Fibrous dysplasia(FD)is a rare,disabling.skeletal disease for which there are no established *** evidence supports inhibiting.the osteoclastog.nic factor receptor activator of nuclear kappa-B lig.nd(RANKL)as a potential treatment *** this study,we investig.ted the mechanisms underlying.RANKL inhibition in FD tissue and its likely indirect effects on osteoprog.nitors by evaluating.human FD tissue pre-and post-treatment in a phase 2 clinical trial of denosumab(NCT03571191)and in murine in vivo and ex vivo preclinical *** analysis of human and mouse tissue demonstrated increased osteog.nic maturation,reduced cellularity,and reduced expression of the pathog.nic g.s variant in FD lesions after RANKL *** sequencing.of human and mouse tissue supported these *** interaction between osteoclasts and mutant osteoprog.nitors was further assessed in an ex vivo lesion model,which indicated that the proliferation of abnormal FD osteoprog.nitors was dependent on *** results from this study demonstrated that,in addition to its expected antiosteoclastic effect,denosumab reduces FD lesion activity by decreasing.FD cell proliferation and increasing.osteog.nic maturation,leading.to increased bone formation within *** finding. hig.lig.t the unappreciated role of cellular crosstalk between osteoclasts and preosteoblasts/osteoblasts as a driver of FD patholog. and demonstrate a novel mechanism of action of denosumab in the treatment of bone disease.

The movement of ＂false antennae＂ in butterflies with ＂false head＂ wing.patterns

In many butterfly species of the family Lycaenidae, the morpholog. and color pattern of the hind wing., tog.ther with certain behaviors, sug.ests the presence of a false head （FH） at the posterior end of the perching.individual. This FH is consi- dered an adaptation to escape from visually oriented predators. A frequent component of the FH are the tails that presumably resemble the antennae, and the typical hind wing. back-and-forth movement along.the sag.ttal plane （HWM） performed while perching.apparently move the tails in a way that mimics antennal movement. By exposing.33 individuals from 18 species of Lycaenidae to a stuffed insectivorous bird, we tested two alternative hypotheses reg.rding.HWM. The first hypothesis proposes that, when the butterfly is observed at close rang., the HWM distorts the shape of the false head thus reducing.its deceiving.effect and, therefore, selection will favor butterflies that stop moving.their wing. when a predator is close by; the second hypothesis says that an increase in the frequency of HWM improves its deflective effect when the butterfly confronts a predator at close rang.. Our results tend to support the second hypothesis because half of the butterflies started to move their hind wing. or increased the rate of HWM when exposed to the stuffed bird; however a substantial proportion of butterflies （30%） stopped moving.their hind wing. or decreased the rate of HWM as expected from the first hypothesis. Our observations also showed that there is g.eat variation in the rates of HWM, and demonstrated the existence of alternative ways of producing.＂vivid＂ movement of the hind wing.tails （the ＂false antennae＂） in the absence of HWM [Current Zoolog. 61 （4）： 758-764, 2015].

Plant root-nodule symbiosis(RNS)with mutualistic nitrog.n-fixing.bacteria is restricted to a sing.e clade of ang.osperms,the Nitrog.n-Fixing.Nodulation Clade(NFNC),and is best understood in the leg.me *** species share many commonalities,explained either by diverg.nce from a common ancestor over 100 million years ag. or by converg.nce following.independent orig.ns over that same time ***,comparative analyses of diverse nodulation syndromes can provide insig.ts into constraints on nodulation—what must be acquired or cannot be lost for a functional symbiosis—and the latitude for Plant Communications g.nomic landscape of nodulation variation in the ***,much remains to be learned about nodulation,especially outside of ***,we employed a larg.-scale phylog.nomic analysis across 88 species,complemented by 151 RNA-seq libraries,to elucidate the evolution of *** phylog.nomic analyses further emphasize the uniqueness of the transcription factor NIN as a master reg.lator of nodulation and identify key muta-tions that affect its function across the *** transcriptomic assessment revealed nodule-specific upreg.lated g.nes across diverse nodulating.plants,while also identifying.nodule-specific and nitrog.n-response *** 70%of symbiosis-related g.nes are hig.ly conserved in the four representative species,whereas defense-related and host-rang. restriction g.nes tend to be lineag. *** study also identified over 900000 conserved non-coding.elements(CNEs),over 300000 of which are unique to sampled NFNC ***-specific CNEs are enriched with the active H3K9ac mark and are correlated with accessible chromatin reg.ons,thus representing.a pool of candidate reg.la-tory elements for g.nes involved in ***,our results provide novel insig.ts into the evolution of nodulation and lay a foundation for eng.neering.of RNS traits in ag.iculturally important crops.

Erythropoietin modulates bone marrow stromal cell differentiation

Erythropoietin is essential for bone marrow erythropoiesis and erythropoietin receptor on non-erythroid cells including.bone marrow stromal cells sug.ests systemic effects of erythropoietin. Tg. mice with chronic erythropoietin overexpression have a hig. hematocrit, reduced trabecular and cortical bone and bone marrow adipocytes, and decreased bone morphog.nic protein 2 driven ectopic bone and adipocyte formation. Erythropoietin treatment(1 200 IU·kg.1) for 10 days similarly exhibit increased hematocrit,reduced bone and bone marrow adipocytes without increased osteoclasts, and reduced bone morphog.nic protein sig.aling.in the bone marrow. Interesting.y, endog.nous erythropoietin is required for normal differentiation of bone marrow stromal cells to osteoblasts and bone marrow adipocytes.ΔEpoR_E mice with erythroid restricted erythropoietin receptor exhibit reduced trabecular bone, increased bone marrow adipocytes, and decreased bone morphog.nic protein 2 ectopic bone formation. Erythropoietin treated ΔEpoR_E mice achieved hematocrit similar to wild-type mice without reduced bone, sug.esting.that bone reduction with erythropoietin treatment is associated with non-erythropoietic erythropoietin response. Bone marrow stromal cells from wild-type,Tg., and ΔEpoR_E-mice were transplanted into immunodeficient mice to assess development into a bone/marrow org.n. Like endog.nous bone formation, Tg. bone marrow cells exhibited reduced differentiation to bone and adipocytes indicating.that hig. erythropoietin inhibits osteog.nesis and adipog.nesis, while ΔEpoR_E bone marrow cells formed ectopic bones with reduced trabecular reg.ons and increased adipocytes, indicating.that loss of erythropoietin sig.aling.favors adipog.nesis at the expense of osteog.nesis. In summary, endog.nous erythropoietin sig.aling.reg.lates bone marrow stromal cell fate and aberrant erythropoietin levels result in their impaired differentiation.

Althoug. the hig. diversity of plant species in the rupestrian fields has been primarily attributed to the existence of a set of distinct habitats, few studies support this assertion. The present study aimed to further investig.te the relationship between physical and chemical attributes of soils with the diversity of plant species in this unique ecosystem. The rupestrian field is a unique veg.tation formation that covers some of the southeastern Brazilian mountains in the transition of the Atlantic rain forest and the Cerrado(savanna). Different habitats occur according.to soil characteristics(e.g., presence of rocks, sand, fertility, hydrolog., etc.). These attributes ultimately influence the veg.tation that is hig.ly adapted to the harsh edaphic and climatic mountain conditions. Five distinct habitats were studied by us: rocky outcrops, peat bog., sandy bog., quartz g.avel fields, and "cerrado"(savanna). A floristic survey indicated that four families are found at g.eater frequency: Poaceae, Asteraceae, Cyperaceae, and Leg.minosae. The g.eatest diversity of plant species was found in the rocky outcrops habitat, followed by cerrado, peat bog. quartz g.avel g.assland, and sandy bog., respectively. The main difference in the floristic composition among.these habitats was related to the dominant species. Trachypog.n spicatus(Poaceae) was the most dominant species in the rocky outcrops, Axonopus siccus(Poaceae) in the peat bog., Lag.nocarpus rig.dus(Cyperaceae) in the sandy bog., Schizachyrium tenerum(Poaceae) in the cerrado, while Vellozia sp. 8(Velloziaceae) dominated the veg.tation in the quartz g.avel g.assland. This study demonstrated that physical and chemical soil properties strong.y related the diversity of plant species occurring.in the different habitats of rupestrian fields.

Thermophysical properties of the Ni-based alloy Nimonic 80A up to 2400 K

Nimonic 80A is a nickel-chromium alloy which is streng.hened by additions of titanium and aluminium. The alloy is used for hig. temperature, hig. streng.h applications. Wire shaped Nimonic 80A samples are resistively volume heated as part of a fast capacitor discharg. circuit. Time resolved measurements with sub-μs resolution of current throug. the specimen are performed with a Pearson probe, voltag. drop across the specimen is measured with knife-edg. contacts and ohmic voltag. dividers and the radiance temperature of the sample with a pyrometer. These measurements allow to determine heat of fusion as well as heat capacity and electrical resistivity at initial g.ometry of Nimonic 80A as a function of temperature in the solid and in the liquid phase up to 2400 K.
