Molecular analysis of hepatitis B virus isolates in Mexico:Predominant circulation of hepatitis B virus genotype H

AIM: To determine the genotypes in Mexican hepatitis Bvirus (HBV) isolates and characterize their precore andcore promoter ***: Forty-nine HBV isolates of Mexico obtainedfrom sera of 15 hepatitis patients, 6 hemodialysis pa-tients, 20 men seeking HIV testing, and 8 AIDS patientswere analyzed. HBV isolates were amplified by PCR,and genotyped by line probe assay (INNO-LiPA HBVGenotyping; INNOGENETICS N V, Ghent, Belgium).HBV genotype confirmation was performed by DNAsequencing part of the sAg region. Precore and core pro-moter mutation characterization was performed by lineprobe assay (INNO-LiPA HBV PreCore; INNOGENETICS NV, Ghent, Belgium).RESULTS: Overall, HBV genotype H was found in 37(75.5%) out of the 49 isolates studied. HBV genotypesG, A, and D were found in 5 (10.2%), 4 (8.2%), and 3(6.1%) isolates, respectively. HBV genotype H was pre-dominant in isolates from hemodialysis patients (100%),hepatitis patients (80%), and men seeking HIV test-ing (75%), and accounted for half of infections in AIDSpatients (50%). Six (12.2%) out of the 49 HBV isolatesshowed both wild type and mutant populations at pre-core codon 28. These mixed wild type and precore mu-tant populations were observed in one HBV genotype Aisolate and in all HBV genotype G isolates. A dual variantcore promoter mutation was observed in 1 (2%) of theisolates, which was genotype ***: HBV genotype H is highly predominantin HBV isolates of Mexico followed by genotypes G, Aand D. A low frequency of precore and core promotermutations is observed in HBV Mexican isolates.

Molecular markers provide a useful method for genotype characterization and allow a high precision determination of the genetic relationship between cultivars and varieties. A system based on DNA sequences—which is known as DNA barcoding—will choose one or several standard loci which can be sequenced and compared to differentiate between species. In this research, the ITS, matK, and trnH-psbA sequences were evaluated for the molecular identification of seven F. carica genotypes, generating complete sequences for the first two loci, but unable to produce bidirectional sequences by using the trnH-psbA sequence. The ITS sequence presented the highest variation rates, while the phylogeny constructed with the matK sequence obtained the highest percentage of solved monophyletic groups. Through Pearson’s correlation analysis, it was possible to determine the existence of a significant correlation between the ITS region and psbA-trnH, and the matK and psbA-trnH sequences, but not between ITS and matK. The phylogenies constructed with the ITS + matK barcodes and ITS + matK + psbA-trnH presented the highest percentage for resolution. However, considering the cost efficiency and the facilitated recovery by using PCR, the matK + ITS combination is recommended.

This paper describes and analyzes floristic, structural and conservation features of the shrubland occupied by Chersophilus duponti in the northwest corner of its world distribution range. This study relates the coverage, floristic composition with estimated lark density in the area. We identified four types of shrublands, three of them varied significantly in floristic composition for the habitats of this species in contrast with those of their world distribution: Cistus ladanifer scrubs;heaths dominated by Ericaceae, and scrubs dominated by the genera Lavandula, Thymus and Cytisus. The fourth type, dominated by aromatic chamaephytes and grass, fits descriptions of the bird’s typical habitat. The areas sampled are subject to different categories of threats, the most significant being the natural succession process due to the cessation of grazing, reforestation measures and the use of flat areas for farmlands or other uses. The density of larks in this area was directly related to coverage-composition of plant community.

AIM: To investigate age, sex, histopathology and Helicobacter pylori (H. pylori) status, as risk factors for gastroduodenal disease outcome in Brazilian dyspeptic *** submitted to upper gastroscopy at Hospital das Clinicas of Marilia, antral biopsy specimens were obtained and subjected to histopathology and H. pylori diagnosis. All patients presenting chronic gastritis (CG) and peptic ulcer (PU) disease localized in the stomach, gastric ulcer (GU) and/or duodenal ulcer (DU) were included in the study. Gastric biopsies (n = 668) positive for H. pylori by rapid urease test were investigated for vacuolating cytotoxin A (vacA ) medium (m) region mosaicism by polymerase chain reaction. Logistic regression analysis was performed to verify the association of age, sex, histopathologic alterations, H. pylori diagnosis and vacA m region mosaicism with the incidence of DU, GU and CG in patients. RESULTS: Of 1466 patients submitted to endoscopy, 1060 (72.3%) presented CG [male/female = 506/554; mean age (year) ± SD = 51.2 ± 17.81], 88 (6.0%) presented DU [male/female = 54/34; mean age (year) ± SD = 51.4 ± 17.14], and 75 (5.1%) presented GU [male/female = 54/21; mean age (year) ± SD = 51.3 ± 17.12] and were included in the comparative analysis. Sex and age showed no detectable effect on CG incidence (overall c 2 = 2.1, P = 0.3423). Sex [Odds ratios (OR) = 1.8631, P = 0.0058] but not age (OR = 0.9929, P = 0.2699) was associated with DU and both parameters had a highly significant effect on GU (overall c 2 = 30.5, P < 0.0001). The histopathological results showed a significant contribution of ageing for both atrophy (OR = 1.0297, P < 0.0001) and intestinal metaplasia (OR = 1.0520, P < 0.0001). Presence of H. pylori was significantly associated with decreasing age (OR = 0.9827, P < 0.0001) and with the incidence of DU (OR = 3.6077, P < 0.0001). AIM: To evaluate if canine models are appropriate for teaching endoscopy fellows the techniques of endoscopic submucosal dissection (ESD). METHODS: ESD was performed in 10 canine models under general anesthesia, on artificial lesions of the esophagus or stomach marked with coagulation points. After ESD, each canine model was euthanized and surgical resection of the esophagus or stomach was carried out according to "The Principles of Humane Experimental Technique, Russel and Burch". The ESD specimens were fixed with needles on cork submerged in a formol solution with the esophagus or stomach, and delivered to the pathology department to be analyzed. RESULTS: ESD was completed without complications using the Hook-knife in five esophageal areas, with a procedural duration of 124 ± 19 min, a length of 27.4 ± 2.6 mm and a width of 21 ± 2.4 mm. ESD was also completed without complications using the IT-knife2 in five gastric areas, with a procedural duration of 92.6 ± 19 min, a length of 32 ± 2.5 mm and a width of 18 ± 3.7 mm. CONCLUSION: ESD is feasible in the normal esopha- gus and stomach of canine models, which are appropriate for teaching this technique.

Preparation of High Purity Neodymium Oxide from Brazilian Monazite by Ion Exchange

A simple and economical chemical process for obtaining high purity neodymium oxide is discussed. The raw material in the form of RE （rare earth） carbonates is produced industrially in Brazil from monazite. Using this concentrate as the feed material, ion exchange with a strong cationic resin, commonly used for water treatment, and without the use of retention ions was used for the fractionating of the REE （rare earth elements）. The eluent was ammonium salt of EDTA （ethylenediaminetetraacetic acid） at pH 4.0. The complex of EDTA-neodymium was transformed into neodymium oxides via oxalate precipitation. We produced 99.9% pure Nd203 in yields greater than or equal to 80%. Molecular absorption spectrophotometry was used to monitor the neodymium content during the process and sector field inductively coupled plasma mass spectrometry was used to certify the purity of the neodymium oxides. The typical neodymium oxides obtained contained the followings contaminants in micrograms per gram： Sc （4.09）; Y （0.39）; La （0.78）; Ce （5.62）; Pr （4.56）; Sm （11.10）; Eu （1.10）; Gd （14.30）; Tb （29.3）; Dy （4.15）; Ho （8.39）; Er （13.80）; Tm （0.23）; Yb （2.29）; Lu （0.39）. High purity neodymium oxides obtained from this procedure replaced the imported product used in research and development work on rare earth at IPEN/CNEN-SP （Instituto de Pesquisas Energeticas e Nucleares/Comissfio Nacional de Eneergia Nuclear-Sao Paulo）.

Accurate information on the thermal preference and specialization of species is needed to understand and predict spe- cies geographical range size and vulnerability to climate change. Here we estimate the position and breadth of species within thermal gradients based on the shape of the response curve of species abundance to temperature. The objective of the study is to compare the measurements of this approach based on abundance data with those of the classical approach using species＇ occur- rence data. The relationship between species＇ relative abundance and minimum winter temperature of 106 bird species wintering in the Iberian Peninsula is modeled at 100 Km2 resolution with quadratic logistic regressions. From these models we calculated the preferred temperature of species as the temperature at which the abundance is maximized, and the thermal breadth of species as the relative area under the temperature-abundance curve. We also estimated the thermal preferences and breadth of species as the average temperature and temperature range of the UTM cells in which the species are present. The abundance-temperature response curves reveal that birds prefer higher temperatures to overwinter, and are more thermally selective, than is measured by the classical approach. Moreover, response curves detect a higher inter-specific variability in both thermal preferences and ther- mal breadth of species. As occurrence data gives the same weight to cells with one or many individuals, the average temperature of the cells in which the species is present roughly reflects the average temperature in the region of study and not the environ- mental preferences of species .

Inadvertent Postoperative Hypothermia at Post-Anesthesia Care Unit: Incidence, Predictors and Outcome

Background: Inadvertent postoperative hypothermia (IPH) is known to be associated with various adverse effects. The aim of this study was to evaluate the incidence, predictors and outcome of core inadvertent hypothermia on admission in the post-anesthesia care unit. Methods: Observational, prospective study in a Post-Anesthesia Care Unit. The study population consisted of adult patients after non-cardiac and non-neurologic surgery. Patients’ demographics, intraoperative and postoperative data were collected. Descriptive analysis of variables was used to summarize data and the Mann-Whitney U test, Fisher’s exact test or Chi-square test was used. Univariate and multivariate analyses were done with logistic binary regression with calculation of an Odds Ratio (OR) and its 95% Confidence Interval. Results: The incidence of IPH on admission was 32%. In univariate analysis: age, body mass index (BMI), high risk surgery, revised cardiac risk index (RCRI), type of anesthesia, use of forced-air warming, amount of intravenous crystalloids administrated, duration of anesthesia, duration of surgery and admission visual analogue scale (VAS) for pain > 3 were considered predictors of hypothermia. In multiple logistic regression analysis, age (OR 1.7, P = 0.045, for age > 65 years), RCRI (OR 3.18, P = 0.041, for RCRI > 2), duration of anesthesia (OR 1.52, P < 0.001) and admission VAS for pain (OR 2.05, P = 0.007) were considered independent predictors of IPH. Patients with IPH at PACU admission stay longer in the PACU. Conclusions: IPH was associated with a longer stay in the PACU. Age, comorbidities duration of anesthesia and pain at PACU admission were considered independent predictors for IPH.
