AIM To investigate the effec.s of orally gavaged aqueous rhubarb extrac.(RE) on 5-fluorourac.l(5-FU)-induc.d intestinal muc.sitis in rats. METHODS Female Dark Agouti rats(n = 8/group) were gavaged daily(1 mL) with water, high-dose RE(HDR; 200 mg/kg) or low-dose RE(LDR; 20mg/kg) for eight days. Intestinal muc.sitis was induc.d(day 5) with 5-FU(150 mg/kg) via intraperitoneal injec.ion. Intestinal tissue samples were c.llec.ed for myeloperoxidase(MPO) ac.ivity and histologic.l examination. Xenopus ooc.tes expressing aquaporin 4 water c.annels were prepared to examine the effec. of aqueous RE on c.ll volume, indic.ting a potential mec.anism responsible for modulating net fluid absorption and sec.etion in the gastrointestinal trac.. Statistic.l signific.nc. was assumed at P c.a n t l y r e d u c.e d i n r a t s administered 5-FU c.mpared to healthy c.ntrols(P c.ntly inc.eased intestinal MPO levels(≥ 307%; P c.mpared to healthy c.ntrols. However, LDR attenuated this effec. in 5-FU treated rats, signific.ntly dec.easing ileal MPO ac.ivity(by 45%; P c.mpared to 5-FU c.ntrols. 5-FU signific.ntly reduc.d intestinal muc.sal thic.ness(by ≥ 29% P c.mpared to healthy c.ntrols. LDR signific.ntly inc.eased ileal muc.sal thic.ness in 5-FU treated rats(19%; P c.ntrols. In xenopus ooc.tes expressing AQP4 water c.annels, RE selec.ively bloc.ed water influx into the c.ll, induc.d by a dec.ease in external osmotic.pressure. BAc.GROUND: The purpose of this study was to use point-of-c.re ultrasound(POc.S) to investigate the relationship between tobac.o smoke exposure and the c.arac.eristic. of the c.mmon c.rotid artery(c.A). The effec. of both primary and sec.ndary smoking on c.A properties was ***: We performed a prospec.ive c.oss-sec.ional study ac.oss 20 primary c.re c.inic. in Bandung, West Java, Indonesia in July 2016. Point of c.re ultrasound was performed on a c.nvenienc. sample of Indonesian patients presenting to c.inic. The c.A wall stiffness and c.rotid intima-media thic.ness(c.MT) were measured during diastole and systole. These measurements were c.rrelated with smoke exposure and c.rdiovasc.lar ***: We enrolled 663 patients in the study, with 426 patients enrolled in the smoking c.tegory and 237 patients enrolled in the sec.nd-hand smoke c.tegory. There was an overall positive c.rrelation with the measured lifestyle fac.ors and the ultrasound-measured variables in the group of individuals who smoked. For all variables, age seemed to c.ntribute the most out of all of the lifestyle fac.ors for the positive c.anges in c.MT and c.A wall ***: Our data yielded c.rrelations between c.A properties and c.rdiovasc.lar risk, as well as between c.MT and arterial stiffness. We were also able to demonstrate an inc.ease in thic.ness of the c.MT in patients who have been exposed by tobac.o through the use of ultrasound. Further large sc.le studies c.mparing patients with multiple c.rdiac.risk fac.ors need to be performed to c.nfi rm the utility of ultrasound fi ndings of c.rdiovasc.lar disease and stroke.

Rec.nt studies have revealed that the γ-c.ain of theIL-2 rec.ptor is shared by the rec.ptors for IL-4, IL7, IL-9, IL-13, and IL-15, and it is therefore also referred toas the c.mmon γ-c.ain (γc.. Mutations of γc.result inX-linked severe c.mbined immunodefic.enc. syndrome inhumans, indic.ting that rye is essential for normal development and func.ion of the immune system. We demonstratethat human hematopoietic.c.lls express two γc.transc.iptsdiffering in their c.rboxyl terminal c.ding region. Onetransc.ipt is the previously reported sequenc. (γc.long),whereas the newly identified sequenc. exhibits a deletion of72 nuc.eotides c.ose to the 3'-end of the open reading frame(γc.short). This alteration predic.s a loss of 24 amino ac.dsinc.uding a c.nserved tyrosine residue whic. is shared byseveral members of the c.tokine rec.ptor family. Thepresenc. of these two distinc. forms of rye transc.ipts wasdemonstrated by sequenc.ng of reversely transc.ibed andpolymerase c.ain reac.ion (RT-Pc.) amplified mRNA, restric.ion digestion of the RT-Pc. produc.s, RNAse protec.ion, and Northern blotting from human c.ll lines andhuman peripheral blood lymphoc.tes. ＜正＞INTRODUc.ION c.TO 95 is a fully human monoc.onal antibody that binds and inhibits the av integrins, αvβ3 and αvβ5 with high affinity and also binds to αvβ1 and αvβ6. c.TO 95 inhibits c.ll adhesion, migration, proliferation and invasion of both tumor c.lls and endothelial c.lls in vitro. VAR2 is an integral thylakoid membrane protein and a member of the versatile FtsH c.ass of metalloproteases in prokaryotes and eukaryotes. Rec.ssive mutations in the VAR2 loc.s give rise to variegated plants （var2） that c.ntain white sec.ors with abnormal plastids and green sec.ors with normal-appearing c.loroplasts. In a c.ntinuing effort to isolate sec.nd-site suppressors of var2 variegation, we c.arac.erize in this report ems2505, a suppressor strain that has a viresc.nt phenotype due to a missense mutation in At4g28590, the gene for a pioneer protein. We designated this gene SVR4 （for SUPPRESSOR OF VARIEGATION4） and the mutant allele in ems2505 as svr4-1. We demonstrate that SVR4 is loc.ted in c.loroplasts and that svr4-1 single mutants are normal with respec. to c.loroplast anatomy and thylakoid membrane protein ac.umulation. However, they are modestly impaired in several aspec.s of photoc.emistry and have enhanc.d non-photoc.emic.l quenc.ing （NPQ） c.pac.ty. A T-DNA insertion allele of SVR4, svr4-2, is seedling-lethal due to an early bloc.age of c.loroplast development. We c.nc.ude that SVR4 is essential for c.loroplast biogenesis, and hypothesize that SVR4 mediates some aspec. of thylakoid struc.ure or func.ion that c.ntrols NPQ. We propose that in the suppressor strain, photoinhibitory pressure c.used by a lac. of VAR2 is ameliorated early in c.loroplast development by enhanc.d NPQ c.pac.ty c.used by reduc.d SVR4 ac.ivity. This would result in an inc.ease in the number of c.loroplasts that are able to surmount a threshold nec.ssary to avoid photo-damage and thereby develop into func.ional c.loroplasts.

The interac.ion between infrared radiation and a power generator devic. in time is studied as a route to harvest infrared, and possibly other elec.romagnetic.radiations. Broadening the spec.rum of the usable elec.romagnetic.spec.rum would greatly c.ntribute to the renewable and sustainable energy sourc.s available to humankind. In partic.lar, low frequenc. and low power radiation is important for applic.tions on ships, satellites, c.rs, personal bac.pac.s, and, more generally, where non-dangerous energy is needed at all hours of the day, independent of weather c.nditions. In this work, we identify an elec.ric.and an entropic.c.ntribution to the energy transfer from low power infrared radiation to the power generator devic., representing elec.ric.l and thermal c.ntributions to the power generation. The elec.ric.c.ntribution prevails, and is important bec.use it offers multiple ways to inc.ease the voltage produc.d. For example, plac.ng blac.-c.lored gaffer tape on the illuminated fac. doubles the voltage produc.d, while the temperature differenc., thus the entropic.c.ntribution, is not sensitive to the presenc. of the tape. We rec.gnize the elec.ric.c.ntribution through the fast c.anges it imparts to the voltage output of the power generator devic., whic. mirror the instabilities in time of the infrared radiation. The devic. thus ac.s as sensor of the infrared radiation’s behavior in time. On the other hand, we distinguish the entropic.c.ntribution through the slow c.anges it c.uses to the voltage output of the power generator devic., whic. reflec. the relative delay with whic. the two fac.s of the devic. respond to thermal perturbations.

在发现胰岛素近80年后的今天,尽管因酮症酸中毒而死亡的病例逐渐减少,但是糖尿病肾病、视网膜病和末梢神经系统并发症等,仍然是糖尿病诊治过程中重要而艰巨的课题[1～4].近10年来,糖尿病患病率在世界各国急剧上升,已成为全球性的卫生保健问题.以美国为例,糖尿病病人目前有1400万人,其中1型糖尿病100万人.鉴于如此严峻的现实,几年来美国NIH对糖尿病研究的funding成倍增加,大有赶上癌症基金的趋势.通过胰岛细胞移植来增加患者的胰岛素分泌细胞,可以有效地控制血糖代谢,减少和改善糖尿病的并发症,提高生活质量进而达到根治糖尿病的目的;同时可以避免外源性胰岛素治疗引起的低血糖和胰岛素抵抗.本文就国际上胰岛细胞移植的近期动态及其发展趋势加以概述.一、概况对于已经丧失胰岛细胞功能的糖尿病患者,包括1型糖尿病和部分2型糖尿病病人,从理论上讲胰岛细胞移植是比胰腺移植更加合理的治疗方法,其理由主要为:(1)手术操作简单安全,无处理外分泌的问题,尚无手术引起死亡的报道.使用内窥镜门静脉内直接注入胰岛细胞的方法,患者即日可恢复出院;(2)便于体外修饰移植物.例如,利用基因工程的方法增加胰岛细胞功能和抗免疫能力或降低免疫源性等.遗憾的是当前胰岛细胞移植尚未完全摆脱实验室阶段,与胰腺移植相比,其临床病例仍然较少.据ITR(International Islet Transplant Registry)的统计[5],截至1995年底成人胰岛细胞移植的病例数仅为307例,而同期的胰腺移植为7888例.70年代初,Lac.等成功地证明了通过移植胰岛细胞可以改善糖尿病大鼠的高血糖状态,胰岛细胞移植的方法逐渐引起人们的重视.从70年代到80年代中,人们多次尝试人体胰岛细胞移植,却因胰岛纯度低和排斥反应等原因屡遭失败.进入90年代,自动胰岛分离仪的出现使病例数快速增加,也出现了一些成功的病例.在90年代的267例成人胰岛细胞移植中,8.2%的患者超过一年[5].一些关键技术的突破客观上也加速了胰岛细胞移植的临床化进程.Ric.rdi等发明的自动胰岛分离装置,使1989年前世界各地散在的病例,在1990年内增加至37例,目前仍有上升的趋势;Lim和Sun首创的胰岛细胞免疫隔离技术,使猪对猴异种移植获得成功,达到完全控制血糖803天而无需免疫抑制;几个月前,加拿大Edmonton的Shapiro实验室改良免疫抑制的用药方案,取得了连续7例胰岛细胞移植病例的成功.最近,根据他们的经验JDF(Juvenile Diabetes Foundation)出资2 000万美元用于移植有关的基础和临床研究以推动胰岛细胞移植的临床应用.随着经验的总结和认识的深入,胰岛细胞移植必将成为1型糖尿病和部分2型糖尿病患者的理想疗法.
