Artemisia vulgaris Induces Tumor-Selective Ferroptosis and Necroptosis via Lysosomal Ca^(2+) Signaling

Objective To evaluate the chemical composition and effects of Artemisia vulgaris(AV)hydroalcoholic extract(HEAV)on breast cancer cells(MCF-7 and SKBR-3),chronic myeloid leukemia(K562)and NIH/3T3 *** Phytochemical analysis of HEAV was done by high-performance liquid chromatography-mass(HPLC)*** and cell death studies were performed using trypan blue and Annexin/FITC-7AAD,***-1(Fer-1)and necrostatin-1(Nec-1)were used to assess the mode of HEAV-induced cell death and acetoxymethylester(BAPTA-AM)was used to verify the involvement of cytosolic calcium in this *** calcium measurements were made using *** HEAV decreased the viability of MCF-7,SKBR-3 and K562 cells(P<0.05).The viability of HEAV-treated K562 cells was reduced compared to HEAV-exposed fibroblasts(P<0.05).Treatment of K562 cells with HEAV induced cell death primarily by late apoptosis and necrosis in assays using annexin V-FITC/7-AAD(P<0.05).The use of Nec-1 and Fer-1 increased the viability of K562 cells treated with HEAV relative to cells exposed to HEAV alone(P<0.01).HEAV-induced Ca^(2+)release mainly from lysosomes in K562 cells(P<0.01).Furthermore,BAPTA-AM,an intracellular Ca^(2+)chelator,decreased the number of non-viable cells treated with HEAV(P<0.05).Conclusions HEAV is cytotoxic and activates several modalities of cell death,which are partially dependent on lysosomal release of Ca^(2+).These effects may be related to artemisinin and caffeoylquinic acids,the main compounds identified in HEAV.

With the expansion of eucalyptus crops to areas with severe water limitations,physiological studies involving eucalyptus clones to identify those that are tolerant to water stress become *** objective of this study was to assess morphological and physiological responses by eucalyptus clones subjected to drought stress and *** experiment consisted of three eucalyptus clones:VC865,I224 and I144 and two water regimes:control and water stress followed by rehydration,with six *** water potential,gas exchange,maximum quantum efficiency of photo systemⅡand plant height and stem diameter were evaluated under drought stress and ***6 d of rehydration,the number of leaves,leaf area and dry mass of root,leaf,stem and their total were *** clones showed intense reduction of gas exchange during the drought stress period,and only VC865 and 1144 showed rapid recovery with 3 d of *** 1224 showed greater reduction in height,stem diameter,number of leaves,water potential at midday(Ψ_(w)_(Midday)),and maximum quantum efficiency of photosystemⅡ(F_(v)/F_(m)).Clones VC865and T144 showed lower reductions inΨ_(wMidday)and F_(v)/F_(m) under ***865 had lower reductions in leaf number,leaf area and higher leaf dry mass,while clone I144 had higher height and lower reduction in root dry mass *** these clones showed higher water use efficiency with 3d of *** different phenotypic plasticities gave the clones VC865 and 1144 efficient mechanisms of acclimatization to stress and more drought tolerance,enhancing their greater capacity for recovery after stress,which allowed lower dry mass *** 1224,however,was more susceptible to drought stress,undergoing greater physiological damage with only partial recovery during rehydration.

AIM:To compare the performance of three commercially available anti-human epidermalgrowth factor receptor 2(HER2)antibodies in whole-tissue sections and tissue microarrays(TMAs)of a series of gastric ***:We present a comparative analysis of three anti-HER2 antibodies(HercepTest,4B5 and SP3)using TMA and whole-tissue sections prepared from the same paraffin blocks of 199 gastric adenocarcinomas operated upon between January 2004 and December2008 at a Brazilian cancer *** data on the patients’age,sex,the anatomical location of the tumor and the Lauren’s histological classification were collected from clinical and pathological *** immunohistochemical(IHC)results were examined by two pathologists and the cases were classified as positive(3+),equivocal(2+)and negative(0 or 1+),according to the criteria of the IHC scoring system of gastric *** and whole-tissue sections were evaluated separately and *** cases yielding discordant IHC results and/or scored as 2+were subjected to dual-color in situ hybridization in order to determine the final HER2 *** determining the sensitivity and predictive value for HER2-positive status,we measured the accuracy of each antibody by calculating the area under the receiver operating characteristic(ROC)*** agreement between the results obtained using the TMAs and those obtained using the whole-tissue sections was assessed by means of Kappa ***:Intratumoral heterogeneity of HER2 expression was observed with all ***2-positive expression(3+)in the whole-tissue sections was observed in 23 cases(11.6%)using the 4B5 antibody,in 18 cases(9.1%)using the SP3 antibody and in 10 cases(5.1%)using the HercepTest *** the TMAs,11 positive cases(5.6%)were identified using SP3 antibody,9(4.6%)using the 4B5 antibody and 6(3%)using the HercepTest *** sensitivity using whole-tissue sections and TMA,respectively,was 95.2%and 42.9%with 4B5,90.5%and 66.7%

BACKGROUND The uncertainties about coronavirus disease 2019(COVID-19),the change in routine,lifestyles and the reduction of physical contact can cause stress,anxiety,emotional overload,poor sleep and even physical health *** To evaluate the scientific publications available on the relationship between COVID-19 and anxiety experienced in the general population,during the period of social isolation,adopted by governmental organizations and public health policymakers as a measure to contain the spread of *** A literature search was performed systematically exploring the PubMed and Medline databases using the following terms classified as MeSH descriptors:(“anxiety”AND“pandemic”AND“COVID-19”).For the search,in the Biblioteca Virtual em Saúde–BVS,***,Web of Science and National Library plat forms, the following keywords were used: ("anxiety" AND "coronavirus" AND"social isolation"). Thirty-seven peer-reviewed articles were found. PRISMA andthe Downs & Black checklist were used for qualitative *** applying the inclusion criteria, seven (n = 7) original scientific articles wereselected. The collated evidence demonstrated increased levels of symptoms ofanxiety and depression during the period of social isolation. The populationbetween 21 to 40 years was most affected. The risk of severe depression was twiceas high at the epicenter of the pandemic. Case Study： Muay Thai Practice with Multiple Sclerosis Patients

MS （multiple sclerosis） is a demyelinating disease that can cause physical, psychological changes. Objective： Verify the influence of practicing the sport of Muay Thai in an MS patient. Method： Case study subject, a 28-yeax-old single man, functional systems Scale （EDSS） 01, type of MS recurring relapsing was evaluated before and after six months of sports practice. The following instruments were applied： the personality factor battery （BFP）; pfister colored pyramids （TPC）; concentrated attention test （D2）; visual selective attention scale （EASV）; social skills inventory （IHS）; a nonverbal intelligence test-Vienna Matxices （WMT-2）; visual face memory test （MVR）; all brazilian psychological test; The Equiscala and Berg scale were performed to evaluate balance; modified fatigue impact scale （MFIS） to assess fatigue; and to evaluate quality of life scale （MSQOL-54）. Result： Concerning the attentional aspects, in D2, the result showed improvement in the percentile of errors. The EASV and MVR results presented a lower average and then an average general classification. The results of BFP and TPC did not change. For the IHS, there was a change in the total score that showed more prudence. The score in both balance evaluation indicated excellent balance. The MFIS and MSQOL-54 showed there was no improvement in the fatigue index and quality of life. There was improvement in the level of nonverbal intelligence, visual memory, selective attention and behavioral change in social skills. Conclusion： After practicing Muay Thai for six months the patient obtained cognitive improvement.

Background:Hemodialysis(HD) per se is a risk factor for *** the growing body of evidence on blood-flow restriction(BFR) exercise in HD patients,identification of possible risk factors related to the prothrombotic agent D-dimer is required for the safety and feasibility of this training *** aim of the present study was to identify risk factors associated with higher D-dimer levels and to determine the acute effect of resistance exercise(RE) with BFR on this ***:Two hundred and six HD patients volunteered for this study(all with a glomerular filtration rate of 70 years old),and HD ***,RE+BFR significantly increases D-dimer after 4 *** with borderline baseline D-dimer levels(400-490 ng/mL) displayed increased risk of elevating D-dimer over the normal range(≥500 ng/mL).Conclusion:These results identified factors associated with a heightened prothrombotic state and may assist in the screening process for HD patients who wish to undergo RE+BFR.D-dimer and/or other fibrinolysis factors should be assessed at baseline and throughout the protocol as a precautionary measure to maximize safety during RE+BFR.

This documentary research aimed to describe the results of an epidemiological survey that sought to determine the prevalence of Infections Related to Health Care (IRHC) in newborns admitted to a Neonatal Intensive Care Unit (NICU) at the city of Rio de Janeiro. For methodology, the existing data in the original research database were used as the primary source. The method used was the case study in a quantitative approach. The data have been statistically arranged by odds ratio, which measured the possibility of IRHC development at the 102 newborns, by exposition at the risk factors. Results: IRHC prevalence with pneumonias, sepsis, navel’s infection and ocular infections predominance rises. Prematures are 4.10 more susceptible. Twelve hours after amniotic rupture there are 1.77 more chances of infections. Babies with more than 10 days of hospitalization are 4.70 more susceptible than those with 10 days. The registers examined were incipient at the IRHC’s characterization, beginning and duration, which make the IRHC’s diagnostic/notification difficult.

A Review of Landsat TM/ETM based Vegetation Indices as Applied to Wetland Ecosystems

A review of vegetation indices as applied to Landsat-TM and ETM+ multispectral data is presented. The review focuses on indices that have been developed to produce biophysical information about vegetation biomass/ greenness, moisture and pigments. In addition, a set of biomass/greenness and moisture content indices are tested in a Mediterranean semiarid wetland environment to determine their appropriateness and potential for carrying redundant information. The results indicate that most vegetation indices used for biomass/greenness mapping produce similar information and are statistically well correlated.

Transcranial Direct Current Stimulation Combined with Peripheral Neuromuscular Stimulation Improves Quality of Life, Fatigue, and Pain in a Patient with Rheumatoid Arthritis and Refractory Radicular Pain Related to Spinal Stenosis

Background: Transcranial direct current stimulation (tDCS) has emerged as an adjuvant noninvasive neuromodulation tool to control fatigue and pain. To date, no studies have assessed the safety and efficiency of tDCS in patients with rheumatoid arthritis and with fatigue, poor quality of life, and refractory radicular pain associated with spinal stenosis. Case Presentation: An 85-year-old woman patient presented with rheumatoid arthritis in remission, refractory radicular pain-associated spinal stenosis, fatigue, and impaired quality of life. The patient underwent 16 daily sessions of tDCS intervention (2 mA, 20 min, positively and negatively charged electrodes were positioned at C1 and Fp2, respectively), in addition to simultaneous peripheral neuromuscular electrical stimulation (frequency of 100 Hz and amplitude of 500 μs). After the intervention, neither disease relapse nor clinical intercurrence occurred. Moreover, there was a significant and sustained improvement in her health-related quality of life, with a reduction in the level of pain and chronic fatigue. Conclusion: The present case report shows that tDCS is safe and may be an adjuvant tool for the treatment of pain and fatigue in patients with systemic autoimmune disease, as well as for improving quality of life. Further studies are required to corroborate this case report.

Using Landsat Images to Determine Water Storing Capacity in Mediterranean Environments

Reservoirs play an important role in water management and are key elements for water *** is needed in order to guarantee the quantity and quality of stored ***,this task is sometimes not *** objective of this study was to develop a procedure for predicting volume of stored water with remote sensing in water bodies under Mediterranean climate *** achieve this objective,multispectral Landsat 7 and 8 images(NASA)were analyzed for the following five reservoirs:La Serena,La Pedrera,Beniarrés,Cubillas and Negratín(Spain).Reservoirs water surface was computed with the spectral angle mapper(SAM)*** that,cross-validation regression models were computed in order to assess the capability of water surface estimations to predict stored water in each of the *** statistical models were trained with Landsat 7 images and were validated by using Landsat 8 *** results suggest a good capability of water volume prediction from free satellite imagery derived from surface water *** free remote sensing images and open source GIS algorithms can be a very useful tool for water management and an integrated and efficient way to control water storage,especially in low accessible sites.
