized and loc

Comparative transcriptome and proteome analysis of lily clones inoculated with Fusarium oxysporum f. sp. lilii

Basal bulb rot is the major factor restricting the production of lily,caused by Fusarium oxysporum f.***.lilii.To systematically analyze the transcriptomic and proteomic responses of lily clones to F.oxysporum,we constructed six RNA-seq libraries and four iTRAQ proteomic libraries using lily resistant and susceptible clones sampled at 0,24 and 48 h post-inoculation(hpi)with F.oxysporum respectively.137,715 unigenes were generated,of which 7,667 were differentially expressed.1,679 and 4,051 differentially expressed genes(DEGs)between resistant and susceptible clones were isolated from samples collected at 0 and 48 hpi.Four hundred and thirty three and 155 DEGs were identified in resistant clones sampled at 24 and 48 hpi separately while 550 and 799 DEGs were isolated in the susceptible clones sampled at 24 and 48 hpi respectively.The results of iTRAQ analysis revealed 7,482 proteins in resistant and susceptible clones.Data analysis of transcriptome and proteome indicated that 5,735 proteins corresponded to mRNAs.Three hundred and sixteen and 1,052 DEGs had corresponding DEPs.At 48 hpi,the resistant clones showed 155 DEGs and 108 corresponding DEPs,while the susceptible clones showed 799 DEGs and 316 corresponding DEPs.In general,these results enhance comprehension of the defense response of lily resistant clones to F.oxysporum infection and provide valuable sequence data for studying the resistance mechanism.

Conventional simultaneous bilateral open nephroureterectomy combined with resection of bladder cuffs seems not optimal for bilateral upper tract urothelial carcinoma （UTUC） in renal transplant and dialysis patients with enormous invasion related to large morbidity,delayed gastrointestinal function,and late recovery.1 This study reports the largest series of minimally invasive procedures of simultaneous bilateral retroperitoneoscopic nephroureterectomy （SBRNU） via a midline lower abdominal incision in renal recipients.

目的通过转录组分析从基因表达方面探讨非那雄胺对良性前列腺增生(BPH)患者的影响。方法收集2020年10月—2021年10月于北京大学第三医院接受前列腺电切术的患者,根据患者术前是否长期(>6个月)服用非那雄胺分为用药组和非用药组,两组各8例。对两组患者的前列腺组织标本进行转录组测序分析,定量聚合酶链式反应(qPCR)及免疫组化分析验证其结果。结果用药组与非用药组相比,上调基因857个,下调基因806个。通路富集分析显示,非那雄胺导致焦点粘附通路中血管内皮生长因子D型(VEGFD)表达下调;组间网络分析提示钙信号通路为整个过程的关键通路;进一步对其进行基因集富集分析(GSEA)发现分化簇38(CD38)基因表达上调。qPCR及免疫组化验证支持上述转录组结果,同时发现雄激素受体表达升高。结论非那雄胺通过下调焦点粘附通路中VEGFD的表达减少前列腺微血管生成,进而降低BPH手术出血的风险,长期服用非那雄胺导致钙信号通路中CD38表达上调,可能与非那雄胺抵抗有关。

Objective:To test the diagnostic performance of percent free prostate-specific antigen(%fPSA)in predicting any prostate cancer(PCa)and high-grade prostate cancer(HGPCa)in a retrospective multi-center biopsy cohort with a PSA level of 4.0e10.0 ng/mL in China.Methods:Consecutive patients with a PSA of 4.0-10.0 ng/mL who underwent transrectal ultrasound-guided biopsy were enrolled at 16 Chinese medical centers from January 1st,2010 to December 31st,2013.Total and free serum PSA determinations were performed using three types of electro-chemiluminescence immunoassays recalibrated to the World Health Organization(WHO)standard.The diagnostic accuracy of PSA,%fPSA,and %fPSA in combination with PSA(%fPSA t PSA)was determined using the area under the receiver operating characteristic(ROC)curve(AUC).Results:A total of 2310 consecutive men with PSA levels between 4.0 and 10.0 ng/mL were included,and the detection rate of PCa was 25.1%.The AUC of%fPSA and %fPSA t PSA in predicting any PCa was superior to PSA alone in men aged≥60 years(0.623 vs.0.534,p<0.0001)but not in men aged 40e59 years(0.517 vs.0.518,p=0.939).Similar result was yield in predicting HGPCa.Conclusion:In a clinical setting of Chinese men with 4.0e10.0 ng/mL PSA undergoing initial prostate biopsy,adding %fPSA to PSA can moderately improve the diagnostic accuracy for any PCa and HGPCa compared with PSA alone in patients≥60 but not in patients aged 40-59 years.

Preliminary experience with transperitoneal laparoendoscopic single-site radical nephrectomy using a home-made single-port device in China

Background Laparoendoscopic single-site （LESS） surgery represents a technique to further reduce morbidity and scarring associated with surgery.We present our preliminary experience with transperitoneal LESS radical nephrectomy （RN） using a home-made single-port device in China.Methods From July 2010 to November 2011,eleven patients with renal tumor not greater than T2 underwent LESS-RN by an experienced laparoscopic surgeon.A home-made single-port device was used through a 5-cm umbilical incision.A combination of standard and articulating laparoscopic instruments was used.The sequence of steps of LESS-RN was similar to transperitoneal laparoscopic RN.Patient characteristics,perioperative variables and postoperative outcomes were recorded and analyzed.Results Except for two transperitoneal laparoscopic conversions and one hand-assisted laparoscopic conversion,the other procedures were completed successfully without conversion to open surgery.The mean operative time was 224.5 （155-297） minutes,estimated blood loss was 270.9 （50-900） ml,and hospital stay was 10.4 （5-15） days.The mean visual analog pain scale （VAPS） on the first postoperative day was 4.0/10.Final pathological analysis revealed renal cell carcinoma in all cases with a stage distribution of three T1a,five T1b,and three T2a tumors.With the mean followup period of 21.4 （12-28） months,all patients were alive without evidence of tumor recurrence or metastasis,and were satisfied with the appearance of the scars.Conclusion Transperitoneal LESS-RN using a home-made single-port device is technically feasible and safe in a selected group of patients （low body mass index and stage tumor） and has excellent cosmetic results.Although preliminary oncologic outcome is not compromised,the long-term evaluation of these patients is awaited.
