3D-printed aerogels as theranostic implants monitored by fluorescence bioimaging

Aerogel scaffolds are nanostructured materials with beneficial properties for tissue engineering *** tracing of the state of the aerogels after their implantation is challenging due to their variable biodegradation rate and the lack of suitable strategies capable of in vivo monitoring the *** nanoparticles(UCNPs)have emerged as advanced tools for in vitro bioimaging because of their fluorescence *** this work,highly fluorescent UCNPs were loaded into aerogels to obtain theranostic implants for tissue engineering and bioimaging applications.3D-printed alginate-hydroxyapatite aerogels labeled with UCNPs were manufactured by 3D-printing and supercritical CO2 drying to generate personalize-to-patient *** physicochemical performance of the resulting structures was evaluated by printing fidelity measurements,nitrogen adsorption-desorption analysis,and different microscopies(confocal,transmission and scanning electron microscopies).Stability of the aerogels in terms of physicochemical properties was also tested after 3 years of *** was evaluated in vitro by different cell and hemocompatibility assays,in ovo and in vivo by safety and bioimaging studies using different murine *** profile,tissue index and histological evaluations of the main organs unveiled an in vivo downregulation of the inflammation after implantation of the ***-decorated aerogels were first-time manufactured and long-term traceable by fluorescence-based bioimaging until 3 weeks post-implantation,thereby endorsing their suitability as tissue engineering and theranostic nanodevices(*** implants).

Neuropsychological features of progranulin-associated frontotemporal dementia: a nested case-control study

The distinction between sporadic and genetic behavioural-variant frontotemporal dementia(bvFTD)regarding some neuropsychological(NP)features remains ***,progranulin(GRN)-associated bvFTD frequently presents with early episodic memory impairment and some degree of parietal dysfunction which are supporters of Alzheimer’s disease(AD)*** this context,we aimed to characterize the NP profile of GRN-bvFTD as compared to sporadic-bvFTD and AD in patients with mild dementia(Mini-Mental State Examination score≥17 and Clinical Dementia Rating Scale score≤*** identified 21 patients at Centro Hospitalar e Universitário de Coimbra,Portugal with GRN mutations belonging to fifteen different *** our focus was bvFTD variants,FTD-related aphasic forms(3 patients)were *** remaining 18 GRN-bvFTD were further matched with 18 sporadic-bvFTD and 18 AD patients according to disease staging,age and *** patients completed the Mini-Mental State Examination,Montreal Cognitive Assessment(MoCA)and a comprehensive NP assessment *** were converted into *** between groups in individual NP measures and NP domains were assessed through non-parametric tests(Kruskal-Wallis test analysis)and eta squared(ŋ2)was calculated as a measure of effect *** comparisons show that GRN patients have worse performances on verbal retrieval processes(P=0.039,ŋ2=0.110)and visuoconstructive abilities(P=0.039,ŋ2=0.190)than sporadic bvFTD *** compared to AD,GRN patients present a higher impairment in frontal(P=0.001,ŋ2=0.211)and parietal(P=0.041,ŋ2=0.129)measures and a better performance in memory tasks(P=0.020,ŋ2=0.120).Sporadic-bvFTD forms are worse than AD in frontal measures(P=0.032,ŋ2=0.200),being better in both memory(P=0.010,ŋ2=0.131)and visuospatial skills(P=0.023,ŋ2=0.231).Considering these results,we conclude that GRN-bvFTD patients present a NP profile that associates the typical patterns of FTD and AD ***

Cancer has nowadays become one of the leading causes of death *** anticancer approaches are associated with different ***,innovative methodologies are being investigated,and several researchers propose the use of remotely activated nanoparticles to trigger cancer cell *** idea is to conjugate two different components,i.e.,an external physical input and *** are given in a harmless dose that once combined together act synergistically to therapeutically treat the cell or tissue of interest,thus also limiting the negative outcomes for the surrounding *** both the properties of the nanomaterial and the involved triggering stimulus,it is possible furthermore to achieve not only a therapeutic effect,but also a powerful platform for imaging at the same time,obtaining a nano-theranostic *** the present review,we highlight the role of nanoparticles as therapeutic or theranostic tools,thus excluding the cases where a molecular drug is *** thus present many examples where the highly cytotoxic power only derives from the active interaction between different physical inputs and *** perform a special focus on mechanical waves responding nanoparticles,in which remotely activated nanoparticles directly become therapeutic agents without the need of the administration of chemotherapeutics or sonosensitizing drugs.

PARTIAL REGULARITY FOR THE NAVIER-STOKes-FOURIER SYSTEM

This paper addresses a nonstationary flow of heat-conductive incompressible Newtonian fluid with temperature-dependent viscosity coupled with linear heat transfer with advection and a viscous heat source term, under Navier/Dirichlet boundary conditions. The partial regularity for the velocity of the fluid is proved for each proper weak solution, that is, for such weak solutions which satisfy some local energy estimates in a similar way to the suitable weak solutions of the Navier-Stokes system. Finally, we study the nature of the set of points in space and time upon which proper weak solutions could be singular.

Reinforced concrete(RC)slabs are characterized by reduced construction time,versatility,and easier space *** structural behavior is not straightforward and,specifically,punching shear strength is a current research *** this study an experimental database of 113 RC slabs without shear reinforcement under punching loads was compiled using data available in the literature.A sensitivity analysis of the parameters involved in the punching shear strength assessment was conducted,which highlighted the importance of the flexural reinforcement that are not typically considered for punching shear *** a discussion of the current international standards,a new proposed model for punching shear strength and rotation of RC slabs without shear reinforcement was *** was based on a simplified load-rotation curve and new failure criteria that takes into account the flexural reinforcement *** experimental database was used to validate the approaches of the current international standards as well as the new proposed *** latter proved to be a potentially useful design tool.

Explicit estimates for Solutions of Nonlinear Radiation-type Problems

We establish the existence of weak solutions of a nonlinear radiation-type boundary value problem for elliptic equation on divergence form with discontinuous leading coefficient. Quantitative estimates play a crucial role on the real applications. Our objective is the derivation of explicit expres- sions of the involved constants in the quantitative estimates, the so-called absolute or universal bounds. The dependence on the leading coefficient and on the size of the spatial domain is precise. This work shows that the expressions of those constants are not so elegant as we might expect.

Stem cell-based approaches: Possible route to hearing restoration?

Disabling hearing loss is the most common sensorineural disability *** affects around 466 million people and its incidence is expected to rise to around 900 million people by 2050,according to World Health Organization *** cases of hearing impairment are due to the degeneration of hair cells(HCs)in the cochlea,mechano-receptors that transduce incoming sound information into electrical signals that are sent to the *** to these cells is mainly caused by exposure to aminoglycoside antibiotics and to some anti-cancer drugs such as cisplatin,loud sounds,age,infections and genetic *** deficits may also result from damage to the spiral ganglion neurons that innervate cochlear *** from what is observed in avian and nonmammalian species,there is no regeneration of missing sensory cell types in the adult mammalian cochlea,what makes hearing loss an irreversible *** review summarizes the research that has been conducted with the aim of developing cell-based strategies that lead to sensory cell replacement in the adult cochlea and,ultimately,to hearing *** main lines of research are discussed,one directed toward the transplantation of exogenous replacement cells into the damaged tissue,and another that aims at reactivating the regenerative potential of putative progenitor cells in the adult inner *** from some of the studies that have been conducted are presented and the advantages and drawbacks of the various approaches discussed.

The importance of trans-generational effects in Lepidoptera

The importance of trans-generational effects in shaping an individuals＇ phenotype and fitness, and consequently even impacting population dynamics is increasingly apparent. Most of the research on trans-generational effects still focuses on plants, mammals, and birds. In the past few years, however, increasing number of studies, especially on maternal effects, have highlighted their im- portance also in many insect systems. Lepidoptera, specifically butterflies, have been used as model systems for studying the role of phenotypic plasticity within generations. As ectotherms, they are highly sensitive to environmental variation, and indeed many butterflies show adaptive phenotypic plasticity in response to environmental conditions. Here, we synthesize what is known about trans-generational effects in Lepidoptera, compile evidence for different environmental cues that are important drivers of trans-generational effects, and point out which offspring traits are mainly impacted. Finally, we emphasize directions for future research that are needed for better understanding of the adaptive nature of trans-generational effects in Lepidoptera in particular, but potentially also in other organisms.

Genetic Identification of Quantitative Trait Loci for Contents of Mineral Nutrients in Rice Grain

In present study, Fe, Zn, Mn, Cu, Ca, Mg, P and K contents of 85 introgression lines （ILs） derived from a cross between an elite indica cultivar Teqing and the wild rice （Oryza rufipogon） were measured by inductively coupled argon plasma （ICAP） spectrometry. Substantial variation was observed for all traits and most of the mineral elements were significantly positive correlated or independent except for Fe with Cu. A total of 31 putative quantitative trait loci （QTLs） were detected for these eight mineral elements by single point analysis. Wild rice （O. rufipogon） contributed favorable alleles for most of the QTLs （26 QTLs）, and chromosomes 1,9 and 12 exhibited 14 QTLs （45%） for these traits. One major effect of QTL for zinc content accounted for the largest proportion of phenotypic variation （11%-19%） was detected near the simple sequence repeats marker RM152 on chromosome 8. The co-locations of QTLs for some mineral elements observed in this mapping population suggested the relationship was at a molecular level among these traits and could be helpful for simultaneous improvement of these traits in rice grain by marker assisted selection.
