Exogenous application of a cytokinin-like compound forchlorfenuron(cPPU)can promote fruit growth,although often at the expense of dry matter(DM),an important indicator of fruit *** chinensis ***‘Hayward’fruit are very responsive to cPPU treatments,but the mechanism underlying the significant fruit weight increase and associated decrease in DM is *** this study,we hypothesised that cPPU-enhanced growth increases fruit carbohydrate demand,but limited carbohydrate supply resulted in decreased fruit *** fruit development,cPPU effects on physical parameters,metabolites,osmotic pressure and transcriptional changes were assessed under conditions of both standard and a high carbohydrate *** showed that cPPU increased fruit fresh weight but the dramatic DM decrease was not carbohydrate *** glucose and fructose concentrations contributed to an increase in soluble carbohydrate osmotic pressure,which was correlated with increased water accumulation in cPPU-treated fruit and up-regulation of water channel aquaporin gene PIP2.4 at 49 days after *** analysis suggested that the molecular mechanism contributing to increased glucose and fructose concentrations was altered by carbohydrate *** standard carbohydrate supply,the early glucose increase in cPPU fruit was associated with reduced starch synthesis and increased starch *** carbohydrate supply was high,the early glucose increase in cPPU fruit was associated with a general decrease in starch synthesis but up-regulation of vacuolar invertase and fructokinase *** conclude that cPPU affected fruit expansion by increasing the osmotically-driven water uptake and its effect was not carbohydrate supply-limited.

In Vitro Activity of Squaramides and Acyclic Polyamine Derivatives against Trophozoites and cysts of Acanthamoeba castellanii

Pathogenic strains of Acanthamoeba cause keratitis (AK), granulomatous amoebic encephalitis (GAE), amoebic pneumonitis (AP), and skin infection in human and animals. The treatment of an Acanthamoeba infection is invariably very difficult and not always effective, and compounds that are amebicidic or amebistatic are frequently toxic and/or irritating for humans. Squaramides and polyamine derivatives have been demonstrated to have antitumor and antiprotozoal activity. The aim of this study was to investigate the activity of 5 squaramides and 5 acyclic polyamines against trophozoites and cysts of A. castellanii Neff. Amoebicidal activity against the trophozoites and cytotoxicity against Vero cells were evaluated with a colorimetric assay, using Alamar Blue?, and chlorhexidine digluconate was assayed as the reference drug. The squaramides 3 and 5 and the acyclic polyamine 6 appeared to be the most active against the trophozoites and their cytotoxicity was low, showing selectivity indexes of 28.3, 26, and 25.7, respectively, similar to the control drug, chlorhexidine digluconate (27.6). But only the squaramide 3 showed complete cysticidal activity at the concentrations of 100 and 200 μM, as the chlorhexidine digluconate. Further studies of the mechanism of action and in vivo assays are needed, but squaramide 3 could be used for developing novel therapeutic approaches against Acanthamoeba infections.

Flavorful meat and great economic importance, crabs and swimming crabs are appreciated throughout the Brazilian coast, where its waste is discarded by fishermen and consumers. In order to verify the centesimal composition, these animals caught in Sepetiba Bay （Rio de Janeiro, Brazil）, 20 samples of Ucides cordatus, cardisoma guanhumi, Menippe nodifrons, callinectes danae and c. exaperatus had their edible and waste parts （shells and hepatopancreas） analyzed in the Laboratory of Bromatology of the UFRRJ. The analysis revealed that the meat of crustaceans is protein （12.68% to 25.56%） and low in fat （0.37% to 0.67%）, sticking to a more demanding consumer market for thin products; the waste has already presented a protein content ranging from 15.75% to 27.64% and fat from 0.75% to 3.27%, with values of calcium （17.96% to 25.23%） and phosphorus （0.93% to 1.55%）, setting a precedent for use in animal feed through future studies in area.

Production of Lipase Using cassava Peel and Sunflower Oil in Solid-State Fermentation： Preliminary Study

Full use of residues from industrial processes is a fundamental necessity of contemporary society, since it avoids impacts to the environment by using residues as inputs for other products of high economic and social importance. In this study, lipase production of the crude enzymatic extracts obtained by Aspergillus niger using cassava peel as substrate and sunflower oil as an inductor was investigated. The optimized cultivation temperature and concentration of inductor were determined using the response surface methodology. The two variables studied exercised influence in the production of lipase in the 95% level of confidence. The response surface obtained indicated that the conditions that maximize lipase activity production were 30.5 ~c and initial concentration of sunflower oil was 2.5% （w/w）. Through this analysis, it is evident that extremes in temperature and concentration of inductor tend to decrease lipase production, since low temperatures decrease metabolism and high temperatures may inactivate the lipase. Optimum lipase yield was 59.8 U/g of dry peel which was fermented for 60 h. Lipase production presents a peak of 61.3 U/g, at 72 h of fermentation. However, this value is statistically equal （p 〉 0.05） of the value of lipase activity obtained for 60 h and 84 h of fermentation.

Bladder Neck Morphologic changes and clinical correlation of Smokers Submitted to Radical Prostatectomy

Introduction: Smoking is an important risky factor to many diseases, affecting arterial system, skin and urogenital system, including bladder neck stenosis. Its effect on bladder neck has not been described. Objective: Evaluate possible morphological changes caused by nicotine in the bladder neck. Material and Method: Fragments of bladder neck of 16 patients were submitted to stereological analysis, and those patients are divided into two groups, one of smokers and the other of non-smokers with 7 and 9 patients, respectively. After 90 days of surgery, they were submitted to free uroflowmetry and data analyzed by T test, having statistical significance with P 0.05. Results: An increase of 63.26% in the amount of fibers in the elastic system of the smokers group was observed, a reduction of 35.96% in the thickness of arteries, as well as an increase of IPSS and decrease of maximum flow in uroflowmetry, all with statistical significance. Discussion: Laboratorial changes are similar to those found in other studies with different tissues, such as skin, in which those findings are related to premature ageing. clinical results, though statistically significant, do not have clinical consistence because the study was meant to morphological analysis. conclusion: Smoking increases the amount of fibers in the elastic system and decreases the thickness of bladder neck arteries.

Biochemical changes of cubiu Fruits (Solanum sessiliflorum Dunal, Solanaceae) According to Different Tissue Portions and Ripening Stages

cubiu fruits (Solanum sessiliflorum Dunal, Solanaceae) are known for their high nutritional value and low caloric content. This work aimed at evaluating biochemical indices of cubiu fruits according to different tissues (peel, pulp and placenta) and ripening stages (green, turning, ripe and fully ripe). The fruits were randomly harvested to investigate sensory aspects (colorimetry, blanching effect, pigments) and biochemical indices (moisture and dry matter, Total Soluble Solids (TSS), pH, Titratable Acidity (TA), TSS/TA ratio, ascorbic acid, Alcohol-Insoluble Solids (AIS), pectinesterase activity and pectin content). The analyses were performed at the laboratory of Instituto Nacional de Pesquisas da Amazonia (INPA), Manaus, Brazil. The blanching process preserved pulp colors at all ripening stages. chlorophylls were higher in green peels;flavonoids and carotenoids, in fully ripe peels. Anthocyanins were entirely absent. Pulp (turning fruits) showed the highest moisture content (91.05), followed by ripe pulp (90.70) and fully ripe pulp (90.62). Pulp TSS changed little whereas placenta TSS declined and were associated with increased TA and pH reduction (fully ripe fruits). Pulp pH and TA varied little whereas placenta pH was notably low and TA was remarkably high (fully ripe fruits). Pulp TSS/TA ratio showed predominance of TSS while in placenta, there was predominance of organic acids. AIS, precursors of pectin, were stable during ripening. The statistical analysis of dietary fiber content showed one modal value in the AIS. In pulp, pectinesterase activity correlated inversely with pectin content. cubiu fruits were significant sources of bioactive compounds, e.g. chlorophylls, flavonoids and carotenoids, predominantly in the peel and the pulp;soluble functional fibers, e.g. pectin (g/100 g fresh weight), particularly in the green peel (1.00) and the fully ripe pulp (1.12), and other versatile molecules, e.g. ascorbic acid (mg/100 g fresh weight), especially in the f

Trichomonas vaginalis, a protist parasite of the urogenital tract in humans, is the causative agent of trichomonosis, which in recent years have been associated with the cervical cancer development. In the present study we analyzed the modifications at the junctional complex level of caco-2 cells after interaction with two isolates of T. vaginalis and the influence of the iron concentration present in the parasite’s culture medium on the interaction effects. Our results show that T. vaginalis adheres to the epithelial cell causing alterations in the junctional complex, such as: (a) a decrease in transepithelial electrical resistance; (b) alteration in the pattern of junctional complex proteins distribution as observed for E-cadherin, occludin and ZO-1; and (c) enlargement of the spaces between epithelial cells. These effects were dependent on (a) the degree of the parasite virulence isolate, (b) the iron concentration in the culture medium, and (c) the expression of adhesin proteins on the parasite surface.

The perceptions of fishers towards the Arrábida Marine Park,a marine protected area(MPA)in the west coast of Portugal,were studied through face-to-face interviews in two different moments of the MPA life ***'perceptions about the MPA and the impact it had on the fishing activity over time were identified just before the full implementation of the zoning and regulations of the management plan and 10 years *** study aimed to investigate fishers'knowledge,acceptance and perceptions about the MPA changed with time,if support for the MPA was linked to the impact of the MPA on the fishing activity,and if fishers'perceptions about the impact of the MPA on the fishing activity match with local landings *** show that although knowledge about the marine park significantly improved over time,fishers'acceptance did not.A decrease on fishers'support was not substantial but *** such as the disagreement with regulations reinforced concerns raised during the implementation of the marine park,particularly in relation to the top-down decision-making,which commonly confers minor participation,recognition and legitimacy to ***,fisheries benefits were still not perceived by local fishers,though they are central for fishers'***,the perceived negative impacts of the park seemed to be more related to social aspects and individual interests than to impacts on *** adequate management,enforcement and participation of local fishers is still possible and are advocated here as to contribute to the expected socioecological outcomes and respective support,leading to the future successful performance of the Arrábida Marine *** fishers’perceptions towards an MPA over time is central and should be included on periodical socioecological monitoring and inform an effective adaptive management.

Lack of Relationship between Testicular Echotexture and Seminal characteristics in Adult Texel Rams

The objective of this study was to determine the minimum representative area to evaluate testicular echotexture parenchyma and to identify the correlations between the intensity of pixels of the testicular parenchyma and the fibrosis score with the physical and morphological characteristics of the ejaculate of Texel rams. The study used 88 Texel rams, aged between 10 and 12 months, and reared in a semi-extensive system. The animals underwent breeding soundness evaluation (BSE) and ultrasound of the testicles. The images were transferred to a computer where they were defined in areas of 400, 1600, 3600, and 6400 mm2, after which the average intensity of the pixels of each image of the testicular regions was evaluated. A testicular fibrosis score was assigned in order to quantify the frequency of fibrotic lesions. In relation to the intensity of pixels of the predetermined regions, only the area of 400 mm2 presented a difference (P mm2 being the smallest area that best represented the testicular parenchyma. There were no correlations between the intensity of pixels of the testicular parenchyma and the fibrosis score with the physical and morphological characteristics of the ejaculate of the rams.

Acromegaly as an Expression of a Rare Disease: Description of an Unusual clinical case of carney complex

Introduction: The carney complex (cc) is an autosomal dominant syndrome, characterized by multiple endocrine neoplasms, pigmented skin lesions, endocrine hyper-activity and myxomas. case study: c. E., 71 years old, was addressed to our center in August 1994, following the finding of a sellar enlargement in a skull radiography performed after an accident. A pituitary tomography showed the presence of a lesion extended up to the optic chiasm, a macroadenoma that was treated with transsphenoidal endoscopic surgery. A histological diagnosis of a “GH secreting macroadenoma” was performed. An echocardiographic study highlighted an atrial isolated myxoma that was subsequently removed. A contrast-enhanced abdominal magnetic resonance imaging (MRI) excluded adrenal lesions. In 2001, in relation to relapse of Acromegaly, the patient started therapy with somatostatin analogues which was replaced, in 2008, by Pegvisomant to obtain biochemical control of the disease. conclusions: At the time of initial screening, it is important not to underestimate the possibility that certain clinical features fall within the context of more complex syndromes. The knowledge of how Acromegaly presents itself in relation to cc and its associated characteristics such as cardiac myxoma or typical skin lesions, will help clinicians to timely diagnose this rare disease and treat it appropriately.
