Quantification and Determination of Starch in Diets Using Different Methodologies:Infrared Spectroscopy with Fourier Transformer,Laser Diffraction and Enzymatic Analysis

A digestibility experiment was conducted in the Animal Nutrition Laboratory of the Veterinary School of the Federal University of Minas Gerais (UFMG), Protocol ethics committee for work with animal experimentation CETEA/ UFMG 212/2010, in order to validate the instrumental techniques Laser Diffraction (Partica) and Infrared Spectroscopy with Fourier Transformer (FTIR) method, as replacement methods of the reference (Enzymatic Analysis) Technique, of starch quantification in diets of growing pigs. The experiment lasted 12 days (7 days of adaptation to the cage and diet and 5 days of total collection). Fifteen barrows with average weight of ±25 kg and 65 days old were used for this study. A completely randomized experimental design was adopted, with split plots and subplots. The plots were the diets (1—Reference, 2—Reference + inclusion of 30% micronized soybean and 3—Reference +30% substitution of plasma) and the subplots were the techniques (Partica, FTIR and Enzymatic Technique, with five repetitions (animals) per diet). The results obtained with the Partica and FTIR techniques were compared to the reference method. There was no statistical difference (p > 5%) between these techniques evaluated in comparison to the reference method to quantify starch. However, it was observed that the FTIR technique was different from the reference method for (p > 5%) overestimating the means of evaluated samples.

More effective approaches are needed in the treatment of blood cancers,in particular acute myeloid leukemia(AML),that are able to eliminate resistant leukemia stem cells(LSCs)at the bone marrow(BM),after a chemotherapy session,and then enhance hematopoietic stem cell(HSC)engraftment for the re-establishment of the HSC ***,we investigate whether light-activatable nanoparticles(NPs)encapsulating all-trans-retinoic acid(RA^(+)NPs)could solve both *** in vitro results show that mouse AML cells transfected with RA^(+)NPs differentiate towards antitumoral M1 macrophages through RIG.1 and OASL gene *** in vivo results further show that mouse AML cells transfected with RA^(+)NPs home at the BM after transplantation in an AML mouse *** photo-disassembly of the NPs within the grafted cells by a blue laser enables their differentiation towards a macrophage *** macrophage activation seems to have systemic anti-leukemic effect within the BM,with a significant reduction of leukemic cells in all BM compartments,of animals treated with RA^(+)NPs,when compared with animals treated with empty *** a separate group of experiments,we show for the first time that normal HSCs transfected with RA^(+)NPs show superior engraftment at the BM niche than cells without treatment or treated with empty *** is the first time that the activity of RA is tested in terms of long-term hematopoietic reconstitution after transplant using an in situ activation approach without any exogenous priming or genetic conditioning of the transplanted ***,the approach documented here has the potential to improve consolidation therapy in AML since it allows a dual intervention in the BM niche:to tackle resistant leukemia and improve HSC engraftment at the same time.

In vitro Metabolomic Approaches to Investigating the Potential Biological Effects of Phenolic Compounds:An Update

Dietary phenolic compounds （PCs） have been receiving intercst lor their presumcd roles ill disease prevention. However, there is a lack of studies on the underlying molecular mechanisnls. In this regard, in vitro metabolomic approaches are suitable for the investigation o1 the molecular changes in response to PC exposure. Up to date, the biological effects of PCs have only been cxalnined for PCs from rosemary （Rosmarinus officinalis）, olive oil, and reSVCl-atrol using cell-based mctabolomic approach, although transcriptomic and/or proteomic studies have also been conducted in the same in vi[ro cell experiment in some cases. Our integral analysis of the reviewed studies suggest that PCs may be involved not only in basic cellular processes or macro-and micro-nutrient meta- bolism, but also in specific metabolic pathways that have been thoroughly investigated. These modulated pathways could have a clinical impact on neurodegenerativc diseases, type 2 diabetes, cancer, and cardiovascular diseases. In conclusion, the in vitro metabolomic approaches provide additional information of the molecular mechanisms involved in disease risk reduction of dietary PCs. In order to elucidate the mechanisms of action of PCs, more metabolomic cell-based studies are nccdcd and testing the physiological conjugated forms of PCs in these cell systems could be of special interest.

Irritable bowel syndrome:A clinical review

Irritable bowel syndrome (IBS) remains a clinical challenge in the 21st century. It’s the most commonly diagnosed gastrointestinal condition and also the most common reason for referral to gastroenterology clinics. Its can affect up to one in five people at some point in their lives, and has a significantly impact of life quality and health care utilization. The prevalence varies according to country and criteria used to define IBS. Various mechanisms and theories have been proposed about its etiology, but the biopsychosocial model is the most currently accepted for IBS. The complex of symptoms would be the result of the interaction between psychological, behavioral, psychosocial and environmental factors. The diagnosis of IBS is not confirmed by a specific test or structural abnormality. It is made using criteria based on clinical symptoms such as Rome criteria, unless the symptoms are thought to be atypical. Today the Rome Criteria III is the current gold-standard for the diagnoses of IBS. Secure positive evidence of IBS by means of specific disease marker is currently not possible and cannot be currently recommended for routine diagnosis. There is still no clinical evidence to recommend the use of biomarkers in blood to diagnose IBS. However, a number of different changes in IBS patients were demonstrated in recent years, some of which can be used in the future as a diagnostic support. IBS has no definitive treatment but could be controlled by non-pharmacologic management eliminating of some exacerbating factors such certain drugs, stressor conditions and changes in dietary *** traditional pharmacologic management of IBS has been symptom based and several drugs have been used. However, the cornerstone of its therapy is a solid patient physician relationship. This review will provide a summary of pathophysiology, diagnostic criteria and current and emerging therapies for IBS.

Analysis of hydrodynamic effects on biofilm thickness in fluidized-bed tapered bioreactors

Fluidized beds in tapered geometries provide advantages such as a broad range of optimum operating ***,the dynamics of the flow inside these bioreactors is significantly more complex,as they promote non-uniform bioparticles *** this end,the CFD technique can give a detailed description of the flow in the ***,the aim of this work is to assess the influence of the hydrodynamic on the steady-state biofilm thickness through numerical simulations of liquid–solid fluidized *** geometries of tapered bioreactors in the operating conditions used to develop *** were run to solve the two-phase RANS equations using the open-source software OpenFOAM in *** SST k−ω model was used to estimate the turbulent features of the *** and bioparticles distribution inside the reactor were analyzed for different inlet velocities,while bioparticles collision frequency and shear stresses were evaluated for various operating *** comparison between the turbulent kinetic energy and the granular temperature indicated that the fluid flow is more turbulent than the bioparticles *** hydrodynamics influence on the steady-state biofilm thickness was assessed,and results led to the conclusion that biofilm is more sensitive to hydrodynamic shear stress than bioparticles collisions.

Targeting neuronal nitric oxide synthase as a valuable strategy for the therapy of neurological disorders

The management of neurological disorders have huge and increasing human and economic costs. Despite this, there is a scarcity of effective therapeutics, and there is an extreme urgency for new and real treatments. In this short review we analyze some promising advancements in the search of new bioactive molecules targeting neuronal nitric oxide synthase （nNOS）, an enzyme deputed to the biosynthesis of nitric oxide （NO）. In different conditions of neuronal damages, this molecule is overproduced, contributing to the pathogenesis and progression of neuronal diseases. Two main approaches to modulate nNOS are discussed： a first one consisting in the direct inhibition of the enzyme by means of small organic molecules, which can be also active against other different targets involved in such diseases. A second section is dedicated to molecules able to prevent the formation of the ternary complex N-methyl-D-aspartate （NMDA）type glutamate receptors, postsynaptic density-95 （PSD95） protein-nNOS, which is necessary to activate the latter for the biosynthesis of NO.

H pylori (CagA) and Epstein-Barr virus infection in gastric carcinomas:Correlation with p53 mutation and c-Myc,Bcl-2 and Bax expression

AIM: To investigate the interrelationship between H pylori and Epstein-Barr virus (EBV) infection in the gastric carcinogenesis having in focus the p53 mutation and the c-Myc, Bcl-2 and Bax expression. METHODS: seventy-one gastric carcinoma tissues were assessed by polymerase chain reaction (PCR) for H pylori and in situ hybridization for EBV. c-Myc, Bcl-2 and Bax expression were detected by immunohistochemistry and single-stranded conformational polymorphism (SSCP) for p53 mutation. RESULTS: The positivity rates for H pylori and EBV were 94.4% and 8.45%, respectively. The majority of the cases displayed only the H pylori presence. All EBV positive cases were also H pylori positive. None infectious agent was observed in 5.55% of the cases. The intestinal type tumor was more frequent in the co-infected and non-infected groups. The female predominated in the non-infected group showing statistical significance (70.4% vs 29.6%, P=0.039). The Bcl-2 was only detected in the group exclusively infected by H pylori. However, c-Myc and Bax were detected in the three groups but with a low frequency in the co-infected group. Mutation of p53 was present in all groups, with the highest frequencies in the H pylori positive groups. CONCLUSION: The frequency of H pylori infection in gastric carcinomas was high. The presented data indicated that gastric carcinogenesis has different pathways depending of the presence of the two investigated infectious agents, suggesting a possible involvement of H pylori with apoptotic process. The low expression of c-Myc and Bax in the EBV-positive groups suggests that EBV may inhibit the expression of these proteins. Nevertheless, p53 mutation shows to be a relevant alteration, independent of both infectious agents.
