Planktonic foraminifera as bio-indicators for monitoring the climatic changes that have occurred over the past 2000 years in the southeastern Tyrrhenian Sea

A high-resolution integrated study has been performed in a super-expanded marine record(sedimentation rate spanning from 11 cm/100 years to 20 cm/100 years)from the continental shelf area of the southeastern Tyrrhenian *** foraminiferal distribution illustrates 6 major environmental changes during the past 2000 years:(i)the Roman Period-Dark Age transition(from herbivorous-opportunistic to carnivorous species);(ii)the Dark Age-MCA transition(from carnivorous to herbivorous-opportunistic species);(iii)the Medieval Classic Anomaly-Little Ice Age transition(a further and definitive change from carnivorous to herbivorous-opportunistic species);(iv)the period during the Maunder event between approximately 1720 AD and 1740 AD(turnover from the carnivorous planktonic foraminifer Globigerinodes ruber to the herbivorous-opportunistic planktonic foraminifer Turborotalita quinqueloba);(v)the Industrial Period(dominance of herbivorous-opportunistic planktonic foraminifera);and(vi)the Modern Warm Period at approximately 1940 AD(the last turnover in favor of herbivorous-opportunistic planktonic foraminifers,associated with an increase in benthic foraminifera).Our studies lead us to link this latter feature to an anthropogenic impact associated with the damming of Sele River(Salerno Gulf)at 1934 AD,which induced a change in the sediment input with a strong decrease in coarse-grained fraction and a probable alteration in nutrient ***δ^(18)O_(***) record of the past 2000 years shows the alternation of warm/wet and cold/dry events related to the Roman Period,the Dark Age,the Medieval Classic Anomaly,the Little Ice Age,the Industrial Period and the Modern Warm *** 5 evidentδ^(18)O_(***) oscillations(between approximately 1325 AD and 1940 AD)coincide with the 5 minima in the solar activity record(Wolf,Spörer,Maunder,Dalton and Damon events).

Apolipoprotein A-IMilano(ApoA-IM)has been shown to significantly reduce coronary atherosclerotic ***,the preparation of cost-effective pharmaceutical formulations of ApoA-IM is limited by the high cost and difficulty of purifying the protein and producing the highly effective dimeric *** aim of this study was to create an expression cassette that specifically drives the expression of dimeric ApoA-IM in the protein bodies of rice *** ApoA-IM protein under control of the 13 kDa prolamin promoter is expressed exclusively in its dimeric form within the seeds,and immunocytochemical and immunogold analyses confirmed its expression in different caryopsis tissue such as seed coat,aleurone cell and endosperm,particularly in amyloplast and storage vacuoles.A plant-based ApoA-IM production system offered numerous advantages over current production systems,including the direct production of the most therapeutically effective dimeric ApoA-IM forms,long-term protein storage in seeds,and ease of protein production by simply growing ***,seeds had the potential to serve as a costeffective source of therapeutic ApoA-IM.

Analysis of the histologic features in the differential diagnosis of intrahepatic neonatal cholestasis

AIM:To compare the histologic features of the liver in intrahepatic neonatal cholestasis(IHNC)with infectious,genetic-endocrine-metabolic,and idiopathic etiologies. METHODS:Liver biopsies from 86 infants with IHNC were *** inclusion criteria consisted of jaundice beginning at 3 mo of age and a hepatic biopsy during the 1st year of *** following histologic features were evaluated:cholestasis,eosinophilia,giant cells,erythropoiesis,siderosis,portal fibrosis,and the presence of a septum. RESULTS:Based on the diagnosis,patients were classified into three groups:group 1(infectious;n=18),group 2(genetic-endocrine-metabolic;n=18),and group 3(idiopathic;n=50).There were no significant differences with respect to the following variables:cholestasis,eosinophilia,giant cells,siderosis,portalfibrosis,and presence of a septum.A significant difference was observed with respect to erythropoiesis,which was more severe in group 1(Fisher's exact test,P=0.016). CONCLUSION:A significant difference was observed in IHNC of infectious etiology,in which erythropoiesis was more severe than that in genetic-endocrine-metabolic and idiopathic etiologies,whereas there were no significant differences among cholestasis,eosinophilia,giant cells,siderosis,portal fibrosis,and the presence of a septum.

Oxidative stress, antioxidants and intestinal calcium absorption

The disequilibrium between the production of reactive oxygen(ROS) and nitrogen(RNS) species and their elimination by protective mechanisms leads to oxidative stress. Mitochondria are the main source of ROS as by-products of electron transport chain. Most of the time the intestine responds adequately against the oxidative stress, but with aging or under conditions that exacerbate the ROS and/or RNS production, the defenses are not enough and contribute to developing intestinal pathologies. The endogenous antioxidant defense system in gut includes glutathione(GSH) and GSH-dependent enzymes as major components. When the ROS and/or RNS production is exacerbated, oxidative stress occurs and the intestinal Ca2+ absorption is inhibited. GSH depleting drugs such as DLbuthionine-S,R-sulfoximine, menadione and sodium deoxycholate inhibit the Ca2+ transport from lumen to blood by alteration in the protein expression and/or activity of molecules involved in the Ca2+ transcellular and paracellular pathways through mechanisms of oxidative stress, apoptosis and/or autophagy. Quercetin, melatonin, lithocholic and ursodeoxycholic acids block the effect of those drugs in experimental animals by their antioxidant, anti-apoptotic and/or anti-autophagic properties. Therefore, they may become drugs of choice for treatment of deteriorated intestinal Ca2+ absorption under oxidant conditions such as aging, diabetes, gut inflammation and other intestinal disorders.

An internal control applied to RT-PCR detection of HCV and HIV-1 in human pooled plasma and plasma-derived medicinal products

A competitive internal control(IC)adapted to RT-PCR in-house assay was developed for HCV RNA detection in human pooled ***,it was applied in a multiplex RT-PCR for the HIV-1 and HCV RNA screening in human pooled plasma and plasma-derived products.A 258-bp PCR product from the 5´non-coding region of HCV genome was obtained.A competitive IC template was constructed by inserting a 52-bp double strand sequence into the NheI site of the 258-bp *** sequence was cloned and the obtained plasmid was used to generate a synthetic *** IC/RNA was incorporated in in-house HCV and/or HIV PCR technique to monitor the efficiency of extraction,reverse transcription,and PCR amplification *** was also used to detect all major genotypes of HCV and HIV-1 strains with similar *** detection limit of the assay for HCV and HIV-1 was 52.7 IU/mL and 164.2 IU/mL,*** techniques have been evaluated in international programs of external quality assurance with highly satisfactory *** IC is an essential reagent in PCR techniques to detect and identify HCV and HIV-1 in pooled plasma samples involved in the manufacture of plasma-derived products as well as in the field of clinical microbiology with limited resources.

Induction of apoptosis on human hepatocarcinoma cell lines by an alkyl resorcinol isolated from Lithraea molleoides

AIM: To study the mechanism of cytotoxicity of a new active 5-alkyl resorcinol [1, 3-dihydroxy-5- (tridec-4’, 7’ -dienyl) benzene] isolated from Lithraea molleoides leaves on liver tumor cells. METHODS: Human hepatocarcinoma cell lines (HepG2 and Hep3B) in culture were treated with inhibitory concentrations, 50% of the compound, for 24 h. The induction of apoptosis was detected in treated cells by analysis of DNA fragmentation, DNA content, and acridine orange and propidium iodide staining. RESULTS: After 24 h of 5-alkyl resorcinol treatment, both cell lines showed: (1) the typical morphological alterations of apoptosis; (2) DNA fragmentation, detected by laddering and appearance of a subG0 population by flow cytometry; and (3) condensed and fragmented nuclei by acridine orange-propidium iodide staining. CONCLUSION: Based on the results, this compound exerts its cytotoxic effect in both hepatocellular cell lines through apoptotic cell death. For Hep3B, cells with mutated p53 and Fas, apoptosis would proceed by p53- or Fas-independent pathways.

Long-lasting effects of unplanned logging on the seed rain of mixed conifer-hardwood forests in southern South America

Phagocytosis(cannibalism) of apoptotic neutrophils by tumor cells in gastric micropapillary carcinomas

AIM:To identify those with a micropapillary pattern,ascertain relative frequency and document clinicopathological characteristics by reviewing gastric ***:One hundred and fifty-one patients diagnosed with gastric cancer who underwent gastrectomy were retrospectively studied and the presence of a regional invasive micropapillary component was evaluated by light *** available hematoxylin-eosin(HE)-stained slides were histologically reviewed and 5 tumors were selected as putative micropapillary carcinoma when cancer cell clusters without a vascular core within empty lymphatic-like space comprised at least5%of the *** tissues from these 5 invasive gastric carcinomas were immunostained using an antimucin 1(MUC1)antibody(clone MA695)to detect the characteristic inside-out pattern and with D2-40antibody to determine the presence of intratumoral lymph *** of intraepithelial neutrophil apoptosis was evaluated in consecutive histological tissue sections by three independent methods,namely light microscopy with HE staining,the conventional terminal deoxynucleotidyl transferase-mediated d UTP-biotin nick end-labeling(TUNEL)method and immunohistochemistry for activated caspase-3(clone C92-605).RESULTS:Among 151 gastric cancers resected for cure,5(3.3%)were adenocarcinomas with a micropapillary *** of the patients died of disease from 6 to 23 mo and one patient was alive with metastases at 9 *** patients had advanced-stage cancer(≥p T2)and lymph node *** MUC1 immunostaining on the stroma-facing surface(inside-out pattern)of the carcinomatous cluster cells,together with negative immunostaining for D2-40 in the cells limiting lymphatic-like spaces,confirmed the true micropapillary pattern in these gastric *** all five cases,several micropapillae were infiltrated by *** staining,TUNEL assay and immunostaining for caspase-3 demonstrated apoptoticneutrophils within cytoplasmic vacuoles of

The influence of canopy-layer composition on understory plant diversity in southern temperate forests

Background： Understory plants represents the largest component of biodiversity in most forest ecosystems and plays a key role in forest *** their importance, the influence of overstory-layer composition on understory plant diversity is relatively poorly understood within deciduous-evergreen broadleaved mixed *** aim of this work was to evaluate how tree overstory-layer composition influences on understory-layer diversity in three forest types（monospecific deciduous Nothofagus pumilio（Np）, monospecific evergreen Nothofagus betuloides（Nb）, and mixed ***（M） forests）, comparing also between two geographical locations（coast and mountain） to estimate differences at landscape ***： We recorded 46 plant species： 4 ferns, 12 monocots, and 30 ***-layer composition influences the herb-layer structure and diversity in two different ways： while mixed forests have greater similarity to evergreen forests in the understory structural features, deciduous and mixed were similar in terms of the specific composition of plant *** pure stands were the most diverse, meanwhile evergreen stands were least *** of exclusive species of mixed forest could represent a transition where evergreen and deciduous communities meet and ***, landscape has a major influence on the structure, diversity and richness of understory vegetation of pure and mixed forests likely associated to the magnitude and frequency of natural disturbances, where mountain forest not only had highest herb-layer diversity but also more exclusive ***： Our study suggests that mixed Nothofagus forest supports coexistence of both pure deciduous and pure evergreen understory plant species and different assemblages in coastal and mountain *** the mixture of canopy patch types within mixed stands will be important for conserving the natural patterns of understory plant composition in southern beech mixe
